Porphyromonas gingivalis, the gram-negative anaerobic oral bacterium, initiates periodontal disease by binding to saliva-coated oral surface. The cholera toxin B subunit (CTB) genetically linked to FimA1 (1-200 aa) or FimA2 (201-337 aa) of the P. gingivalis fimbrial antigen were introduced into Solanum tuberosum cells by Agrobacterium tumefaciens-mediated transformation method. The integration of CTB-FimA1 or CTB-FimA2 fusion genes were confirmed in the chromosome of transformed leaves by genomic DNA PCR amplification method. Synthesis and assembly of the CTB-FimA fusion proteins into oligomeric structures with pentamer size was detected in transformed tuber extracts by immunoblot analysis. The binding activities of CTB-FimA fusion proteins to intestinal epithelial cell membrane receptors were confirmed by GM1-ganglioside enzyme-linked immunosorbent assay (GM1-ELISA). The ELISA showed that the expression levels of the CTB-FimA1 or CTB-FimA2 fusion proteins were 0.0019, 0.002% of the total soluble protein in transgenic tuber tissues, respectively The synthesis of CTB-FimA monomers and their assembly into biologically active oligomers in transformed potato tuber tissues demonstrates the feasibility of using edible plants for the production of enterocyte targeted fimbrial antigens that could elicit mucosal immune responses.
The present study was conducted to compare the relative efficiency of two different haploid breeding methods in tobacco varietal development. A single F t hybrid plant from cross of two flue-cured cultivars of Nicotiana tabacum L., Bright Yellow4(BY4) and NC 95, was used to develop the 30 dihaploid lines by anther culture(F1-ADH) and maternally-derived doubled haploid utilizing Nicotiana africana(F1-MDH), respectively. As compared with mid-parent, ADH lines showed increasing in number of leaves, delaying in days to flower and narrowing in leaf width. However, no significant differences in the other characters investigated were recognized. MDH lines also showed narrow leaf width, while no significant differences in the other characters were observed. The variations of the characters investigated were generally greater in ADH than MDH lines. MDH lines had higher plant height and shorter days to flower than ADH lines, while other characters did not show remarkable differences. The degree of heritability for each of the characters observed between ADH and MDH was almost the same. The characters showing high heritability value were plant height, leaf number, days to flower, and yield, while those showing relatively low value were leaf length, leaf width, and total alkaloid content. Predicted gains from selection for increased yield were calculated for both populations(F1-ADH, F1-MDH) and correlated responses associated with selection for yield were estimated. Plant height, leaf width, days to flower, percent reducing sugar and disease resistance would be expected to improve with selection for yield much faster in the MDH population than in the ADH.
Pot experiment was conducted to investigate the effects of fly ash on growth responses and on accumulation of the heavy metals in soil. Two kinds of fly ash, anthracite and bituminous coal, were treated with different levels of 0, 0.4, 0.8, and 1.2 kg/pot(20L). Tobacco growth was better by application of fly ash than that by the control. However, the early stage of growth by application of bituminous coal, 1.2 kg/pot, was decreased due to the boron toxicity occurred by fly ash. Generally, tobacco yield was significantly increased with applying fly ash, showing the highest yield at 1.2 kg/pot for anthracite and at 0.8 kg/pot for bituminous coal. The content of total nitrogen in leaves was higher with fly ash than that of the control, while the content of calcium in leaves was low, Contents of heavy metal and the other minerals were not significantly different between the control and the treatment of fly ash. Soil pH after experiment was linearly increased with application level of fly ash, indicating that the application of bituminous coal was more effective than that of anthracite. Contents of available phosphate, exchangeable $Ca^{2+}$+ and $Mg^{2+}$ in soil were increased by application of fly ash, especially with bituminous coal. Contents of Cu, Cr, and Ni were increased with application level of bituminous coati even if the contents were still lower than critical levels for farming land. The other heavy metals were similar between the control and the application of fly ash.

