황색종 연초 돌연변이 계통 KF8832-85의 특성 (Characteristics of A New Flue-cured Tobacco Mutant Line KF 8832-85)

  • A new flue-cured tobacco mutant line BU 8832-85 was developed at Taegu Experiment Station, Korea Ginseng and Tobacco Research Institute in 1994. KF 8832-85 was resulted from a cross of flue-cured cultivars NC 95$\times$NC 2326, and developed by a pedigree system of breeding ; initial selection was made by plant type and resistance to bacterial wilt(BW) disease(heudomonas solanaceamm) in the F2 generation under the natural field conditions infested with the pathogen. One white flowered plant was occurred by spontaneous mutation in a certain line among the F3 generatioin while the others were pink. Six plants from the seeds by selfing were selected at the field infested with the pathogen among 240 populations with white flowering in the F4, KF 8832-85 was selected based on yield and leaf quality trials among 6 lines in Fs generation. BCF 8832-85 was compared with its Parent for certain agronomic and chemical characteristics at Taegu Experiment Station in 1993 and 1994. The results showed that KF 8832-85 have white flower, the stalk height was approximately that of NC 2326 but averaged about loom taller than NC 95. It produced ground suckers as much as NC 95, and did not breakdown leaf at the same as WC 2326. KF 8832-85 have high resistance to bacterial wilt disease. Yield of KF 8832-85 was 10 and 18% higher then that of NC 2326 and WC 95, respectively. Price per Kg was equal to that of NC 2326. The contents of nicotine and reducing sugar did not differ significantly from NC 95, while total nitrogen was significantly lower than NC 95. Therefore, the new mutant line is genetically stable for agronomic and chemical characteristics and provides a source of bacterial wilt disease resistance for use in breeding resistant flue-cured cultivars. I. Effect of Acidity(pH), Moisture(pF) and Anions (Cl-, SO4-) in Soil on Grwth and Ionic Balance of Tobacco)

  • 토양(土壤)의 몇가지 물리적(物理的), 화학적성질(化學的成質)이 연초식물(煙草植物)의 생육(生育) 및 Ionic Balance에 어떤 영향을 미치고 있는가를 연구(硏究)하기 위하여 폿트실험을 하였다. 공시(供試)된 토양(土壤)은 pH를 3.5와 5.8인 두가지로 조정하였고 이에 2종류의 음(陰)이온을 ($NH_4Cl$의 Cl, $(NH_4)_2SO_4$$SO_4$) 처리하였고, 4수준(水準)의 pF치(値)(1.5-2.0-2.5-3.5)를 두었다. 그 결과(結果)를 요약(要約)하면 다음과 같디. 1. 산도(酸度)가 높은 토양(土壤)(pH3.5)에 $NH_4Cl$이 가(加)해진 경우 연초(煙草) Cl의 함량(含量)이 높았으며, $NH_4$와 Cl의 해작용(害作用)뿐아니라, Mn의 독작물(毒作用)까지 받았으므로 거의 생장(生長)하지 못하고 대단히 비정상적(非正常的)인 생육(生育)을 하였다. 한편 ($(NH_4)_2SO_4$를 많이 흡수하므로 Mg와 Ca의 결핍증(缺乏症)을 보였다. 그러나 산도(酸度)가 낮은 토양(pH5.8)에 가(加)해진 $NH_4-N$은 질산화작용(窒酸化作用)에 의하여 모두 $NO_3-N$로 변하였으며, $NH_4-N$와 함께 가(加)해진 Cl이나 $SO_4$에 관계없이 연초(煙草)는 건전한 생육(生育)을 하였으며 Cl 보다는 $SO_4$가 가(加)해진 토양(土壤)에서 자란 식물(植物)이 건물중(乾物重)도 더 높고 성숙도(成熟度)도 더 빨랐다. 2. 토양(土壤)의 수분함량(水分含量)은 식물(植物)이 인산(燐酸)을 흡수하는데 제한인자(制限因子)가 되는 것으로 생각되며, 지상부(地上部)와 뿌리의 건물수량(乾物收量)은 pF 1.5>2.0>2.5>3.5>의 순서이었고 pF3.5은 연초(煙草)의 일시계절점으로 생각되었다. 3. 연초식물(煙草植物)의 화학분석치(化學分析値)와 건물수량(乾物收量)은 이 식물(植物)이 정상적(正常的)인 (C-A) 개념(槪念)에 잘 적용된다는 것을 보여주었으며, 정상적(正常的)인 생육(生育)을 하고 있는 연초식물(煙草植物)에 있어서는 지상부(地上部)의 Carboxylate의 함량(含量)은 (C-A) 값과 거의 비슷함을 보여 주었다. 만약 식물체중(植物體中)에 $NH_4$함량(含量)이 상당히 많은 양(量)으로 존재(存在)한다면 $NH_4$를 분석(分析)하여 그 값을 (C-A)의 값을 계산하는데 사용해야 할것이다. Al은 가동성(可動性)이 매우 낮은 물질이므로 식물(植物)의 지상부(地上部)의 해(害)를 줄정도의 양(量)으로는 존재(存在)하지 않고 많은 양(量)이 뿌리의 내부나 그 주위에 존재(存在)한다. 식물체(植物體)가 Al을 많이 함유(含有)하고 있을 경우에도 (C-A) 값의 산출에 Al의 함량(含量)을을 고려해야 한다.

