T cell phenotype and intracellular $IFN-{\gamma}$ production in peritoneal exudate cells and gut intraepithelial lymphocytes during acute Toxoplasma gondii infection in mice

  • Although there are many reports on the splenic (systemic) T cell response after Toxoptasma gondii infection, little information is available regarding the local T cell responses of peritoneal exudate cells (PEC) and gut intraepithelial Iymphocytes (IEL) following peroral infection with bradyzoites. Mice were infected with 40 cysts of the 76K strain of T. gondii, and then sacrificed at days 0, 1, 4, 7 and 10 postinfection (PI). The cellular composition and T cell responses of PEC and IEL were analyzed. The total number of PEC and IEL per mouse increased after infection, but the ratio of increase was higher in IEL. Lymphocytes were the major component of both PEC and IEL. The relative percentages of PEC macrophages and neutrophils/eosinophils increased signiflcantly at day 1 and 4 PI, whereas those of IEL did not change significantly. The percentage of PEC NK1.1 and ${\gamma\delta}T$ cells peaked at day 4 PI (p < 0.0001), and CD4 and $CD8{\alpha}T$ cells increased continuously after infection. The percentages of IEL $CD8{\alpha}$ and ${\gamma\delta}T$ cells decreased slightly at first, and then increased. CD4 and NK1.1 T cells of IEL did not change significantly after infection. $IFN-{\gamma}-producing$ PEC NK1.1 T cells increased significantly from day 1 PI, but the other T cell subsets produced $IFN-{\gamma}$ abundantly thereafter. The proportion of IEL $IFN-{\gamma}-producing$ $CD8{\alpha}$ and ${\gamma\delta}T$ cells increased significantly after infection, while IEL NK1.1 T cells had similar $IFN-{\gamma}$ production patterns. Taken together, CD4 T cells were the major phenotype and the important $IFN-{\gamma}$ producing T cell subsets in PEC after oral infection with T. gondii whereas $CD8{\alpha}T$ cells had these roles in IEL. These results suggest that PEC and IEL comprise different cell differentials and T cell responses, and according to infection route these factors may contribute to the different cellular immune responses.

가미청폐화담탕이 천식 유발 병태 모델에서 천식 관련 활성 면역세포에 미치는 영향 (Effects of Gamicheungpyehwadam-tang on Immune-cell Regulation in Association with Bronchial Asthma in OVA-induced Mouse Model)

  • These studies were investigated the effects of Gamicheungpyehwadam-tang (CPHDT) on immune-cell regulation in association with bronchial asthma in OVA-induced mouse model. The administration of 400 mg/kg CPHDT significantly reduced the number of total cells in lung, peripheral lymph node and spleen in OVA-induced bronchial asthma mouse model. The administration of 400 mg/kg CPHDT significantly reduced $CD3^+,{\;}CD19^+$and $CD3^+,{\;}CD69^+$ cell numbers separated from lung, peripheral lymph node and spleen in OVA-induced bronchial asthma mouse model. CPHDT significantly reduced $CD3^+/CCR3^+,{\;}CD4^+,{\;}B220^+/IgE^+$, and $CD3^+/DX5^+$ cell numbers separated from lung, peripheral lymph node and spleen in OVA-induced bronchial asthma mouse model in a dose dependent manner, However, CPHDT significantly reduced $CD8^+$ cell numbers from only lung and spleen. The administration of CPHDT significantly reduced $NK^+$ cell numbers separated from lung of OVA-induced bronchial asthma mouse model in all concentrations, but 200 mg/kg CPHDT reduced $NK^+$ cell numbers separated from peripheral lymph node. These results suggest that CPHDT has anti-asthma and anti-allergy effects. In addition to, CPHDT may be useful treatment of asthma based on the further studies about the individual efficacy search of the components of CPHDT and the adding of variety drugs to CPHDT.

