생체판의 제작 및 실험 (A Study on the Manufacture of the Artificial Cardiac Tissue Valve)

  • Treatment of valvular heart disease with valve replacement has been one of the most popular procedures in cardiac surgery recently. Although, first effort was directed toward the prosthetic valve, it soon became popular that bioprosthesis, the valvular xenograft, was prefered in the majority cases. Valvular xenograft has some superiority to the artificial prosthetic valve in some points of thromboembolism and hemolytic anemia, and it also has some inferiority of durability, immunologic reaction and resistance to Infection. Tremendous efforts were made to cover the inferiority with several methods of collection, preservation, and valve mounting of the porcine valve or pericardium of the calf, and also with surgical technique of the valvular xenograft replacement. Auther has collected 320 porcine aortic valves immediately after slaughter, and aortic cusps were coapted with cotton balls in the Valsalva sinuses to protect valve deformity after immersion in the Hanks' solution, and oxidation, cross-linking and reduction procedures were completed after the proposal of Carpentier in 1972. Well preserved aortic valves were suture mounted in the hand-made tissue valve frame of 19, 21, and 23 mm J.d., and also in the prosthetic vascular segment of 19 mm Ld. with 4-0 nylon sutures after careful trimming of the aortic valves. Completed valves were evaluated with bacteriologic culture, pressure tolerance test with tolerane gauge, valve durability test in the saline glycerine mixed solution with tolerance test machine in the speed of 300 rpm, and again with pathologic changes to obtain following results: 1. Bacteriologic culture of the valve tissue in five different preservation method for two weeks revealed excellent and satisfactory result in view of sterilization including 0.65% glutaraldehyde preservation group for one week bacteriologic culture except one tissue with Citobacter freundii in 75% ethanol preserved group. 2. Pressure tolerance test was done with an apparatus composed of V-connected manometer and pressure applicator. Tolerable limit of pressure was recorded when central leaking jet of saline was observed. Average pressure tolerated in each group was 168 mmHg in glutaraldehyde, 128 mmHg in formaldehyde, 92 mmHg in Dakin's solution, 48 mmHg in ethylene oxide gas, and 26 mmHg in ethanol preserved group in relation to the control group of Ringer's 90 mmHg respectively. 3. Prolonged durability test was performed in the group of frame mounted xenograft tissue valve with 300 up-and-down motion tolerance test machine/min. There were no specific valve deformity or wearing in both 19, 21, and 23 mm valves at the end of 3 months (actually 15 months), and another 3 months durability test revealed minimal valve leakage during pressure tolerance test due to contraction deformity of the non-coronary cusp at the end of 6 months (actually 30 months) in the largest 23 mm group. 4. Histopathologic observation was focussed in three view points, endothelial cell lining, collagen and elastic fiber destructions in each preservation methods and long durable valvular tolerance test group. Endothel ial cell lining and collagen fiber were well preserved in the glutaraldehyde and formaldehyde treated group with minimal destruction of elastic fiber. In long durable tolerance test group revealed complete destruction of the endothelial cell lining with minimal destruction of the collagen and elastic fiber in 3 month and 6 month group in relation to the time and severity. In conclusion, porcine xenograft treated after the proposal of Carpentier in 1972 and preserved in the glutaraldehyde solution was the best method of collection, preservation and valve mounting. Pressure tolerance and valve motion tolerance test, also, revealed most satisfactory results in the glutaraldehyde preserved group.

Antioxidative and Probiotic Properties of Lactobacillus gasseri NLRI-312 Isolated from Korean Infant Feces

  • We selected a Lactobacillus spp. from Korean healthy infant feces based upon their antioxidant activity. This strain was identified as Lactobacillus gasseri by 16S rDNA sequencing, and named Lactobacillus gasseri NLRI-312. In the present study, we investigate the protective effect of this strain on the $H_2O_2$ induced damage to cellular membrane lipid and DNA in Jurkat cells. To estimate the extent of cellular lipid peroxidation inhibition, MDA (malondialdehyde) was measured, and DNA damage was tested by the comet assay. We also examined probiotic properties including tolerance to acid and bile, antibiotic resistance. From the results obtained, the supplementation of Jurkat cells with NLRI-312 decreased in DNA damage, while no effect was shown on MDA decrease. In probiotic properties, this strain was resistance to both acid and bile, showed considerably higher survival when incubated in pH 2 or 1% bile salts (w/v). We concluded that the NLRI-312 could be used as potential probiotic bacteria, with the effect of reducing DNA damage induced by $H_2O_2$.

