Functional Activity of Water and Ethanol Extracts from Red Pepper (Capsicum annuum L.) Seeds (고추씨의 물과 에탄올 추출물의 생리활성)

  • This study was carried out to investigate the extract yield, total polyphenolic compounds content, electron donating activity (EDA) and nitrite scavenging activity (NSA) of various red pepper (Capsicum annuum L.) seeds. The water and ethanol extracts showed yields in the ranges of $11.30{\sim}18.93%$ and $3.00{\sim}5.25%$, respectively. Ethanol extract yield was higher than water. In the total polyphenol content, water and ethanol extracts were in the range of $10.22{\pm}1.29{\sim}25.98{\pm}0.55mg/$ and $8.00{\pm}0.57{\sim}33.99{\pm}0.09mg/g$, respectively. Also, nitrite scavenging activities were $70.11{\pm}0.71{\sim}94.07{\pm}0.86%$ and $81.93{\pm}1.77{\sim}99.90{\pm}0.70%$ for water and ethanol extracts, respectively. In the electron donating activity, water extracts showed lower activity than ethanol extracts. The electron donating activity of water extracts was in the range of $0.04{\pm}0.07{\sim}14.31{\pm}0.06%$ while that of NSA of ethanol extracts was $35.48{\pm}0.23{\sim}73.83{\pm}0.04%$.

Anti-bacterial Effects of Aqueous Extract Purified from the Immature Cone of Red Pine (Pinus densiflora) (미성숙 솔방울 열수추출물의 항균성)

  • Red pine (Pinus densiflora) is widely used traditional medicine, pharmacological and nutritional values from which the phytochemical compounds are derived. The present study was aimed to examine the antibacterial effects in the absence and presence of a immature red pine cone extract against 13 microorganisms. The components in the aqueous extract from immature red pine cone were identified by GC-MS. About 1.4% of total polyphenolic compound was measured in aqueous extract collected from immature red pine cone. Also, the high concentration of ${\beta}$-phellenandrene, ${\alpha}$-pinene, limonene, bornyl acetate and aldehyde was detected in total ion chromatograms. Of total 13 microorganisms, 4 microorganisms including Pseudomonas aeruginosa, Vibrio cholera, Listeria monocytogenes, Klebsiella pneumonia were effectively killed by aqueous extract of immature red pine cone. The highest anti-bacterial effect was detected in P. aeruginosa, followed by V. cholera, L. monocytogenes and K. pneumonia. In case of P. aeruginosa, the largest diameter of inhibition zone was maintained to 1/2 solution treated cells and slightly decreased at 1/4 and 1/8 solution treated cells. Also, in test used V. cholera and L. monocytogenes, the inhibition zone was strongly formed in only 1 and 1/2 solution treated cells, while K. pneumonia showed the very small diameter of inhibition zone in all concentrations. Therefore, these results suggested that the aqueous extracts of immature red pine cone should be considered as a new and potentially important anti-bacterial substrate to effectively prevent the microbial infection and penetration.

Antioxidant Compounds and Activities of Methanolic Extracts from Oat Cultivars (품종별 귀리 메탄올 추출물의 항산화 성분 및 항산화 활성)

  • Oats (Avena sativa L.) were evaluated in recent years as a promising crop for increasing the nutritional quality of foods, due to their abundance in many bioactive compounds. The objectives of this study were to determine antioxidant compounds and antioxidant activities of nine oat cultivars and investigate the relationships between antioxidant compounds and antioxidant activities in oat cultivars. The contents of total polyphenolics and vitamin E in oat cultivars were analyzed by spectrophotometric methods and normal phase HPLC, respectively. 2,2'-Azino-bis-(3-ethylbenzothiazoline- 6-sulphonic acid) (ABTS) radical and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities, reducing power, and chelating effects were used to compare the relative antioxidant activities of oat cultivars. In this study, total polyphenolic and vitamin E contents of oat cultivars ranged from 83.41 to 130.59 mg gallic acid equivalents/100 g and 0.54 to 1.90 mg/100 g, respectively. High levels of ABTS radical (116.14 mg Trolox equivalent antioxidant capacity (TEAC)/100 g) and DPPH radical (38.58 mg TEAC/100 g) scavenging activities and reducing power ($A_{700}=0.922$) were found in Daeyang. Significant correlation was evaluated between total polyphenolic contents and their antioxidant activities, namely ABTS and DPPH radical scavenging activities and reducing power. A low correlation was observed between vitamin E contents and their antioxidant activities. The results of this study could have an impact on oat consumption by enhancing consumer awareness of the health benefits of oats.

