Effects of Supplements with Different Protein Contents on Nutritional Performance of Grazing Cattle During the Rainy Season

  • The objective of this study was to evaluate the effects of supplements with different crude protein (CP) contents on grazing cattle intake, digestibility, ruminal fermentation pattern, and nitrogen (N) metabolism characteristics during the rainy season. Five ruminal and abomasal cannulated Holstein${\times}$Zebu steers (296 kg body weight, BW) were used in a $5{\times}5$ Latin square design. The animals grazed five signal grass paddocks (0.34 ha). The five treatments evaluated were: Control (no supplement) and 1.0 g of supplement/kg BW with 0, 330, 660, and 1,000 g of CP/kg as-fed. The supplement was composed of starch, soybean meal, urea, and ammonium sulphate. There was a positive linear effect ($p{\leq}0.033$) of the CP content in the supplements on the organic matter (OM), CP, and digested OM intakes. The provision of supplements did not increase ($p{\geq}0.158$), on average, total and ruminal digestibilities of OM and CP. However, the increase in CP content in the supplements caused a positive linear effect ($p{\leq}0.018$) on ruminal digestibilities of OM and CP. Additionally, a quadratic effect of the CP contents of the supplements were observed (p = 0.041) for the ruminal digestibility of neutral detergent fiber corrected for ash and protein, with the highest estimate obtained with the CP content of 670 g/kg. The supply of supplements increased (p<0.001) the ruminal ammonia N concentration, which also changed linearly and positively (p<0.001) according to increase in CP content in the supplements. The apparent N balance and relative N balance (g/g N intake) were not, on average, changed ($p{\geq}0.164$) by the supplements supply. However, both showed a tendency of a linear increase ($p{\leq}0.099$) with increasing supplement CP content. The supplements increased (p = 0.007) microbial N production in the rumen, which also changed linearly and positively (p = 0.016) with increasing supplement CP content. In conclusion, protein supplementation in grazing cattle during the rainy season, while stimulating voluntary forage intake, results in higher efficiency of N utilization when compared to energy supplementation. This is a possible response to increased microbial protein synthesis in the rumen and improved N status in the animal body.

Effects of amino acid composition in pig diet on odorous compounds and microbial characteristics of swine excreta

  • Background: Major amino acids in pig diets are Lys, Met, Thr, and Trp, but little is known about the requirements for the other essential amino acids, especially on odorous compounds and microbial characteristics in feces of growing-finishing pigs. To this end, different levels of amino acid composition added to diets to investigate the effects of amino acid composition on microbial characteristics and odorous compounds concentration. Methods: A total eight (n = 8) barrows (Landrace ${\times}$ Yorkshire ${\times}$ Duroc) with an average bodyweight of $89.38{\pm}3.3kg$ were individually fed diets formulated by Korean Feeding Standards 2007 (old version) or 2012 (updated with ideal protein concept) in metabolism crates with two replication. After 15-day adaptation period, fresh faecal samples were collected directly from pigs every week for 4 weeks and analysed for total volatile fatty acids (VFA), phenols and indoles by using gas chromatography. The nitrogen was determined by Kjeldahl method. Bacterial communities were detected by using a 454 FLX titanium pyrosequencing system. Results: Level of VFA tended to be greater in 2012 than 2007 group. Among VFAs, 2012 group had greater (p < 0.05) level of short chain fatty acids (SCFA) than control.Concentration of odorous compounds in feces was also affected by amino acid composition in pig diet. Levels of ammonium and indoles tended to be higher in 2012 group when compared with 2007 group.Concentration of phenols, p-cresol, biochemical oxygen demand, and total Kjeldahl nitrogen, however, were lower (P < 0.05) in 2012 treatment group compare to 2007. The proportion of Firmicute phylum were decreased, while the Bacteriodetes phylum proportion increased and bacterial genera includingCoprococcus, Bacillus, and Bacteroides increased (p < 0.05) in 2012 compare to 2007 group. Conclusion: Results from our current study indicates that well balanced amino acid composition reduces odor by modulating the gut microbial community. Administration of pig diet formulated with the ideal protein concept may help improve gut fermentation as well as reduce the odor causing compounds in pig manure.

Characteristics of Anaerobic Acid Fermentation with Food waste leachate by Reactor Type of Retention Time for Landfill Site Injection (매립지 주입을 위한 음폐수 산발효 시 반응기 형태와 체류시간에 따른 특성)

  • In order to increase landfill gas (LFG) production with food waste leachate, this study was confirmed to be acidogenetic conditions for landfill site injection. Thereby, it was conducted for acidogenetic treatments to determine the decrease in viscosity and VFA production. After acidogenesis treatments, solubility of food waste leachate increased approximately 15%, and as a result, UASB and CSTR were similar by reactor type using the change of retention time. Based on the result of the change in viscosity by reactor type, efficiency of UASB showed approximately 11.38% of higher decrease in viscosity as $76.95{\pm}3.27%$ vs. CSTR. Also, VFA production showed the higher increase of 2.01 times (UASB) and 1.76 times (CSTR) respectively at the point of increasing retention time from 3 to 5 days. From the above results, efficiency of UASB in a reactor was relatively higher because large molecular lead to longer retention time than small molecular due to having screen effect in the fixed media.

