Taenia saginata Infection Misdiagnosed as Acute Cholecystitis in a Tibetan Patient, in China

  • The present study reports a rare case of Taenia saginata infection, which was initially diagnosed as acute cholecystitis in a Tibetan patient at the Qinghai-Tibetan Plateau pastoral area, China. A 45-year-old female was initially diagnosed with acute cholecystitis at a hospital in China. She had a slight fever, weight loss and constipation and complained of pain in the upper abdomen and left back areas. Increase of monocyte, eosinophil and basophil levels were shown. Taenia sp. eggs were detected in a fecal examination. An adult tapeworm approximately 146 cm in length, whitish-yellow color, was collected from the patient after treatment with traditional Chinese medicine. The adult tapeworm had a scolex and proglottids with genital pores. The scolex was rectangular shape with 4 suckers and rostellum without hooklet. The cox1 gene sequence shared 99.5-99.8% homology with that of T. saginata from other regions in China. The patient was diagnosed finally infected with T. saginata by morphological and molecular charateristics.

Gamijiya-tang Decreases the Dimethylnitrosamine-Induced Hepatic Fibrosis in the Rats

  • Gamijiyu-tang (GJT) described originally in the Dong Eui Bo Gam, a traditional reference for oriental medicine in the Korea, has been clinically used for treatment of chronic liver disease. In order to evaluate scientifcally a hepatoprotective effect of GJT in the liver fibrotic disease, the present study investigated how GJT improves a hepatic function in the dimethylnitrosamine (DMN)-treated rat. DMN treatment caused a significant increase of relative liver weight to the body at 28 days after DMN induction. Administration of with a clinical dose decreased significantly the sAST level $(158.8{\pm}7.76\;IU/L)$ elevated by DMN in jection (p<0.01). A similar phenomenon was also observed at change of both Salt and Salt level in the GJT and/or DMN-treated animal (p<0.01, p<0.05, respectively). A remarkable increase of hydroxyproline was observed by treatment of DMN with comparing to the normal rat $(361.9{\pm}7.35\;vs.\;1278.1{\pm}52.9\;{\mu}g/g\;tissue,\;p<0.01)$. This was significantly reduced by a simultaneous treatment of GJT with DMN for 21 days (p<0.05), but not recovered completely to its normal value. In addition. GJT administration ameliorated conspicuously the DMN-induces histopathological changes of liver such as hemorrhage. Cell necrosis and fibrosis. Tak'en together, results described here demonstrated scientifically in first the medicinal efficacy of GJT by using in vivo animal model, indicating that GJT improves the DMN-induced hepatic injury through reducing an excessive accumulation of collagen and histopathological changes. The decreased collagen content may be a pivotal process for GJT to improve hepatic function in the DMN-induced liver fibrosis. The present study suggests that GJT may be useful for and applicable to the treatment of hepatic fibrosis in chronic liver disease.

Analysis of the Components of Guibitang and Fermented Guibi-tang and their Ability to Inhibit Angiotensin-converting Enzyme

  • Guibi-tang is a traditional medicine used for the treatment of colds. We investigated the levels of several compounds in Guibi-tang before and after fermentation with Lactobacillus and tested their ability to inhibit angiotensin-converting enzyme. Six known compounds (decursin, decursinol angelate, nodakenin, liquiritin, formononetin, and 6-gingerol) and 2 unidentified compounds were detected in Guibi-tang (GB) and fermented Guibi-tang (FGB) by an established HPLC-DAD method. The levels of the 6 known compounds were decreased after fermentation. FGB showed more potent inhibition of angiotensin-converting enzyme activity than GB. In conclusion, fermentation with Lactobacillus affects the content of several compounds in GB and improves its angiotensin-converting enzyme inhibitory activity.

A 90-Day Repeated Oral Dose Toxicity Study of Alismatis Rhizoma Aqueous Extract in Rats

  • Alismatis rhizoma (AR), the dried rhizome of Alisma orientale (Sam.) Juzep, is a well-known, traditional medicine that is used for the various biological activities including as a diuretic, to lower cholesterol and as an anti-inflammatory agent. The present study was carried out to investigate the potential toxicity of the Alismatis rhizoma aqueous extract (ARAE) following 90-day repeated oral administration to Sprague-Dawley rats. ARAE was administered orally to male and female rats for 90 days at 0 (control), 500, 1,000 and 2,000 mg/kg/day (n = 10 for male and female rats for each dose). Additional recovery groups from the control group and high dose group were observed for a 28-day recovery period. Chromatograms of ARAE detected main compounds with four peaks. Treatment-related effects including an increase in the red blood cells, hemoglobin, hematocrit, albumin, total protein, and urine volume were observed in males of the 2,000 mg/kg/day group (p < 0.05). However, the diuretic effect of ARAE was considered, a major cause of hematological and serum biochemical changes. The oral no-observed-adverse-effect level (NOAEL) of the ARAE was > 2,000 mg/kg/day in both genders, and no target organs were identified.