고속충격에 의한 A1 5052-H34 합금의 관통거동에 관한 연구 (A Study on perforation behavior of Aluminum 5052-H34 alloy by high velocity impact)

  • In order to investigate the fracture behaviors(perforation modes) and resistance to perforation during ballistic impact of aluminum alloy plate, ballistic tests were conducted. Depth of penetration experiments with 5.56mm-diameter ball projectile launched into 25mm-thickness Al 5052-H34 targets were conducted. A powder gun launched the 3.55g projectiles at striking velocities between 0.6 and 1.0 km/s. radiography of the damaged targets showed different penetration modes as striking velocities increased. Resistance to perforation is determined by the protection ballistic limit($V_{50}$), a statistical velocity with 50% probability for complete perforation. Fracture behaviors and ballistic tolerance, described by perforation modes, are respectfully observed at and above ballistic limit velocities, as a result of $V_{50}$ test and Projectile Through Plates (PTP) test methods. PTP tests were conducted with $0^{\circ}$ obliquity at room temperature using 5.56mm ball projectile. $V_{50}$ tests with $0^{\circ}$ obliquity at room temperature were conducted with projectiles that were able to achieve near or complete perforation during PTP tests. The effect of various impact velocity are studied with depth of penetration.

CGMMV-CP 형질전환 수박대목의 CGMMV 내성시험 및 계통확보 (CGMMV Tolerance Test of CGMMV-CP Trangenic Watermelon Rootstock and Establishment of Transgenic Line)

  • 수박작물의 대목용으로 사용하는 수박공대에 CGiMMV-CP 유전자를 도입하여 개발된 LM 수박공대의 CGMMV 내성 정도를 격리하우스와 노지 포장내에서 조사하였다. 격리온실에서의 $T_{3}$ 형질전환 수박대목의 CGMMV 내성은 접종 후 70일까지 유지되는 반면 대조구는 접종 후 20일에 전부 이병되었다. 인위적 토양전염 포장에서 형질전환체는 접종후71일까지 약 40%의 내성률을 보였으며, 대조구는 접종 후 37일에 모두 이병되었다. 인위적 접촉전염 포장에서 형질전환체는 대조구에 비해 약 10일 정도 지연효과를 보였다. 따라서 CGMMV-CP 형질전환체는 CGMMV에 저항성을 가진 것이 아니라 감염시기를 지연시키는 부분 내성으로 나타났다. CGMMV-CP homozygous T 세대를 진전시켜서 형질전환 수박공대 계통을 $BC_{1}T_{5}$ 세대에서 선발하였다. 또한 LM 수박공대에 형질전환 되지 않은 접수 (슈퍼금천수박)를 접목하여 non-LM 수박을 생산하고 CGMMV-CP 유전자에 관련된 물질의 이동 여부를 조사하였다. PCR, northern, western 분석한 결과 수박공대 대목에서 형성되는 DNA, RNA, protein 물질이 접수로 이동되지 않음을 확인하였다.

방사선 유도 내염성 증진 사료용 옥수수 돌연변이체 특성 분석 (Characterization of a Gamma Radiation-Induced Salt-Tolerant Silage Maize Mutant)

  • 식물은 다양한 환경 스트레스에 적응하기 위해 스트레스 내성 유전자의 발현과 자연 돌연변이를 통해 외부 환경 및 자극에 대한 반응 특성을 강화시켜 왔다 본 연구는 사료용 옥수수를 대상으로 감마선을 이용하여 돌연변이 집단을 구축하고 내염성이 증대된 계통을 개발하고자 수행되었다. 140RS516은 NaCl 처리 조건에서 대조군인 KS140과 비교하여 증가된 염 스트레스 내성을 보였다. 감마선에 의한 다양한 유전변이를 보인 140RS516 식물체는 염 스트레스 조건에서 대조군보다 높은 발아율과 생장, 기공전도도 그리고 proline함량을 나타냈으며, 내염성에 관여하는 유전자들의 발현이 증가하였다. 따라서 본 연구를 통해 개발된 140RS516 옥수수는 간척지 염화토양과 같이 불량한 환경에서 작물 재배 및 생산이 가능한 내염성 품종을 개발하기 위한 육종 소재로 활용될 수 있을 것이다.

