Studies for Reestabilishment of Approval Toxin Amount in Paralytic Shellfish Poison-Infested Shellfish 5. Comparison of Toxicity and Toxin Composition of Paralytic Shellfish Poison between Blue mussel, Mytilus edulis and Oyster, Crassostrea gigas

  • The toxicity and toxin composition between blue mussel, Mytilus edulis and oyster, Crassostrea gigas collected at Woepori in Ko je island in South Coast of Korea in 1996 and 1997 were compared. The highest toxicity score was about 10 times higher in blue mussel than oyster (blue mussel, 8,670 $\mu\textrm{g}$; oyster, 860$\mu\textrm{g}$ in 1996, blue mussel, 5,657 $\mu\textrm{g}$/100g in 1997). The blue mussel also retained its toxicity for slightly longer period than oyster. In the both shellfish, PSP was composed almost exclusively of C toxicity (Cl and C2, 20~65%) and gonyautoxins (GTXl, 2, 3, and 4, 38~78%). In the early period of toxin accumulation, the ratio of 11$\beta$-epimer toxins (C2, GTX4) whose amount was 25~56 mole% (5th March to 12th April in 1996) and 25~80 mole% (18th March to 7th April in 1997), were higher than that of 11-epimer toxins (Cl, GTX2) whose amount was 41~57 mol%(27th May to 3rd June in 1996) and 25~56 mole% (29th April to 12th May in 1997), became higher than that of 11-epimer toxins. The toxin compositions in the both samples changed on a daily basis, presumably owing to metabolism of the toxin in the bivalves.

Comparison of Toxicity and Deposition of Nano-Sized Carbon Black Aerosol Prepared With or Without Dispersing Sonication

  • Nanotoxicological research has shown toxicity of nanomaterials to be inversely related to particle size. However, the contribution of agglomeration to the toxicity of nanomaterials has not been sufficiently studied, although it is known that agglomeration is associated with increased nanomaterial size. In this study, we prepared aerosols of nano-sized carbon black by 2 different ways to verify the effects of agglomeration on the toxicity and deposition of nano-sized carbon black. The 2 methods of preparation included the carbon black dispersion method that facilitated clustering without sonication and the carbon black dispersion method involving sonication to achieve scattering and deagglomeration. Male Sprague-Dawley rats were exposed to carbon black aerosols 6 hr a day for 3 days or for 2 weeks. The median mass aerodynamic diameter of carbon black aerosols averaged $2.08{\mu}m$ (for aerosol prepared without sonication; group N) and $1.79{\mu}m$ (for aerosol prepared without sonication; group S). The average concentration of carbon black during the exposure period for group N and group S was $13.08{\pm}3.18mg/m^3$ and $13.67{\pm}3.54mg/m^3$, respectively, in the 3-day experiment. The average concentration during the 2-week experiment was $9.83{\pm}3.42mg/m^3$ and $9.08{\pm}4.49mg/m^3$ for group N and group S, respectively. The amount of carbon black deposition in the lungs was significantly higher in group S than in group N in both 3-day and 2-week experiments. The number of total cells, macrophages and polymorphonuclear leukocytes in the bronchoalveolar lavage (BAL) fluid, and the number of total white blood cells and neutrophils in the blood in the 2-week experiment were significantly higher in group S than in normal control. However, differences were not found in the inflammatory cytokine levels (IL-$1{\beta}$, TNF-${\alpha}$, IL-6, etc.) and protein indicators of cell damage (albumin and lactate dehydrogenase) in the BAL fluid of both group N and group S as compared to the normal control. In conclusion, carbon black aerosol generated by sonication possesses smaller nanoparticles that are deposited to a greater extent in the lungs than is aerosol formulated without sonication. Additionally, rats were narrowly more affected when exposed to carbon black aerosol generated by sonication as compared to that produced without sonication.

