Expression of the Promoter for the Maltogenic Amylase Gene in Bacillus subtilis 168

  • An additional amylase, besides the typical $\alpha-amylase,$ was detected for the first time in the cytoplasm of B. subtilis SUH4-2, an isolate from Korean soil. The corresponding gene (bbmA) encoded a malto­genic amylase (MAase) and its sequence was almost identical to the yvdF gene of B. subtilis 168, whose function was unknown. Southern blot analysis using bbmA as the probe indicated that this gene was ubiquitous among various B. subtilis strains. In an effort to understand the physiological function of the bbmA gene in B. subtilis, the expression pattern of the gene was monitored by measuring the $\beta-galactosidase$ activity produced from the bbmA promoter fused to the amino terminus of the lacZ struc­tural gene, which was then integrated into the amyE locus on the B. subtilis 168 chromosome. The pro­moter was induced during the mid-log phase and fully expressed at the early stationary phase in defined media containing $\beta--cyclodextrin\;(\beta-CD),$ maltose, or starch. On the other hand, it was kept repressed in the presence of glucose, fructose, sucrose, or glycerol, suggesting that catabolite repression might be involved in the expression of the gene. Production of the $\beta-CD$ hydrolyzing activity was impaired by the spo0A mutation in B. subtilis 168, indicating the involvement of an additional regu­latory system exerting control on the promoter. Inactivation of yvdF resulted in a significant decrease of the $\beta-CD$ hydrolyzing activity, if not all. This result implied the presence of an additional enzyme(s) that is capable of hydrolyzing $\beta-CD$ in B. subtilis 168. Based on the results, MAase encoded by bbmA is likely to be involved in maltose and $\beta-CD$ utilization when other sugars, which are readily usable as an energy source, are not available during the stationary phase.

Activation of Fibrinolytic System during Open Heart Surgery (개심술중 섬유소융해계의 활성화에 관한 연구)

  • Hemorrhagic tendency observed in open heart surgery patients has been attributed, among other causes, to increased fibrinolytic activity during extracorporeal circulation. But the exact mechanism of enhanced fibrinolytic activity which occurs during extracorporeal circulation is still unknown. So, we studied and compared the changes of parameters of fibrinolytic and protein C system according to time obtained from the plasma of 31 adult open heart surgery patients[EGG group] and 10 adult general thoracic surgery patients[control group], in order to confirm the hypothesis that the activated protein C system might affect the fibrinolytic system during extracorporeal circulation. In ECC group, the nature of the enhanced fibrinolytic activity that evolved during extracorporeal circulation was characterized by significant increase in fibrin degradation products[P < 0.01] and significant decrease in plasminogen and alpha2-antiplasmin[P < 0.05, P < 0.01] in spite of adequate amount of heparin administration. These changes were most pronounced in the early phase of extracorporeal circulation and normalized after termination of extracorporeal circulation. The results of these observations were the same after volume correction with the value of hematocrit. The change of volume corrected protein C ratio during extracorporeal circulation revealed similar pattern to those of plasminogen and alpha2-antiplasmin [P < 0.01], but volume corrected ratio of free protein S showed significant increase after the commencement of extracorporeal circulation then decreased after extracorporeal circulation. Although the above mentioned changes occur similarly in both bubble type oxygenator-used and membrane oxygenator-used patients groups, but the degree of decrease was more severe in membrane oxygenator-used patients group [P < 0.01] and showed much slower recovery to reach to the preextracorporeal circulation level. These results confirm the hypothesis that the enhanced fibrinolysis during extracorporeal circulation might be caused by the activation of protein C system and the activation is possibly linked to the appearance of thrombin from contact activation of blood after wide exposure to the synthetic surfaces of extracorporeal circuit. A Novel High Performance Scan Architecture with Dmuxed Scan Flip-Flop (DSF) for Low Shift Power Scan Testing

  • Power dissipation during scan testing is becoming an important concern as design sizes and gate densities increase. The high switching activity of combinational circuits is an unnecessary operation in scan shift mode. In this paper, we present a novel architecture to reduce test power dissipation in combinational logic by blocking signal transitions at the logic inputs during scan shifting. We propose a unique architecture that uses dmuxed scan flip-flop (DSF) and transmission gate as an alternative to muxed scan flip-flop. The proposed method does not have problems with auto test pattern generation (ATPG) techniques such as test application time and computational complexity. Moreover, our elegant method improves performance degradation and large overhead in terms of area with blocking logic techniques. Experimental results on ITC99 benchmarks show that the proposed architecture can achieve an average improvement of 30.31% in switching activity compared to conventional scan methods. Additionally, the results of simulation with DSF indicate that the powerdelay product (PDP) and area overhead are improved by 28.9% and 15.6%, respectively, compared to existing blocking logic method.

