Milk mineral과 α-tocopherol 첨가가 유화형 소시지의 지방산화와 무기물 함량에 미치는 영향 (Effects of Addition of Milk mineral and α-tocopherol on the Lipid Oxidation and Mineral Contents of Emulsion-type Sausages)

  • 본 연구는 유화형 소시지 제조 시 milk mineral과 α-tocopherol을 첨가하여 소시지를 제조한 다음 소시지의 지방산화와 무기물 함량에 미치는 영향을 비교하고, 또 이들의 시너지 효과를 구명하고자 실시하였다. 소시지의 pH값은 milk mineral 첨가와 milk mineral과 α-tocopherol 혼합 첨가 소시지가 전 저장기간 동안 대조구와 α-tocopherol 첨가 소시지에 비해 높았다(P<0.05). POV값은 milk mineral과 α-tocopherol 혼합 첨가 소시지가 대조구와 α-tocopherol 첨가 소시지에 비해 낮았다(P<0.05). TBARS는 milk mineral 첨가와 α-tocopherol과 milk mineral 혼합 첨가 소시지가 대조구와 α-tocopherol 첨가 소시지에 비해 낮았다(P<0.05). 마그네슘, 칼슘 및 인 함량은 milk mineral과 α-tocopherol 혼합 첨가 소시지가 대조구, milk mineral 및 α-tocopherol 첨가 소시지에 비해 현저히 높았지만, 나트륨과 칼륨 함량은 α-tocopherol 첨가 소시지가 높았다(P<0.05).이상에서 milk mineral과 α-tocopherol 혼합 첨가 소시지는 대조구와 α-tocopherol 첨가 소시지에 비해 POV와 TBARS가 낮아 지방산화 억제 효과가 있었지만, 현저한 시너지 효과는 없었다. 또한 milk mineral과 α-tocopherol 혼합 첨가 소시지는 각종 무기물 함량을 증진시킬 수 있는 육제품 생산이 가능하다고 사료된다.

  • To observe the effect of the different level of PUFA and marginal tocopherol supplement on HDL-chol, tissue tocopherol content and fatty acid composition, the rats were supplied either safflower oil or conconut oil with or without tocopherol supplement to the experimental diet. Plasma tocopherol level was not greatly influenced by the different dietary fat and similar effect was observed in the liver but not in the adipose tissue. HDL-chol level was reduced in the high PUFA diet regardless of tocopherol content. No effect by tocopherol supplement was observed in the fatty acid composition of liver and adipose tissue lipid in both dietary PUFA levels . There was also no increase in the content of tissue polyenoid acid by tocopherol in the high PUFA diet . Fatty acid composition of tissue lipid was rather more influenced by dietary fat. Lauric and myristic acid contents were higher in the low PUFA diet and linoleic acd and total polynoic acid content were higher in the high PUFA diet. With tocopherol supplement tocopherol /PUFA ratio of tissue was increased but the ratio of high PUFA diet was significantly lower than that of low PUFA diet. Marginal tocopherol supplement could not reduce the peroxidizability index of high PUFA diet.

한국 성인 남자의 혈장 Tocopherol 수준과 관련 요인 분석 (Association between Plasma Tocopherol Levels and Related Factors in Middle-Aged Korean Men)

  • Vitamin E in the body system plays an important role in preventing chronic diseases by decreasing the oxidative stress by free-radicals. However, there are not enough researches on analyzing the primary factors affecting vitamin E levels in the blood in Korean adults. Therefore, the purpose of this research was to examine blood tocopherol levels and the primary factors affecting the status. A complete lifestyle survey was performed on 314 Korean adult men and surveyed their smoking, drinking and exercising habits. The average plasma level of ${\alpha}-\;and\;{\gamma}-tocopherol$ showed similar mutual relations with plasma total cholesterol (TC), triglyceride (TG), or low density lipoprotein cholesterol (LDL-C) levels (p<0.001). Plasma ${\alpha}-tocopherol$ level of the subjects did not show any difference as smoking, drinking and exercising habits changed. However, ${\gamma}-tocopherol$ per TG showed much lower figure in smokers than non smokers (p < 0.05). Amongst diet factors, plasma ${\alpha}-tocopherol$ level showed negative correlations with Vitamin E intake, while ${\gamma}-tocopherol$ level showed positive correlations with Vitamin E intake. Erythrocyte superoxide dismutase (SOD) activity and plasma tocopherol showed negative correlations, and catalase activity and plasma ${\alpha}-tocopherol$ showed positive correlationship. The level of cell DNA damage of Iymphocyte and plasma ${\alpha}-\;or\;{\gamma}-tocopherol$ showed negative correlations. As a result of this research, the factors that affect Korean adult men's plasma ${\alpha}-tocopherol$ level are plasma TG, LDL-C and cell DNA damage in Iymphocyte, while the factors that affect ${\gamma}-tocopherol$ level are plasma TG, LDL-C and vitamin E intake based on multiple regression analysis. These findings implies that the level of different types of tocopherol depends on slightly different factors. A further research is needed on the factors involved in the differentiation of the types of tocopherol.