Johne's disease, caused by Mycobacterium avium subsp. paratuberculosis (MAP), is one of the most widespread and economically important diseases in cattle. Current diagnostic methods are based on the detection of anti-MAP antibodies in serum or isolation of the causative agent. However, these techniques are often not applicable for cases of subclinical infection due to relatively low sensitivity. Therefore, finding new antigen candidates that strongly react with the host immune system had been attempted. To effectively detect infection during the subclinical stage, several antigen candidates were selected based on previous researches. Characteristics of the selected antigen candidates were analyzed using bioinformatics-based prediction tools. A total of nine antigens were selected (MAP0862, MAP3817c, MAP2077c, MAP0860c, MAP3954, MAP3155c, MAP1204, MAP1087, and MAP2963c) that have MAP-specific and/or high immune responses to infected animals. Using a transmembrane prediction tool, five of the nine antigen candidates were predicted to be membrane protein (MAP3817c, MAP3954, MAP3155c, MAP1087, and MAP1204). Some of the predicted protein structures identified using the I-TASSER server shared similarities with known proteins found in the Protein Data Bank database (MAP0862, MAP1204, and MAP2077c). In future studies, the characteristics and diagnostic efficiency of the selected antigen candidates will be evaluated.
Offshore structures are exposed to low- and high-frequency responses due to environmental loads, and fatigue damage models are used to calculate the fatigue damage from these. In this study, we tried to optimize the main parameters used in fatigue damage calculation to derive a new fatigue damage model. A total of 162 bi-modal spectra using the elliptic equation were defined to describe the response of offshore structures. To calculate the fatigue damage from the spectra, time series were generated from the spectra using the inverse Fourier transform, and the rain-flow counting method was applied. The considered optimization variables were the size of the frequency increments, ratio of the time increment, and number of repetitions of the time series. In order to obtain optimized values, the fatigue damage was calculated using the parameter values proposed in previous work, and the fatigue damage was calculated by increasing or decreasing the proposed values. The results were compared, and the error rate was checked. Based on the test results, new values were found for the size of the frequency increment and number of time series iterations. As a validation, the fatigue damage of an actual tension spectrum found using the new proposed values and fatigue damage found using the previously proposed method were compared. In conclusion, we propose a new optimized calculation process that is faster and more accurate than the existed method.
Purpose: The goal of this study is to investigate the correlation of orthodontic treatment with self-esteem and how orthodontic treatment has an effect on university student's self-esteems by investigating and analyzing the difference in self-esteem of each general characteristic and the difference in self-esteem according to the existence or nonexistence of malocclusion and orthodontic treatment for adult university students. Methods: This study conduced a survey for 420 university students including dental related department, department of health and department of non-dental health in two universities located in Taegu, Gyeongbuk-do province from May 15, 2012 to May 30, 2012, and in 420 questionnaires of the investigated 420 target students, total 400(95.2%) questionnaires except for the questionnaires with falsely or erroneous responses were used for study analysis. Results: In the relations of general characteristic with self-esteem, for self-esteem by each gender, male student showed higher, for self-esteem by residential area, the university student residing at metropolitan area showed higher, for self-esteem by financial status, high class and middle class students in home financial status showed higher, for self-esteem by face shape, the student with triangular face shape showed higher, and in the relation of self-esteem with orthodontic treatment, the university student who is getting orthodontic treatment showed higher self-esteem statistically significantly. Conclusion: For gender, male university student, for residential area, the university student residing at metropolitan area, the high class university student in financial status, the university student with triangular face shape, and the university student who is getting orthodontic treatment showed statistically significantly higher self-esteem.