부품이 실장된 전자회로보드의 RLC 병렬회로 검사기법에 대한 연구 (A Study on the Test Method of RLC Parallel Circuits on the Device-Mounted Electronic Circuit Board)

  • In the existing ICT technique, the mounted electronic devices on the printed circuit board are tested whether the devices are good or not by comparing and measuring the value of the devices after separating the devices to be tested from around it based on the guarding method. But, in case that resistance, inductor and capacitor are configured as a parallel circuit on the circuit pattern, values for each device can not be measured because the total impedance value of the parallel circuit is measured. Accordingly, it is impossible to test whether the parallel circuit is good or not in case that the measured impedance value is within the tolerance error. Also, it is difficult to identify that which device among R, L and C of the parallel circuit is bad in case that the measured impedance value is out of the tolerance error. Accordingly, this paper proposes a test method which can enhance the quality and productivity by separating and measuring accurately R, L and C components from the RLC parallel circuits on the device-mounted printed circuit board. First, the RLC parallel circuit to be test is separated electrically from around it using three-terminal guarding technique. And then R, L and C values are computed based on the total impedance values and phase angles between voltage and current of the parallel circuit measured from two AC input signals with other frequency, Finally, the availability and accuracy of the proposed test method is verified by reviewing the simulation results.

주요경제수종(主要經濟樹種)의 내음성(耐陰性) 및 광선요구도(光線要求度)와 수분특성(水分特性)에 관한 연구(硏究)(II) -인공피음(人工被陰)이 활엽수(闊葉樹) 4종(種)의 수분(水分)포텐셜 및 기공증산저항(氣孔蒸散低抗)에 미치는 영향(影響)- (Studies on the Shade Tolerance, Light Requirement and Water Relations of Economic Tree Species(II) -Effects of Artificial Shade Treatment on the Water Potential and Stomatal Diffusive Resistance of Four Deciduous Hardwood Species-)

  • 전광처리구에서 2-6%의 상대투광량을 보이는 최강피음구까지 총 5수준의 인공피음 처리 하에서 자라는 자작나무, 느티나무, 고로쇠나무, 산벚나무 1년 생 묘목의 수분 특성을 밝히기 위해 기공증산저항과 수분포텐설의 일 변화와 계절 변화를 조사하였다. 활엽수 4종에 대해서 측정한 기공증산저항은 오전 9시에 1.7 ~25.1s/cm, 오후 1시 2.3~33.3s/cm, 오후 5시에 1.1~36.8s/cm의 범위 내에서 수종과 계절에 따라서 피음처리 수준별로 다양한 변화를 보였다. 기공증산저항은 조사된 대부분의 수종에서 피음도가 강해질수록 증가하였으며 6월이나 7월에 비해 9월에 더 높았다. 느티나무, 산벚나무와 고로쇠나무에서는 전광처리구에 비해 최강피음구의 증산저항 측정치가 약 3~4배 높았다. 수분포텐셜 또한 오전 9시에 -0.17~-1.20MPa, 오후 1시 -0.30~-2.03MPa, 오후 5시에 -0.18~-1.34MPa의 범위 내에서 수종과 계절에 따라서 피음처리 수준별로 다양한 변화를 보였다. 수분포텐셜은 조사된 대부분의 수종에서 9월의 측정치가 6월이나 7월의 측정치보다 낮은 값을 보였으며 그 계절적인 차이는 다른 수종에 비해 느티나무에서 현저하게 커지고 있었다. 수분포텐셜은 전광처리구에 비해 최강피음구에서 약 2~3배 더 높았지만 그 차이는 기공증산저항에서 보이는 차이보다는 다소 작은 것으로 분석되었다. 5 수준의 피음도 구배에 따른 수분포텐셜의 차이는 자작나무의 경우에 다른 수종보다 상대적으로 작았다. 내음성이 다소 안정되는 고로쇠나무나 산벚나무에서는 전광처리구에서 상대투광량 22-28% 의 중피음구까지 피음도를 높이면서 한낮을 중심으로 수분포텐셜이 급속하게 증가되었으나 최강피음구에 접근하면서 수분포텐셜의 증가는 둔화되었다. 양수에 속하는 자작나무나 느티나무의 경우에는 전체적으로 피음수준이 높아지면서 수분포텐셜이 점진적으로 증가하였으며 그 증가폭은 자작나무보다는 느티나무에서 더 컸다.

