우리나라 일부 소금의 중금속 함량에 대한 조사연구 (A Study on the Heavy Metal Contents of Common Salts in Korea)

  • In order to determine the content levels of trace metals in common salts, 64 bay salt samples were collected from three producing districts and 33 bay salt samples, 32 remade salt samples and 5 fine salt samples were collected from 7 major cities in Korea, from August to September 1987. These were analysed for content levels of Pb, Cd, Cu, Zn and Mn using Atomic Absorption Spectrophotometer. The results were as follows: 1. Lead contents in three type salts were N.D. - 1081.9 $\mu$g/kg and fourteen percent of the 114 samples exceeded the World Health Organization(WHO) criteria of 100$\mu$g/kg. Cadmium contents of samples were N.D.- 382C.5 $\mu$g/kg and five percent of the 114 samples were over the Spanish criteria of 500 $\mu$g/kg. Copper contents of samples were 8,9-214.9 $\mu$g/kg and there was not a sample over the World Health Organization(WHO) criteria of 500 $\mu$g/kg. Zinc contents ranged N.D. - 342.9 $\mu$g/kg and Manganese contents ranged N.D.- 8.31 mg/kg. 2. The comparison of heavy metal contents among the bay salts from three producing districts was significantly different in Pb, Cd and Cu contents. 3. The comparison of heavy metal contents between the bay salts and remade salts was not significantly different in Pb, Cd and Cu contents. 4. The contents of Pb, Cd, Zn and Mn in fine salts were much lower than those of bay salts and remade salts.

The Digestibility of Organic Trace Minerals along the Small Intestine in Broiler Chickens

  • An experiment was conducted to evaluate the effects of low concentrations of organic and inorganic dietary trace minerals on broiler performance and trace mineral digestibility along the small intestine of 35-day-old broiler chickens reared under floor-pen conditions. Eight hundred male, day-old Cobb broiler chickens were randomly allocated to 4 dietary treatments (25 birds per pen with 8 replicates per treatment). Broilers fed diets supplemented with 4, 20, 40 and 30 mg/kg, respectively, of Cu, Fe, Mn and Zn from organic chelates and inorganic salts achieved the same body weight gain as those supplemented at the NRC levels (8 mg Cu, 40 mg Fe, 60 mg Mn and 40 mg Zn/kg, respectively) from inorganic salts. However, birds fed a control diet without any supplementation at dietary levels of 7.4-8.8, 60.1-69.2, 14.6-15.4 and 19.1-20.6 mg/kg of Cu, Fe, Mn and Zn, respectively, had decreased feed intake and growth rate. There was no significant difference in the digestibility of Cu in all regions of the small intestine. Throughout the small intestine the apparent absorption of Mn from both organic and inorganic sources was small, whereas the digestibility of Zn seemed to be more complex, exhibiting differences in the apparent absorption due to both mineral source and intestinal site. Therefore, the digestibility of organic Zn was improved (p<0.01) in the ileum compared to inorganic Zn. The digestibility of Zn in the duodenum was smaller (p<0.05) than that in the ileum.

미생물발효에 의한 L-Phenylalanin생산에 미치는 배지성분의 영향 (Effects of Medium Components on Microbial Production of L-Phenyralsnine)

  • 본 연구에서는 매생물발효에 의한 L-phenylalanine생산에 미치는 배지조성의 영향을 살펴보았다. 탄소원의 선택은 L-phenylalanine생산에 큰 영향을 미침을 알았다. 자당, 포도당, 과당 혹은 그 혼합물을 탄소원으로 사용할때에 비해 당밀을 사용하는 것이 L-phenylalapine악 생산성을 크게 향상시켜 주었다. 최소배시의 사용시 미량의 염류들이 요구됨을 확인하였으나, 과량으로 첨가하면 더이상의 효고는 없었다. 또한 세포의 증식과 아미노산의 생산에 있어 적정량의 biotin과 thiamine이 필요함도 알I 수 있었다. Corynehacterium 균주외에 L.phenylalanine생산주로 알려진 조절기작을 상실한 영양요구성 변이주인 Brev-ihacterium lactofermenticum과 Arthrohacter citreus의 생산능을 조사해본 결과, Corynehacterium에 비해 생산성이 낮았으며 역시 영양요구 성질을 잃어버린 것으로 확인되었다.

