Amino Acid, Amino Acid Metabolite, and GABA Content of Three Domestic Tomato Varieties

  • To determine the nutritional value of domestic tomatoes, the levels of amino acids, amino acid metabolites, and the bioactive compound ${\gamma}-aminobutyric-acid$ (GABA) were analyzed in three domestic tomato varieties (Rafito, Momotaro, and Medison). Eighteen free amino acids were found, and total free amino acid content was 3,810.21~4,594.56 mg/100 g (dry weight). L-glutamic acid (L-Glu) was the most abundant amino acid, ranging from 1,866.60 mg/100 g for Momotaro to 2,417.45 mg/100 g for Medison. The next most abundant amino acids were L-glutamine (L-Gln) and L-aspartic acid (L-Asp). The three tomato varieties had a good balance of all the essential amino acids except tryptophan. Total essential amino acid content was 274.26~472.71 mg/100 g (dry weight). The following amino acid metabolites were found: L-carnitine (L-Car), hydroxylysine (Hyl), o-phosphoethanolamine (o-Pea), phosphoserine (p-Ser), ${\beta}-alanine$ (${\beta}-Ala$), N-methyl-histidine (Me-His), ethanolamine (EtNH2),and L-citrulline(L-Cit). Large quantities of GABA were found in all three varieties: 666.95-868.48 mg/100g (dry weight). These results support the use of these tomato varieties as nutritious food materials.

Characterization of Bioactive Compounds of Domestic Tomato Varieties (국내산 토마토 품종의 생리활성 물질의 특성)

  • The aim of this study was to investigate bioactive compounds from three domestic tomato varieties (Rafito, Momotaro TY Winner, and Medison). Lycopene, ${\beta}-carotene$ and polyphenols were quantified and identified using HPLC and LC-MS/MS. The levels of lycopene ranged from 28.36 mg/100 g to 60.18 mg/100 g. The content of ${\beta}-Carotene$ ranged from 2.00 mg/100 g to 2.92 mg/100 g. Ten kinds of polyphenol compounds were identified: caffeic acid-hexose isomer (I), caffeic acid-hexose isomer (II), 3-caffeoylquinic acid, 5-caffeoylquinic acid, caffeoylquinic acid isomer, quercetin-3-apiosylrutinoside, quercetin-3-rutinoside, di-caffeoylquinic acid, tri-caffeoylquinic acid, and naringenin chalcone. The level of 5-caffeoylquinic acid was the highest in domestic tomato varieties, ranging from 12.71 mg/100 g to 28.40 mg/100 g. The content of quercetin-3-rutinoside ranged from 3.74 mg/100 g to 17.64 mg/100 g. The contents of 3-caffeoylquinic acid and quercetin-3-apiosylrutinoside were 1.01~2.31 mg/100 g and 5.84~6.83 mg/100 g, respectively. Arrestingly naringenin chalcone was found only in Medison variety (36.82 mg/100 g). These results revealed that domestic tomato can be a good source of bioactive compounds for human health.

Effect of the Clay Mineral Illite on the Growth of Cherry Tomato in the Bed Soil (상토에서 점토광물 일라이트 (illite) 처리에 따른 방울토마토 생육 효과)

  • This study was initiated to investigate the physical and chemical properties of the clay mineral illite and its effect on the growth of cherry tomato (Hongjak) as affected by the forms and concentrations of illite using the bed soil in the glass house. The composition of illite was analyzed by SEM(scanning electron microscope). The cherry tomato was cultivated in the bed soil in the glass house at Chungbuk National University for 10 weeks of total duration. The application rates of illite used as particulate and powder forms were standard 1:20 (w/w), two times 1:10 (w/w), four times 1:5 (w/w) of standard application, respectively. The surface of illite used in the study appeared to be hexagonal sheet and it included K and the variety of other elements. The growth lengths of cherry tomato were 11-23% greater in the pots treated than in the untreated pots. Based on the analyses of the major cations, K, Ca, and Mg from the cherry tomato the amounts of K taken in the leaf and stem of the cherry tomato were 8-45% and 3-30% greater than untreated pots, respectively. The amount of Ca in the leaf and stem taken up by the cherry tomato were 6-39% and 11-40% greater than untreated pots, respectively. The amounts of Mg in the leaf and stem taken up by the cherry tomato were 17-62% and 19-54% greater than untreated pots, respectively. Based on the analysis for the amount of cations, such as K, Ca, and Mg taken up by cherry tomato in treated and untreated bed soil, it appears that the illite treatment enhanced the early growth of cherry tomato.

