Influence of Tether Length in the Response Behavior of Square Tension Leg Platform in Regular Waves

  • The tension leg platform (TLP) is a vertically moored structure with excess buoyancy. The TLP is regarded as moored structure in horizontal plan, while inherit stiffness of fixed platform in vertical plane. In this paper, a numerical study using modified Morison equation was carried out in the time domain to investigate the influence of nonlinearities due to hydrodynamic forces and the coupling effect between surge, sway, heave, roll, pitch and yaw degrees of freedom on the dynamic behavior of TLP's. The stiffness of the TLP was derived from a combination of hydrostatic restoring forces and restoring forces due to cables and the nonlinear equations of motion were solved utilizing Newmark's beta integration scheme. The effect of tethers length and wave characteristics such as wave period and wave height on the response of TLP's was evaluated. Only uni-directional waves in the surge direction was considered in the analysis. It was found that for short wave periods (i.e. 10 sec.), the surge response consisted of small amplitude oscillations about a displaced position that is significantly dependent on tether length, wave height; whereas for longer wave periods, the surge response showed high amplitude oscillations about that is significantly dependent on tether length.

Inhibitory Effect of Medicinal Plant Extract on Cell Toxicity and Interleukin-8 Production Induced by Helicobacter pylori (Helicobacter pylori에 의한 세포독성 및 Interleukin-8 생성에 미치는 생약혼합물의 영향)

  • The effects of Helicobacter pylori and medicinal plants extract (Leweifang) on the viability and interleukin(IL)-8 production of gastric epithelial cell were investigated. Cells viability was significantly decreased when they incubated with H. pylori or H. pylori toxin. Co-incubation with Leweifang increased H. pylori or H. pylori toxin-inhibited cell growth in a concentration-dependent manner. The production of IL-8 was greatly increased in H. pylori-infected KATO III gastric epithelial cells in a concentration- and time-dependent manner. The increased production of IL-8 was significantly inhibited by Leweifang $(1,000{\sim}5,000{\mu}g/ml)$. These results indicate that Leweifang has protective effect on H. pylori-inhibited cell growth and H. pylori-induced gastric mucosal cell inflammation by suppressing the production of inflammatory cytokine (IL-8) from gastric epithelial cells.

Anti-proliferative effect of Sam-nueng(Sparganii Rhizoma) extract on MCF-7 cells (삼릉(三稜) 추출물의 인간 유방암 세포 성장 억제 효과)

  • Purpose : This investigation was undertaken to evaluate the antiproliferation, atoptosis of Sam-nueng(Sparganii Rhizoma) extract using MCF-7 human breast cancer cells. Methods : MCF-7 cells were cultured in Dulbecco's modified Eagle's medium/F12(DMEM/F12) supplemented with 10 % fetal bovine serum(FBS; Gibco) and antibiotics. At varying times after extract treatment, cells were harvested with scraper and processed for analysis of protein expression, proliferation, cytotoxicity and apoptosis. Results : Our results show that the extract of Sam-nueng(Sparganii Rhizoma) strongly inhibits the proliferation of MCF-7 cells in a dose and time-dependent manner. Sulforhodamine B showed that the addition of Sam-nueng(Sparganii Rhizoma) extract reduced the viability of MCF-7 cells in a dose-dependent manner. Poly[ADP(ribose)] polymerase(PARP) which serves as a marker of cells undergoing apoptosis, a major substrate for caspase-3 was extensively cleaved in the Sam-nueng(Sparganii Rhizoma)-treated cells. Conclusion : So, we can conclude that Sam-nueng(Sparganii Rhizoma) can have an inhibitive effect on MCF-7 human breast cancer cells by variable mechanisms.

The mechanism of apoptosis induced by eugenol in human osteosarcoma cells

  • Eugenol is commonly used in dentistry for the sedation of toothache, pulpitis, and dental hyperalgesia. This study was performed to investigate the apoptotic effect of eugenol to human osteosarcoma (HOS) cells and the potential use of this compound in osteosarcoma cells. Eugenol showed the apoptotic effect in HOS cells in dose- and time-dependent manner. Fragmentation and condensation of DNA were showed by TUNEL assay, Hemacolor stain and Hoechst stain. In the DNA electrophoresis analysis, cells showed DNA degradation characteristic of apoptosis with a ladder pattern of DNA fragments. Apoptosis-related factors were analyzed by western blotting. Cells treated with eugenol showed caspase-3, PARP, lamin A and DFF-45 cleavage. Eugenol treatment induced caspase-3 cleavage and activation. Cleavages of PARP, DFF-45 and lamin A were accompanied with activation of caspase triggered by eugenol in HOS cells. Though this study needs more investigations, these results suggest that eugenol induce apoptosis via caspase dependent pathway in HOS cells and eugenol may constitute a potential antitumor compound against osteosarcoma cells.