Pichia pastoris에서 메탄올 유도시 첨가물이 재조합 HBsAg 생산에 미치는 영향 (Effect of Various Additives on the Production of Recombinant HBsAg during Methanol Induction in Pichia pastoris)

  • 본 연구에서는 P. pastoris를 이용한 유전자 재조합 HBsAg 생산에서 메탄올 유도시 여러 가지 첨가물들의 영향에 대해서 알아보았다. 회분식 배양에서 탄소원으로 글리세롤을 사용하다가 유가식 배양의 공급 탄소원으로 글리세롤이 아닌 당알콜인 sorbitol로 대체하였을 때 단백질 발현이 향상된 결과를 보였다. 또한 메탄올 유도시에 적당량의 아미노산 혼합물 첨가는 세포증식에는 영향이 없었지만 단백질 발현율은 크게 증가시켰다. 계면활성제인 Trition X-100의 첨가는 세포증식과 단백질 발현을 현저히 감소시켰지만, Pluronic F-68를 첨가했을 경우 세포증식의 저해영향 없이 단백질 발현율을 향상시켰다. 배지 부피의 0.01%(v/v)으로 oleic acid를 첨가하면 플라스크 배양에서는 단백질 발현에 긍정적인 효과를 보였으나, 5 L 발효조 배양에서는 메탄올 유도 시간이 지속되면서 첨가하지 않은 경우에 비해 발현율이 낮아지는 결과를 보였다. 마지막으로 trace salts는 첨가량에 따라 세포증식에는 영향이 없으며 단백질 발현에는 소량 trace salts 첨가로 단백질 발현에 긍정적인 효과를 보였다. 하지만 첨가량이 많아질수록 단백질 발현에는 부정적인 영향을 보임을 확인할 수 있었다.

Effects of hypochlorite exposure on morphology and trace organic contaminant rejection by NF/RO membranes

  • The impacts of membrane degradation due to chlorine attack on the rejection of inorganic salts and trace organic contaminants by nanofiltration (NF) and reverse osmosis (RO) membranes were investigated in this study. The rejection of trace contaminants was examined at environmentally relevant concentrations. Changes in the membrane surface morphology were observed as a result of chlorine exposure. A small increase in rejection was consistently observed with all four membranes selected in this study after being exposed to a low concentration of hypochlorite (100 ppm). In contrast, a higher concentration of hypochlorite (i.e., 2000 ppm) could be detrimental to the membrane separation capacity. Membranes with severe chlorine impact showed a considerable decrease in rejection over filtration time, possibly due to rearrangement of the polyamide chains under the influence of chlorine degradation and filtration pressure. The reported results indicate that loose NF membranes are more sensitive to chlorine exposure than RO membranes. The impact of hypochlorite exposure (both positive and negative) on rejection is dependent on the strength of the hypochlorite solution and is more significant for the neutral carbamazepine compound than the negatively charged sulfamethoxazole.

Effect of Various Factors on the Operational Stability of Immobilized Cells for Acrylamide Production in a Packed Bed Reactor

  • The effect of concentrations of phosphate buffer and acrylonitrile, pH, and various salts on the operational stability of the immobilized cells of Brevibacterium CH2 in a packed bed reactor were investigated. The effects of salts and carriers on the swelling of the immobilized beads during hydrolysis in a columnreactor were also investigated. Immobilization of the cells in Ba-alginate was more desirable than those in polyacrylamide and Ca-alinate for the swelling of the immobilized beads and the desired quality of the acrylamide produced. High quality acrylamide was produced using the Ba-alginate beads in a recycle fed-batch reactor without using an isotonic substrate. The conversion yield was nearly 100%, including a trace amount of acrylic acid produced as a by-product.

Influence of a Single Dose of Fe Dextran Administration with Organic Trace Mineral Supplementation on the Performance of Piglets

  • This study was designed to evaluate the influence of a single or double dose of Fe dextran with organic trace mineral supplementation on the performance of piglets from dams fed diets with either inorganic (ITM) or organic trace minerals (OTM). It also determined the effect of the source of the trace minerals on the reproductive performance of sows. The trace mineral premixes were prepared using metal proteinates and the corresponding inorganic salts for the OTM and the ITM, respectively. Each mineral premix provided 100 ppm Fe/175 ppm Fe, 35 ppm Cu/170 ppm Cu, 90 ppm Zn/120 ppm Zn, and 40 ppm Mn/35 ppm Mn when added at 0.20% in sows /weaned pigs' diets, respectively. The first dose of Fe dextran was administered to piglets at 3 d and the second dose at 10 d after birth. One dose of Fe dextran supplied 100 mg of Fe. A total of 16 gestating sows (Landrace${\times}$Yorkshire) in parities 2 to 4 were randomly allocated to four treatments: 1) diet with ITM/one dose of Fe dextran to piglets, 2) diet with ITM/two doses of Fe dextran to piglets, 3) diet with OTM/one dose of Fe dextran to piglets, and 4) diet with OTM/two doses of Fe dextran to piglets. The total born alive, weaned, body weight at birth and at weaning were not affected by the sow's dietary treatment. Although organic trace mineral supplementation tended to increase the milk Fe content (p<0.10) at 7 d postpartum, piglets in all treatments performed equally from birth to weaning. The double doses of Fe dextran neither improved the average daily gain (ADG) nor influenced the survival of piglets from birth to weaning (21 d). Results suggest that a single dose of Fe dextran given to suckling pigs is adequate to sustain their needs for growth throughout the lactation period (21 d). Furthermore, there was a 21% improvement in both the ADG and the average daily feed intake (ADFI) (p<0.05) in weaned pigs fed diets with OTM. Cu and Fe in the liver (p<0.01), and Zn in both the bone (p<0.01) and the serum (p<0.01) were higher in piglets fed OTM than in those fed ITM. It would be concluded that single dose of Fe dextran administration with organic trace mineral supplementation show similar growth performance compared to 2 dose Fe dextran administration with inorganic mineral supplementation in young pigs.