The inhibitory mechanism of crude saponin fraction from Korean Red Ginseng in collagen-induced platelet aggregation

  • Background: Korean Red Ginseng has been used as a traditional oriental medicine to treat illness and to promote health for several thousand years in Eastern Asia. It is widely accepted that ginseng saponins, ginsenosides, are the major active ingredients responsible for Korean Red Ginseng's therapeutic activity against many kinds of illness. Although the crude saponin fraction (CSF) displayed antiplatelet activity, the molecular mechanism of its action remains to be elucidated. Methods: The platelet aggregation was induced by collagen, the ligand of integrin ${\alpha}_{II}{\beta}_I$ and glycoprotein VI. The crude saponin's effects on granule secretion [e.g., calcium ion mobilization and adenosine triphosphate (ATP) release] were determined. The activation of mitogen-activated protein kinases (MAPKs), including extracellular signal-regulated protein kinase 1/2 (ERK1/2), c-Jun N-terminal kinases (JNKs), and p38 MAPK, and phosphoinositide 3-kinase (PI3K)/Akt was analyzed by immunoblotting. In addition, the activation of integrin ${\alpha}_{II}b{\beta}_{III}$ was examined by fluorocytometry. Results: CSF strongly inhibited collagen-induced platelet aggregation and ATP release in a concentration-dependent manner. It also markedly suppressed $[Ca^{2+}]_i$ mobilization in collagen-stimulated platelets. Immunoblotting assay revealed that CSF significantly suppressed ERK1/2, p38, JNK, PI3K, Akt, and mitogen-activated protein kinase kinase 1/2 phosphorylation. In addition, our fraction strongly inhibited the fibrinogen binding to integrin ${\alpha}_{IIb}{\beta}_3$. Conclusion: Our present data suggest that CSF may have a strong antiplatelet property and it can be considered as a candidate with therapeutic potential for the treatment of cardiovascular disorders involving abnormal platelet function.

하수오 물 추출물이 골형성 및 파골세포에 미치는 영향 (Effects of Polygoni Multiflori Radix (PMR) on Osteogenesis and Osteoclasts in vitro)

  • Polygoni Multiflori Radix (PMR) a dried root tuber of Polygonum multiflorum Thunberg with bioactivities in bone metabolism is one of the most famous tonic traditional medicines in China, Japan and Korea.To observe the anti-osteoporotic effect of PMR, we observed the effects on proliferation and alkaline phosphatase (ALP) activity of primary osteoblasts, bone nodule formation, pit formation of osteoclasts and osteoclastogenesis in vitro. As the result of that, although PMR extracts have no meaningful changes on the the proliferation of primary osteoblasts, significant (p < 0.01) increases of ALP acitivity of osteoblasts were dose-dependently observed in all PMR extracts treated groups tested compared to that of vehicle control. It means that it enhanced the differentiation (ALP activity) and bone nodule formation of osteoblast. In addition, PMR extracts dose-dependently inhibits the pit formation and the number of multinucleated osteoclast-like cells (OCLs), osteoclastogenesis in vitro. Therefore these results show the possibility of PMR extracts as another anti-osteoporotic agents.

Inhibitory Effects of Total Saponin Korean Red Ginseng on Thromboxane A2 Production and P-Selectin Expression via Suppressing Mitogen-Activated Protein Kinases

  • Ginseng has been widely used for traditional medicine in eastern Asia and is known to have inhibitory effects on cardiovascular disease (CVD) such as thrombosis, atherosclerosis, and myocardial infarction. Because, platelet is a crucial mediator of CVD, many studies are focusing on inhibitory mechanism of platelet functions. Among platelet activating molecules, thromboxane $A_2$ ($TXA_2$) and P-selectin play a central role in CVD. $TXA_2$ leads to intracellular signaling cascades and P-selectin plays an important role in platelet-neutrophil and platelet-monocyte interactions leading to the inflammatory response. In this study, we investigated the inhibitory mechanisms of total saponin fraction from Korean red ginseng (KRG-TS) on $TXA_2$ production and P-selectin expression. Thrombin-elevated $TXA_2$ production and arachidonic acid release were decreased by KRG-TS dose (25 to $150{\mu}g/mL$)-dependently via down regulation of microsomal cyclooxygenase-1 (COX-1), $TXA_2$ synthase (TXAS) activity and dephosphorylation of cytosolic phospholipase $A_2$ ($cPLA_2$). In addition, KRG-TS suppressed thrombin-activated P-selectin expression, an indicator of granule release via dephosphorylation of mitogen-activated protein kinases (MAPK). Taken together, we revealed that KRG-TS is a beneficial novel compound inhibiting $TXA_2$ production and P-selectin expression, which may prevent platelet aggregation-mediated thrombotic disease.

