The mechanisms underlying the actions of the antioxidants upon reactive oxygen species (ROS) generation by NADPH oxidase complex have remained uncertain. In this study, we investigated NADPH oxidase activity and the role of antioxidant enzymes upon the generation of ROS during hypoxic stress. ROS generation was found to increase in the mouse kidney under hypoxic stress in a time-dependent manner. Moreover, we found in MCT cells that hypoxia-induced hydrogen peroxide production was decreased by NAC pretreatment. We further analyzed HIF-$1{\alpha}$, PHD2 and VHL expression in the NAC-pretreated MCT cells and assessed the response of antioxidant enzymes at the transcriptional and translational levels. SOD3 and Prdx2 were significantly increased during hypoxia in the mouse kidney. We also confirmed in hypoxic $Prdx2^{-/-}$ and SOD3 transgenic mice that erythropoietin (EPO) is transcriptionally regulated by HIF-$1{\alpha}$. In addition, although EPO protein was found to be expressed in a HIF-$1{\alpha}$ independent manner in three mouse lines, its activity differed markedly between normal and $Prdx2^{-/-}$/SOD3 transgenic mice during hypoxic stress. In conclusion, our current results indicate that NADPH oxidase-mediated ROS generation is associated with hypoxic stress in the mouse kidney and that SOD3 and Prdx2 cooperate to regulate cellular redox reactions during hypoxia.

To elucidate the toxic effect of oxygen free radicals on cultured mouse spinal motor neurons damaged by hydrogen peroxide(H₂O₂)-induced neurotoxicity, we examined the neurotoxicity induced by oxygen radicals by NR assay when cultured spinal motor neurons were grown in the medium containing various concentrations of H₂O₂ for 6 hours. In addition, neuroprotective effects of herb extracts such Rhizoma Gastrodiae(RG), on H₂O₂-induced neurotoxicity in cultured spinal motor neurons were evaluated after cultured spinal motor neurons were preincubated with various concentrations of herb extract, RG for 2 hours before 50uM H₂O₂ for 6 hours. H₂O₂ decreased remarkably cell viability in dose-and time-dependent manner in these cultures, and also herb extract, RG increased cell viability of spinal motor neurons damaged by H₂O₂ in these cultures. From the above results, it is suggested that H₂O₂ was toxic in cultured spinal motor neurons derived from mouse, and RG was effective in blocking the neurotoxicity induced by oxidative stress in these cultures.
This study aimed to investigate the interactive effects of individual's cultural disposition (individualism, collectivism, horizontal disposition and vertical disposition) and cognitive style (analytic vs. holistic thinking) combination on differences in global precedence. For this purpose, the participants were divided into 4 different cultural disposition groups based on Singelis et al.'s(1995) INDCOL scale, and 2 cognitive style groups based on Choi et al.'s(2007) AHS, and required to respond selectively to global or local property of two types of compound stimuli (compound figure and compound letter). Reaction time and error rates for both global or local property were analyzed and compared as the dependent measurement. The results can be summarized as followings. First, faster responses were observed in compound figure condition than in compound letter condition, and to global property than to local property. Second, contrasting to individualism-cognitive style or horizontal disposition-cognitive style combinations, significant interaction effects both between collectivism and cognitive style, and between vertical disposition and cognitive style on global/local processing were found. This result indicated that combinations of collectivism-holistic thinking and vertical disposition-holistic thinking can induced larger global precedence effect than other combinations of cultural disposition and cognitive style.
Allergic contact dermatitis (ACD) is an inflammatory skin disease and regarded as a prototype of T-cell mediated delayed-type hypersensitivity reactions. Aroma Oil Complex (AOC) is composed of lavender true oil, chamomile roman oil and tea tree oil. This study was performed to assess the effects of AOC in a canine model of ACD. ACD was induced on the back of dogs induced by sensitization and repeated application by 2,4-dinitro-1-chlorobenzene (DNCB). Topical treatment of AOC was applied once a day for 8 days and skin biophysical parameters including transepidermal water loss (TEWL), skin hydration, skin thickness and erythema index, were measured every two days during experimental periods. Histopathology and immunohistochemistry were performed to evaluate the anti-inflammatory effect. In skin biophysical parameters, TEWL, skin hydration, skin thickness and erythema index were significantly increased, with a maximum increase appeared on day 2 (p<0.05). After the completion of AOC treatment, skin biophysical parameters were significantly reached those of baseline in a time-dependent manner (p<0.05). In histopathology, marked increases of epidermal thicknesses were induced after DNCB challenge with numerous inflammatory cell infiltrations and edematous changes, decreases of connective tissue occupied regions in dermis. In addition, marked increases of cytokine - tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interferon-${\gamma}$ (IFN-${\gamma}$)-immunoreactivities in the dermis and of apoptotic markers - caspase-3 and PARP-immunoreactivities in the epidermis were observed in DNCB control as compared with intact control, respectively (p<0.01). The decrease of infiltrated inflammatory cells and related decreases of pro-inflammatory cytokine immunoreactivities were observed in AOC treated skin (p<0.01). Based on these findings, AOC may have anti-inflammatory and alleviatory effects in the allergic contact dermatitis.

