Effects of Powdered Pine Needle (Pinus densiflora seib et Zucc.) on Serum and Liver Lipid Composition and Antioxidative Capacity in Rats Fed High Oxidized Fat (솔잎분말이 과산화지질을 급여한 흰쥐의 혈장 및 간장의 지질구성과 항산화능에 미치는 영향)

  • Effects of powdered pine needle (Pinus densiflora seib et Zucc) on serum and liver lipid composition and antioxidative capacity were investigated in rat fed high oxidized fat. Sixty male Sprague-Dawley rats weighing 161.25$\pm$2.51 g were blocked into four groups according to body weight and raised seven weeks with basal diet (normal group, I), basal diet and 10% oxidized fat (control group, II), basal diet, 10% oxidized fat and 2% powdered pine needle (2% powdered pine needle group, III) and basal diet, 10% oxidized fat and 3% powdered pine needle (3% powdered pine needle group, IV). Food intake, body weight gain and feed efficiency were not significantly different among oxidized fat diet groups. The level of plasma total cholesterol showed a tendency to decrease, whereas the plasma HDL-cholesterol concentration revealed a tendency to increase in pine needle groups. However plasma triglyceride level showed no significant differences in the treatment groups. Intake of the oxidized fat has increased the levels of liver cholesterol and triglyceride. The powdered pine needle showed a tendency to decrease thiobarbituric acid values in plasma and liver. The pine needle samples have also decreased the plasma GOT and GPT activities, whereas they have increased the liver glutathione peroxidase activity.

Effect of Korean Native Plant Diet on Lipid Metabolism, Antioxidative Capacity and Cadmium Detoxification in Rats (한국 고유의 산채류 첨가 식이가 흰쥐의 지방 대사 및 항산화능과 Cadmium 제독에 미치는 영향)

  • This study was performed to investigate the effects of Korean native plant diets on lipid and cadmium (Cd) metabolisms and also antioxidative enzyme activities in rats. Seventy male Sprague-Dawley rats weighing 210.3$\pm$2.7g were blocked into ten groups according to body weight. Rats were raised for four weeks with diets containing 0 or 0.04%(w/w) cadmium chloride and 5%(w/w) plant powder-Ssook(Artemisia princeps var. orientalis HARA), Chamchwi(Aster scaber THUNB), Gomchwi(Lingularia fischeri TUBCZ). or Solbeerum (Portulaca oleracea LINNE). Food intake, weight gain, food efficiency ratio, and weights of liver, kidney, spleen and epididymal fat were significantly lower in Cd-exposed groups, food efficiency ratio, and weights of liver, kidney, spleen and epididymal fat were significanlty lower in Cd-exposed groups. Plasma total lipid level, liver total lipid, cholesterol and triglycerde concentrations, and fecal total lipid, cholesterol and triglyceride excretions were decreased by Cd administration. Total lipid and triglyceride concentration in plasma and liver were lower in Chamchwi groups together with increased fecal total lipid and triglyceride excretions. Cholesterol concentration in plasma and liver were lower in Gomchwi groups with increased fecal cholesterol excretions. Activities of red-blood-cell superoxide dismutase(SOD), and liver catalase, SOD and glutathione peroxidase (GSH-px) were decreased significantly by Cd administration. Thiobarbituric acid reactive substance(TBARS) concentrations of plasma and liver were increased by Cd administration. Thiobarbituric acid reactive substance(TBARS) concentration of plasma and liver were increased by Cd administration. Administration of plant diets decreased plasma and liver catalase, SOD and GSH-px activities in Cd-free diet groups and increased them in Cd-administered groups. Plasma and liver TBARS concentrations were decreased in animals fed plant diets, with Chamchwi showing the most effective antioxidative capacity. The concentration of Cd accumulated in blood and tissue decreased in Cd-exposed groups fed plant diets. Liver Cd concentration were lower in the Chamchwi and Gomchwi groups, and kidney Cd levels were lower in the Ssook and Chamchwi groups. Among them, Chamchwi was the most effective in Cd detoxification. In conclusion, Chamchwi and Gomchwi were effective in lowering tissue lipid levels, with Chamchwi having the greatest antioxidative and detoxifying effects.

The Effect of Dietry Se Levels and Alcohol Administration on the Lipid Hyperoxidation and the Lipid Metabolism in the Rats (식이내 Se수준과 알콜 섭취가 흰쥐의 지질과산화와 지방대사에 미치는 영향)