에탄올과 토코페롤이 간조직의 지질산화와 미토콘트리아 산화능에 미치는 영향 (Effects of Ethanol and Tocopherol on Hepatic Peroxidation and Mitochondrial Respiration in the Rat)

  • To investigate effects of chronic alcohol consumption and tocopherol on lipid peroxidation and mitochondrial respiration 48 male rats of Sprague-Dawley strain were divided into 4 groups. Each group received for 3 weeks one of 4 experimental diets: tocopherol deficient control (TDC), tocopherol deficient-ethanol (TDE), tocopherol-supplemented control (TSC) and tocopherol-supplemented-ethanol (TSE). Composition of the diets was based on the Lieber and Decarli liquid diet and $\alpha$-tocopherol was supplemented at the level of 30mg/liter of diet, and ethanol supplied 36kcal%. TDC and TSC were pair-fed to TDE and TSE, respectively. Increase of body weight of tocopherol deficient-ethanol group was the lowest and the effect was diminished with tocopherol supplementation. Respiration of liver mitochondria was depressed in ethanol-administered groups and the effect became larger with tocopherol deficiency. Hepatic lipid peroxide level was not influenced by ethanol, but hepatic tocopherol content decreased with ethanol treatment. The result indicated that, although lipid perroxide level was unchanged with chronic ethanol consumption, oxidative stress exists in tissues of rate administered ethanol and may be relieved by tocopherol supplementation.

$\alpha$-tocopherol의 항산화작용에 미치는 인삼사포닌분획의 영향 (The Effect of Ginseng Saponin Fraction on Antioxidant Activity of $\alpha$-Tocopherol)

  • 인삼 사포닌 분획이 $\alpha$-tocopherol의 항산화작용에 미치는 영향을 시헌관내 실험과 동물실험으로 관찰하였다. 흰쥐 (Wister, 수컷, 180~200g)의 간 미크로좀 분획을 $Fe^{3+}$, NADPH, ADP, $\alpha$-tocopherol($50{\mu}M}$), 인삼 사포닌 분획(최종농도 $10^{-4}$%, 대조군은 사포닌 분획 대신 같은 부피의 물)을 함유한 혼합물에서 반응을 진행시키고 생성된 malondialdehyde를 분석한 결과, 사포닌은 $\alpha$-tocopherol의 항산화작용을 상승적으로 증가시켰다. 인삼 saponin 분획이 함유된 $\alpha$-tocopherol의 alcohol용액을 쥐에게 경구투여한 후 적당 시간 후에 간에서의 과산화수소량을 분석하여 대조군과 비교한 결과, 인삼 saponin 은 $\alpha$-tocopherol의 흡수를 촉진하므로서 $\alpha$-tocopherol의 항산화활성이 더욱 효율적으로 일어나게 하는 것으로 해석된다.

Tocopherol과 Ascorbate 투여가 사염화탄소로 유도한 흰쥐의 효소활성 및 항산화적용에 미치는 영향 (The Effect of Tocopherol and Ascorbate on the Enzyme Activity and Antioxidation in $CCl_4$ Induced Rats)

  • The enzyme activities and antioxidative of tocopherol and ascorbate were investigated at the levels of liver, kidney homogenates and sera of SD-rats intoxicated with tetrachlorocarbon$CCl_4$. GOT and GPT activities of sera in the $CCl_4$ group were 3, 6 times increased compared to normal group. But they tended to decrease significantly in tocopherol and ascorbate administered group. As for BUN and total cholesterol they were the same. HDL-cholesterol in the $CCl_4$group was 42% decreased compared to normal group. HDL-cholesterol was about 26% increased in the tocopherol group and tocopherol ascorbate group compared to $CCl_4$ group. MDA and SOD activities in the liver and the kidney tissue homogenates were significantly increased in $CCl_4$ group compared to normal group. But they were decreased significantly in the tocopherol group and tocopherol-ascorbate group compared to $CCl_4$ group. In view of this study tocopherol and ascorbated were ascorbate were effective on the detoxication of liver and kidney injury.

$\omega$6/$\omega$3계 불포화지방산을 투여한 후 혈장의 지질조성과 Tocopherol, Malondialdehyde 형성 및 적혈구의 Hemolysis 변화에 대한 Kinetic 연구 (Kinetic Study on Variations of Lipids, Tocopherol and Malondialdehyde Levels of Plasma and Red Blood Cell in Young Women fed Dietary $\omega$6/$\omega$3 Polyunsaturated Fatty Acids)

  • Ten college women were treated with either corn oil(CO< as a source of C18:2, $\omega$6) or fish oil concentrates(FO, as a source of C20:5+C22:6, $\omega$3) with experimental diet for 7 days and then returned to normal home made diet. Kinetic changes of plasma and RBC lipids, tocopherol, MDA levels and hemolysis were observed at day-0, -3, -7, -8, -10, -14 and -21 of experimental periods. Plasma and RBC tocopherol contents were significantly increased at day-3 and -7 by tocopherol tocopherol supplement in dietary oil, but there was no significant difference between CO and FO diet with large dose of tocopherol supplement. After stop experimental regimen at day-7, plasma tocopherol content was sustained at high level until day-10 but drastically decreased at day-14 and remained at low level at day-21. However, RBC tocopherol level was not greatly responded to the dietary intake of tocopherol and was varied in the very narrow range. MDa levels of plasma and RBC were not responded in the same way as tocopherol content of plasma. MDa content of RBC was very low compared to that of plasma. RBC hemolysis by incubation in hypotonic solution was negatively correlated to plasma tocopherol level and was not correlated to the level of MDa in either plasma or RBC. There were no significant change in the levels of plasma cholesterol, HDL-chol, triglyceride, phospholipid, and lipoprotein pattern throught experimental periods.