Treatment outcome of localized prostate cancer by 70 Gy hypofractionated intensity-modulated radiotherapy with a customized rectal balloon

  • Purpose: We aimed to analyze the treatment outcome and long-term toxicity of 70 Gy hypofractionated intensity-modulated radiotherapy (IMRT) for localized prostate cancer using a customized rectal balloon. Materials and Methods: We reviewed medical records of 86 prostate cancer patients who received curative radiotherapy between January 2004 and December 2011 at our institution. Patients were designated as low (12.8%), intermediate (20.9%), or high risk (66.3%). Thirty patients received a total dose of 70 Gy in 28 fractions over 5 weeks via IMRT (the Hypo-IMRT group); 56 received 70.2 Gy in 39 fractions over 7 weeks via 3-dimensional conformal radiotherapy (the CF-3DRT group, which served as a reference for comparison). A customized rectal balloon was placed in Hypo-IMRT group throughout the entire radiotherapy course. Androgen deprivation therapy was administered to 47 patients (Hypo-IMRT group, 17; CF-3DRT group, 30). Late genitourinary (GU) and gastrointestinal (GI) toxicity were evaluated according to the Radiation Therapy Oncology Group criteria. Results: The median follow-up period was 74.4 months (range, 18.8 to 125.9 months). The 5-year actuarial biochemical relapse-free survival rates for low-, intermediate-, and high-risk patients were 100%, 100%, and 88.5%, respectively, for the Hypo-IMRT group and 80%, 77.8%, and 63.6%, respectively, for the CF-3DRT group (p < 0.046). No patient presented with acute or late GU toxicity ${\geq}$grade 3. Late grade 3 GI toxicity occurred in 2 patients (3.6%) in the CF-3DRT group and 1 patient (3.3%) in the Hypo-IMRT group. Conclusion: Hypo-IMRT with a customized rectal balloon resulted in excellent biochemical control rates with minimal toxicity in localized prostate cancer patients.

  • Obejectives : The electroacupuncture(EA) on regional fat is often used by oriental medicine doctors in Korea. However, there have been few studies about safety and biocompatibility of its application. Therefore we investigated the safety and change of mechanical character of electroacupuncture after its application on localized fat deposit. Methods : Online surveys were completed from Aug 21 2008 to Aug 28 2008, by email, the online survey data were obtained from nearly 10,002 Korean medical doctors who listed their email address on the website of association of korean oriental medicine. This study includes an experiment on the physical strength and biological property of long needles. We represent each method which were collected in advance survey, then sealed the needles off and did laboratory experiment, which includes surface observation, analysis, vickers hardness test, and also biocompatibility and toxicity test. Results : There was no considerable difference about the physical property after applying electric current, the cell survival rate did not change in comparison with controlled group either. Conclusions : Although it seems there are no considerable acupuncture-related problem, we still need additional studies about clinical effectiveness difference related operation condition. Besides, it will need research on the actual condition, standardization, and criteria about the comsumed needle in Korea.

Comparison of cytotoxicities and wound healing effects of diquafosol tetrasodium and hyaluronic acid on human corneal epithelial cells

  • This study aimed to compare the cellular toxicities of three clinically used dry eye treatments; 3% diquafosol tetrasodium and hyaluronic acid at 0.3 and 0.18%. A methyl thiazolyltetrazoiun (MTT)-based calorimetric assay was used to assess cellular proliferation and a lactate dehydrogenase (LDH) leakage assay to assess cytotoxicity, using Human corneal epithelial cells (HCECs) exposed to 3% diquafosol tetrasodium, 0.3% hyaluronic acid (HA), or 0.18% HA or 1, 6 or 24 h. Cellular morphology was evaluated by inverted phase-contrast light microscopy and electron microscopy, and wound widths were measured 24 h after confluent HCECs were scratched. Diquafosol had a significant, time-dependent, inhibitory effect on HCEC proliferation and cytotoxicity. HCECs treated with diquafosol detached more from the bottoms of dishes and damaged cells showed degenerative changes, such as, reduced numbers of microvilli, vacuole formation, and chromatin of the nuclear remnant condensed along the nuclear periphery. All significantly stimulated reepithelialization of HCECs scratched, which were less observed in diquafosol. Therefore, epithelial toxicity should be considered after long-term usage of diquafosol and in overdose cases, especially in dry eye patients with pre-existing punctated epithelial erosion.

