Quantitative analysis of the activities of transport ATPases as well as alkaline phosphatase of the mouse uterus was carried out during the estrous cycle. Even though the proportional patterns of the enzyme activities were similar each another between the stages of estrous cycle, the absolute activities of the enzymes except $K^+$-dependent and $Na^+$, $K^+$-activated ATPases at the time of estrus were significantly (p<0.025) higher than that at any other time of the estrous cycle. That is, the activities of $K^+$-dependent and $Na^+$, $K^+$-activated ATPases were negligible during the period of time from diestrus to estrus while the little activities (0.04 $\\sim$ 0.05$\\mu$M/mg protein/hr in average, $6\\sim7$% of the total enzyme activity) of these enzymes appeared at the time of metaestrus. On the other hand, at the time of estrus, the activities of $Mg^++$-dependent phosphatase, transport ATPase and alkaline phosphatase were rapidly and tremendously increased to be 0.69 (35%), 0.42 (21%) and 1.58 (79%), respectively. The activity of alkaline phosphatase was in the range of 0.60 $\\sim$ 1.58 (79 $\\sim$ 90%) and predominant throughout the estrous cycle. The activity of $Mg^++$-dependent alkaline phosphatase was estimated as 12 $\\sim$ 16% of the total enzyme activity. Therefore, it is assumed likely that $K^+$-dependent and $Na^+$, $K^+$-activated ATPases are not the main factors to control the fluid accumulation at the time of estrus, but may be the factors to reabsorb the luminal fluid into the uterine epithelium at the time of metaestrus, and that $Mg^++$-dependent phosphatase, transport ATPase and alkaline phosphatase must be closely involved in the secretion of luminal fluid from the epithelial cells of the mouse uterus.
Twenty-one dogs(male 11 heads, female 10 heads) which were about 16 months ($16.3{\pm}3.5$) old and 10kg($10.1{\pm}2.0$) body weight, were allotted randomly into four groups as follows. Group I consisted of five dogs whose muscles were operated for sham muscle injuries. Group II consisted of seven dogs treated for cystic duct obstruction. Group III consisted of five dogs treated for the union fracture model. Group IV consisted of five dogs treated for the non-union fracture model. Radiographical and histological observations were carried out to determine bone alkaline phosphatase(BALP) and total alkaline phosphatase(TALP) values of each group for 20 weeks after the treatments with the condition of new bone formation. And also the applicability of percentage of BALP values to TALP (B/T) was studied after BALP was compared respectively with TALP. The level of TALP was increased without any relation to bone formation in group II, and all levels of BALP and B/T were increased in group III. The mean of B/T was high in statistical significance, due to varied levels of B/T and BALP. The changes of rates of B/T were significantly increased only in the case of the active new bone formation in group III, union fracture model. It was recognized that the mean values of B/T were statistical significant of the high applicability of the B/T ratio as an index of bone formation.
The total and heterotrophic bacterial distributions, compositions and alkaline phosphatese actibities were analyzed in Lake Soyang from Sep. 1987 to Aug. 1988. The heterotrophic bacteria was small portion, 0.07-2.63% of total bacterial number which ranged from $3.2{\times}10^{5}$ to $3.2{\times}10^{6}$ cells/${\mu}\ell$. The composition of bacterial community was less diverse in summer and at the fish farm site and Peridinium blooming site. Pseudomonas and Flavo bacterium were the dominant genera in all sites. The highest proportion and activity of alkaline phsophatase was appeared in Flavobacterium, while Pseudomonas was the most predominant group.
The activity of acid and alkaline phosphatases of pine moth, Dendrolimus spect abilis Butler was measured in a serier of developmental stages ranging from the larva to the adult. The activity of both enzymes increased gradually with age of larvae, and then decreased in the prepupal stage. Acid enzyme was at a maximum in the pupal early stage and alkaline enzyme in the 8th instar larva, respectively. And in the prepupal stage there were no significant differences between both acid and alkalnie phosphatases. However, their activities were far lower than in the 8th instar larva. In the pupal early stage there occurs a increase in the acitivity of acid enzyme followed by a decrease in the pupal later stage, and in the adult stage its activity increased again.
We investigated variations in alkaline phosphatase (APase) activity and alkaline phosphatase hydrolyzable phosphorus (APHP) in northern Gamak Bay from September to December 2009. Dissolved inorganic nitrogen (DIN) and dissolved inorganic phosphorus (DIP) decreased gradually, and the DIN/DIP ratio was higher than the Redfield ratio (16) based on molecular concentrations during most of the observation period. The total APase (T-APase) activity increased with decreasing DIP concentration; i.e., the Relationship between T-APase and DIP showed a high negative correlation (r=-0.80, P<0.001), with APase activity being a good indicator of DIP limiting the Redfield ratio. The T-APase was positively correlated with the concentration of chlorophyll a (r=0.73, P<0.001). This suggests that a major portion of APase activity in northen Gamak Bay seawater is attributed to phytoplankton. The proportion of APHP among dissolved organic phosphorus (DOP) was low in September and high in November. Thus, APase-producing phytoplankton may be able to grow by utilizing APHP as a phosphorus source in autumn when DIP is limiting. Thus, APase activity and the use of DOP by phytoplankton may play an important role in the growth of phytoplankton under DIP limiting conditions such as those of northern Gamak Bay.