Effect of MEM Vitamins Supplementation of In vitro Maturation Medium and In vitro Culture Medium on the Development of Porcine Embryos

  • This study was carried out to examine the influence of minimum essential medium (MEM) vitamins supplementation to in vitro maturation medium and in vitro culture medium on the development of porcine embryos. Porcine embryo development was investigated following cultivation in both in vitro maturation and culture medium with the supplementation of MEM vitamins (0, 0.1, 0.2 and 0.4%) using immature oocytes collected from the ovary of prepubertal gilts. Embryo development was observed and the total cell number in each blastocyst generated under the culture conditions was quantified following supplementation of the medium. The embryonic development rate of the blastocyst and hatched blastocyst was higher, but not significantly so, when 0.4% MEM vitamins were supplemented to the in vitro maturation medium of the porcine oocyte. Interestingly, the total number of cells in the blastocyst was significantly higher in the in vitro maturation MEM vitamins supplemented group compared to either the untreated group or the group which had MEM vitamins supplemented to both in vitro maturation and in vitro culture medium simultaneously (p<0.05). Therefore, the supplementation of 0.4% MEM vitamins to the in vitro mature medium has a beneficial effect on the embryonic development of in vitro produced blastocysts derived from the immature porcine oocytes.

인천대교 민자구간의 대구경 현장타설 말뚝기초의 LRFD 설계 적용 사례 (Design of Drilled Shafts Foundation by LRFD in Incheon Bridge Project)

  • Incheon bridge project is to construct total 12km long bridges on the sea consist of 800m span length cable stayed bridge, approach bridge and viaduct bridge based on LRFD design specification. To design pile foundations by RCD of each bridge unit, total 4 number of preliminary full scale pile load tests with Osterberg cell method were carried out on the piles for testing. The test load was planned to more than the expected design ultimate capacity and about 29,000tons maximum load was recorded. From the interpretation of test results, design parameters are evaluated and applied to the design. Preliminary pile load test plan and detailed execution of pile load tests are introduced and summarized. The resistance factors are presented for pile design of Incheon Bridge Project in LRFD considering variation of ground conditions and number of test piles.

생쥐배의 핵치환에 관한 연구 III. 핵치환 생쥐의 생산 (Studies on Nuclear Transplantation in Mouse Embryos III. Production of Nuclear Transplanted Mice)

  • These experiments were carried out to develop the technique of nuclear transplantation necessary for elevating utilization efficiency of high quality embryos and the production of clone animals. Embryos of pronucleus stages were obtained from ICR mice. Removal of pronuclei and their transfer to recipient embryos were carried out by micromanipulation and virus mediation. The results obtained in these experiments were summarized as follows ; 1. Total 337 pairs of pronuclear stage embryos were subjected to nuclear transplantatin and 247 pairs(73.3%) of them were successfully transplanted and the number of fused embryos between transplanted nucleus and cytoplasm was 188 pairs(55.8%). 2. Of the 188 fused embryos cultured in vitro, 174(92.4%), 131(69.7%) and 117(62.2%) embryos were developed to 2-cell, morula and blastocyst stages, respectively. 3. When total 104 nuclear transplanted embryos were transferred to uteri of recipient mice on day 2-3 of pseudopregnancy, 4 of 12 recipient mice were pregnant and the number of embryos developed to young was 28(26.9%).

진도견 혈액내 기생충과 호산구 관계 조사 (Studies on Relation of Heartworm and Eosinophils in Blood of Jindo Dogs)

  • The present study was conducted in order to get the relationship of heartworm, Dirofilaria immitis and eosinophils in blood of Jindo dogs. Blood samples were taken from 141( man 31, womam 110) of yellow, 44 ( man 17, woman27) of white Jindo dogs in Jindo area. The results obtained in this study were summarized as follows : 1. There were no significant differences of the hematological values in comparison with others reported values, but the mean corpuscular hemoglobin concentration and the number of eosinophils were high in comparison with other reported values. 2. The mean values of total white blood cell count(P<0.05), neutrophil(P<0.05) and monocyte(P< 0.05) were that the yellow dogs and a tendency toward fisher values than the white dogs, but no significant differences were observed. 3. Age-related differences were detected for the total erythrocyte count(P<0.05), hemoglobin content (P<0.001), PCV(P<0.001) and the number of eosinophils(P<0.001). 4. The direct and indirect eosinophil counts were increased with age, there were no significant differences of the values between two groups. 5. Dirofilaria immitis was found in 34(18.4%), increased with age and were relatively significant differences in areas of the 185 Jindo dogs. 6. Survey for hematological values of Dirofilaria immitis infected dogs with direct and indirect method, mean values of eosinophil were 2,788/ ${\mu}l,$ 3,021/ ${\mu}$l, respectively. They showed that eosinophil had a tendency toward higher values than others, but others were no significant differences.