Studies on the Juice Manufacturing Properties of Various Tomato Varieties (토마토의 품종별 쥬스제조 적성에 관한 연구)

  • For the elucidation of some informations on juice manufacturing properties of tomato, physical, chemical and sensory characteristic were analyzed for two groups of cultivation pattern-nonproped cultivation (5 varieties) and proped cultivation (4 varieties). The proped cultivation showed higher sugar content of juice than the nonproped on. Among the proped Jeokpung and Master 2 showed high value of sugar content. The lowest value of sugar content was observed in 79078$\times$ARC of the nonproped. There was no difference in acidity between cultivation groups but high value was observed in TM103, Jinhong from the nonproped and Jeokpung from the proped. There was no difference in pH between tomoto juice. High content of vitamin C was observed in 79078$\times$CL1561 from the nonproped and 76Moll-3-2-2 from the proped. High viscosity was observed in 79078$\times$CL1561 from the nonproped and Horgju from the proped. The nonproped showed greater value in dominant wavelength than the proped. Good Hope showed the greatest value in dominant wavelength, color score of panel test and yield. It was found that Good Hope from the nonproped and Jeokpung from the proped were suitable for juice processing. But better quality of juice can be possibly made by using various varieties rather than using single variety because each variety has each goodness of juice preparation separately.

Comparison of Feeding Behavior of B and Q biotypes of Bemisia tabaci (Hemiptera: Aleyrodidae) against Red Pepper and Tomato Varieties (고추와 토마토 품종별 담배가루이 B와 Q biotype의 섭식행동 비교)

  • Nymphal development of the B and Q biotypes of Bemisia tabaci was normal on all seven tomato varieties tested. However, their nymphal development was different on red pepper varieties. B biotype was not normally developed on nine red pepper varieties tested. On the contrary, Q biotype was normally developed, but its adult emergence rate was very low in Nokkwang variety than in other eight varieties. The EPG analysis of the feeding behavior of Bemisia tabaci showed that B and Q biotypes had different duration of phloem phases on red pepper. Q biotype showed longer phloem phases than B biotype. On Nokkwang variety, Q biotype had short phloem phases and did not prefer to feed on Nokkwang variety. Interestingly, Q biotype was found to have long duration of phloem phases on eight red pepper varieties, but B biotype did not prefer to feed on red pepper varieties. However, both biotypes did not show any difference in feeding time on tomato varieties.

Breeding on High Lycopene and Beta Carotene with Multi-Disease Resistance in Tomato

  • This study was carried out to breed and develop high quality and functional nutrient tomato with multi disease resistance as well as a stable growing adaptation for fresh market usage under protected plastic houses cultivation. The materials were used 5 inbred lines and their 6 hybrids of large tomato group, which have been bred and developed from 1999 to 2007 in Division of Plant Resource Department of Chungnam National University. Fruit weight showed hybrid vigor effect that $F_1$ hybrids weighed more than their parent lines, fruit shape formed three type of oblate, deep oblate and globe shape, in firmness and pericarp thickness have got a high significant correlation, inbred DN611 line was measured the most firm fruit with 6.04 mm pericarp thickness. In fruit color at maturity, pink color crossed to red color appeared all red fruit color in the $F_1$ hybrids, it means red skin color is a dominant gene compared to pink skin color is a recessive gene in tomato, while between fruit skin color and shoulder part color showed no any co-relationship. The sugar content and titratable acid of $F_1$ hybrids inherited an intermediate data of their parent lines, the flavor of KP543 inbred line and the hybrid (JB535 x KP543) revealed the better taste with high brix and proper titratable acid content$^{*}$. In beta carotene content DN611 line showed 2~3 times higher than other materials so that its 3 hybrids contained an increased level of beta carotene, lycopene content was not so much difference among inbred lines and $F_1$ hybrids, of them MD508 contained higher of 8.72 mg and hybrid (JB535 x JA517) had 8.05 mg lycopene content per 100 g fruit, overall pink skin color and red skin color measured a higher lycopene content than yellow and orange skin color at ripe stage. In disease resistance test by PCR marker for Fusarium race2 (I2), Nematode (Mi1), ToMV ($Tm2^2$), Cladosporium (Cf9), (JB535 x JA517) hybrid have got multi-resistance with homozygote band in Nematode, ToMV, Cladosporium and heterozygote band in Fusarium race2. Through this breeding program we could select high quality and functional nutrient with multi resistant $F_1$ hybrids and inbred lines in tomato which are two best hybrids (JB535 x MD508), (JB535 x JA517), additionally developed high beta carotene inbred line DN611 and increased the level of lycopene inbred line MD508. These results will be very useful to make a high quality tomato variety continuously.