Probabilistic design methods can consider uncertainties of design variables and are widely used in the design of vertical breakwaters. The probabilistic design methods include a partial safety factor method, reliabilitybased design method, and performance-based design method. Especially the performance-based design method calculates the accumulated sliding distance during the lifetime of the breakwater or during a design storm. Recently a time-dependent performance-based design method has been developed based on the first-passage probability of individual sliding distance during a design storm. However, because the allowable criteria in the first-passage probability method are not established, the stability of structures cannot be quantitatively evaluated. In this study, the allowable first-passage probabilities for two limit states are proposed by calculating the first-passage probabilities for the cross-sections designed with various water depths and characteristics of extreme wave height distributions. The allowable first-passage probabilities are proposed as 5% and 1%, respectively, for the repairable limit state (allowable individual sliding distance of 0.03 m) and ultimate limit state (allowable individual sliding distance of 0.1 m). The proposed criteria are applied to the evaluation of the effect of wave-height increase due to climate change on the stability of the breakwater.
$Ce^{3+}$-doped yttrium aluminum gallium garnet (YAGG:$Ce^{3+}$), which is a green-emitting phosphor, was synthesized by solid state reaction using ${\alpha}$-phase or ${\gamma}$-phase of nano-sized $Al_2O_3$ as the Al source. The processing conditions and the chemical composition of phosphor for the maximum emission intensity were optimized on the basis of emission intensity under vacuum UV excitation. The optimum heating temperature for phosphor preparation was $1550^{\circ}C$. Photoluminescence properties of the synthesized phosphor were investigated in detail. From the excitation and emission spectra, it was confirmed that the YAGG:$Ce^{3+}$ phosphors effectively absorb the vacuum UV of 120-200 nm and emit green light positioned around 530 nm. The crystalline phase of the alumina nanoparticles affected the particle size and the luminescence property of the synthesized phosphors. Nano-sized ${\gamma}-Al_2O_3$ was more effective for the achievement of higher emission intensity than was nano-sized ${\alpha}-Al_2O_3$. This discrepancy is considered to be because the diffusion of $Al^{3+}$ into $Y_2O_3$ lattice is dependent on the crystalline phase of $Al_2O_3$, which affects the phase transformation of YAGG:$Ce^{3+}$ phosphors. The optimum chemical composition, having the maximum emission intensity, was $(Y_{2.98}Ce_{0.02})(Al_{2.8}Ga_{1.8})O_{11.4}$ prepared with ${\gamma}-Al_2O_3$. On the other hand, the decay time of the YAGG:$Ce^{3+}$ phosphors, irrespective of the crystalline phase of the nano-sized alumina source, was below 1 ms due to the allowed $5d{\rightarrow}4f$ transition of the $Ce^{3+}$ activator.
Circadian timing system plays a major role in a wide range of reproductive function. However it is plausible idea that other environmental and/or internal cue might be simultaneously participated in the optimal regulation of reproductive system. In the present study we extended the reverse feeding (RF) time regimen up to 8 weeks, then measured the general and reproductive indices of the animals. The animals of ad libitum feeding group (Control, CON) have free access to food for 4, 6 and 8 weeks, respectively. The day feeding animals (reverse feeding, RF group) have restricted access to food during daytime (09:00-18:00) for 4, 6 and 8 weeks, respectively. When the feeding schedules were over, key indices were measured. After 4 weeks and 8 weeks of feeding, body weights of animals were not significantly different. However, body weights of 6 weeks RF animals were significantly smaller than those of control animals (CON : RF = $333.46{\pm}12.71$ g : $289.91{\pm}8.31$ g, p<0.01). The blood glucose levels of 4 weeks RF animals were significantly decreased compared to the levels of control animals (CON : RF = $161.4{\pm}2.7$ mg/dL : $176.7{\pm}5$ mg/dL, p<0.01) while the levels of 6 weeks RF and 8 weeks RF animals were not different form those of control animals. Reproductive and non-reproductive tissue weights from 6 weeks RF group were significantly lowered than those from CON group (testis, CON : RF = $1.4714{\pm}0.0174$ g : $1.3724{\pm}0.0168$ g, p<0.001; epididymis, CON : RF = $0.3574{\pm}0.0059$ g : $0.3243{\pm}0.0068$ g, p<0.001; seminal vesicle, CON : RF = $0.1655{\pm}0.0068$ g : $0.1328{\pm}0.0054$ g, p<0.001; prostate, CON : RF = $0.3350{\pm}0.0231$ g : $0.2528{\pm}0.0143$ g, p<0.01). After 4 weeks and 8 weeks of reverse feeding, sperm counts in RF animals were markedly reduced than those in control animals[CON 4W : RF 4W = $121.17{\pm}9.96\;({\times}10^6)$ : $50.86{\pm}9\;({\times}10^6)$, p<0.001; CON 8W : RF 8W= $138.69{\pm}9.8\;({\times}10^6)$ : $108.94{\pm}4.22\;({\times}10^6)$, p<0.001]. Present study indicates that RF may induce an adaptable metabolic stress and cause impairment of androgen-dependent reproductive tissues. On-going longitudinal studies will allow a better understanding of the how does mealtime shift affect the reproductive function and exact nature of adaptation.

