Characteristics of Nursery Plants Influenced by Leaflet and Raising Method for Soft-Nodal cuffing in Cherry Tomato (토마토 절간(節間)을 이용한 마디삽목 시(時) 삽수의 절위(節位)와 삽수의 소엽(小葉)부착 유무가 묘(苗) 소질에 미치는 영향)

  • This study was carried out to investigate the role of attached leaflet on rooting and growth by one nodal cutting in tomato. As cutting sources 4 to 6 nodes could be obtained from one nursery seedling. Medium for cutting was a mixture with perlite plus peat moss (1 : 1, v/v), and each plug capacity was 30mL in the 72 cells-plug tray. Plant height and the number of leaves were significantly increased by attached leaflet cutting in 'Pepe' and 'Coco' cherry tomato. Dry weights of top and root were increased as much as 3 to 15 times in the cutting attached leaflet. Rooting percentage was 93.5% in one node cutting and 86% in the cutting with hypocotyl node part. At 20 to 24 days after nodal cutting, healthy nursery plant could be produced to transplant in field. The nursery plants by nodal cutting (NPNC) showed more fibrous roots and less tap roots than that of seedling. In terms of rooting pattern, NPNC rooted at the whole ground stem part, while rooting of seedling occurred at basal part of hypocotyl.

Effect of Scion Age on Survival Rate and Initial Yield of the Grafted Tomato Seedlings (토마토 접목묘 생산시 접수연령이 활착율과 초기 수확량에 미치는 영향)

  • This research was conducted to know the appropriate leaf number of scion for tube grafting in context with survival rate and quality of seedlings, and early yield of $1^{st}$ and $2^{nd}$ cluster of cherry tomato plants. Scions were grafted when having leaves 1~2 (LF-2), 2~3 (LF-3), or 3~4 (LF-4). The treatment LF-3 showed the highest survival rates and the best results in 6 factors among 12 seedling quality factors. In case of early yield, the treatment LF-2 showed higher yield than others with the big difference. Economic analysis showed that LF-3 is desirable in case of nursery while LF-2 is better in case of farmers. The compromising solution is that farmers buy a little expensive seedlings grafted at the leaf stage of 1~2 from nurseries in terms of win/win strategy.

Determination of the Optimal Nitrogen Concentration in Pre-planting Fertilizers for the Cultivation of Tomato Plug Seedlings

  • This study investigated the effect of pre-planting nitrogen (N) fertilization levels added to a soilless root medium on the growth of 'Dotaerang Dia' tomato seedlings. The N levels were varied for a total of 7 treatments: 0, 100, 250, 500, 750, 1,000, or $1,500mg{\cdot}L^{-1}$. The pH of the root media gradually rose in all treatments as the seedlings grew; however, the differences in the pH were not significant among the treatments. The electrical conductivity (EC) of the root media was significantly different among the treatments from sowing to week three, then drastically decreased after week four, which diminished the differences in the EC among the treatments. At week six, plant height, leaf length, leaf width, number of leaves, and fresh and dry weights of the shoot were highest for the treatment with $500mg{\cdot}L^{-1}N$. In contrast, the treatment with $1,500mg{\cdot}L^{-1}N$ had the lowest results for all growth measurements. The fresh weight was 67% heavier in the $500mg{\cdot}L^{-1}N$ treatment compared to the $1,500mg{\cdot}L^{-1}N$ treatment. The total N content in the tissues was lowest in the treatment with $0mg{\cdot}L^{-1}N$ and highest in the treatment with $1,000mg{\cdot}L^{-1}N$. The contents of calcium (Ca), magnesium (Mg), and metal micronutrients in the tissues were highest in the $250mg{\cdot}L^{-1}N$ treatment. A previous study demonstrated that adjusting the fertilization level to promote growth to over 90% of the maximum growth is a good strategy for lowering production costs and preventing damage due to excessive fertilizer absorption by crops. Our results indicated that the optimal pre-planting N fertilization level for tomato plug seedlings should be lower than $500mg{\cdot}L^{-1}$ and the optimum tissue N contents should be around 3.21% to 4.60%.

