Inhibition of c-FLIP by RNAi Enhances Sensitivity of the Human Osteogenic Sarcoma Cell Line U2OS to TRAILInduced Apoptosis

  • To study effects of cellular FLICE (FADD-like IL-$1{\beta}$-converting enzyme)-inhibitory protein (c-FLIP) inhibition by RNA interference (RNAi) on sensitivity of U2OS cells to tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL)-induced apoptosis, plasmid pSUPER-c-FLIP-siRNA was constructed and then transfected into U2OS cells. A stable transfection cell clone U2OS/pSUPER-c-FLIP-siRNA was screened from the c-FLIP-siRNA transfected cells. RT-PCR and Western blotting were applied to measure the expression of c-FLIP at the levels of mRNA and protein. The results indicated that the expression of c-FLIP was significantly suppressed by the c-FLIP-siRNA in the cloned U2OS/pSUPER-c-FLIP-siRNA as compared with the control cells of U2OS/pSUPER. The cloned cell line of U2OS/pSUPER-c-FLIP-siRNA was further examined for TRAILinduced cell death and apoptosis in the presence of a pan-antagonist of inhibitor of apoptosis proteins (IAPs) AT406, with or without 4 hrs pretreatment with rocaglamide, an inhibitor of c-FLIP biosynthesis, for 24 hrs. Cell death effects and apoptosis were measured by the methods of MTT assay with 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and flow cytometry, respectively. The results indicated that TRAIL-induced cell death in U2OS/pSUPER-c-FLIP-siRNA was increased compared with control cells U2OS/pSUPER in the presence or absence of AT406. Flow cytometry indicated that TRAIL-induced cell death effects proceeded through cell apoptosis pathway. However, in the presence of rocaglamide, cell death or apoptotic effects of TRAIL were similar and profound in both cell lines, suggesting that the mechanism of action for both c-FLIP-siRNA and rocaglamide was identical. We conclude that the inhibition of c-FLIP by either c-FLIP-siRNA or rocaglamide can enhance the sensitivity of U2OS to TRAIL-induced apopotosis, suggesting that inhibition of c-FLIP is a good target for anti-cancer therapy.

User′s Effects on Avifauna in Bukhan Mountain National Park (북한산 국립공원의 이용객이 야생조류에 미치는 영향)

  • This study was conducted to investigate avifauna and user's effect on the avifauna in Bukhan Mountain district of Bukhan Mountain National Park. Avifauna were investigated bimonthly from April to October 1987 over four districts which were main trail and sub-trail in each valley and ridge-midslope region by line transect methods. The observed birds in Bukhan Mountain districts was 34 species and 753 individuals and these consist of 19 species for residents. 11 species for summer visitor. 2 species for passage migrant and 2 species for winter visitor. The dominant species were Parus major (22.7%), Paradoxornis webbina(10%), Parus palustris (10.8%), Pica pica (10.0%) in order. No. of species and species diversities was higher in valley region than in ridge-midslope region. No. of species, individuals and species diversities were lower in main trail of high user's density than in sub-trail. Especially avifauna in main trail was severely decreased in August in valley region in which users were concentrated. But there was no change in similarity indices according to user's intensities. Therefore avifauna protection policy such as restoration and rehabilitation of edge species along trail, user's control in valley region during breeding periods, etc., shall be made for Bukhan Mountain National Park.

User′s Effects on Avifauna in Chiak Mountain National Park (치악산 국립공원의 이용객이 야생조류의 서식에 미치는 영향)

  • This study was conducted to investigate the user's effects on the avifauna in Chiak Mountain National Park. The survey was carried over 5 districts which were 3 main trail (valley) by line transect method from May to October 1988. The observed birds were 44 species and 613 individuals. these consist of 22 species for residents, 13 species for summer visitor, 5 species for winter visitor and 4 species for passage migrant. The average density of birds was 2.96 ea/ha, and main dominant species were Panus minar, Paradoxomis webbiana fulvicanda, Panus palustris hellmayri and Parus ater amurensis in order. The observed frequency of wild-birds was 6.77 times/km in average. No. of species, species diversities and observed frequency were lower in main trail of high user's density than sub trail and closed trail, especially in summer. But similarity indices were affected by inhabitation environments than user's density. Therefore, from now, in Chiak Mountain National Park management, the enjoyment oriented activities will be regulated and the control of trail use on main trail are necessary during breeding periods for wild birds protection.

A Study on Visitors' Characteristics and Cognition in Regard to Damage of Nature Environment in Different Types of Ecotourism Destination (생태관광지 유형별 탐방객 특성 및 자연환경 훼손에 대한 인식 비교 연구)

  • For understanding the tourists' characteristics of several types of ecotourism sites, we surveyed and analyzed the demographic characteristics, travel motivations and cognition of damage in Bukhansan trail, Upo marshes, Jirisan trail and Uljin Keumgang pine tree trail. The field survey were conducted through face-to-face interviews. In the case of Bukhansan trail, many people came to build up their health, so the ratio of eco-tourists and the awareness of environmental damage was low. In Upo marshes, there were many group tourists who want to enhance friendship between the colleagues. And the ratio of eco-tourists, enjoying several activities such as observation or experience about the ecosystems of wetland and bird watching appeared high, but the cognition of tourists about the damage of natural environment was low because the flatland type tour site, Upo marshes has less damage factors comparing with mountain type tour sites. In Jirisan trail, eco-tourists, nature tourists and mass tourists appeared mixed and the cognition of damage was higher than that of flatland type. In the case of Uljin Keumgang pine tree trail, almost indices showed higher values than other sites because the tourists who visited there have high cognition of eco-tourism and nature conservation by limiting entrance. Tourists were generally accepted positively the control method such as collection of entrance fee and visitor reservation system when natural damages are expected.