Non-spinning syndrome of Bombyx mori has been serious issue in sericulture industry near Kyungbuk area. This study was focused on the analysis of the mechanism and on screening candidate chemicals inducing the anti-metamorphosis of the silkworms. Rearing temperatures or initial body weight of the final instar larvae did not affect a normal larval to pupal metamorphosis of B. mori. However, pyriproxyfen (a juvenile hormone (JH) agonist) induced follicle patency significantly even at its 10$\^$-8/ M concentration and inhibited metamorphosis of B. mori in both developmental time and dose dependent manners. Pyriproxyfen induced JH esterase (JHE) activity and downregulated expression of JH binding protein of 5. mori. These results suggests that pyriproxyfen induced JHE activity as a JH agonist and that the elevated JHE activity degraded endogenous JH and resulted in JHBP gene expression. Based on the fact that the JH agonist induced follicle patency and inhibited metamorphosis of B. mori, follicle patency bioassay suggested that three commercial pesticides including simazine, molinate or alachlor were proved to give potent JH agonistic effect on B. mori. Further direct exposure experiments to these candidates are required to determine the chemicals responsible for the non-spinning syndrome of 8. mori.

The microstructure of ceramic composite has been found to be governed by the type and amount of the secondary phase, the sintering aid, and the sintering conditions such as sintering temperature, pressure and holing time. Moreover, tribological properties are strongly dependent on microsturcture of composite and operating conditions. In this study, silicon nitride with various amount of magnesia as a sintering aid were prepared and sintered by a hot pressing (HP) technique. Microstructure, mechanical properties (hardness, strength, and fracture toughness), and tribological properties in different environments of $Si_{3}N_{4}$ (in air, water, and paraffine oil) were investigated as a function of MgO content in $Si_{3}N_{4}$. As increasing the amount of MgO in $Si_{3}N_{4}$, the glassy phase in the grain boundaries enlarged the $\beta$-phase elongated grains, and also degraded the Hertzian contact damage resistance. Tribological behaviors in air was seemed to be determined by fracture toughness of $Si_{3}N_{4}$, and those in water and paraffin oil was seemed to be determined by hardness as well as strength. Since glassy grain-boundary phase (MgO) in $Si_{3}N_{4}$ expected to be reacted with water during sliding, such tribochemical reaction reduced wear. In paraffin oil under a higher applied load, the initial sliding dominated wear rate because of Hertzian contact damage.
Objective: The purpose of this study was to determine how exercise intensity affects muscle activity and kinematic variables during squat. Method: Fifteen trainers with >5 years of experience were recruited. For the electromyography (EMG) measurements, four surface electrodes were attached to both sides of the lower extremity to monitor the rectus femoris (RF) and biceps femoris. Three digital camcorders were used to obtain three-dimensional kinematics of the body. Each subject performed a squat in different conditions (40% one-repetition maximum [40%1RM], 60%1RM, and 80%1RM). For each trial being analyzed, three critical instants and two phases were identified from the video recording. For each dependent variable, one-way analysis of variance with repeated measures was used to determine whether there were significant differences among the three different conditions (p<.05). When a significant difference was found, post hoc analyses were performed using the contrast procedure. Results: The results showed that the average integrated EMG values of the RF were significantly greater in 80%1RM than in 40%1RM during the extension phase. The temporal parameter was significantly longer in 80%1RM than in 40%1RM and 60%1RM during the extension phase. The joint angle of the knee was significantly greater in 80%1RM than in 40%1RM at flexion. The range of motion of the knee was significantly less in 80%1RM than in 40%1RM and 60%1RM during the flexion phase and the extension phase. The angular velocity was significantly less in 80%1RM than in 40%1RM and 60%1RM during the extension phase. Conclusion: Generally, the increase of muscle strength decreases the pace of motion based on the relation between the strength and speed of muscle. In this study, we also found that the increase of exercise intensity may contribute to the increase of the muscle activity of the RF and the running time in the extension phase during squat motion. We observed that increased exercise intensity may hinder the regulation of the range of motion and joint angle. It is suitable to perform consistent movements while controlling the proper range of motion to maximize the benefit of resistance training.