Optimization of an Industrial Medium and Culture Conditions for Probiotic Weissella cibaria JW15 Biomass Using the Plackett-Burman Design and Response Surface Methodology

  • The objective of this study was to optimize industrial-grade media for improving the biomass production of Weissella cibaria JW15 (JW15) using a statistical approach. Eleven variables comprising three carbon sources (glucose, fructose, and sucrose), three nitrogen sources (protease peptone, yeast extract, and soy peptone), and five mineral sources (K2HPO4, potassium citrate, ⳑ-cysteine phosphate, MgSO4, and MnSO4) were screened by using the Plackett-Burman design. Consequently, glucose, sucrose, and soy peptone were used as significant variables in response surface methodology (RSM). The composition of the optimal medium (OM) was 22.35 g/l glucose, 15.57 g/l sucrose, and 10.05 g/l soy peptone, 2.0 g/l K2HPO4, 5.0 g/l sodium acetate, 0.1 g/l MgSO4·7H2O, 0.05 g/l MnSO4·H2O, and 1.0 g/l Tween 80. The OM significantly improved the biomass production of JW15 over an established commercial medium (MRS). After fermenting OM, the dry cell weight of JW15 was 4.89 g/l, which was comparable to the predicted value (4.77 g/l), and 1.67 times higher than that of the MRS medium (3.02 g/l). Correspondingly, JW15 showed a rapid and increased production of lactic and acetic acid in the OM. To perform a scale-up validation, batch fermentation was executed in a 5-l bioreactor at 37℃ with or without a pH control at 6.0 ± 0.1. The biomass production of JW15 significantly improved (1.98 times higher) under the pH control, and the cost of OM was reduced by two-thirds compared to that in the MRS medium. In conclusion, OM may be utilized for mass producing JW15 for industrial use.

Effects of Germinated and Fermented Unmarketable Soybean on Laying Performance and Egg Quality in Laying Hens (발아, 발효 처리한 비상품성대두 급여가 산란계의 생산성과 계란의 품질에 미치는 영할)

  • This study was conducted to investigate the effects of germinated and fermented unmarketable soybean (GFS) on laying performance and egg quality in laying hens. A total of two hundred laying hens were divided into 5 groups (5 treatment $\times$ 4 replication $\times$ 10 birds each) and fed with the experimental diets for 8 wk as follows: control, GFS free; T1, GFS 0.15%; T2, GFS 0.3%; T3, GFS 1%; T4, GFS 2%. The laying performance, egg quality, blood profiles, cecal microbial population, isoflavone content in egg yolk were investigated. There were no significant differences laying performance, relative liver and spleen weights, egg yolk color, eggshell color among groups. Eggshell strength in groups fed with diets containing GFS increased, but not significantly. Eggshell thickness significantly increased in the GFS-supplemented group. No significant differences were observed in the blood profiles and intestinal microflora after supplementation. The decrease of Haugh unit during storage was alleviated by feeding of GFS (p<0.05). The concentrations of malondialdehyde in groups fed with GFS were decreased as compared with control (p<0.05). Isoflavones in the egg yolk were detected in group fed with diet containing 2% GFS. These results showed that unmarketable GFS could be used as a favorable feed additive and feedstuff for production of quality enhanced and isoflavone fortified eggs.

Quality Properties of Commercial Chungkukjang in Korea (국내 시판 청국장의 품질 및 특성)

  • This study aimed to improve the quality with acomparison of quality characteristics by collecting Chungkukjang in the market, and to establish the foundation data for starter, which support to manufacture standardized Chungkukjang products. Firstly, Chungkukjang was collected from the industrial company and home-made production which located in the several regions of Korea. Moisture contents of collected Chungkukjang were ranged from 56.3 to 62.1% and salinity was 0.23 to 11.51%. Total nitrogen and amino nitrogen contents of Chungkukjang ranged from 5.82 to 8.76% and 173.71 to 2,700.66 mg%, respectively. Nitrogen degradation ratio, which means the proportion of amino nitrogen to total nitrogen, was 2.88~26.93%. Collected Chungkukjang AG and Z have predominant value in histamine and tyramine contents as 755.40 and 1913.51 mg/kg, respectively, and viable cells count in collected Chungkukjang, maximum was $9.6{\times}10^9CFU/g$. As a result, most products showed various properties in flavor and nutrition, and it is supposed to be caused on use of different soybean species and unstandardized fermentation process.

