Effects of Static Magnetic Fields on Early Seed Germination and Radish Sprouts Growth (정적 자기장이 무순(radish sprouts)의 초기 발아와 성장에 미치는 영향)

  • In order to prepare for weakening of the productivity of agriculture due to serious climate change, the researches on promoting the growth of plant are required. Although the method of using magnetic field for improving the growth of plant was introduced, the effective method of imposing the field on the plant have yet to be studied thoroughly in the literatures. In this paper, we investigated the effects of static magnetic field on the growth and the early seed germination of radish sprouts according to the strength, direction of excitation and the expose time of the magnetic field, and present the effective method of imposing magnetic fields. From the measurements, it was found that the radish sprouts exposed in the field shows a rapid germination of about 3~4 days than those which had no field, and in order to the effective growth, the strength of the magnetic field should be properly selected, and the excitation direction of magnetic field has little effect on the radish sprouts growth.

Influence of Rainfall During the Ripening Stage on Pre-Harvest Sprouting, Seed Quality, and Longevity of Rice (Oryza sativa L.)

  • The influence of rainfall during the ripening stage on pre-harvest sprouting, seed viability, and seed quality was investigated in two Korean rice cultivars, Shindongjin and Hopum. When the rainfall was artificially treated in a greenhouse, HP started to pre-harvest sprouting at three days of rainfall treatment (DRT), but Shindongjin did not show pre-harvest sprouting at 40 DAH treatment and just 0.3~0.8% at 50 DAH, which was much lower than 15.3~25.8% of Hopum in the same treatment. After harvest, the seed germination of Hopum decreased about 10~25% compared to non-treated seeds, but that of Shindongjin decreased much little rate than that of Hopum. The seed longevity tested by accelerated aging decreased with prolonged rainfall period in both cultivars, but the varietal difference was clear; Shindongjin could withstand longer accelerated aging than Hopum. Shindongjin maintained its germination (>50%) ability after 15 days of accelerated aging regardless of the rainfall treatment period and time, but Hopum dropped below 50% germination ability after only 5 days of accelerated aging. In conclusion, rainfall during the ripening stage induced not only pre-harvest sprouting, but also reduced seed quality and longevity during storage, which varied between two cultivars.

Varietal Differences and Drying Storage Effects for Some Treatment Conditions of EMS and MMS on Rice Seeds (수도종자에 EMS와 MMS의 처리조건에 따른 건조저장효과와 품종간차이)

  • In order to study the biolgiocal effects of dry back following EMS (Ethyl methane sulfonate) treatment on seeds of 'Dunghan Shali' that belongs to indica-type rice with various presoaking(24, 36, and 40 hrs respectivily) and treatment time(l, 2 and 3 hrs respectivily), this experiment was done. Seedling heights of wet seeds presoaked for 36 hrs was only greatly decreased with EMS treatment compared with other periods. In germination rate of wet seeds, all of these damage was not changed. For the drying back seeds, otherwise, seedling height and germination rate were greatly reduced depend upon the time of treatment and presoaking as well as storage effects after drying back. Reduction patterns, both seedling height and germination rate, between 1 week and 8 weeks after dry back were quite similar. The other experiment was carried out in order to study for the influences on the seedling growth as biological damage with EMS and MMS(Methyl methane sulfonate) treatments among different varieties. The indica-type rice varieties; Taichung Native No.1, Dunghan Shali and IR-8, were used as the experimental materials. From this trial some results were obtained as follows; 1. Both seedling growth and germination rate on all varieties, used, MMS, treatment showed more toxic effects than those of EMS treatment. 2. Seeding growth injury of rice seeds dried back was increased gradually, and then was approached the maximum at 6-8 days after storing. In IR-s variety, otherwise, growth damage was appeared a little. Germination rate of these were quite smilar, even though chemical used is different and storage period was prolonged. 3. It was showed clearly that varietal differences of chemical mutagene treatments were recognized.

Measurement of and Changes in L-carnitine Levels in Developing Cucumber Cotyledon (오이 떡잎의 발달 과정에서 carnitine의 검출과 변화)

  • Mobilization of storage lipids is critical for the germination of oil seeds, as they supply carbon and energy until photosynthesis commences in cotyledons. In this study, we determined the levels of plant carnitine and associated changes in these levels from seed germination to cotyledon senescence. We also examined changes in the content of unsaturated fatty acids throughout seedling development. Carnitine levels peaked on day 3 at 14.5 nM in cotyledons and decreased sharply to 7.2 nM on day 4. On development day 3 carnitine levels were maintained at around 3 nM until day 7. The unsaturated fatty acid content dropped by half at the same time as carnitine peaked (day-3), and storage lipids were almost depleted by day 5. Thereafter, carnitine was hardly detected until the second stage of cotyledon senescence, at which stage the carnitine content was 6.8 nM, similar to that on day 4 at the time of fatty acid depletion in the cotyledons. Unsaturated fatty acids levels remained constant in green cotyledons but slightly increased in the senescing cotyledons. The latter can be explained by intracellular breakdown of membrane lipids. This is the first such discovery in developing cotyledons and may offer clues regarding other roles of the acetyl unit transport system in plants. The expression of BOU was closely associated with carnitine metabolism during seed germination and cotyledon development. The results provide support for the possibility of carbon re-routing during the glyoxylate cycle in the supply of energy for early germination and development.

