The Effect of Blanching and Lactic Acid Bacterial Inoculation on the Quality of Kimchi (열처리 및 젖산균 접종이 김치의 품질에 미치는 영향)

  • The object of this study was to investigate the effects of blanching and lactic acid bacterial inoculation on the quality of kimchi. The pHs of the group added Leuconostoc mesenteroides were rapidly decreased, and then kept almost steady states. However, the pHs of the groups added Bifidobacterium bifidum were gradually decreased. Blanching treatment reduced the number of viable cells. At the beginning of the fermentation, the total organic acid contents of the blanched groups were lower than those of the non-blanched groups, but later on they were higher. With fermentation, the contents of malic, citric and fumaric acid were decreased in the control group, but increased in the cultured groups and all blanched groups. The cutting forces of the blanched groups were higher than those of the non-blanched groups during the whole fermentation period. The inoculation of Leu. mesenteroides was effective on the preservation of ascorbic acid. Blanching and the inoculation of Leu. mesenteroides gave good effect on the sensory acceptability. The acceptability of the groups added Bifidobacterium bifidum was low in initial fermentation period, but increased during the late fermentation period.

BIOASSAY OF HUMNA TOOTH PROTEIN BLOTTED POLYVINYLIDENE DIFLUORIDE(PVDF)MEMBRANE (사람치아 단백질을 분리 흡착한 PVDF막의 생체반응에 관한 연구)

  • Purpose: Human tooth proteins are highly heterogeneous, comprising diverse proteins derived from a number of genes. The attempts to identify protein for activity of tooth matrix proteins have been defied by several factors. First, the amount of proteins within teeth is very small relative to many extracellular matrix proteins of other tissues. Second, the bioassay system is tedious and needed for long time. Therefore we tried to find easy techniques, which increase the product rate, and an assay of small proteins, with which amino acid sequence is possible without additional procedures. Materials and Methods: Total protein were extracted from 300 g enamel removed teeth and 600 g teeth with 4 mol/L guanidine HCl and purified by gel chromatography. Aliquot of proteins was implanted into muscle pouches in Sprague-Dawley rats for bioassay. By SDS-PAGE and membrane blotting, molecular weight of each protein was estimated and a partial amino acid sequence was obtained. Each fraction blotted on the membrane was cut out and inserted in rat ectopic model. Results: In dissociative method, total tooth proteins were obtained 1mg/ml from enamel removed teeth and 3.5 mg/ml from teeth. In SDS-PAGE, four clear bands at the sites corresponding to 66, 40, 20 and 18 kD. Especially The 66 kD band was clearly exhibited. Amino acid sequencing from tooth could be possible using PVDF membrane blotting technique. In amino acid sequencing, 66 kD protein was identified as albumin. Conclusion: Compared with conventional method for extraction of teeth protein and bioassay of proteins, the methods in this study were easy, time-saving and more productive technique. The matured tooth proteins omitting additional procedure of mechanical removal of enamel were simply analyzed using blotted PVDF membrane. This method seems to make a contribution as a technique for bioassay and amino acid sequencing of protein.

Lipase-Catalyzed Synthesis of Structured Lipids with Capric and Conjugated Linoleic Acid in a Stirred-Batch Type Reactor (대두유로부터 Lipase를 이용한 재구성 지질의 합성 및 특성)

  • Structured lipid (SL) was produced from soybean oil with molar ratio of 1:2:2 (soybean oil:capric acid:CLA) using Chirazyme L-2 lipase (4% by weight of total substrates). The reaction was conducted for 24 hr at 55$^{\circ}C$ in a 1 L stirred-batch type reactor. SL-soybean oil contained 4.9 mol% capric acid and 4.1 mol% CLA, respectively. Iodine value of SL-soybean oil was reduced than that of soybean oil due to the incorporated capric acids. Tocopherol content in SL-soybean oil was 18.2 mg/l00 g. SL-soybean oil appeared more yellowish color than soybean oil. Reverse-phase HPLC showed that SL-triacylglycerol species containing capric acid consisted of about 12.6 area%.