멸치젓 첨가량이 부추김치의 발효숙성에 미치는 영향 (Effect of Various Level of Salted Anchovy on Puchukimchi Fermentation)

  • This study was undertaden to investigate the effect of salted anchovy during Puchukimchi fermentation, Puchukimchi is a kind of Korean kimchi made with Puchu(Allium odorum L.). For this purpose, the Puchukimchi with 10%, 15% and 30% salted anchovy was investigated by measuring physicochemical, microbiological and sensory characteristics during fermentation at 5$\pm$2$^{\circ}C$ up to 60 days, In the palatable period, pH was remarkably decreased to the range of 4.8-5.16 and total acidity was increased 2-3 times more than that of initial period. Reducing sugar contents in Puchukimchi increased up to 22days and remarkably decreased. The changing pattern of vitamin C drawed sigmoidal curve, invreasing significantly in the palatable period. The content of lactic acid was a little in the initial fermentation period but increased 3-13 times more than that of the initial fermentation period in the palatable period. The content of malic acid and citric acid were decreased as the fermentation progressed. The contents of Ca, Mg and K was gradually decreased and Na was increased as the fermentation progressed in the same period. There were similiar changes in the number of lactic acid bacteria and total cell number. The score of overall taste, overall acceptability in evaluation during fermentation was high in order of 15%, 10%, 30% level of salt fermented anchovy.

Anatomy teaching in Saudi medical colleges- is there necessity of the national core syllabus of anatomy

  • Curricular updates in medicine resulted in changes in gross anatomy teaching. We aim to find the trends and methods of gross anatomy teaching in medicine programs in Saudi Arabia. Further, examine whether the data would help to discuss whether a core Anatomy syllabus is required. A survey questionnaire based on the earlier studies, was sent to the anatomy faculty of 25 medical colleges to collect the data on the pedagogic and dissection/laboratory approaches, inclusion of radiological, clinical, surface anatomy sessions, and the total number of hours allocated for anatomy education. A total of 15 responses were received from different medical colleges of which nine provided complete details. A wide variation in the component and mode of delivery of anatomy was observed. The number of hours for the anatomy course ranged from 89 to 388 hours. These data will provide an update on gross anatomy teaching approaches, which will help in making informed decisions in course revisions and adopting the best practices. The variations in anatomy course with short duration raises concern about whether the essential learning outcomes are achieved to prepare a skillful and safe clinician? do we require a core syllabus of Anatomy to be adopted at the national level to achieve the essential learning outcomes? The Anatomical Society, UK has developed core syllabi of Anatomy for undergraduate medical, dental, nursing, and pharmacy students, which can serve as a guide in developing the core syllabus of Anatomy for medicine in Saudi.

단무지 제품의 보관온도 설정을 위한 저장 온도별 품질 특성 (Monitoring of Quality Characteristics of Danmooji Product Storage Temperatures for Storage Temperature Establishment)