Effects of Rootstocks and Nitrogen Levels on Plant Growth, Fruit Quality and Infection of Root Rot Fusarium Wilt Disease in the Grafted-Tomato Plant (대목의 종류와 질소비요의 시용량이 접목 토마토의 생장, 과실의 품질 및 뿌리썩음시들음병발생에 미치는 영향)

  • Tomato (lycopersicon esculentum Mill.‘Sunroad’) seedlings grafted onto the four different rootstocks, ‘Anchor-T’, ‘Kagemushia’, ‘Joint’ and ‘Vulcan’, and fed with different amount of nitrogen were investigated for their susceptibility to the root rot Fusarium wilt (Fusarium oxysporum Schl. f. sp. lycopersici Snyder et Hansen, race J$_3$) disease, plant growth, and fruit quality. The flowering of the first flower cluster was promoted in grafted plants (GP) as compared to non-grafted (NG) ones. The plant height was increased by the grafting, and the growth and stem diameter were enhanced with increased nitrogen levels. Grafted plants were little affected by the Fusarium wilt disease except ‘Anchor-T’rootstock (64.7% infection). The rate of malformed, underdeveloped, and gray-mold infected fruits were reduced in the grafted plants regardless of the rootstocks. Soluble solid content, sugars, ascorbic acids and organic acids did not show any difference between grafted and non grafted plants, but ascorbic acid tends to be reduced in the high nitrogen treatment. Content of N, Ca and Mg in leaves were greatly increased in the GP grown with high nitrogen level (30kg.㏊$^{-1}$ ).

Effects of Diurnal Alternation of Nutrient Solution Salinity on Growth and Fruit Quality of Tomatoes Hydroponically Grown in NFT System (NFT 수경재배시스템에서 주/야 양액농도변환이 토마토의 생육 및 품질에 미치는 영향)

  • This experiment was conducted to investigate the effects of diurnal alternation of nutrient solution salinity on growth and fruit quality of tomatoes (Lycoperisicon esculentum cv. 'House momotaro') hydroponically grown in root intercept bag-NFT (RIB-NFT) system. Plant height was the lowest in the high concentration during daytime (6/1 $dS\;m^{-1}$, day/night). Yield was very high in the concentration of 1/1 $dS\;m^{-1}$, it decreased with increasing the concentration of nutrient Yield was higher at low concentration (4/1 $dS\;m^{-1}$) at nighttime compared to the same concentration (4/4 $dS m^{-1}$) at daytime and nighttime, and the reverse (1/4 $dS\;m^{-1}$) was similar to the control (perlite culture). Yield was greatly reduced by higher concentration at daytime than nighttime, and the decrease was alleviated by lower concentration at nighttime. With increasing the concentration of nutrient solution during daytime, sugar content of tomato fruit was increased, but yield was decreased. In the other experiment, tomato plants were hydropoically cultured in NFT system diurnally alternated between Aichi's solution and $Ca(NO_3)_2$ solution. $Ca(NO_3)_2$ solution was supplied for 4 hours from 10:00 to 14:00 at daytime and from 22:00 to 2:00 at nighttime, respectively, and Aichi's solution was supplied for the time except the 4 hours. Ca content of leaves and sugar content of fruit were increased by supplying $Ca(NO_3)_2$ solution at daytime compared to nighttime, but plant growth was greatly suppressed by supplying $Ca(NO_3)_2$ solution with the concentration of 4 $dS\;m^{-1}(4/4^{Ca}\;dS\;m^{-1})$ at nighttime.

Morphology and Leaf Color Changes of Grafted Tomato Plug Seedlings Irradiated by Different Wavelengths of Photosynthetically Active Radiation during Low Light Irradiation Storage (저광 조사 저온 저장 중 PAR의 각 파장에 의한 토마토 플러그 묘의 형태 및 엽색의 변화)

  • To investigate the effects of different wavelengths of photosynthetically active radiation on the morphology and leaf color changes of a single tomato (Lycopersicon esculentum) seedling, we stored the seedling at $10{\pm}0.5^{\circ}C$ under eight different wavelengths (peak wavelengths; 405, 450, 505, 545, 600, 645, 680, and 700 nm) with a constant photosynthetic photon flux of $3{\mu}mol\;m^{-2}s^{-l}$ for 28 d. Under the 405, 450, and 505 nm wavelength conditions, the leaves of the seedlings showed vigorous shape with an upright morphology. Rachis elongation was suppressed and hence compact appearance was observed under the 450 and 505 urn conditions. Although the difference in leaf color between before storage and on 28 days after storage was observed under all wavelength conditions, the 405 and 700 um irradiations changed the leaf color to light green. Application of light-emitting diode (LED) light irradiated from around 450 to 545 nm can contribute to vigorous shape with an upright morphology of tomato seedlings during low light irradiation-low temperature storage.

