Clostridium difficile present in feces of food animals may contaminate their meats and act as a potential source of C. difficile infection (CDI) to humans. C. difficile resistance to antibiotics, its production of toxins and spores play major roles in the pathogenesis of CDI. This is the first study to evaluate C. difficile prevalence in retail raw animal meats, its antibiotics susceptibilities and toxigenic activities in Al-Jouf, Saudi Arabia. Totally, 240 meat samples were tested. C. difficile was identified by standard microbiological and biochemical methods. Vitek-2 compact system confirmed C. difficile isolates were 15/240 (6.3%). Toxins A/B were not detected by Xpect C. difficile toxin A/B tests. Although all isolates were susceptible to vancomycin and metronidazole, variable degrees of reduced susceptibilities to moxifloxacin, clindamycin or tetracycline antibiotics were detected by Epsilon tests. C. difficile strains with reduced susceptibility to antibiotics should be investigated. Variability between the worldwide reported C. difficile contamination levels could be due to absence of a gold standard procedure for its isolation. Establishment of a unified testing algorithm for C. difficile detection in food products is definitely essential to evaluate the inter-regional variation in its prevalence on national and international levels. Proper use of antimicrobials during animal husbandry is crucial to control the selective drug pressure on C. difficile strains associated with food animals. Investigating the protective or pathogenic potential of non-toxigenic C. difficile strains and the possibility of gene transfer from certain toxigenic/ antibiotics-resistant to non-toxigenic/antibiotics-sensitive strains, respectively, should be worthy of attention.
The antifungal activity of isolated lactic acid bacteria (LAB) from a locally fermented cereal, "Kunu", was tested against toxigenic Aspergillus flavus. The liquid refreshment, "Kunu", was prepared under hygienic condition using millet, sorghum, and the combination of the two grains. The antifungal potential of isolated LAB against toxigenic A. flavus was carried out using both in vitro and in vivo antifungal assays. The LAB count from prepared "Kunu" ranged from $2.80{\times}10^4CFU/mL$ to $4.10{\times}10^4CFU/mL$ and Lactobacillus plantarum, Lactobacillus delbrueckii, Lactobacillus fermentum, Pediococcus acidilactici, and Leuconostoc mesenteroides were the isolated bacteria. Inhibitory zones exhibited by LAB against toxigenic A. flavus ranged from 5.0 mm to 20.0 mm. The albino mice infected with toxigenic A. flavus showed sluggishness, decrease in body weight, distortion of hair, and presence of blood in their stool, while those treated with LAB after infection were recovered and active like those in control groups. Except for the white blood cell that was increased in the infected mice as $6.73mm^3$, the packed cell volume, hemoglobin, and red blood cell in infected animals were significantly reduced (P<0.05) to 29.28%, 10.06%, and 4.28%, respectively, when compared to the treated mice with LAB and control groups. The antifungal activity of LAB against toxigenic A. flavus can be attributed to the antimicrobial metabolites. These metabolites can be extracted and used as biopreservatives in food products to substitute the use of chemical preservatives that is not appealing to consumers due to several side effects.

The spatio-temporal distribution of dinoflagellate resting cysts was investigated by palynological processing to understand the harmful algal bloom (HAB) potential in the sediment of the Saemangeun area in 2003. In total, thirty-two dinoflagellate species were identified, and their concentrations were in the range of $6-1,618\;cysts{\cdot}g^{-1}$ (dry weight). The concentrations of resting cysts were higher in the spring (26 species, $64-1,101\;cysts{\cdot}g^{-1})$ and summer (30 species, $81-1,618\;cysts{\cdot}g^{-1})$ than in the autumn (32 species, $6-1,150\;cysts{\cdot}g^{-1})$ and winter (24 species, $25-728\;cysts{\cdot}g^{-1}).$ The composition rate of the heterotrophic dinoflagellate species to the total, which is closely related to the eutrophication process, to ranged from 6 to $29\%$ in the study area. The most dominant species was toxic Alexandrium tamarense/catenella $(25\%)$ followed by Gonyaulax scrippsea $(9\%)$ and toxic Protoceratium reticulatum $(5\%).$ Given the high abundance of the toxigenic dinoflagellate species, the Saemangeun area is considered to have the great potential for HABs in the future.