Evaluation of Antifungal Activities and Safeties of 6-[(N-2,4-Dibromophenyl) amino]-7-Chloro-5,8-Quinolinedione (6-[(N-2,4-디브로모페닐)아미노]-7-클로로-5,8-퀴놀린디온의 항진균작용 및 안전성 평가)

  • 6-[(N-3,4-Dibromophenyl)amino]-7-chloro-5,8-quinolinedione(FCK13) was tested for antifungal activities. The MIC values were determined by the two-fold dilution method. The therapeutic potential of RCK13 had been assessed in comparison with ketoconazole and fluconazole against systemic infections with candida albicans in normal mice. RCK13 had ED50,0.80$\pm$0.21 mg/kg but ketoconazole had ED50, 8.00$\pm$0.73 mg/kg respectively. And administered RCK13 at the ED50 for 14 days improved survival rates as well as ketoconazole. Acute oral toxicity studies of RCK13 were carried out in ICR mice of both sexes. These acute oral toxicities of RCK13 were low and LD50 values were over 2,850 mg/kg in ICR mice. The genotoxicities of RCK13 had been evaluated. RCK13 was negative in Ames test with Salmonella typhimurium and chromosomal aberration test in CHL cells. The clastogenicity was tested on the RCK13 with in vivo mouse micronucleus assay. RCK13 did not show any clastogenic effect in mouse peripheral blood and was negative in mouse micronucleus assay. These results indicate that RCK13 has no genotoxic potential under these experimental conditions.

  • Using dispersants is known to be an effective solution to accelerate the natural dispersion and being an appropriate oil spill response strategy. By breaking up large oil chunks into small droplets, dispersants are generally intended to help reducing further oil exposures and slicks. Collecting property data of circulating crude oil in South Korea and understanding the interaction between crude oils and dispersions need to be preceded for the effective dispersant use. This paper provides an property analysis of three selected oils which have the same composition of spilt oils from the Herbei Spirit Incident and conducts an emulsification and toxicity experiment with selected domestic and foreign dispersants. Results will present a direction of future domestic dispersants' development which aimed at eco-friendly and safety.

  • This study was Performed to evaluate the effective solidification of Cd sludge using cement and power plant fly ash as cement admixture, to identify the leaching characteristics of the heavy metal Cd sludge solidified, and to develope proper KLT(Korean Leaching Test) of hazardous waste. KLT was compared with EPT(Extraction Procedure Toxicity) and TCLP(Toxicity Characteristics Leaching Procedure). Fly ash contents ranged from 0% to 30% of cement weight. The experimental results showed that the optimum amount of fly ash replaced was 10% to 15% and KLT was less appropriate than EPA and TCLP. Also the purpose of the study was to suggest the modification factors on the leaching test currently used, based on the above mentioned aspects. The effects of pH, leaching time, leaching for agitating intensity, and leaching solvent were investigated. As the result of test, the leaching potential was relatively high when the pH and agitation intensity of leaching solution were 5 and 150rpm, respectively. Leaching time of six hours was found to be sufficient and the use of acetic acid as a leaching solvent is more useful in landfill site particularly.

  • Objectives: This study was to investigate the signs and notations of skin absorption, carcinogenicity, germ cell mutagenicity, and reproductive toxicity in the occupational exposure limits of Korea and of other advanced countries. Methods: Information on occupational exposure limits in Korea, the USA, the UK, Germany, and Japan was investigated through the Internet, and items marked as carcinogenicity and skin absorption were compared by country. Results: Legal occupational exposure limits have been greatly simplified. However, in the case of HSE WEL, skin absorption, carcinogenicity classification, sensitization, and in the case of DFG MAK, skin absorption, carcinogenicity, pregnancy risk group, germ cell mutagenicity, airway and skin sensitization, photo contact sensitization, and vapor pressure were provided. Conclusions: It is desirable to indicate the carcinogenicity and skin absorption within permissible limits, and to include information on critical effects in chemical substance exposure limits to uphold the right to know of industrial hygienists and workers in Korea. It is also necessary to clarify the precautions, limitations and protections for skin absorption.

  • The present study was carried out to investigate the effect of Ni on germination, cell elongation, ${\alpha}-amylase$ activity, contents of chlorophyll and protein in radish were determined in the water culture. As the concentration of Ni was increased in the water culture, germination of radish was 55% by Ni 10 mg/kg and 30% by Ni 20 mg/kg. The ratio of cell elongation injury was 50%, by two days after Ni 20 mg/kg treatment. The injury ratio of ${\alpha}-amylase$ activity was 45% in the same condition and as the time goes on, inhibition of ${\alpha}-amylase$ activity were slightly decreased. Contents of chlorophyll a and b were decreased two days after treatment and chlorophyll a was more inhibited than chlorophyll b. Also changes of the protein contents was slightly decreased. Activity of ${\alpha}-amylase$ was decreased at germination stage, contents of chlorophyll a and b were decreased at growing stage.