Characteristics of brown rice Makgeolli brewed at different temperatures and mixing ratios of Nuruk (누룩비율과 온도에 따른 현미막걸리의 품질특성)

  • Brown rice Makgeolli was brewed by using the Saccharomyces cerevisiae strain SRCM102596 under different fermentation conditions: temperatures at 20 and 25℃ and Nuruk ratios of 5, 10, and 15%. There were no significant differences in the pH and total acidity between samples. The alcohol content at the different nuruk ratios varied significantly by the days in the fermentation process. The major free sugars were maltose, glucose, and fructose, and they gradually reduced with fermentation. The major organic acids in the brown rice Makgeolli were oxalic acid, citric acid, malic acid, succinic acid, and acetic acid. The lactic acid content increased with the number of days in the fermentation process. Among the 24 different free amino acid contents identified, the total free amino acid content of, especially, threonine, serine, and alanine were high in the brown rice Makgeolli, at 20℃ and nuruk ratio of 10%.

A study on manufacturing of red ginseng Makgeolli using the red ginseng starch and changes of physicochemical components of red ginseng Makgeolli during storage periods (홍삼 전분을 이용한 홍삼막걸리의 제조 및 이화학적 성분 변화)

  • This study was performed to develop the maufacturing processes of Makgeolli using red ginseng starch (RGS). After the fermentation of RGS with koji, nuruk, and yeast, the different temperature effects on the number of the yeast cells, the content of organic acid, free sugars, and total acid, and pH were investigated. There were no changes in the composition of the yeast cell number and content of organic acid amd during 20 days at $4^{\circ}C$. The content of free sugars (sucrose, glucose and mannose) and the pH value of red ginseng Makgeolli decreased during storage at $4^{\circ}C$. This meant that the total acid content and pH value increased after organic acid was produced from fermentation. Therefore, red ginseng Makgeolli is highly acidic and sour. Since high acidity helps improve storage conditions, so this developed red ginseng Makgeolli is considered safe for consumption. Furthermore, the total content of ginsenoside was 2.47 mg/mL, which was differentiate Makgeolli using red ginseng starch, with others. Therefore, new red ginseng Makgeolli is rich in organic acid, free sugars, and ginsenoside. As a result, its storage, taste, and flavor improved.

A Follow-up Study on the Nutritional Status of Korean Fatty Liver Patients with Different Conditions (한국인 지방간 환자의 병세변화에 따른 영양상태에 관한 Follow-up연구)

  • The purpose of this study is to investigate dietary factors which influence the conditions of Korean fatty liver patients. The subjects were 83 fatty liver patients living in city, ages of 10 to 60 yrs old, and they were the same patients studied 9 months ago in previous study. The patients were groupsed into 3 groups(improved, similar, worsened groups) according to the changes in disease condition. Nutrient and alcohol intakes and health status were investigated. In improved group, the patients tended to lose weight, the number of exercising patients was increased, the consumptions of calorie, carbohydrate, calcium, phosphorus were decreased. And alcohol intake was decreased markedly and serum total amino acids(AA), EAA, NEAA, BCAA and AMAA contents, EAA/NEAA and BCAA/AMAA ratios were increased. In worsened group, patients tended to gain weight, the number of exercising patients decreased, the consumptions of calorie, protein, carbohydrate, calcium, phosphorus and niacin decreased but vitamin. A intake increased. And also, in this group, alcohol intake of male patients decreased and that of female slightly increased, and serum EAA, BCAA and AMAA contents, EAA/NEAA and BCAA/AMAA ratios, and total fatty acid contents increased and total AA and NEAA contents decreased. In conclusion, it is desirable to reduced weight, stress, alcohol, salt and animal fat consumptions, and to exercise, and to take adequate amount and quality of protein to improve conditions of fatty liver patients.

