Effects of 2,4-D and Kinetin on the Production of Saponin in Ginseng Tissue Culture (인삼근 유래 칼루스조직의 사포닌 함량에 미치는 2,4-D와 키네틴의 영향)

  • In the present study effects of 2,4-D and kinetin on the callus tissue growth of Korean ginseng(Panax ginseng C. A. Meyer), in relation to the synthesis of saponin were investigated. The saponin synthesis in the callus culture of ginseng root was enhanced by 2,4-D and kinetin. The total saponin content of callus grown on the optima growth conditions, that is, 5mg/l of 2,4-D and 2mg/l of kinetin, was about three times as high as that of the 6 year-old ginseng roots commercially used as herbs. The kinetin specifically increased the synthesis of protopanaxadiol group ginsenoside and decreased the syntehsis of protopanaxatriol gropu in callus cultures, while 2,4-D caused to an increase in the synthesis of protopanaxatriol group ginsenoside and decrease the synthesis of protopanaxadiol group.

  • Aloe in vitro culture was attempted to induce callus and regeneration ability from different explant sources onto MS medium with 0.5mg/l NAA plus 1.0mg/l BA. Anthers that no developed any callus and plant regeneration, while only four out of 274 filament explants induced calli at cut edge without regenerated plants. Twenty ovary explants regenerated four direct plantlets without via callus from the base of epidermal tissues. Regenerated plants on the root tip gave 2n=14 of chromosome numbers.

Production of Giant Hyssop Oil by Plant Tissue Culture (식물조직배양에 의한 배초향유의 생산)

  • Callus was derived from the seedlings of Agastache rugosa(Labiatae). The growth rate of callus and the production of essential oil were studied with the variation of culturing conditions. 2, 4-D 2ppm in the medium was more effective for the production of essential oil than NAA 2ppm. The growth rate of callus and the production of essential oil were inhibited by the illumination of the light. The essential oils from Agastache rugosa and the callus cultivated on the medium containing 2, 4-D 2 ppm and kinetin 0.2 ppm were analysed by TLC, gas chromatography and mass spectrometry. These two oils showed different compositions. The main component of the plant oil, methyl chavicol was not contained in the callus oil.

  • In order to investigate the biocompatibility of base metal alloys in dental prosthesis, the 3 types of Ni-Cr alloys and the 3 types of Co-Cr alloys were collected and each alloy was extracted in the culture medium. L-cells derived from the subcutaneous tissue of mouse and extracted medium were cultivated. The relative growth rate of L-cells in the tissue culture was calculated with N-R assay. The obtained results were as follows: 1. In the case of the Co-Cr alloys tested, its cytotoxicity proved weak. 2. In the case of the Ni-Cr alloys tested, there was no significant degree of cytotoxicity, especially 25% Victory was proved noncytotoxicity.

  • The in vitro construction of osteo-articular large implants combining biomaterials and cells is of great interest since these tissues have limited regeneration capability. But the development of such organoids is particularly challenging, especially in the later time of the culture, when the extracellular matrix has almost filled the initial porous network. The fluid flow needed to efficiently perfuse the sample can then not be achieved using only the hydraulic driving force. In this paper, we investigate the interest of using an electric field to promote mass transport through the scaffold at the late stage of the culture. Based on the resolution of the electrokinetics equations, this study provides an estimation of the necessary electric driving force to reach a sufficient oxygen perfusion through the sample, thus analyzing the feasibility of this concept. The possible consequences of such electric fields on cellular activities are then discussed.

Production of Essential Oils by Tissue Culture of Schizonepeta tenuifolia (형개의 조직배양에 의한 정유생산)

  • The callus was induced from the seedlings of Schizonepeta tenuifolia Brig. (Labiatae) and the effects of culturing conditions on growth rate and essential oil formation of the callus were experimented. It was found in the experiments, that the proper culturing temperature is $23^{\circ}C$ and the addition of biosynthetic precursors(leucine, mevalonic acid lactone) inhibits the growth of the callus. The growth rate of the callus and the amount of essential oils of the callus in the medium containing NAA were higher than the medium containing 2,4-D. The essential oils from the callus and the leaves of the cultivated Schizonepeta tenuifolia showed different GC pattern, but pulegone was found in both oils.

  • This study aims at detecting the three dimensional gripping points for the transplanting robot system to grip in the process of developing transplanting robot system, which is one of the automation systems for transplanting tissue culture. The stereo vision system equipped with two cameras has been used to detect the gripping points of the plant stem. The method for matching the plants of the image information which came from two cameras was to measure the total numbers of pixels, leaves, and the heights of the plants. The gripping points were detected near the roots after extracting the stem parts by the standard deviation of the X axis according to the Y axis. The performance test of the developed program showed that the detecting errors of the gripping points were 0∼1mm for X axis and 1∼2mm for Y & Z axis. The mean running time of the program was about 3 seconds.

  • Tissue culture systems to optimize regeneration plant species of Ocimum spp were evaluated as a method to micropropagate individual plants and to better study their biology in vitro. Ocimum species were also evaluated for the production of natural plant products during and following the regeneration process. The primary goal of this project was to enhance the regeneration efficiency of basil. Several factors were examined using different Ocimum species and commercial varieties. The effect of cytokinin combination, activated charcoal, gelling agents, and different carbon sources were investigated. Anthocyanin callus spots were produced only in four varieties among six tested. 'Sweet Dani' showed the best results on anthocyanin accumulation, while 'African beauty', 'Tree basil' and 'Methylcinnamate' produced only a few spots. Shoot regeneration was only achieved from 'Sweet Dani' explants. As the activated charcoal concentration increased, callus formation rate decreased respectively compare to the controls for all varieties. There was a decrease in callus growth with increasing concentration of agar and phytagel.

  • Permanent cell culture lines derived from human cancer tissue are important experimental models in the study of human cancer cell proliferation. The in vitro effects of C. militaris and its extracted fractions on the human breast cancer (SK-BR3), liver cancer (SK-Hep1, HepG2), kidney cancer (p15), lymphoma (Jurkat) were studied. F1 (CCCA, crude cordycepin containing adenosine), F2 (ethanol precipitation), F3 (ethanol soluble supernatant) and F4 (fraction of through SK-1B) significantly stimulated in vitro cytotoxic in human cancer cell lines.