Effect of Feeding Yogurt Using Enterococcus faecium KHM-11 on the Growth in Piglet (Enterococcus faecium KHM-11를 이용한 요구르트 급여가 자돈의 성장에 미치는 영향)

  • A total of 70 colonies were isolated from the Korean human milk samples on the BCP plate count agar. These LAB isolates were subcultured in 10% reconstituted skim milk, and two strain thereof were finally selected for their highest acid productions. These strains were identified as Enterococcusfaecium based on 16S rDNA sequencing data, named as Enterococcus faecium KHM-11. Sugar utilization of E. faecium KHM-11 was investigated by API 50CH kit, and 19 different sugars including D-arabinose, L-arabinose, galactose, D-glucose, D-fructose, and D-mannose were utilized. For fermentation profiles, a yogurt inoculated by E. faecium KHM-11 after 15 hour, reached at pH 4.09, titratable acidity at 1.10% and average viable counts $1.30{\times}10^9\;CFU/mL$. Effects of the administration of yogurt 0.5% of piglet diet to piglets were investigated for growth rate, analysis of blood and incidence of diarrhea. 24 heads of piglets were divided into two groups: the experimental and the control of 12 animals each. The average growth rate in the yogurt-fed group was higher for 21.67%, compared with control (p<0.05). There were no differences in the concentrations of blood glucose, cholesterol, albumin and globulin between the two treatments. Incidence of diarrhea was no in pigs fed yogurt as compare to control.

Characteristics of Nutrient Removal and Membrane Fouling in a Membrane Bioreactor using Food Waste as an Additional Carbon Source (음식폐기물 응축수를 보조탄소원으로 이용하는 막 결합 생물 응조에서의 질소, 인 제거와 막 오염 특성)

  • Due to the low C/N ratio of domestic wastewater characteristic, addition of external carbon source for the effective N and P removal is necessary. High organic content of food waste can be used for the external carbon source in biological nutrient removal processes, The applicability of condensate of food waste (CFW), which is produced during the high-rate fermentation process, was examined in membrane bioreactor for the nutrient removal. Under the various operating conditions, nutrient removal efficiencies and membrane fouling characteristics were evaluated using synthetic wastewater. From nitrate utilization rate (NUR) test, denitrification rate was 0.19 g $NO_3-N/g$ VSS/day. With the addition of CFW increased, average removal efficiencies of T-N and T-P could be increased up to 64% and 41%, respectively. Also the optimal retention time was 3 hr/5 hr for anoxic/aerobic reactor. When applied to real sewage, membrane fouling resistance was increased up to 60%, which could be reduced from $10.4{\times}10^{12}m^{-1}$ to $5.9{\times}10^{12}m^{-1}$ with the control of influent suspended solid concentration. In summary, it was suggested that CFW could be used as an economical and effective carbon source for membrane assisted biological N and P removal.

Effect of Calcium Acetate on the Dough Fermentation and Quality Characteristics of Bread (Calcium Acetate의 첨가가 반죽의 발효와 빵의 품질특성에 미치는 영향)

  • This study was conducted to investigate the quality characteristic of bread added with 0, 2, 4 and 8% liquid calcium acetate(LCA-breads) to the dough prepared with brown rice, vinegar and ash of black snail replacing wheat flour. The pH of the dough was 5.38 in control and 5.39∼5.42 in the LCA-broads, which showed that the higher the content of LCA, the higher the pH. There was no big difference of the baking loss between control (10.73%) and LCA-breads(10.11∼10.81%). The loaf volume index was 7.12 in the control, 7.18 in the 2% LCA-bread, while the index was 6.22 in the 4% LCA-bread, 5.80 in the 8% LCA-bread. Calcium content was 16 mg% in the control, 30, 60 and 120 mg% in 2, 4 and 8% LCA-bread, respectively. The hardness, gumminess and brittleness of LCA-breads were higher, while springiness and cohesiveness were lower than that of the control. In the LCA-breads, $L^{*}$ values were lower and $b^{*}$ value was higher than those of the control, while there were no significant difference in af values. Increasing the LCA, air cells were bigger and irregular, and starch matrix was unstable. There were no significant difference in sour and bitter taste. The scores of stickiness of the 2∼4% LCA-breads were similar to that of the control and higher in the 8% LCA-bread. The scores of the flavor and overall taste in the LCA-breads were similar to the control, while the values were lower in the 4∼8% LCA-breads. The shelf-life evaluated by the number of moldy spots was not different between the control and 2% LCA-bread, while it was extended two times in 4% and three times in the 8% LCA-bread.ead.

  • This study was conducted to obtain practical information on the efficient drying of animal manure under the sunny dry condition. The effects of the height of manure pile (5, 10, 15, and 20cm), stirring times (0, 1, 2, and 4 times/day), the addition of dried manure (30%, w/w), and the type of drying bed on the removal of water from fresh pig manure were investigated in a plastic house. Pig manure was dried very well by lowering the height of manure pile and the drying efficiency was the highest at 10cm height. Water evaporation rate was the greatest at the twice stirring per day treatment. The addition of dried manure as bulking material enhanced the water evaporation rate of wet pig manure. The amounts of water removed for 19 days under the condition of 10cm height of manure pile and twice-stirring in spring, summer, autumn and winter were 75.6, 73.3, 54.6 and $32.6kg/1.2m^2$, respectively.