This study was carried out to compare distribution and isozyme pattern of nonspecific esterase, acid phosphatase and alkaline phosphatase on developing sparganum and adult of Spinometra erinacei by using enzyme-histochemical method and electrophoresis The sparganum and adult were recovered from rats and cat that were infected by sparganum. The results obtained were as follows: Nonspecific esterase had a strong activity in the parenchymal musculature of sparganum and adult, but no detectable level in ihe tegument. A total of 7 and 8 nonspecific esterase bands were detectable in sparganum and adult, respectively. Of these bands, band 3 and 4 were major bands in sparganum and adult. Acid phosphatase had a strong activity in the tegument and the epidermal musculature of sparganum, but no detectable level in the parenchymal musculature. A total of 3 bands were detectable in sparganum and adult. Of these bands, band 3 was major band in sparganum and adult. Alkaline phosphatase had a strong activity in the tegument and the epidennal musculature of sparganum and of adult, but no detectable level in the parenchymal musculature. A total of 2 and 4 bands were detectable in sparganum and adult. Of these bands, band 2 was major band in sparganum and adult. Based on the present results isozyme band patterns showed qualitative and quantitative changes in each tissues of sparganum and of adult during the development.
In clinical chemistry tests, the interfering substances such as hemoglobin, lipid, bilirubin, and drugs, etc. can cause the changes of test results performed by spectrophotometrical methods. We evaluated the effects of interfering substances on the test results by adding interfering substances on the samples in the 19 kinds of clinical chemistry tests such as aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase, gamma-glutamyltransferase, total protein, albumin, glucose, total cholesterol, total bilirubin, triglyceride, uric acid, calcium, inorganic phosphours, high density lipoprotein cholesterol, low density lipoprotein cholesterol, creatinine, blood urea nitrogen, and C-reactive protein using newly implemented automatic chemical analyzer Toshiba TBA-C8000 under the direction of CLSI EP07-A guideline. Hemolytic samples show increased concentration of total protein, aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase and reduced concentration of total bilirubin, alkaline phosphatase by interfering effect. Hyperlipemic samples show increased concentration of total protein and alkaline phosphatase and reduced concentration of low density lipoprotein cholesterol. The samples with conjugated bilirubinemia show increased concentration of inorganic phosphours, otherwise the samples with unconjugated bilirubinemia show no interference or allowable range in the test result.
The distribution and some properties of alkaline phosphatase (ALP) were investigated in the midgut of the earthworm, Eisenia andrei. The ALP activity appeared to be highly polarized toward the luminal side of epithelium, with minor ALP activity in chloragogue tissue. The epithelial and chloragogeneous tissues contained approximately 85 and 15% of total intestinal ALP activity, respectively. The optimal temperature was approximately 37$^{circ}C$ and isoelectricpoint was estimated to be 4. The treatment of neuraminidase and PtdIns-PLC failed to change the migration rate of ALPs. Also, these ALPs appeared to have a wide range of substrate specificity. The relationship between the properties and physiological significances of the midgut ALPs in Eisenia andrei was discussed.
The concentration of amino acids, total nitrogen, trehalose, lipids and the activities of respiratory, acid$\cdot$alkaline phosphatase, glutamic oxalozcetic transaminase and glutamic pyruvic transaminase during larval stage in Pine leaf gall midge, Thecodiplosis janensis Uchi. et Inouye were measured using Paper chromatographic method, micro-Kjeldahl method, Thin layer chromatographic method, Warburg's manometric method, Bessey-Lowry method and Reitman-Frankel method, respectively. Healthy specimens )yore chosen as samples of each larval stages; alrva in gall and larva in soil. Amino acids present in the alcoholic extracts were alanine, glutamic acid, glycine, histidine, methionine, proline, threonine, tryptophan and valine. The total nitrogen concentration reached to 31.348mg/g during the larva in gall and the larval stage in soil of the value was decreased to 29.027mg/g. The hemolymph sugar, trehalose value for larva in soil was about two times of the value for larva in gall. Total lipid, phospholipid, monoacylglycerol, triacylglycerol, sterol, free fatty acid and ester cholesterol were identified at larval stages in gall and soil. Triacylglycerol concentration reached high level in contrast with other lipid contents during larvae in gall and larva in soil. Free fatty acid, sterol except decreased lipids during larval stage in soil. Endogenous respiration, succinate of respiratory activities decreased at larval stage in soil compare with larva in gall. The activities of acid phosphatase decreased larval stage in soil but the activities of alkaline phosphatase increased remarkably. The activities of glutamic oxaloacetic transaminase and glutamic pyruvic transaminase reached high level of the larva in gall.
The quantitative analyses of the phosphatase activity in the endometrium of the rat ovariectomized on Day 2 of pregnancy was carried out in comparison with the intact one, in order to investigate the hormonal dependency of the uterus prior to the implantation, and to study the phosphatase activity in the endometrial tissues in vitro incubated in different acidity of the medium. The results obtained were as follows: 1. The activity of the total phosphatase was the highest at Day 3 of pregnancy of the intact animals irrespective of acidity of the medium. However, the ovariectomized rat showed its peak somewhat delayed. The time of the highest activity of the enzymes was matched with the time of high secretion of the ovarian hormones. 2. The activity of acid phosphatase in the endometrium was twice or four times as much high as that of neutral or alkaline phosphatase, respectively. 3. The activity of alkaline phosphatase was rather steady in Day 3 through Day 5 of the pregnancy of the rat intact or ovariectomized but with low level compared to those of other phosphatase. 4. The present re~lt indicated more important role by $Mg^{2+}$-dependent phosphatase than by $K^+$-dependent one for the preparation for decidualization.