부탄올 내성 미생물의 분리, 동정 및 변이주의 개발 (Isolation, Identification and Mutant Development of Butanol Tolerance Bacterium)

  • 부탄올 용매에서 생존하는 부탄올 내성 미생물을 분리하였다. 분리된 미생물들의 세포성장은 부탄올 농도가 증가함에 따라 감소하였으며, 그 중에서 BRS02가 12.5 g/L에서 가장 높은 내성도를 나타내었다. 또한, UV를 이용하여 BRS02균의 변이를 유도하여 고농도 부탄올 내성균 BRS251을 개발하였다. 부탄올 생산 모델균주로 대장균과 함께 부탄올, 프로판올 및 펜탄올에 대한 내성도를 비교한 결과, 대장균은 7.5 g/L 부탄올과 20 g/L 프로판올, 2 g/L 펜탄올 농도까지 생육이 가능한 반편, BRS251은 더 고농도인 17.5 g/L 부탄올과 32.5 g/L 프로판올, 6 g/L 펜탄올 농도까지 생육이 가능하였다. 분리된 세균을 동정하기 위해서 그람염색 후 광학현미경으로 관찰한 결과 그람양성의 구균으로 확인이 되었으며, 6.5% NaCl에서 생육이 가능하였다. 생화학적 특성을 분석한 결과, arginine dihydrolase, ${\alpha}$-glucosidase, urease 효소활성을 가지고 있었으며, 호기적인 조건에서 D-galactose, Dmaltose, D-mannitol, D-mannose, methyl-${\beta}$-D-glucopyranoside, D-ribose, sucrose, D-trehalose를 탄소원으로 자화하여 산을 생성할 수 있었으며, bacitracin, vibriostatic agent O/129 및 optochin에 대한 항생제 내성을 나타내었다. 16S rRNA 유전자 서열을 결정하고 계통발생도 분석을 통해 BRS02는 최종적으로 Staphylococcus sp.임을 동정하였다.

Insulin secretory activity and mechanism of compound K

  • Purpose: Panaxadiols are more potent than panaxatriols as far as insulin secretory activity is concerned. In this study, we examined insulin secretory activity and mechanism of compound K (CK), a major intestinal bacterial metabolite of ginsenosides. Method: Insulin secretory activity of CK was examined using pancreatic beta cells and in Oral Glucose Tolerance Test assay. In addition, insulin secretory mechanism was studied in terms of calcium dependent or independent pathways. Results: In vitro, CK enhanced the insulin secretion concentration-dependently when compared to glucose-stimulated control cells. Insulin secretory mechanism of CK seems to block ATP sensitive K channels, which was confirmed by diazoxide (K channel opener) but, insulin resistance ameliorating activity of CK can't be ruled out. In vivo, CK showed hypoglycemic effect in OGTT.

False Positive SOD Activity of Bifidobacterium spp. Grown in MRS Medium

  • The superoxide dismutase (SOD) activity of seven Bifidobacterium spp. strains was examined by an indirect SOD assay method. Some Bifidobacterium spp. showed significant levels of SOD activity. However, we could not observe any significant differences between anaerobic and aerobic cultures. Furthermore, although several Bifidobacterium spp. exhibited some degree of tolerance to paraquat which produces superoxide radicals, the apparent SOD activity of these strains was not correlated with their resistance to paraquat. In addition, when we added increasing amounts of manganese or iron to MRS medium which had been prepared without either of the metal ions, the apparent SOD activity of cell free extracts (CFEs) was increased with increasing concentration of both metal ions. To our surprise, the heat-denatured CFEs also showed nearly identical correlative patterns. Based on these results, the apparent SOD activity was likely due to a nonenzymatic dismutation. These results strongly suggest that high concentration of divalent metal ions ($Mn^{2+}$, $Fe^{2+}$) in MRS medium result in nonenzymatic dismutation which can lead to false positive SOD activities in Bifidobacerium spp.