  • 단무지는 단체급식 및 외식업체에서 소비되는 부식재료로 소비량이 점점 늘어나고 있는 추세이다. 더운 여름철 상온에서 대량 소비처로 운반되는 단무지의 경우 미생물에 의한 부패와 효소작용에 의한 연화현상은 단무지 생산자들에게 해결해야할 가장 큰 문제가 되고 있다. 따라서 향후 소비가 더욱 증가될 것으로 예상되는 단무지에 대한 품질개선 방안을 마련하기 위한 기초자료로 활용하고자 포장된 단무지 제품의 저장온도에 따른 품질변화와 특성을 비교하였다. 단무지의 저장 중 색도, 물성, 미생물 변화와 관능적 특성을 알아보기 위하여 $5^{\circ}C$$25^{\circ}C$의 저장온도로 각각 나누어 3주간 조사 하였다. 단무지와 단무지 여액의 명도와 적색도는 저장기간이 길어짐에 따라 큰 차이는 없었지만 유의적으로 나타났으며, 황색도의 경우 $5^{\circ}C$$25^{\circ}C$ 모두 저장기간에 따라 감소하는 경향을 나타냈다. 단무지의 Hardness는 $5^{\circ}C$의 경우 7일에서 가장 큰 값을 나타냈고, $25^{\circ}C$의 경우는 14일에서 가장 큰 값을 나타냈다. 저장기간 중 단무지의 총 균수를 조사한 결과 저장기간 동안 $10^5CFU/g$로 거의 균수변화가 없었지만, 단무지 여액의 경우 저장후기부터 균수가 늘어났다. 효모곰팡이 수는 단무지의 경우 $5^{\circ}C$, $25^{\circ}C$ 모두 저장 7일까지는 증가하다가 단무지의 균수는 이후 변화가 없었고 단무지 여액의 균수는 계속 증가 하였다. 단무지 저장별 관능적 특성은 맛의 전체적 기호도는 저장 14, 21일($5^{\circ}C$)에서 가장 높았으며, 향의 전체적 기호도는 저장 7일($5^{\circ}C$)에 보관한 단무지가 높았으나 유의적 차이는 없었다. $25^{\circ}C$에 보관한 단무지의 Texture, Color 기호도는 저장기간이 길어짐에 따라 유의적으로 감소하였고(p<0.05), $25^{\circ}C$에 보관한 단무지의 전체적 기호도는 저장기간이 길어짐에 따라 유의적으로 감소하였다(p<0.05). 전체적으로 $5^{\circ}C$에 보관한 단무지의 기호도가 유의적 차이는 없었지만 $25^{\circ}C$에 보관한 단무지의 기호도보다 높게 나타나 단무지 제품의 보관온도는 $25^{\circ}C$보다는 $5^{\circ}C$가 적합하다고 사료된다.

혈소판 농축혈장이 조골세포의 초기부착과 증식 및 활성에 미치는 생물학적 영향 (Biological Effect of Platelet Rich Plasma on the Initial Attachment, Proliferation and Cellular Activity of Osteoblast)

  • For reconstruction of the bony defect, various artificial substitutes were developed. Among them, there has been a study of calcium phosphate coated bone substitutes for increasing attachment of osteoblasts in vivo. The purpose of this study was to evaluate the effects of serum and platelet-rich plasma (PRP) on calcium phosphate coated culture plate for the initial attachment, proliferation and activity of osteoblasts. After sampling the blood from white rats and concentrating by centrifugation, the amount of attachment of PDGF-BB and $TGF-{\beta}$ on the calcium phosphate coated culture plate was measured. Cultured HOS and ROS 17/2.8 cell was measured on attachment level and proliferation rate of osteoblasts. Alkaline phosphatase activity of HOS and ROS 17/2.8 cell was measured for studying on the activating rate of osteoblast. 1. Counting the amount of platelets of seperated plasma and PRP, the average number of platelets was 177,003 $cell/{\mu}l$ in plasma, and 1,656,062 $cell/{\mu}l$ in PRP, which was about 9 times as high as in plasma. 2. Amount of PDGF-BB deposited at calcium phosphate coated plate had increased by the total amount of plasma and PRP on the culture plate, whereas $TGF-{\beta}$had been deposited on the plate only when treated by $50{\mu}{\ell}$ of PRP(p<0.01). 3. After plating serum and PRP for 3 hours, we attached with HOS and ROS17/2.8 cell for 1 hour and 4 hours. There were no significant difference of the attachment between serum and control group, whereas there were significantly difference of the attachment between depositioning of PRP and control group. 4. After attaching plasma and PRP for 3 hours, cell number has much increased when HOS and ROS17/2.8 cell had been cultured for 48 hours(p<0.05). 5. After attaching plasma and PRP for 3 hours, concentration of alkaline-phosphatase has increased when HOS and ROS17/2.8 cell had been cultured for 48 hours(p<0.01). These results suggested that PRP affected on initial cell attachment rather than proliferation and activation of osteoblasts at calcium phosphate coated plate.