Analysis of Trans Fatty Acid Content in Retort Food, Powdered Milk, Biscuit and Pizza Products (레토르트식품, 분유, 비스킷 및 피자 내에 함유되어 있는 트랜스지방산 함량 분석)

  • The consumption of foods containing trans fatty acids (TFAs) is a matter of concern at present. According to many studies, trans fatty acids (TFAs) may cause illnesses such as the coronary heart disease, diabetes mellitus, large intestine cancer, and breast cancer. They can also raise low density lipoprotein (LDL) cholesterol and reduce high density lipoprotein (HDL) cholesterol. TFAs can also inhibit the synthesis of phospholipids containing polyunsaturated fatty acids in arterial cells. As a consequence the Food and Drug Administration has deemed that saturated fatty acid, cholesterol and trans fatty acid levels be listed on food labels as of 2006. The Korea Food and Drug Administration also has required the listing of trans fatty acid content on food labels since 2007. The aim of this study was to determine the total lipid and trans fatty acid (TFA) contents in retort food, powdered milk, biscuit and pizza products. The number of samples examined were 2 retort food, 6 powdered milk, 7 biscuit and 3 pizza products. The extraction of total lipids in retort food and powdered milk followed the chloroform methanol method. The extraction of total lipids in biscuit and pizza was by the acid digestion method. All samples were analyzed by gas chromatography (GC) using a SP-2560 capillary column and a flame ionization detector. The TFA contents per 100g of sample were 1-2.8% (1.9%) in retort foods, 0.4-2.4% (1.37%) in powdered milk products, 0-2.9% (1.23%) in biscuits, and 2.8-3.45% (3.03%) in pizzas.

Quality Characteristics of Ginseng Jung Kwa and Jung Kwa Solution on Jung Kwa Process (인삼정과 제조과정에 따른 품질학적 특성)

  • This study was carried out to investigate the quality characteristics of ginseng Jung Kwa and Jung Kwa solution prepared according to boiling frequency on Jung Kwa process. Ginseng Jung Kwa was made as follows: washed ginseng 8 kg was boiled 5 min with water 16 kg and then boiling water 8 kg was removed. Sucrose 10 kg was added to boiled ginseng soaking in hot water. Jung Kwa was boiled down for 60 min in sugar syrup and soaked for 24 hrs. According to the above process, Jung Kwa was boiled down 5 times. Moisture and pH of Jung Kwa and Jung Kwa solution decreased as the number of boiling time increased and $^{\circ}Brix$ of Jung Kwa solution increased. Crude saponin content of last Jung Kwa (GJ 6) increased to 4 times of raw ginseng. Content of Rf and Rd, component of ginsenosides, increased 77 and 16 times on Jung Kwa, respectively and content of crude saponin on last Jung Kwa solution (GJS 5) was 61.88 mg/g. The other ginsenosides on Jung Kwa decreased. As the number of boiling time increased, concentration of total sugar, glucose and fructose of Jung Kwa and Jung Kwa solution increased. Content of total organic acid on Jung Kwa decreased, especially, succinic acid, main organic acid of ginseng, decreased rapidly. L value of Jung Kwa decreased whereas a and b values increased.

Effect of Niacin Supplementation on Rumen Metabolites in Murrah Buffaloes (Bubalus bubalis)