  • KA 101 and Xanthi-Basma were cultivated in Korea and Greece to compare the respective responses to environment and cultivation method on their growth characteristics and some properties of harvested leaves during 1984 and 1985. The contents of clay, calcium, organic matter and pH were higher in the soil of greece than that of Korea. Sunshine hours and precipitation in Greece during harvest and curing season were 8.6 hours per day and 53.3mm compared to that of 3.8 hours per day and 320mm in Korea, respectively. The plant height, leaf size and yield decreased, and days to flower was shorter 8-11 days in greek grown plants. The dehydration and curing process in Greece was linear and rapid due to the constant daily change of temperature and relative humidity with dry weather during curing season during curing season. But those process in Korea was variable and tardy due to rainy weather. Cured leaf ratio increased with upper stalk leaves in Greece but it was rather lower at top stalk position in Korea. The harvested green leaf cultivated in Greece showed higher contents of petroleum ether extract, total nitrogen and nicotine, but lower contents of reducing sugar and lower ratio of protein nitrogen to total nitrogen than in Korea.

급격한 광도 변화가 담배 잎에서 반사되는 Normalized Difference Vegetation Index에 미치는 영향 (Effect of a Sudden Increase in Light Intensity on Normalized Difference Vegetation Index (NDVI) Reflected from Leaves of Tobacco)

  • Normalized Difference Vegetation Index (NDVI) has played an important role in assessing green plant biomass through remote sensing on global scale since the early 1970s. The concept of NDVI is based on the fact that green plants show higher reflection in near-infrared region than in visible region of the electromagnetic spectrum. However, it is well known that the relocation of chloroplasts in plant leaf cells may dramatically change the optical properties of plant leaves. In this study I traced the changes in the reflectance and transmittance properties of Tobacco leaves at the wavelengths of 660 and 800 nm after a sudden increase in light intensity. The results showed that NDVI of leaves gradually decreased from 72.7% to 69.9% when exposed to a sudden increase in light intensity from 30 to $1,200{\mu}mol/m^2{\cdot}s$. This means that the error resulting from the physiological status of the plant should be accounted for a more precise understanding of ground truth corresponding to the data from the remotely acquired images.

Structural Characteristics of Two Wheat Histone H2A Genes Encoding Distinct Types of Variants and Functional Differences in Their Promoter Activity

  • To investigate the regulation of plant histone H2A gene expression, we isolated two H2A genes (TH254 and TH274) from wheat, which encode different types of variants. Both genes had an intron in the coding region. In the promoters, some characteristics sequences, such as Oct and Nona motifs, which are conserved among plant histone genes were also found, and they were located in a short region (about 120 bp) upstream from the putative TATA box. Analyses of promoter activity with H2A-GUS fusion genes in the transient system using tobacco protoplasts revealed novel types of positive cis-acting sequences in the TH254 promoter: a direct repeat of a 13-bp sequence (AGTTACATTATTG) and a stretch composed of an AT-rich sequence (ATATAGAAAATTAAAA) and a G-box (CACGTG). A quantitative S1 assay of the mRNA amounts from the TH254/GUS and TH274/GUG chimeric genes in stably transformed and cell cycle-synchronized tobacco cell lines showed that the promoters of both genes contained at least one cis-acting element responsible for S phase-specific expression. Histochemical analysis of transgenic tobacco plants carrying the chimeric genes showed that the promoters of the two H2A genes were both active in developing seedlings and flower organs but regulated in different manner.

An Active Site Arginine Residue in Tobacco Acetolactate Synthase

  • Acetolatate synthase(ALS) catalyzes the first common step in the biosynthesis of valine, leucine, isoleucine in plants and microorganisms. ALS is the target of several classes of herbicides, including the sulfonylureas, the imidazolinones, and the triazolopyrimidines. To elucidate the roles of arginine residues in tobacco ALS, chemical modification and site-directed mutagenesis were performed. Recombinant tobacco ALS was expressed in E. coli and purified to homogeneity. The ALS was inactivated by arginine specific reagents, phenylglyoxal and 2,3-butanedione. The rate of inactivation was a function of the concentration of modifier. The inactivation by butanedione was enhanced by borate, and the inactivation was reversible on removal of excess butanedione and borate. The substrate pyruvate and competitive inhibitors fluoropyruvate and phenylpyruvate protected the enzyme against inactivation by both modifiers. The mutation of well-conserved Arg198 of the ALS by Gln abolished the enzymatic activity as well as the binding affinity for cofactor FAD. However, the mutation of R198K did not affect significantly the binding of FAD to the enzyme. Taken together, the results imply that Arg198 is essential for the catalytic activity of the ALS and involved in the binding of FAD, and that the positive charge of the Arg is crucial for the interaction with negatively charged FAD.