Visitors' Evaluation of the Forest Trail Facilities by Applying Universal Design Concepts (유니버설디자인을 적용한 숲길 보행시설에 대한 이용객 평가)

  • As 'design for all' and 'adaptable design', universal design is highly demanded to be adopted in neighborhood forest trail. The research areas were Mt. Doeum in Pohang, Gyeongsangbuk Province and Mt. Anma in Chuncheon, Gangwon Province which were equipped based on 7 principles of universal design. The purpose of this research is providing baseline data for effective formulation of forest trail using universal design through visitors' evaluation on forest trail facilities by importance-performance analysis. Visitor's evaluation was achieved through questionnaire survey, and total 247 pieces of subjects were used for the analysis, 114 pieces from Mt. Doeum in Pohang and 113 pieces from Mt. Anma in Chuncheon. The residential areas of the respondents were mainly near each mountain - Mt. Doeum and Mt. Anma. According to the responses, nature study and education were the biggest reasons for the respondents' visits to Mt. Doeum, and so was the health to Mt. Anma, which are representative types of visiting behavior to nearby forest trails. The mean of importance was shown to be 3.87 while that of performance was 4.04, being a little higher than importance's, in importance-performance analysis of Mt. Doeum. This result suggests that the responders placed positive value on the forest trail applying universal design. It is necessary to improve the safety and convenience of guidance-safety handrail, make information sign easy to understand, and introduce an interpretive sign meeting the visiting purposes of visitors. Also, the management to make the road surface flat is in demand for the accessibility to forest trail. The mean of importance was 3.92 and that of performance was 3.99 in Mt. Anma, which suggests the necessity of improvement in safety of pavement material and boundary safety facilities.

  • The term 'greenways' is a fairly new one. The term greenways refer to linear corridors which set aside pieces of nature. The objective of this study was to find out the feasibility of developing greenways in Taegu. Sample of 446 trail users were asked to describe their use patterns and as well as their perceptions of the benefits and problems associated with trails through on-site interviews conducted during the months of July and August of 1999. Trail users in Taegu were predominately male(61%) and over half of the respondents used private vehicle to access to the study areas as compared to 2% of respondents used bicycle. Trail users responded that health & fitness and open space preservation were the two most important benefits, and the provision of trail access to disabled persons was ranked last. As problems at study areas, lack of interpretive information was the biggest problem followed by lack of drinking water, poor trail marking/signs and not enough information. Most(94.1%) of the trail users surveyed indicated that they would support greenways development and 96.8% of respondent would use after greenways development. Willingness to pay for developing greenways was assessed with hypothetical scenario. Average willingness to pay in the survey was 10,200 won as a tax. The development/application of greenways in Taegu would work best when establish a network of trails with environmentally sensitive manner.

Comparing Effects of Driving Simulator and Dynavision Training on Cognitive Ability and Driving Performance After Stroke (뇌졸중 이후 운전 시뮬레이터와 Dynavision 훈련이 인지 및 운전 수행 능력에 미치는 효과 비교)

  • Objective : The purpose of this study was to compare with the effects of driving simulator and Dynavision training after stroke through the test of cognitive ability and driving performance. Methods : Twenty-one stroke patients were randomly classified to the driving simulator training group (N=11) and Dynavision training group (N=10), and were carried out respectively training for 15 times. The driving performances was measured by the driving simulator test, and cognitive-perceptive abilities was measured by the DriveABLE Cognitive Assessment Tool, Trail Making Test-A, Trail Making Test-B and Mini Mental State Examination-K. Results : The driving simulator training group showed significant changes in all cognitive tests and most of driving performances. The Dynavision training group also showed significant changes in all cognitive tests except for Trail Making Test-A and some driving performances. The significant differences on both groups were found regarding the estimated degree of results on the on-road evaluation, the number of off road accidents and collisions. In addition, the causal influence of the two training methods on these variables was analyzed to be more than 20%. Conclusion : The driving simulator and Dynavision training were found to be effective intervention in the driving rehabilitation after stroke. In particular, it was confirmed that the driving simulator is an effective training to improve overall driving ability of stroke patients. In addition, the difference in training effect between the two training methods was found to be more than 20%.

Estimating the Impact of DMZ Punchbowl Trail as a National Forest Trail on Local Economy using the Regional Input-Output Model (지역산업연관모델을 이용한 국가숲길의 지역경제 파급효과 분석: DMZ펀치볼둘레길을 중심으로)

  • This study was conducted to identify the usage characteristics of the DMZ Punchbowl Trail (DPT) as a national forest trail (NFT) and to estimate its ripple effects on the local economy. The objective of this study is to provide policy implications for sustainable operational management. Out of the 500 questionnaires distributed, 215 respondents provided their complete travel itineraries and expenditures. The respondents, mainly aged 50 and above and residing in the Seoul Metropolitan Area, spend 3.5 hours of travel time to the DPT. Together with their families, the respondents typically spend approximately 4 hours for leisurely activities, primarily appreciation of scenic views and relaxation by visiting the "O-yubatgil." Furthermore, they extend their travels to other parts of Gangwon Province, where the DPT is situated. Within Gangwon Province, Yanggu County is the most visited destination. The respondents reported a notably higher average expenditure per visitor compared with the typical local walking tourists. Estimates show that the DPT generates an annual average of KRW 2.1 billion in direct expenditure (based on an average of 10,000 visitors for over five years), KRW 2.8 billion in production, and KRW 1.3 billion in added value, and it has created 40 jobs in Gangwon Province. The results of this study lies in empirically determining the specific economic scale and ripple effects of DPT as an NFT in the major sector, which occupies a significant portion of the Gangwon Province's local economy. The results will be instrumental in validating NFT policies and informing policy making for sustainable forest utilization.