  • Glyphosate (N-[phosphonomethyl]glycine) applied to the assimilate-exporting leaves or sprayed to the whole plants of tomato(Lycopersicon esculentum Mil var. Moneymaker) induced the rapid inhibition of 5-enolpyruvyl skimic acid 3-phosphate synthase(EPSP-synthase). It shows that EPSP-synthase activity precedes chlorophyll loss. There is no difference in EPSP-synthase activity between in vivo tomato meristem and cell suspension culture if glyphosate is not applied. The EPSP-synthase activity is in a range of 4 to 6 nkat per mg protein. The inhibition of EPSP-synthase action is induced within 36 h after glyphosate application while the Chl contents were reduced 48 h after the application. In cell suspension culture of tomato and Corydalis (Corydalis sempervirens), a sublethal concentration of glyphosate retards the fresh weight increase and prolonged lag phase. The fresh weight is reached maximal about 14 days after the subculture in the presence of glyphosate. The inhibitory effect of glyphosate on EPSP-synthase is remarkably induced in lag phase.

Introduction of Shiva Gene into tobacco and Potato Using Tissue-Specific Tomato PAL Promoter (조직특이성 promoter를 이용한 Shiva 유전자의 식물체내 도입)

  • In this study we tried to transform an antimicrobial peptide gene (Shiva) under the promoter of tomato phenylalanine ammonia-lyase (tPAL5) into tobacco and potato plants. Antimicrobial peptide gene was isolated originally from giant silk moth (Hyalophora cecropia) and modified ie nucleotide sequence to increase antimicrobial activity. Transgenic tobacco plants were regenerated and their seeds were tested on the media containing kanamycin (500 mg/L). The results of PCR amplification and genomic Southern blot hybridization confirmed the integration of construct (tPAL5 promoter-Shiva-NOS-GUS-NOS) into chromosome. We observed that one of the transgenic tobacco plants showed chromosome rearrangement when integrated. In case of potato transformation, the efficiency of regeneration was maximized at the medium containing Zeatin 2mg/L, NAA 0.01mg/L, GA$_3$ 0.1mg/L. We also observed the high expression of GUS (${\beta}$-glucuronidase) enzyme which was located next to the terminator sequence of nopaline synthase gene (NOS) in the vascular tissue of stem, leaves of transgenic potatoes. This result suggested that a short sequence of Shiva gene (120 bp) and NOS terminator sequence might be served as a leader sequence of transcript when translated.

Identification of Brassinosteroid-Related Protein, BAK1 from Nutrition Deficient Tomato Cultivated by Soilless Cultivation System (수경재배 영양결핍토마토에서 브레시노스테로이드관련 신호전달 단백질 BAK1의 동정)

  • Brassinolide insensitive associated receptor kinase 1(BAK1) is a critical component that play an important roles in signaling of brassinosteroid biosynthesis. Brassinosteroid-deficient and -insensitive mutants showed the characteristic of dwarf symptom. The nutrient deficient tomato showing stunt phenomenon was selected from soiless cultivation system using modified Sonneveld hydroponic solution. Twenty eight protein spots showing different expression levels compared to the control were isolated from extracts of stunted tomato leaves by 2D PAGE analyses. Significantly down-regulated 6 protein spots out of 28 protein spots were analyzed and sequenced by MALDI-TOF mass spectrometry. The protein spot having pI=4.5 and MW=24 kDa was identified as a signal protein, BAK1, which is directly related to brassinosteroid biosynthesis. In addition, five other protein spots were identified as BCK1, cystein proteinase, sulfutase, peroxidase and zinc finger factor respectively, and they were also signal proteins related to brassinosteroid biosynthesis. Furthermore, amplification of 500bp of BAK1 mRNA by RT-PCR using a primer set of peptide matched regions was inhibited conpared to that of the wild type. The results sugested that the BAK1 might be regulated at the transcription level in response to nutrition applications.

  • A severe blight disease of lily (Lilium spp.) caused by Botrytis elliptica was found in a vinyl house in Taejon for the first time in 1987. Initial symptom of the disease was brown spotting on leaves and stems of the plants. The spots gradually increased in size and coalesced to form bigger spots and blights on the leaves. Plants with severe damage became brown and sometimes died. In the inoculation experiments, the fungus was found to be pathogenic only to lily, but not to pepper, tomato, ginseng, onion and welth onion.