Inhibition of Cellular Proliferation by CURCUMAE LONGAE Rhizoma Extracts on MCF-7 (강황(薑黃) 추출물이 유방암 세포주 MCF-7 증식 억제에 미치는 영향)

Inhibitory Effect of Curcumin on Nitric Oxide Production in Lipopolysaccharide-Stimulated RAW264.7 Cells and Its Suppressive Mechanism (대식세포주 RAW264.7 세포에서 Curcumin의 Lipopolysaccharide에 의한 Nitric Oxide 생성 억제 효과)

  • Curcumin, a polyphenolic antioxidant purified from turmeric, has been known to possess various biological activities such as anti-oxidative, anti-inflammatory and anti-cancer effects. In this study, we have explored anti-inflammatory effect of curcumin using Gram (-) bacterium-derived endotoxin (lipopolysaccharide: LPS) and macrophage cell line RAW264.7. Curcumin suppressed NO production in LPS-activated RAW264.7 cells in a dose-dependent manner, Curcumin also blocked the activation of $NF-{\kappa}B$ but not AP-1 according to luciferase assay. Furthermore, this compound suppressed the phosphorylation of a series of intracellular signaling components such as Src, JAK-2, Akt, IKK and $I{\kappa}B{\alpha}$ under LPS stimulation in a time dependent manner, Therefore, our data suggest that curcumin was able to protect the host from Gram(-) bacterial-infection-mediated inflammatory symptoms.

Investigation of decontamination characteristics of a serial multiple pool scrubber system for consequence mitigation of severe accidents

  • A pool scrubber is often used as a wet-type design to mitigate the consequence of a severe nuclear accident. While studies indicated higher decontamination performance of a deeper pool, utilizing a very tall pool can be problematic due to potential structural stability and water backflow issues. This study proposes, as an alternative to a single pool system, a pool scrubber system composed of serially connected multiple pools with lower heights. Since large fraction of aerosol removal takes place in the injection region, serially connected pool scrubber system is expected to enhance the overall decontamination capability of a pool scrubber system. To support the analysis of the proposed system's decontamination capability, a new computer model was developed in the study to describe the bubble size dependent effect on aerosol removal including the effect of pool residence time. The accuracy of the new model was examined against experimental data for its validation. The proposed scrubber system composed of serially connected multiple shorter pools is found to have much improved decontamination performance over the current single pool system design.

Pharmacological Profile of KR-31125, an Orally Active AT1 Receptor Antagonist (안지오텐신 수용체 리간드 KR-31125의 생체 내 활성에 관한 연구)

  • In vivo studies of KR-31125 (2-butyl-5-dimethoxymethyl-6-phenyl-7-methyl-3-[[2'-(1H-tetrazol-5-yl) biphenyl-4-yl]methyl]-3H-imidazo[4,5-b]pyridine) were performed in pithed rats, conscious angiotensin II (AII) challenged normotensive rats, renal hypertensive rats (RHRs) and furosemide-treated beagle dogs. KR-31125 induced a non-parallel right shift in the dose-pressor response curve to AII ($ID_{50}$: 0.095 mg/kg) with a dose-dependent reduction in the maximum responses in pithed rats. Compared to losartan, this antagonistic effect was about 18 times more potent, presenting competitive antagonism. Other agonists such as norepinephrine and vasopressin did not alter the responses induced by KR-31125. Orally administered KR-31125 had no agonistic effect and dose-dependently inhibited the pressor response to AII with a slightly weaker potency ($ID_{50}$: 0.25 and 0.47 mg/kg, respectively) in the AII-challenged normotensive rat model, but with a more rapid onset of action than losartan (time to $E_{max}$: 30 min for KR-31125 and 6 hr for losartan). KR-31125 produced a dose-dependent antihypertensive effect with a higher potency than losartan in RHRs, and these effects were confirmed in furosemide-treated dogs where they presented a dose-dependent and long-lasting (>8 hr) antihypertensive effect with a rapid onset of action (time to $E_{max}$: 2-4 hr), as well as a 20-fold greater potency than losartan. These results suggest that KR-31125 is a potent, orally active $AT_1$ receptor antagonist that can be applied to the development of new diagnostic and research tools as an added exploratory potential of $AT_1$ receptor antagonist.

  • The model for the combined refraction and diffraction of water waves, the so-called mild-slope equation, was first developed by Berkhoff (1972) and has been studied by many coastal engineers because the model is able to consider the combined effect of refraction and diffraction of water waves and eliminate the problem of ray crossing which may happen in the previously developed ray theory.   • This paper investigated, using a flexible approach, the effects of the psychological type of the decision maker and the advanced format of information presentation on decision maker performance in a computer-simulated production game. The current sutdy was guided by a model derived from a general model developed by Chervany, Dickson, and Kozar(1972). The experimental model had two dependent variables; total profit and decision making time. Three independent variables representing the psychological type of the decision maker, the report format, and decision aids were used in this study.