Biochemical and Antioxidant Activity of Yogurt Supplemented with Paprika Juice of Different Colors

  • Paprika is known to contain polyphenolic compounds that have good antioxidant properties. This study was conducted to investigate the benefits of adding paprika juice of different colors to yogurt and to determine how paprika affects the quality characteristics of yogurt. Stirred yogurt samples supplemented with different levels of red, orange, or yellow paprika juice were inoculated with mixes of Streptococcus thermophilus and Lactobacillus delbrueckii ssp. bulgaricus. Paprika addition decreased the pH but increased titratable acidity, lactic acid bacteria (LAB) counts, total polyphenol content (TPC), levels of vitamin A and C, and antioxidant activity. Proteolysis and viscosity of paprika-containing yogurts were significantly higher than those of the control yogurt without paprika juice (p<0.05). In particular, the viscosity of red paprika yogurt was significantly higher than that of yogurts containing 5% orange and yellow paprika juices (each p<0.05). The antioxidant activity of the methanol extract of the yogurt containing 2.5% orange paprika juice was the highest. Storage at 4℃ for 15 days only slightly altered LAB counts, antioxidant activity, and TPC of paprika yogurt. These results indicate that paprika could be used as a natural food additive for the development of functional yogurts.

Antioxidant Activity of Gamma-Irradiated Asparagus cochinchinensis (Asparagi radix) (Lour.) Merr. Extract and Inhibition Effect on Lipid Oxidation of Emulsion-Type Pork Sausage

  • The objective of this study was to determine the antioxidant activity of gamma-irradiated Asparagus cochinchinensis (Asparagi radix) (Lour.) Merr. Extract (ARE) and its inhibition effect on food lipid oxidation using emulsion-type pork sausage as a model. ARE was prepared from dried Asparagi radix root and ARE solution (1.0 g/mL) was gamma-irradiated with designated doses at 5, 10, and 20 kGy. Antioxidant activity of ARE solution was determined by measuring 1,1-diphenyl-e-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-9-sulphonic acid) (ABTS) radicals. Activities of DPPH and ABTS radicals were decreased, whereas total phenolic contents increased after gamma irradiation with a dose dependence. Addition of gamma-irradiated ARE dose-dependently retarded lipid oxidation of emulsion-type pork sausage during storage at $4^{\circ}C$. These results indicated that gamma-irradiated ARE might have antioxidant activity more than non-irradiated ARE due to increase of the content of polyphenolic compounds by ionizing radiation.

Protective effects of Paeoniae Radix Alba and Puerariae Radix combination on alcoholic liver disease (알코올성 간 손상 동물 모델에서 芍藥 葛根 복합물의 간 손상 보호 효과)

  • Objective : Alcoholic liver disease (ALD) is caused by excess alcohol intake. In the liver, alcohol breakdown results formation of toxic byproducts that lead to damage to tissue. This study is to investigate the therapeutic effects of Paeoniae Radix Alba and Puerariae Radix combination (PP) on ALD. Methods : PP was analyzed for polyphenolic compounds and free radical scavenging activity. ALD mouse model was induced by feeding ethanol and water (Control), silymarin (50 mg/kg), low-dose (PP: 100 mg/kg) or high-dose (PP: 200 mg/kg) was orally administrated to ALD mice for 14 days. The serum was assessed with levels of AST, ALT, total bilirubin, total cholesterol, and triglyceride. Liver tissues were evaluated for ROS levels, degree of liver damage and protein expression. Results : The 3:1 (Paeoniae Radix Alba:Puerariae Radix) ratio showed the best antioxidant values for the experiment. In ALD model, levels of AST, ALT, total bilirubin, total cholesterol, and triglyceride were significantly increased in the Control and the levels were decreased by treatment of PP. In addition, increased ROS, ONOO- and MDA levels in the Control were reduced in the PP groups. Western blot analysis figured out that proteins related to ROS and cholesterol metabolism were higher in ALD than in PP-treated ALD. Antioxidant enzyme expression was low in the control group and increased by PP treatment. Conclusion : Our results suggest that PP has the potential to be a medicine in ALD in terms of regulating oxidative stress and adjusting lipid metabolism.

Antioxidant Activities and Antioxidant Compounds of Commercial Red Wines (국내 시판되는 적포도주의 항산화효과 및 항산화성분)

  • The objectives of this study were to determine antioxidant activities and antioxidnat compounds of 13 imported and 4 domestic red wines and to investigate relationships between antioxidant activities and antioxidant compounds in the selected red wines. The concentrations of total polyphenolics and anthocyanins in the samples were investigated by spectrophotometric methods. ABTS (2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid) and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity, and reducing power have been used to compare the relative antioxidant activities of the selected red wines. In this study, total polyphenolic contents of the red wines were ranged from 250 to 2,298 mg gallic acid equivalents/L and the anthocyanin contents were ranged from 11 to 349 mg cyanidin-3-glucoside equivalents/L, respectively. As expected, all the red wines exhibited excellent antioxidnat activities (ABTS and DPPH radical scavenging and reducing power) except three domestic red wines. The correlation coefficient between total polyphenolic content and their antioxidant activities, namely ABTS radical scavenging activity, DPPH radical scavenging activity and reducing power, were 0.9784, 0.9905 and 0.8580, respectively. No correlation was observed between total anthocyanin content and their antioxidant activities.