Mineral- and Tissue-Specific Metabolic Changes in Tomato (Lycopersicon esculentum L.) Plants Grown under NPK-Starved Conditions

  • Specific metabolic network responses to mineral starvation are not well-defined. We examined a detailed broad-scale identification of metabolic responses of tomato leaf and root to N, P or K starvation. Tomato plants were grown hydroponically under optimal (5 mM N, 0.5 mM P, or 5 mM K) and starved (0.5 mM N, 0.05 mM P, or 0.5 mM K) conditions and metabolites were measured by LC-MS and GC-MS. Overall, the levels of metabolites (lipids, nucleotides, peptides and secondary metabolites) presented in this paper largely showed mineral- and tissue-specific responses. Most strikingly, G3P (glycerol-3-P), GPC (glycerol-P-choline) and choline phosphate responded differently to a type of mineral; an increase in N or K starvation and a decrease in P starvation. A dramatic increase in the levels of secondary metabolites, in particular, rutin and chlorogenate in both tomato tissues during N starvation were observed. Based on these data, it is necessary to clearly elucidate an unknown event taking place in a variety of abiotic impacts, and we are now studying to expand our knowledge on metabolic- and proteomic-responses using GS-MS and LC-MS.

Mineral Uptake and Soluble Carbohydrates of Tomato Plants as Affected by Air Temperatures and Mineral Treatment Levels

  • Both low and high temperatures affect plant growth and development at whole plant level, tissue and even cell level through a variety of metabolic changes. Temperature stress is one of frequently occurring problems in greenhouse crops in summer and winter seasons due to the wide-spread year-round cultivation. In the present study, we investigated the extent of the inhibition of growth, macro-element uptake and soluble carbohydrate production, and the effect of extra-supply of minerals as a means of the recovery from temperature damage. Tomato plants were grown five different growth temperatures (15/8, 20/13, 28/21, 33/23 and $36/26^{\circ}C$), and extra-supply of minerals was composed of 1.5- and 2.0-fold stronger than the standard nutrition (1/2 strength of Hoagland's solution). Temperature stress significantly adversely affected tomato growth and mineral uptake, whereas soluble carbohydrate accumulation represented temperature-dependent response, more accumulation at low temperature and more consumption at high temperature. The soluble sugars in leaves and stems were mostly declined with the supply of extra-minerals at low and optimal temperatures, whereas remained unchanged at high temperature. The starch levels also remained unchanged or slightly decreased.

Lifespan Elongation of Bombus terrestris and Economic Effect by Regular Pollen Supplement to Its Hives Released on Beefsteak-tomato Varieties (완숙토마토 품종별 서양뒤영벌(Bombus terrestris) 방사시 추가 화분공급에 의한 봉군수명 연장과 경제적 효과)

  • About 50,000 hives of Bombus terrestris are used for pollination of tomato and other economical crops (fruit vegetables and fruit trees) in Korea. Therefore this study was conducted to find out the effective use of B. terrestris and its economic effect in the major beefsteak-tomato cultivation areas from February to August in 2008. The pollinating activities, rate of bite-mark and lifespan of B. terrestris's hive according to all beefsteak-tomato varieties (Super Momotaro, Zeus42, Mascara and Super Sunroad, 500 $m^2$, $9,600{\sim}12,000$plants) were apparent by regular supplement of pollen ($5{\sim}6g$/3 days) to the hive of B. terrestris. Especially, Super Momotaro and Super Sunroad variety were most distinctive in the effects by regular pollen supplement.

Identification and Characterization of Novel Biocontrol Bacterial Strains

  • Because bacterial isolates from only a few genera have been developed commercially as biopesticides, discovery and characterization of novel bacterial strains will be a key to market expansion. Our previous screen using plant bioassays identified 24 novel biocontrol isolates representing 12 different genera. In this study, we characterized the 3 isolates showing the best biocontrol activities. The isolates were Pantoea dispersa WCU35, Proteus myxofaciens WCU244, and Exiguobacterium acetylicum WCU292 based on 16S rRNA sequence analysis. The isolates showed differential production of extracellular enzymes, antimicrobial activity against various fungal or bacterial plant pathogens, and induced systemic resistance activity against tomato gray mold disease caused by Botrytis cinerea. E. acetylicum WCU292 lacked strong in vitro antimicrobial activity against plant pathogens, but induced systemic resistance against tomato gray mold disease. These results confirm that the trait of biological control is found in a wide variety of bacterial genera.