The Use of a Tobacco mosaic virus-Based Expression Vector System in Chrysanthemum

  • Chrysanthemums (Chrysanthemum morifolium) are susceptible to tobacco mosaic virus (TMV). TMV-based expression vectors have been used in high-throughput experiments for production of foreign protein in plants and also expressing green fluorescent protein (GFP) to allow visualization of TMV movement. Here, we used TMV expressing the GFP to examine the infection of chrysanthemum by a TMV-based expression vector. Viral replication, movement and GFP expression by TMV-GFP were verified in upper leaves of chrysanthemums up to 73 days post inoculation (dpi) by RT-PCR. Neither wild-type TMV nor TMV-GFP induced symptoms. GFP fluorescence was seen in the larger veins of the inoculated leaf, in the stem above the inoculation site and in petioles of upper leaves, although there was no consistent detection of GFP fluorescence in the lamina of upper leaves under UV. Thus, a TMV-based expression vector can infect chrysanthemum and can be used for the in vivo study of gene functions.

형질전환 연초(Nicotiana tabacum L.)의 Mouse Adenosine Deaminase 유전자 발현 (Expression of Mouse Adenosine Deaminase Gene in Transgenic Tobacco (Nicotiana tabacum L.))

  • 동물유전자인 mouse adenosine deaminase (ADA)유전자가 안정적으로 연초 형질전환체에서 발현되었다. ADA cDNA 을 연초에서 강력히 발현시키기 위해서 35S/35S/AMV promoter을 부착시켰으며 식물세포에 도입을 위해서 binary vector인 pRD400을 이용하였다. 연초의 형질전환은 Tri- parental mating에 의해서 도입된 ADA 유전자 함유 binary vector와 disarmed Ti-plasmid을 함유하고 있는 Agrobacterium tmefacience MP9O을 사용하였다. 동시배양은 연초의 잎 disc을 이용해서 kanamycin이 첨가된 배지로부터 직접 shoots을 선발하여 형질전환체로 유도하였다. 형질전환 체에 ADA 유전자의 삽입여부는 PCR을 이용하였으며, 실험결과 ADA 유전자를 확인할 수 있었다. 또한 도입된 mouse ADA 유전자로부터 mRNA 및 단백질 합성여부를 각각 northern blot 및 immunoblot 분석한 결과 형질전환체 에서는 공히 확인되었다.

RPS5A Promoter-Driven Cas9 Produces Heritable Virus-Induced Genome Editing in Nicotiana attenuata

  • The virus-induced genome editing (VIGE) system aims to induce targeted mutations in seeds without requiring any tissue culture. Here, we show that tobacco rattle virus (TRV) harboring guide RNA (gRNA) edits germ cells in a wild tobacco, Nicotiana attenuata, that expresses Streptococcus pyogenes Cas9 (SpCas9). We first generated N. attenuata transgenic plants expressing SpCas9 under the control of 35S promoter and infected rosette leaves with TRV carrying gRNA. Gene-edited seeds were not found in the progeny of the infected N. attenuata. Next, the N. attenuata ribosomal protein S5 A (RPS5A) promoter fused to SpCas9 was employed to induce the heritable gene editing with TRV. The RPS5A promoter-driven SpCas9 successfully produced monoallelic mutations at three target genes in N. attenuata seeds with TRV-delivered guide RNA. These monoallelic mutations were found in 2%-6% seeds among M1 progenies. This editing method provides an alternative way to increase the heritable editing efficacy of VIGE.

Acibenzolar-S-Methyl(ASM)-Induced Resistance against Tobamoviruses Involves Induction of RNA-Dependent RNA Polymerase(RdRp) and Alternative Oxidase(AOX) Genes

  • Tobamoviruses are the major viral pathogens of tomato and bell pepper. The preliminary results showed that Acibenzolar-Smethyl(ASM; S-methylbenzo(1,2,3) thiadiazole-7-carbothiate) pre-treatment to tomato and tobacco plants reduces the concentration of Tomato mosaic tobamovirus(ToMV) and Tobacco mosaic tobamovirus(TMV) in tomato and bell pepper seedlings, respectively. Pre-treatment of the indicator plant(Nicotiana glutinosa) with the ASM followed by challenge inoculation with tobamoviruses produced a reduced number and size of local lesions(67 and 79% protection over control to TMV and ToMV inoculation, respectively). In order to understand the mechanism of resistance the gene expression profiles of antiviral genes was examined. RT-PCR products showed higher expression of two viral resistance genes viz., alternative oxidase(AOX) and RNA dependent RNA polymerase(RdRp) in the upper leaves of the ASM-treated tomato plants challenge inoculation with ToMV. Further, the viral concentration was also quantified in the upper leaves by reverse transcription PCR using specific primer for movement protein of ToMV, as well as ELISA by using antisera against tobamoviruses. The results provided additional evidence that ASM pre-treatment reduced the viral movement to upper leaves. The results suggest that expressions of viral resistance genes in the host are the key component in the resistance against ToMV in the inducer-treated tomato plants.