Optimization of Extraction Conditions of Polyphenolic Compounds from Amaranth Leaf using Statistically-based Optimization (통계학적 최적화를 이용한 아마란스 잎으로부터 폴리페놀 열수추출조건 최적화)

  • This study examined the optimization of hot-water extraction conditions for maximizing the total polyphenol compounds (TPC) extracted from amaranth leaf. The effects of three independent variables, including extraction temperature, extraction time and ethanol concentration on TPC were investigated using central composite design (CCD). The concentration of TPC increased with increased levels of extraction temperature and time. The extraction temperature and the ethanol concentration showed the significant effect on TPC production (p<0.05). The predicted values at the optimized condition were acceptable when compared to the experimental values ($R^2=0.9566$). The optimum extraction conditions were as follows: temperature of $90.1^{\circ}C$, time of 50 min and ethanol concentration of 61.6% (v/v) for the maximum TPC of 12.6 mg GAE/g DM.

Antitumoral and Antioxidant Potential of Egyptian Propolis Against the PC3 Prostate Cancer Cell Line

  • It has been shown previously that nutritional supplements rich in polyphenolic compounds play a significant role in prostate cancer chemoprevention. Propolis is a natural, resinous hive product that has several pharmacological activities including antimicrobial, antioxidant, anti-inflammatory, and antitumoral activities. The aim of this study was to compare the cytotoxic, antioxidant and antitumoral activities of an ethanolic extract of Egyptian propolis (EEP) in vitro with an established chemotherapeutic drug such as doxorubicin (DOX), and the effects of their combination against the PC3 human prostate cancer cell line. Cellular viability and $IC_{50}$ levels with EEP, DOX and their (v/v) combination were detected by sulphorhodamine-B (SRB) assay after incubation of PC3 cells for 72h with different doses (0, 0.01, 0.1, 1, 10 and $100{\mu}g/ml$). Two selected doses of $IC_{50}$ and $IC_{25}$ were applied to cells for 24h for antitumor evaluation assay of treatment compounds. EEP and its (v/v) combination with DOX showed significant antitumor potential besides high antioxidant properties of superoxide dismutase (SOD), total antioxidant capacity (TAC), catalase (CAT), nitric oxide (NO) and reduced glutathione (GSH) levels when compared with the control untreated cells. DNA fragmentation assay and semi quantitative RT-PCR analyses for p53 and Bax genes showed that EEP activated cellular apoptosis and increased the mRNA expression levels more than other treatment. In conclusion, EEP alone or in combination with DOX at both doses used here showed greater antioxidant, antiproliferative and apoptotic effects against the PC3 cell lines as compared to treatment with DOX alone. Therefore, EEP could be considered as a promising candidate for prostate cancer chemotherapy.

A Study on Antioxidant Activity and Antioxidant Compound Content by the Types of Tea (다류의 유형별 항산화 성분함량 및 항산화 활성 연구)

  • The purpose of this study was to investigate the contents of antioxidant compounds and antioxidant activities in teas. A total of 99 teas were tested for their antioxidant activities based on their ability to scavenge DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical. Antioxidant activity was expressed as mg of ascorbic acid equivalents per 100 g tea sample (L-ascorbic acid equivalent antioxidant capacity, AEAC) and was expressed as mg per 1 serving size (free radical scavenging activity, $FSC_{50}$). The total polyphenol contents of the extracts was quantified using the Folin-Ciocalteu method and the total flavonoid contents of the extracts was determined using a modied method of Davis. Vitamin C was analyzed by HPLC method. According to the AEAC value, mate tea, green tea, black tea, oriental raisin tea, chamomile tea and burdock tea showed relatively high antioxidant activities. Polyphenolic compounds were the major naturally occurring antioxidant compounds found in teas and the high concentrations of total polyphenol compounds were observed in black tea, green tea and mate tea. The high concentrations of total flavonoid compounds were observed in mate tea, black tea and yam tea and the amount of vitamin C contents were found to be high in citron tea and green tea. As a result, steeping tea was much higher in antioxidant activity than either powdered tea or liquefied tea. According to the study, a high correlation was demonstrated between the total polyphenol contents and antioxidant activities in teas (r = 0.846) and correlations between the total flavonoid contents and antioxidant activities was statistically significant in teas (r = 0.625). It was found that also the proportional relationship established among the total polyphenol content and antioxidant activities. That is, antioxidant activity of teas has been confirmed to have been caused by the total polyphenol.