  • An experiment was conducted on 3 male rumen fistulated adult buffaloes fed on wheaten straw and concentrate mixture in a Latin square design to study the impact of niacin supplementation on rumen metabolites. Three animals were fed wheaten straw+concentrate mixture (group I, control), wheaten straw+concentrate mixture+100 ppm niacin (group II), and wheaten straw +concentrate mixture+200 ppm niacin (group III). After 21 days feeding, rumen liquor was drawn for 3 consecutive days at different time intervals (0, 2, 4, 6 and 8 h) to study the various rumen metabolites i.e., rumen pH, ammonia-N, total-N, trichloroacetic acid precipitable-N, non-protein nitrogen, total volatile fatty acids, their fractions and number of protozoa. Mean pH values in strained rumen liquor (SRL) of animals in 3 groups were 6.64, 6.71 and 6.67, indicating no statistically significant difference. Results revealed a significant (p<0.01) increase in TVFA concentration among the supplemented groups (group II and III) in comparison to control group. Mean TVFA concentration (meq/dl) was 9.75, 10.97 and 11.44 in 3 groups respectively. The highest concentration of TVFA was observed at 4 h and minimum at 0 h in all the 3 groups. The percentage of acetic, propionic, butyric and isobutyric acid was statistically similar among the three groups. The mean ammonia-N concentration (mg/dl SRL) was significantly (p<0.01) lower in group II (16.38) and group III (15.42) than group I (18.14). Ammonia-N concentration was higher (p<0.01) at 4 h as compared to all the time intervals. The mean total-N concentration (mg/dl SRL) was higher (p<0.01) in group II (74.16) and group III (75.47) as compared to group I (62.04). Total-N concentration was higher (p<0.01) at 4 h as compared to other time intervals and lowest value was recorded at 0 h.Concentration of TCA-ppt-N (mg/dl SRL) was significantly (p<0.01) lower in control group as compared to niacin supplemented groups. Mean value of NPN (mg/dl SRL) was significantly (p<0.01) lower in group III (23.21) as compared to group I (25.71), whereas groups I and II, and groups II and III were similar to each other. Total protozoa number (${\times}10^4$/ml SRL) ranged from 18.06 to 27.41 in group I, 20.89 to 38.44 in group II and 27.61 to 39.45 in group III. The mean protozoa number was significantly (p<0.01) higher in SRL of group II (27.60) and III (30.59) as compared to group I (22.48). It can be concluded from the study that supplementation of niacin in the diet of buffaloes had improved the rumen fermentation by decreasing the concentration of ammonia-N and increasing protein synthesis.

Characteristics of Volatile Flavor Compounds in Kochujang Prepared with Commercial Enzyme During Fermentation (효소제를 사용한 개량식고추장의 숙성과정 중 휘발성 향기성분의 특성)

  • Kochujang was prepared for this study with raw material inoculated by commercial enzyme of amylase and protease. Volati1e compounds of Kochujang were analyzed using a purge and trap method during fermentation and identified with GC-MSD. Total 54 kinds of volatile flavor components like 16 kinds of alcohol, 16 kinds of ester, 7 kinds of acid, 4 kinds of aldehyde, 2 kinds of alkane, 1 kind of benzene, 3 kinds of ketone, 1 kind of alkene, 2 kind of amine, 1 kind of phenol, other 1 were found. Total number of volatile flavor detected right after manufacturing were 23 kinds like 3 kinds of alcohol, 6 kinds of ester, 3 kinds of aldehyde. After 30 days storage, total number of volatile flavor went up to 31 kinds with addition of 4 kinds of alcohol, 1 kind of ester. The total number of volatile flavor after 120 days storage were increased to 49 kinds. Volatile flavor compounds detected during the storage period were total 20 kinds like 6 kinds of alcohol such as 2-methyl-1-propanol, ethanol, 3-methyl-1-butanol, 5 kinds of ester such as ethyl acetate, isoamyl acetate, ethyl butyrate, 3 kinds of aldehyde such as butanal, acetaldehyde and 6 kinds of others. Even though peak area % of flavor compound varied depends on fermentation period, ethanol, ethyl acetate, ethyl butyrate, ethenone, 2-methyl-1-propanol, 3-methyl-1-butanol were the main compounds that consisted of flavor from Kochujang which was made with enzyme treatment. Ethly acetate showed the highest result in the treatment of right after manufacturing, 3-methyl-1-butanol had up to 90th day and ether were the other days.