석탄회를 이용한 염류집적 토양 개선과 작물 생육 증진 (Improvement of Salt Accumulated Soil and Crop Growth using Coal Ash)

  • 본 연구는 폐자원 재활용 측면에서 농업적 유용활용(beneficial use)이 가능할 것으로 판단되는 영가철과 석탄회로 제조한 토양개량제를 투입하여 토양 염류 농도와 작물 생산량에 미치는 영향을 배양시험과 재배시험으로 평가하였다. 토양개량제는 석탄회를 구형으로 제형하고 영가철이 함유된 용액(0.1, 1.0%)을 표면에 살포하여 제조하였다. 토양개량제는 밭토양으로 충진된 와그너포트에 40 g/10a과 400 g/10a를 처리하였고, 30일 간 배양한 후 토양 EC와 염 농도를 분석하였다. 토양의 EC는 400 g/10a 실험구에서 대조구 대비 약 50% 감소한 것으로 나타났다. 국내 대표적인 염류인 유효인산의 경우 개량제의 처리 후 1/3 수준으로 감소되었다. 개량제 처리에 따른 토양 내 EC와 염 농도의 감소는 매우 유의한 수준으로 분석되었다(r=0.672~0.985). 또한 개량제 처리 후 뿌리와 줄기의 생육은 약 10% 증가였고, 건중량 또한 대조구에 비해 줄기와 뿌리에서 각각 30-50%와 60-75% 유의하게 증가하였다. 석탄회와 영가철을 농업용 토양개량제로 재활용할 경우 농업 환경 개선뿐만 아니라 농업 생산성 증대에도 기여할 수 있을 것으로 판단된다.

고체상미량분석법(SPME-GC/FID)에서 실험계획법을 이용한 디젤첨가제 미량분석의 최적화 연구 (Optimization Study of Trace Analysis of Potential Diesel Oxygenate Using the Design Of Experiment (DOE) in Solid-Phase Microextraction with GC/FID)

  • 본 연구에서는 액상에서 EGBE, DGBE, DBM, TGME의 효율적인 분석을 위해 이용되던 액상추출법을 대신할 수 있는 방법으로 GC/FID를 이용한 SPME법 실험 연구를 진행하였다. 그리고 실험계획법(DOE)을 사용하여 EGBE를 포함한 디젤첨가제 미량분석의 최적조건을 설명하였다. 실험은 통계분석결과 뿐만 아니라 요인 수의 최적화에 따른 중심합성설계에 의한 완전요인 설계법을 사용하였다. 반응표면분석은 추출 효율이 주 영향인 염농도, 흡착 온도, 흡착 시간과 sonication 시간에 따른 2차 다항식에 의해 설명될 수 있음을 보여주었다. 실험계획법(DOE)을 사용하는 것은 액상 시료에서 디젤첨가제의 정량분석을 개선하는 자료분석법이 될 것이다.

Optimization of media components for cordycepin production from Paecilomyces japonica

  • 본 연구는 P. japonira의 액체배양을 통해 HPLC를 이용하여 유용물질인 cordycepin의 농도를 분석하였다. 배지성분중 무기질소원, 아미노산, 무기염류 및 금속이온의 영향을 검토하였다. 그 결과 균사체의 경우에서는 cordycepin의 농도가 매우 낮음을 알 수 있었고, 배양여액의 경우에서는 $(NH_4)_2HPO_4$. histidine. $CaCO_3$ 등을 첨가했을 때 control에 비해 높은 농도의 cordycepin의 생성을 확인할 수 있었다.