  • The purposes of this studies were to investigate the effect of dietry Se levels and alcohol administration on the lipid hyperoxidation and the lipid metabolism in the rat. Seventy two male rats of Sprague-Dawley Strain weighting about 58~629 were divided into 12groups. The dietary Se levels were 10, 0.4 and Omg, and the dietary a-tocopherol levels were 150 and 0mg per kg diet, respectively. Alcohol-administrated groups received drinking water solution containing 10% of ethanol from the 3-weeks of experimental periods. The obtained experimental results are summarized as follows. 1. Food intake, body weight gain and food efficiency ratio were significantly lower in H-, L-and alcohol administrated groups(-A) by administrated Se and alcohol in diet. The weight of liver and spleen tended to be greater in H- and alcohol administrated groups. 2. The glutathione values in liver tend to be lower in alcohol or Vit. E administrated groups than nonadministrated groups. Also there were higher in H- and L- than C-groups, but the increasing range decreased due to administrated alcohol. The lipid peroxide values In liver were significantly higher in alcohol groups, and L- and tocopherol groups were higher values. Specially the increasing of lipid peroxide values were significantly effected by alcohol in low Se and Vit. E groups. 3. The contents of total glyceride in plasma were higher in alcohol groups, there were significantly higher values in alcohol administrated groups under low Se and Vit. E groups. The contents of total cholesterol and HDL-cholesterol In plasma were significantly higher in alcohol groups. 4. The contents of total lipid in liver were higher alcohol groups, and slightly higher values in low Se groups(L-groups) than other groups, also higher values in low Vit. E groups. Those of total glyceride in liver were significantly higher in alcohol groups, appeared highest values when alcohol was administrated in low Se and Vit. E groups. The increasing of total glyceride content was significantly effected by alcohol in low Se groups than that in C-groups.

Inhibitory Effects of Opuntia humifusa on 7, 12-Dimethyl-benz[a]anthracene and 12-O-tetradecanoylphorbol-13-acetate Induced Two-stage Skin Carcinogenesis

  • Opuntia humifusa, member of the Cactaceae family, was previously demonstrated to have radical scavenging, anti-inflammatory and anti-proliferative effects in in vitro models. It was suggested that O. humifusa could function in the prevention of carcinogenesis. To investigate the in vivo chemopreventive effect of O. humifusa, mice were fed a diet containing either 1% or 3% following 7, 12-dimethylbenz[a] anthracene (DMBA) and 12-O-tetradecanoylphorbol-13-acetate (TPA) induction of skin carcinogenesis. Significant decrease in the numbers of papilloma and epidermal hyperplasia were observed in mice fed with O. humifusa, compared to the control group. O. humifusa also upregulated high total antioxidant capacity and level of phase II detoxifying enzyme such as superoxide dismutase and glutathione S-transferase activity in the skin. Lipid peroxidation activity level was measured in skin cytosol and significantly inhibited in 3% OH fed group compared to the control group. These results suggest that O. humifusa exerts chemopreventive effects on chemical carcinogenesis in mouse skin and that prevention effects are associated with reduction of oxidative stress via the modulation of cutaneous lipid peroxidation, enhancing of total antioxidant capacity especially in phase II detoxifying enzyme system and partial apoptotic influence.

GSTT1 Null Genotype Distribution in the Kumaun Region of Northern India

  • Glutathione S-transferases (GSTs) constitute a multigene family of multifunctional phase II metabolic enzymes. GSTT1, an important member of this group has a wide range of substrates including carcinogens. Total homozygous deletion or null genotype resulting in total lack of enzyme activity exists in populations for this enzyme. Since the null genotype may contribute to lower detoxification of carcinogens, this genotype is expected to increase cancer risk. The frequency of the GSTT1 null genotype is known to vary significantly among populations. However, little is known about its distribution in the hilly Kumaun region of northern India. Therefore, in this study, we determined the prevalence of the GSTT1 null polymorphism in the Kumaun popilation by conducting duplex PCR in 365 voluntary healthy individuals. The GSTT1 null genotype was detected in 18.4% of the individuals. Since GSTs play significant role in xenobiotic metabolism, the present data on GSTT1 genotype distribution should contribute in understanding genetic association with cancer risk in this understudied population.

Effect of Dietary Level of $\beta$-Carotene on Antioxidants Contents and Hepatic Morphology in Rats ($\beta$-Carotene 의 수준별 공급에 따른 흰주의 항산화 물질과 간조직의 형태적 변화)

  • This study was designed to investigate the effect of dietary level of $\beta$-carotene on level of antioxidant nutrients of rat tissues. Male Sprague Dawley rats at the age of 30days were fed on diets containing different levels of $\beta$-carotene(0, 10, 120, 1200, 12000mg per kg diet). Body weight gain of rats fed with 12000mg $\beta$-carotene diet was significantly decreased, but liver and heart weights were not significantly different among groups, The content of total glutathione tended to decrease significantly in 12000mg $\beta$-carotene diet group when compared to $\beta$-carotene restriction group(BC O). However, total vitamin C content of liver showed the tendency to increase by $\beta$ -carotene supply up to 1200mg. But this tendency was not found in plasma, The content of zinc in liver and plasma was significantly decreased by $\beta$-carotene restriction. Alkaline phosphatase activity was significantly higher in 12000mg diet group. In case of $\beta$-carotene restriction group, fibroblasts were proliferated in portal endothelium, and the vacuolar size was enlarged more than the nuclear, In 12000mg diet group, hepatic vacuoles were extended, but their size was regular and the lysis of hepatocytes was observed. Also, fibroblasts were proliferated in portal endothelium and the regular vacuolar size was extended.