There has been an impetus in the development of biocontrol agents (BCAs) with the removal of a number of chemical compounds in the market, especially in the European Union. This has been a major driver in the development of Integrated Pest Management systems (IPM) for both pest and disease control. For control of mycotoxigenic fungi, there is interest in both control of colonization and more importantly toxin contamination of staple food commodities. Thus the relative inoculum potential of biocontrol agent vs the toxigenic specie sis important. The major bottlenecks in the production and development of formulations of biocontrol agents are the resilience of the strains, inoculum quality and formulation with effective field efficacy. It was recently been shown for mycotoxigenic fungi such as Aspergillus flavus, under extreme climate change conditions, growth is not affected although there may be a stimulation of aflatoxin production. Thus, the development of resilient biocontrol strains which can may have conserved control efficacy but have the necessary resilience becomes critical form a food security point of view. Indeed, under predicted climate change scenarios the diversity of pests and fungal diseases are expected to have profound impacts on food security. Thus, when examining the identification of potential biocontrol strains, production and formulation it is critical that the resilience to CC environmental factors are included and quantified. The problems in relation to the physiological competence and the relative humidity range over which efficacy can occur, especially pre-harvest may be increase under climate change conditions. We have examined the efficacy of atoxigenic strains of A. flavus and Clanostachys rosea and other candidates for control of A. flavus and aflatoxin contamination of maize, and for Fusarium verticillioides and fumonisin toxin control. We have also examined the potential use of fluidized-bed drying, nanoparticles/nanospheres and encapsulation approaches to enhance the potential for the production of resilient biocontrol formulations. The objective being the delivery of biocontrol efficacy under extreme interacting climatic conditions. The potential impact of climate change factors on the efficacy of biocontrol of fungal diseases and mycotoxins are discussed.
In order to develop a protocol to quantify cyanobacteria and Microcystis simultaneously, the primers and probe were designed from the conserved regions of 16S rRNA gene sequences of cyanobacteria and Microcystis, respectively. Probe match analysis of the Ribosomal Database Project showed that the primers matched with over 97% of cyanobacterial 16S rRNA genes, indicating these can be used to amplify cyanobacteria specifically. The TaqMan probe, which is located between two primers, matched with 98.2% of sequences in genus GpXI, in which most Microcystis strains are included. The numbers of cyanobacterial genes were estimated with the emission of SYBR Green from the amplicons with two primers, whereas those of Microcystis spp. were measured from the fluorescence of CAL Fluor Gold 540 emitted by exonuclease activity of Taq DNA polymerase in amplification. It is expected that this method enhances the accuracy and reduces the time to count cyanobacteria and potential toxigenic Microcystis spp. in aquatic environmental samples.
Bacillus thuringiensis microbial insecticide products have been applied worldwide. Although a few cases of B. thuringiensis foodborne illness have been reported, little is known about the toxigenic properties of B. thuringiensis isolates. The aims of this study were to estimate the pathogenic potential of B. thuringiensis selected from microbial insecticide products, based on its possession of toxin genes and production of enterotoxins. Fifty-two B. thuringiensis strains selected from four kinds of microbial insecticide products were analyzed. PCR assay for detection of toxin genes and immunoassay for detection of enterotoxins were performed. The hemolysin BL complex as a major enterotoxin was produced by 17 (32.7%), whereas the non-hemolytic enterotoxin complex was detected in 1 (1.9%) of 52 B. thuringiensis strains. However, cytK, entFM, and ces genes were not detected in any of the tested B. thuringiensis strains. The potential risk of food poisoning by B. thuringiensis along with concerns over B. thuringiensis microbial insecticide products has gained attention recently. Thus, microbial insecticide products based on B. thuringiensis should be carefully controlled.
Cyanobacteria are well known for their production of a multitude of highly toxic and / or allelopathic compounds. Among the photosynthetic microorganisms, cyanobacteria, belonging to the genus Nostoc are regarded as good candidate for producing biologically active secondary metabolites which are highly toxic to humans and other animals. Since so many reports have been published on the poisoning of different animals from drinking water contaminated with cyanobacteria toxins, it might be assumed that bioactive compounds are found only in aquatic species causes toxicity. However, the discovery of several dead dogs, mice, ducks, and fish around paddy fields, prompted us to study the toxic compounds in a strain of Nostoc which is most abundant in the paddy fields of Iran, using polymerase chain reaction and liquid chromatography coupled with a diode array detector and mass spectrophotometer. Results of molecular analysis demonstrated that the ASN_M strain contains the nosF gene. Also, the result of ion chromatograms and $MS^2$ fragmentation patterns showed that while there were three different peptidic compound classes (anabaenopeptin, cryptophycin, and nostocyclopeptides), there were no signs of the presence of anatoxin-a, homoanatoxin-a, hassallidin or microcystins. Moreover, a remarkable antifungal activity was identified in the methanolic extracts. Based on the results, this study suggests that three diverse groups of potentially bioactive compounds might account for the death of these animals. This case is the first documented incident of toxicity from aquatic cyanobacteria related intoxication in dogs, mice, and aquatic organisms in Iran.
Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous in the environment. They are highly toxigenic and carcinogenic. Probiotic bacteria isolated from fermented foods were tested to check their ability to degrade and/or detoxify PAHs. Five probiotic bacteria with distinct morphologies were isolated from a mixture of 26 fermented foods co-cultured with benzo(a)pyrene (BaP) containing Bushnell Haas minimal broth. Among them, B. velezensis (PMC10) significantly reduced the abundance of BaP in the broth. PMC10 completely degraded BaP presented at a lower concentration in broth culture. B. velezensis also showed a clear zone of degradation on a BaP-coated Bushnell Haas agar plate. Gene expression profiling showed significant increases of PAH ring-hydroxylating dioxygenases and 4-hydroxybenzoate 3-monooxygenase genes in B. velezensis in response to BaP treatment. In addtion, both live and heat-killed B. velezensis removed BaP and naphthalene (Nap) from phosphate buffer solution. Live B. velezensis did not show any cytotoxicity to macrophage or human dermal fibroblast cells. Live-cell and cell-free supernatant of B. velezensis showed potential anti-inflammatory effects. Cell-free supernatant and extract of B. velezensis also showed free radical scavenging effects. These results highlight the prospective ability of B. velezensis to biodegrade and remove toxic PAHs from the human body and suggest that the biodegradation of BaP might be regulated by ring-hydroxylating dioxygenase-initiated metabolic pathway.
Objectives: Microcystin (MC) produced during cyanobacterial blooms is a worldwide problem presenting a serious health threats to humans and ecosystems. During July through October of 2013, the Ilwol Reservoir experienced a high biomass of phytoplankton (maximum $211.7mg/m^3$ of Chlorophyll-a) containing the toxigenic cyanobacterium Oscillatoria sp. The aim of this study is to analyze MC concentration in the reservoir water, as well as in representative fish species (Carassius cuvieri, Carassius auratus, Channa argus). We also evaluated the human health risk of exposure to MCs accumulated in the fish. Methods: Concentrations of MCs in the water and fish samples were analyzed by liquid chromatography with a triple quadrupole tandem mass spectrometer (LC/MS/MS) and enzyme-linked immunosorbent assay (ELISA). Results: The total levels of four MC variants, including MC-LR, MC-RR, MC-YR and MC-LA were below the WHO drinking water guideline limit (1 ug MC-LR per liter) both for the dissolved and particulate fraction present in the water samples. The mean MC concentrations in the livers of all species were significantly higher than in the gills (p < 0.01) and muscles (p < 0.05). The values of estimated daily intake of MCs in muscles, the edible part of the fish, would be only $0.005-0.015{\mu}g/kg{\cdot}day$, much lower than WHO's provisional tolerable daily intake of $0.04{\mu}g/kg{\cdot}day$. Conclusion: This study suggests that, owing to the spatial distribution or temporal variation of MC, there is a need for careful monitoring of cyanotoxin in reservoir water and aquatic animals to protect public health.

A total of 110 Fusarium graminearum isolates were obtained from corn and barley samples which were collected from Kangwon province and the southern part of Korea, respectively. The isolates were tested for trichothecene and zearalenone (ZEA) production in rice culture. The incidences of trichothecene production by 51 isolates of F. graminearum from corn were 64.7% for deoxynivalenol (DON), 7.8% for 3-acetyldeoxynivalenol (3-ADON),33.3% for 15-acetylde-oxynivalenol (15-ADON), 21.6% for invalenol (NIV), and 13.7% for 4-acetylnivalenol (4-ANIV). DON producers frequently co-produced 15-ADON rather than 3-ADON. On the other hand, the incidences of trichothecene production by 59 isolates of F. graminearum from barley were 71.2% for NIV, 61.0% for 4-ANIV, and only one isolate produced DON and 3-ADON. The incidences and mean levels of ZEA producers were 32.0% and 71.$\mu$g/g for the isolates from corn, and 29.0% and 74 .$\mu$g/g for the isolates from barley. There was a great regional difference in trichothecene production of F. graminearum isolates between Kangwon province and the southern part of Korea.