  • We propose a new machine learning algorithm which analyzes daily activity patterns of users from multi-modal wearable sensor data. The proposed model learns and extracts activity patterns using input from wearable devices in real-time. Inspired by cue integration of human's property, we constructed gated multi-modal neural networks which integrate wearable sensor input data selectively by using gate modules. For the experiments, sensory data were collected by using multiple wearable devices in restaurant situations. As an experimental result, we first show that the proposed model performs well in terms of prediction accuracy. Then, the possibility to construct a knowledge schema automatically by analyzing the activation patterns in the middle layer of our proposed model is explained.

Cytotoxic and Apoptotic Activites of Echinomycin Derivative (Echinomycin-7) on P388 Murine Leukemia Cells

  • Echinomycin-7 is an echinomycin derivative, Smethylated sulfonium perchlorate of echinomycin. We studied the in vitro cytotoxicity and in vivo antitumor activity of echinomycin-7 against P388 leukemia cells and compared the results with echinomycin. With respect to the cytotoxic effects, echinomycin-7 had cell line-dependent $IC_{50}$ values while echinomycin had similar values to several tumor cell lines. Also, in vivo antitumor activities were observed in tumor-bearing mice treated with both agents, which showed that echinomycin-7 had a broad therapeutic dose range. We also observed the apoptosis on leukemia cells treated with echinomycin-7 which exihibited the ladder pattern of DNA on electrophoresis. In addition to apoptosis, echinomycin-7 arrested $G_1/S$ phases of the cell cycle at the same time. We then examined the signaling pathway of echinomycin-7-induced apoptosis and showed that ERK of the MAP kinase family was activated and translocated into the nucleus by echinomycin-7 stimulation. This study suggests that echinomycin-7 acts as an antitumor agent through in vitro cytotoxicity and has in vivo antitumor activity against leukemia cells, and that the echinomycin-7- induced apoptosis might involve signal transduction via MAP kinases.

An Analysis of Saengmaegsan's Ingredients and a Comparison Study on Anti-Oxidation Effects According to Kinds of Extract (생맥산(生脈散)과 구성 약물의 성분 분석 및 항산화(抗酸化) 효능 비교)

  • Objectives: The experiment was to estimate the value of Saengmaegsan as a therapeutic anti-aging agent with an analysis of ingredients and their bio-activating effects by enzymologic methods. Methods: We analyzed anti-aging effects of the ingredients of Saengmaegsan being Liriope Platyphylla, Schizandrae Chinensis, and Schizandrae Chinensis. Results: In Liriope Platyphylla and Schizandrae Chinensis, reducing sugar concentration was shown at the highest level. Of mineral contents, K+ and Na+ were the highest in Saengmaegsan and each ingredient also, and then Ca++ and Al. The amount of polyphenol showed was in order Schizandrae Chinensis > Saengmaegsan > Panax Ginseng > Liriope Platyphylla. The electron donating ability of Schizandrae Chinensis doubled that of Saengmaegsan and Panax Ginseng, at the same time that one per 1 ml in Saengmaegsan was significantly found the greatest level as compared to each single ingredient. SOD-like activity was high in Saengmaegsan and Schizandrae Chinensis. SOD-like activity of Saengmaegsan was higher than in single components. Nitric oxide inhibition in pH 1.2 was high in Saengmaegsan and Schizandrae Chinensis, and for per 1 ml it showed the same pattern as above. In pH 3.0 the result was not different from in pH 1.2. Xanthine oxidase inhibition was high in Schizandrae Chinensis and for per 1 ml showed the greatest activity in Liriope Platyphylla. Tyrosinase inhibition in Omija was the most noticeable, and for per 1 ml was Liriope Platyphylla more than any others. Conclusions: With this analysis of ingredients, it is proven that Saengmaegsan and each component fosters antioxidation. On the whole, the composite prescription Saengmaegsan was superior to each individual component.