Tocopherol Content and Composition in Peanut

  • The content and composition of tocopherol were analyzed in peanuts produced in Korea. The tocopherol assay was consisted of an extraction with n-hex-ane:isopropanol mixture followed by high pressure liquid chromatography with fluorescence detection. Tocopherol content was higher in leaves and seeds but lower in seed coats and shells. $\alpha$-/${\gamma}$- Tocopherol ratio was as low as 0.53 in seeds and as high as 22.5 in shells. Tocopherol content in seeds of nine varieties ranged from 142 to 220 ng/mg dry weight, and the $\alpha$-/${\gamma}$-tocopherol ratio from 0.40 to 0.75. Tocopherol content decreased by 18% but $\alpha$-/${\gamma}$-tocopherol ratio increased by 44% in roasted seeds. The results indicate that the level of tocopherol in Korean peanut varieties is moderate and the ratio of $\alpha$- to ${\gamma}$-tocopherol is low.

Alpha-Tocopherol Contents of Peanut Seeds Depend on Varieties and Processing Methods

  • The purpose of this study is to establish an extraction and analysis method for $\alpha$-tocopherol, and to then distinguish among varieties. The $\alpha$-tocopherol contents of 22 varieties of peanut seeds were analyzed by HPLC. Peanut seeds of cv. Palkwang were processed in two ways, by boiling and roasting. The $\alpha$-tocopherol contents of the two types of peanuts were compared with fresh seeds without any processing. $\alpha$-Tocopherol was detected at a retention time of 2.95 minutes. Five standards of $\alpha$-tocopherol covering a range of $20{\sim}100{\mu}g/ml$ were made up in 2% isopropyl alcohol/n-hexane and analyzed in duplicate. The $\alpha$-tocopherol contents differed according to extraction temperature. The contents were $85{\mu}g/g$ or less at $10^{\circ}C\;and\;20^{\circ}C$ and $94{\mu}g/g$ at $30^{\circ}C$, but they were decreased at $40^{\circ}C$ or higher. The $\alpha$-tocopherol contents in 22 peanut varieties were $61.36{\sim}96.80{\mu}g/g$ according to variety. Fresh peanuts contained $106.7{\mu}g/g$ of $\alpha$-tocopherol, while boiled peanuts contained $108.8{\mu}g/g$ of $\alpha$-tocopherol, and roasted peanuts contained $109.2{\mu}g/g$ of $\alpha$-tocopherol.

  • 한국산 인삼(Panax ginseng C. A. Meyer) 뿌리에서 추출한 사포닌 성분이 ${\alpha}$-tocopherol의 항산화 활성에 미치는 영향을 실험관 내 실험과 동물실험으로 관찰하였다. 흰쥐(Sprague-Dawley, male, $18{\~}200g$)의 마이크로좀 분획을 NADPH, $Fe^{+3}$, ATP $\alpha^{-+}$-ocopherol 과 인삼 사포닌 분획 (대조군은 사포닌 분획대신 같은 부피의 물)을 함유한 혼합물에서 30분간 반응을 진행시키고 생성되는 molondialdehyde를 분석한 결과 사포닌 분획은 ${\alpha}$-tocopherol의 항산화작용을 상승적으로 증가시켰다. 그리고 ${\alpha}$-tocopherol의 항 산화작용에 상승적 효과는 반응액에서의 인삼 사포닌 분획의 농도가 $10^{-5}\%$ 전후에서 특히 현저하였다. 술로 과산화물을 유발케 한 쥐에게 ${\alpha}$-tocopherol만을 또는 ${\alpha}$-tocopherol과 인삼 사포닌 분획의 혼합물을 경구투여하고 일정 시간 후에 간의 과산화 지질을 thiobarbituric acid 방법으로 분석한 결과 사포닌 분획이 동물체내에서도 ${\alpha}$-tocopherol의 항 산화작용을 크게 촉진시키는 것으로 관찰되었다. 또한 사포닌 분획은 ${\alpha}$-tocopherol의 흡수를 촉진시키는 것으로 보고되고 있으며 본 실험에서도 이것을 확인하였고 이와 같은 사포닌 분획의 ${\alpha}$-tocopherol흡수 촉진은 ${\alpha}$-tocopherol의 항 산화작용을 더욱 효과적으로 촉진 시킬 것으로 생각된다.