Propagation of asymbiotically germinated seedlings with liquid culture for endangered lady's slipper orchid (Cypripedium macranthos Sw.) (액체배양 방법을 이용한 멸종위기종 복주머니란 종자 무균발아 및 증식)

  • We had already reported the successful germination for green pods of purple lady's slipper orchid (Cypripedium macranthos Sw.). The green pod methods is to take immature seeds in green capsules, sterilize the capsule, and take out the sterile seeds. This method, however, needs very critical time of harvest. The critical time of seed harvest changes depending upon the species, condition of the specimen, and climatic influence, and the right time lies between 5 and 12 weeks after fertilization. In this study, the mature seeds were collected after 120-130 days with hand-polination of lady's slipper orchids. Mature seeds are usually dormant and it has to be overcome, either with hormone or storing the seeds near freezing for two or three months to break dormancy. The seeds were first surface sterilized with 70% ethanol and then transferred 1% NaOCl for 10-15 minutes, followed by rinses 3 times with sterilized distilled water. The cypripedium seeds consists of an embryo within a seed coat known as a testa. The testa is water repellent and the seed has a large air space between the embryo and testa so the seed tends to float on water. We had resolved the problems with vacuum pump to soak water into the testa before sterilization. The seeds were placed on liquid or agar solidified germination media. Cultures were incubated at $24{\pm}1^{\circ}C$ in dark. The seeds were germinated in 6-8 weeks in liquid suspension culture (germination percentage over 18%); however, the seeds on agar solidified media took more than 5 months to germinate and the germination percentage less than 5%. The most effective media for liquid culture was 1/4 strength Murashige and Skoog (MS) medium with 50 ml/l coconut water ($4brix^{\circ}$) at pH 5.8.

Establishment of Herbicide Screening Methods for Reed (Phragmites communis Trin.) Control - I. Propagation of Reed (갈대(Reed, Phragmites communis Trin.)의 방제를 위한 제초제 스크리닝방법의 확립 - I. 갈대의 육묘)

  • This experiment was conducted to find out an effective propagation method for reed(Phragmites communis Trin.), ensuring a continuous herbicide screening for reed control. Reed propagation methods were compared under a greenhouse condition using tour different materials; seeds, rhizomes, depressed stolons of P. japonica Steud., and stem cuttings. Although reed seeds were easy to harvest and store, their germination rate(${\leq}$5%) was very low and seedling growth from the seeds was slow. Rhizomes were difficult to harvest and their harvest time was limited from November to March. Furthermore, reed propagation using rhizomes had problems of a relatively low germination rate(46%), no uniformity in size and shape, individual differences at the early stage of growth, and difficulties in material storage. Rate of reed growth from rhizomes was higher in commercial soil mix(Boo Nong soil) than in sand or in sand+upland soil(1:1). Depressed stolons of P. japonica had a moderate germination rate(65%) and were relatively easy to harvest. However, their harvest time was limited only from August to September. Propagation method using stem cuttings had several advantages over the above methods using other materials. Reed plants could uniformly be propagated from the stem cuttings with a relatively high germination rate(75%). Stem cuttings of central nodes showed a higher germination rate compared to those of upper or lower nodes. Stem cuttings from the field should be used immediately after harvest, since their germination rate decreased rapidly when they were stored under a wet- or a dry-refrigerated condition. Furthermore, the germination of stem cuttings tended to decrease when they were collected from the field after August. This indicates that there is a limitation of harvest time for stem cuttings. However, a year-round propagation of reed using stem cuttings is possible if parent plants are grown in a greenhouse, and thus herbicide screening for reed control could continuously be performed.