Purpose: This study is to examine the relationship between safety awareness and safety accident occurrence with elementary school students. Materials and methods: It selected 10 elementary schools under the supervision of Jeonnam Board of Education and sampled 700 students randomly from 2-3 classes in the 5th and the 6th grade. One preliminary survey was conducted to revise and complement the contents and forms of questionnaire with 70 students of a school from Apr. 1 to 6, 2004 and this study was conducted from Apr. 10 to 30, 2004. Total 700 questionnaires were distributed and 681 were collected (97%) and 602 were used for final analysis except 79 lacking responses. Data collected were analyzed with SPSS statistical program. Results: The average score of safety awareness of subjects by area was high as 2.72 points out of a possible 3. Area showing the highest safety awareness was safety from fire. Girl students had higher safety awareness than boy students. The fifth graders had higher safety awareness than the sixth graders. In safety awareness by the number of siblings, single son or single daughter showed the highest safety awareness. 53.2% of the subjects experienced accident for one year, the frequency of accident was mostly once and most of accidents were occurred at school. Accidents for the last year were higher in boy students, the sixth graders. According to relationship between safety awareness and safety accident, group with lower safety awareness in school safety(P < .022), traffic safety(P < .016), fire safety(P < .019), home safety(P < .007) and accident treatment and first aid(P < .003) had higher safety accidents than that with higher safety awareness. Conclusion: Development of safety education program by grade, that of parents safety education program, field experience and practice with students, teachers and parents and various safety education programs should be substantially and repeatedly accomplished for prevention education of safety accidents. In addition, to prevent and cope with safety accidents, safety education should be reflected on normal education, safety education textbook be developed, special safety education teachers be employed for safety education.
The purpose of this study was to investigate students' mental models and alternative conceptions about the Earth's interior. A total of 126 10th grade students participated in this study. They were requested to draw Earth's interior tasks and were interviewed about their mental models. A coding frame was designed to classify the students' drawings and interview responses, and then a four step cognitive model was established based on the frame of coding. In addition, the relationship of mental models was compared in terms of students' gender, and the type of alternative conceptions was analysed. The research results showed a variety of alternative conceptions on the interior of the Earth. The classified mental models showed naive mental model (11%), unstable mental model (81%), conceptual model (8%), while 69% of all sampled students accounted for the static process. The gender difference in the cognitive model showed no statistical significance.
As homework, a total of 32 seventh grade science-gifted students were asked to observe the moon at daily intervals for one month duration. They were also asked to take the photos and to record relevant variables in a given format. The purpose of the task was to investigate what students thought, as they observed the moon. The results show that students paid attention mainly to the position of the moon relative to other variables such as the date in the lunar calendar, the observer's position on the earth, and the position of the sun. Overall students' response implied that students did not observe the lunar phase in relation with relevant variables. Some reponses from students show common misconceptions such as the cause of the lunar phase to reflect the shadow of the earth. However, some responses reveal students' idea that has been rarely reported in the previous researches. For example, some students drew the moon to revolve in the opposite direction. Significant number of students drew the sun's position to be due west before the full moon and due east after the full moon. Few students recognized the relation between the time of observation and observer's position on the earth. The results of current research suggest that not only the education but also the research needs to be expanded to consider informal environment such as the actual field conditions.

The effects of formalin on mixed function oxygenase (MFO) system and stress-response were investigated in olive flounder (Paralichthys olivaceus). Olive flounder was exposed to formalin at the concentration of 300 ppm for 1, 2, 4 and 16 h. Levels of stress-response enzymes together with total protein, glucose and osmolality were quantitatively determined in blood, and the activities of phase I (cytochrome P450, ethoxyresorufin deethylase) and phase II (glutathione S-transferase) hepatic enzymes were also determined. Since the formalin-exposure for 16 h resulted no significant changes in aspartate aminotransferase and alanine aminotransferase, specific enzymes for liver damage, it was thought that it did not cause hepatic tissue damage at the concentration of 300 ppm. However, hepatic MFO system was induced at 1 to 4 h, and stress response was induced after 16 h of exposure. Moreover, it is considered that the depression of MFO activity after 16 h of exposure may not be adaptation to formalin, but toxic response. These results suggest that low concentration of formalin does not cause hepatic tissue damage of fish, but could induce MFO and stress response.