Anti-ulcer and wound healing activity of Ruta graveolens

  • The effects of ethanol extract of Ruta graveolens on ulceration induced by ethanol and wound healing properties were investigated in mice. Oral administration of the extract reduced the incidence of ulcer, ulcer index and ulcer length produced by ethanol. The gastroprotective effects of R. graveolens were observed in a dose-dependent manner. However, this activity was statistically less potent than the anti-ulcer drug, omeprazole. When the extract applied on the wound, it promoted wound healing in mice. The wound contraction was faster in these mice when compared to untreated wounds. Based on these present findings, R. graveolens possesses anti-ulcer activity and promotes incision wound healing in mice.

돼지에서 두 가지 Cefquinome 2.5% 제제의 생물학적 동등성 평가 (Bioequivalence Evaluation of Two Cefquinome 2.5% Injectable Products in Piglets)

  • Cefquinome은 제4세대 cephalosporin으로 동물전용의약품으로 개발되었으며, ${\beta}$-lactamases에 대해서 매우 안정하고, 그람 음성 세균 및 양성 세균에 대한 광범위한 살균력을 가지고 있다. 본 연구는 현재 시판중인 cefquinome 주사제를 이유 자돈에 2 mg/kg 용량으로 근육 주사한 후 약물동태학적 특성을 파악하여 두 제제의 생물학적 동등성을 평가하였다. Cefquinome의 혈중 농도는 액체크로마토그래프/질량분석기를 이용하여 분석하였으며, 생물학적 동등성을 판정하기 위한 약물동태학적 인자로는 혈중 최고 농도 ($C_{max}$)와 혈장 농도 곡선하 면적 ($AUC_{0{\rightarrow}{\infty}}$)을 사용하였다. 시험약과 대조약의 혈중 최고 농도는 $4.34{\pm}0.58{\mu}g/mL$$4.22{\pm}0.47{\mu}g/mL$로 각각 나타났으며, 혈장 농도 곡선 하 면적은 $10.43{\pm}1.96{\mu}g{\cdot}h/mL$$10.25{\pm}2.98{\mu}g{\cdot}h/mL$로 관찰되었다. 로그변환한 약물동태학적 인자의 평균비율의 90% 신뢰 구간은 $C_{max}$의 경우 0.941-1.115이었고, $AUC_{0{\rightarrow}{\infty}}$의 경우 0.927-1.172로서 생물학적 동등성 기준인 0.8-1.25를 모두 만족시켰다. 이상의 결과로 시판중인 두 cefquinome 제제는 생물학적으로 동등하다고 판단된다.

Radioprotective Effect of Lifukang, a Chinese Medicinal Plants Prescription

  • We have examined in vitro and in vivo radioprotective effect of a Chinese medicinal plants prescription, Lifukang. Micronucleus assay was employed to evaluate in vitro radioprotective effect of Lifukang. In the presence of Lifukang, the frequencies of miconuclei were greatly reduced from 7.2 to 2.9, 1.6 and 1.6% at the concentrations of Lifukang from 0 to 2, 10 and $50{\mu}g/ml$, respectively. For in vivo assay, we monitored the incidences of apoptotic cells in mouse small intestine crypts and endogeneous spleen colonies. When Lifukang was administered to mice P.O. Or I.P. at doses of 1 mg/ml in drinking water for 7 days or 0.3 mg/mouse 24 hrs prior to irradiations, respectively, the average numbers of apoptotic cells were reduced to 3.1 or 2.3, respectively, as compared to 4.4 acquired from untreated control experiments. In addition, in spleen colony assay, Lifukang increased the number of hematopoietic spleen colonies. When samples were administered after irradiation, better results were obtained. The numbers of spleen colonies were increased from 14 colonies to 18.3 or 19.6 colonies when Lifukang was given through P.O. (1 mg/ml in drinking water for 11 days) or I.P. (0.3 mg/ mouse) after irradiation, respectively.