An enhanced root system developmental responses under drought by inoculation of rhizobacteria (Streptomyces mutabilis) contributed to the improvement of dry matter production in rice

  • Drought limits rice production under upland condition. This study quantified the effect of rhizobacteria inoculation on rice root system developmental response to drought and its role in maintaining high soil water use, and dry matter production under drought using NSIC Rc192 (rainfed lowland rice variety). The source of inoculant was Streptomyces mutabilis, a recently isolated rhizobacteria containing plant growth promoting compounds such as ACC deaminase, indole-3-acetic acid and phosphatase (Cruz et al., 2014, 2015). In the first experiment, pre-germination inoculation of seeds with S. mutabilis significantly increased the shoot and root (radicle) length as well as root hair lengths, relative to the non-inoculated control. In the second experiment, rice plants inoculated with S. mutabilis and grown in rootbox with soil generally had greater total root length under drought regardless of the timing of inoculations, relative to the non-inoculated control. Consequently, improved root system development contributed to the increase in soil water uptake under drought and thus, dry matter production. Among inoculation treatments, one-time inoculation of S. mutabilis either at pre-germination or pre-drought stress at 14 days after sowing (DAS), had significantly greater shoot dry matter production than three-time inoculation at pre-germination, at thinning (3 DAS) and at pre-drought (14 DAS). This study demonstrated the effectiveness of rhizobacteria (S. mutabilis) containing growth promoting compounds for enhancing drought dehydration avoidance root traits and improving the growth of rice plants under drought condition.

Relationship between Seed Vigour and Electrolyte Leakage in Rice Seeds with Different Grain-filling Period

  • The conductivity test is a measure of electrolytes leakage from plant tissue. The shorter the maturation period after heading was the greater electrical conductivity (EC) of rice seed. The polymer-coated seed was not different in EC compared with non-coated seed. As soaking time of rice seed increased, EC increased gradually. The EC varied from 9.9 to 20.7$\mu$S $cm^{-1}g^{-l}$ for control plots and from 21.3 to 41.7$\mu$S $cm^{-1}g^{-l}$ for heat-killed seeds which were produced by autoclaving seeds at 121$^{\circ}C$ for 20 minutes. The germination speed (the rate of 5th day) of rice seed was 94% at control plot, 83% at low temperature and 20% at high temperature. Besides, germination percentage was 95% for the control, 92% for the low temperature treatment and 39% for the high temperature treatment. The EC was negatively correlated (r=-0.771$^{**}$) with germination percentage at low temperature. Water uptake in seeds of 30, 40, 50 days after heading (DAH) was greater than that of 20 DAH. Plant height of seedlings was 9.84 cm for the control but 4.32 cm for the high temperature treatment, and the tallest for polymer-coated seed. Dry weight of seedlings was 0.841 g for the control and 0.287 g at high temperature. Besides, the polymer-coated seed was heavier than non-coated seed. The number of roots was largest from 40 to 50 DAH and polymer-coated seed, but was decreased from 20 to 30 DAH. The length of roots was 20.52 cm at control plot and 19.89 cm polymer-coated seed but 8.68 cm for the low temperature treatment and 7.28 cm for the high temperature treatment.

  • This research was carried out to promote the seed germination of Styrax japonica, by several pretreatments and to examine its seedling growth and seed component variations according to time and pretreatment. The results are as follows ; 1. Styrax japonica seeds are usually germinated after more than one year cold stratification. However, when the seeds were stratificated for 5 months after dipped in the solution of 70% $H_2SO_4$ for 30 minutes and also treated with $GA_3$(3000ppm), they showed 55% germination. This demonstrated that the seeds could be germinated in one year when pretreated. 2. The growth of one-year-old seedlings after pretreatment was the same as that of two-year-old seedlings when compared 160 days after seeding. 3. Before seeding, one-year-old, pretreated seeds of Styrax japonica showed increased moisture content but decreased crude fat, carbohydrate and exude ash, which were similar to the components of two-year-old seeds.

Kernel Characteristics and Germination Rate during . the Grain Filling in Super Sweet Corn (초당옥수수 등숙시기에 따른 종실특성 변화와 발아율)

  • The higher sugar and lower starch in super sweet corn may be due to modified endosperm genes sh, bt series, but its seeds have major limiting factors causing low germination and low seedling vigor, This study was conducted to determine what measurable kernel characteristics during the grain tiling period might be more useful as a guide to optimize harvest date for good seed quality in hybrid super sweet corn production. Artificial crossing in super sweet corn hybrid (Chodangok 1) was made on the same day, and ears were harvested from 18 days to 53 days after pollination at weekly intervals. Kernel weight, moisture content, hardness, endosperm rate, seedling height and storage nutrients such as sugars, protein, starch, and germination rate were measured for the kernels at each harvest. Super sweet corn hybrid, Chodangok 1 presented satisfactory germination rate above 70% when harvested on 39 to 46 days after pollination. Its storage nutrients at that time were 23.7-24.2% in starch content 5.5-5.9% in total sugars, 38.9-46.6% in kernel moisture, and 62.7-64.2% in endosperm rate. Germination rate was extremely high when harvested on 39 days after pollination. The black layer of Chodangok 1 could not be used as an indicator for seed harvest. These results suggested that optimum harvest date seems to be 39 days, and kernel moisture and starch content could be used as indications of kernel maturity in deciding when to harvest fer good seed quality in super sweet corn.