  • This paper proposes a low power resource allocation and scheduling algorithm that minimized power consumption such as DSP circuit in high-level synthesis process. In this paper, we have used list-scheduling method for low power design in scheduling step. Also, it increase possibility to reuse input through resource sharing when assign resource. After scheduling, the resources allocation uses the power function in consideration of the result of calculating average hamming distances and switching activity between two input. First, it obtain switching activity about input value after calculate average hamming distances between two operator and find power value make use of bit pattern of the input value. Resource allocation process assign operator to minimize average hamming distance and power dissipation on all occasions which is allocated at each control step according to increase control step. As comparing the existed method, the execution time becomes fast according to number of operator and be most numberous control step. And in case of power that consume, there is decrease effect from 6% to 8% to be small.

Influence of Boreal Summer Intraseasonal Oscillation on the 2016 Heat Wave over Korea (한반도 2016년 폭염에 여름철 계절안진동이 미친 영향)

  • Severe and long-lasting heat waves over Korea and many regions in the Northern Hemisphere (NH) during the 2016 summer, have been attributed to global warming and atmospheric teleconnection coupled with tropical convective activities. Yet, what controls subseasonsal time scale of heat wave has not been well addressed. Here we show a critical role of two dominant boreal summer intraseasonal oscillation (BSISO) modes, denominated as BSISO1 and BSISO2, on modulating temporal structure of heat waves in the midst of similar climate background. The 2016 summer was characterized by La Nina development following decay of strong 2015/2016 El Nino. The NH circumglobal teleconnection pattern (CGT) and associated high temperature anomalies and heat waves were largely driven by convective activity over northwest India and Pakistan during summer associated with La Nina development. However, the heat wave event in Korea from late July to late August was accompanied by the phase 7~8 of 30~60-day BSISO1 characterized by convective activity over the South China Sea and Western North Pacific and anticyclonic circulation (AC) anomaly over East Asia. Although the 2010 summer had very similar climate anomalies as the 2016 summer with La Nina development and CGT, short-lasting but frequent heat waves were occurred during August associated with the phase 1~2 of 10~30-day BSISO2 characterized by convective activity over the Philippine and South China Sea and AC anomaly over East Asia. This study has an implication on importance of BSISO for better understanding mechanism and temporal structure of heat waves in Korea.

  • In the era of digital media, one of the most distinctive characteristics is the activity what audience play. This characteristic can be particularly demonstrated in the case of PPV(Pay-Per-View) use. Since this service is related with payment by audience according to each behaviour of their own media use. In doing so, this paper explores the characteristics and behavioral pattern in PPV use. In particular, the study tries to predict from the result that which trait of characteristic can explain the adoption of PPV. The analytical result shows that PPV users are much more active in gathering information as well as achieving the diversity in terms of program choice compared to non PPV users. At the same time, PPV users are prone to be unsatisfied against the other media and naturally willing to adopt the new media. PPV users are much more keen to use toward age related programme classification system in order to protect their children from inappropriate programmes. The variable of participational behaviour is the most relevant variable to predict the degree of PPV use, however, it fails to predict to distinguish the PPV users group from non PPV users group. It maybe given as a conclusion that PPV users are much more active in not only choosing their own program they like to watch but also planning ahead for maximising their pleasure from viewing.

Principles for Government Involvement in Urban Logistics Planning (도시물류정비를 위한 공공부문의 역할에 관한 연구)

  • The pattern of urban transport activity has two major components: the movement of people and the movement of goods. Roads in urban areas carry large number of trucks laden with goods that support urban economy and lifestyle. Therefore urban goods movement plays a vital role, but at the same time, urban goods movement are an important source of congestion, and a major contributor to the adverse environmental and social impacts of urban transport. In addition, the demand for goods movement is growing at a faster rate than personal travel in many cities, due to changes in industry logistics and consumer purchasing patterns. As a result, the significance of urban freight activity is increasing in terms of both its role in urban economy and its adverse impacts on urban amenity. This study is focused on the role and involvement scope of metropolitan government in urban logistics aimed at increasing its efficiency and reducing its negative impacts. The major suggestions of this study are (a) development of a measure of urban goods movement activity, (b) evaluation of data requirements and availability, (c) dialogue between the public sector and private sector, and (d) the role of metropolitan government.