The Effects of Persicae Semen Herbal-acupuncture at GB34(Yangneungcheon) on a Rat Model of Hyperlipidemia (양릉천 도인 약침이 고지혈증 병태 모델 흰쥐에 미치는 영향)

  • Objective: The purpose of this study is to observe the effects of Persicae Semen herbal-acupuncture at GB34(Yangneungcheon) on hyperlipidemia of rats caused by high-fat diet. Methods: The author performed several experiments to measure the DPPH radical scavenging activity of PS-HAS,to analyze various components and enzymes in serum and liver, and to observe the histological changes of liver and aorta. Results: 1. Persicae Semen herbal-acupuncture solution increased the DPPH radical scavenging activity in rat's liver cells. 2. In the Persicae Semen herbal-acupuncture group, compared with control group, the level of total cholesterol in serum and the ratio of total cholesterol to HDL-cholesterol decreased significantly. 3. In the Persicae Semen herbal-acupuncture group, compared with the control group, the level of glutathione in liver cells increased significantly. 4. In the Persicae Semen herbal-acupuncture group, compared with the control group, the pathological changes in liver and aorta were reduced. Conclusion: The above results suggest that Persicae Semen herbal-acupuncture at GB34 has good effects on preventing and treating hyperlipidemia.

Effects of Apium graveolens Extract on the Oxidative Stress in the Liver of Adjuvant-Induced Arthritic Rats

  • Apium graveolens Linn. (Apiaceae) is an indigenous plant of the North and South Americas, Southern Europe, and Asia and has been widely used as a food or a traditional medicine for treatment of inflammation and arthritis. The purpose of this study was to investigate the antioxidant effects of a methanolic extract of A. graveolens (AGE) against liver oxidative stress in an adjuvant-induced arthritic rat model. The AGE (250, 500, and 1,000 mg/kg) was given orally for 24 consecutive days after induction by injecting complete Freund's adjuvant. Liver and spleen weights were recorded. The superoxide anion level, total peroxide (TP), glutathione peroxidase (GPx) activity, superoxide dismutase (SOD) activity, total antioxidant status, and oxidative stress index (OSI) were also measured. AGE treatment significantly decreased the levels of the superoxide anion, TP, and OSI whereas the GPx and SOD activities significantly increased in the liver of the arthritic rats. These results indicated that AGE showed an ameliorative effect against liver oxidative stress in adjuvant-induced arthritic rats by reducing the generation of liver free radicals and increasing the liver antioxidant enzyme activity.

Effects of taeeumjoweetang Herbal-Acupuncture on plasma and liver lipid composition and antioxidative capacity in rat fed high fat diet (태음조위탕(太陰調胃湯) 약침(藥鍼)이 고지방 급여 흰쥐의 혈장 및 간장의 지질구성과 항산화능에 미치는 영향)

  • Objectives : In order to measure the Effects of taeeumjoweetang Herbal- Acupuncture at joksamri(ST36) and kwanweun(CV4) on liver and plasma lipid composition and antioxidative capacity in rat fed high fat diet. Results : Concentrations of plasma triglyceride, total cholesterol and LDL-cholesterol showed a tendency to increase in the high fat diet group. However Herbal-Acupuncture groups showed a lower values than control groups. HDL-cholesterol showed a tendensy to decrease in high 131 fat diet groups and in high fat diet groups, these values showed no significantly different. Liver total cholesterol values showed no significantly different in all treatment groups. Triglyceride concentration showed a high value in control group and other treatment groups showed no significantly different. plasma GOT and GPT values showed a tendency to increase in high fat diet group. however these values decreased in Herbal-Acupuncture group. The concentration of TBARS in liver and plasma showed a high values in high fat diet group, however these values showed a tendency to decrease in aqua- acupuncture group. Glutathione peroxidase, superoxide dismutase and catalase activity values showed a low values in high fat diet group, however these values showed a tendency to increase in Herbal-Acupuncture group.

Effects of acupuncture at Gansoo(B-18) and (Chungwan (CV-12) on Serum Lipid Composition and Antioxidative Capacity in Rats Fed Peroxidized Lipids (간수(B-18) 및 중완(CV-12) 자침이 과산화지질(過酸化脂質)을 급여(給與)한 흰쥐의 혈청지질(血淸脂質)강하 및 항산화능(抗酸化能)에 미치는 영향(影響))

  • Objectives : To investigate the effects of acupuncture at Gansoo (B-18) and Chungwan (CV-12) on lipid metabolism in rats fed peroxidized lipid diet. Methods : Effects of acupuncture at Gansoo and Chungwan on plasma and liver lipid composition and antioxidative capacity were investigated in rats fed peroxidized lipids. Results : Although the level of plasma total cholesterol and triglyceride showed a tendency to decrease in the acupuncture group, the plasma HDL-cholesterol concentration showed no significant difference. The level of liver total cholesterol and triglyceride showed no significant difference in all treatment groups. Thiobarbituric acid( TBARS ) values in plasma and liver showed a tendency to decrease in the acupuncture groups. The plasma GOT and GPT activities showed low values in the acupuncture groups. The liver glutathione peroxidase, superoxide dismutase and catalase activity showed high values in the acupuncture groups.
