Serological Detection of Borrelia burgdorferi among Horses in Korea

  • Lyme disease is a tick-borne zoonotic infectious disease caused by Borrelia burgdorferi. The present study assessed the infection status of B. burgdorferi among horses reared in Korea using ELISA and PCR. Between 2009 and 2013, blood samples were collected from 727 horses throughout Korea. Data for each animal including age, gender, breed, and region of sample collection were used for epidemiological analysis. Overall, 38 (5.2%; true prevalence: 5.5%) of 727 horses were seropositive by ELISA. There were statistically significant differences according to breed and region (P<0.001) whose differences might be attributed to the ecology of vector ticks and climate conditions. Using 2 nested PCR, none of the samples tested positive for B. burgdorferi. Thus, a positive ELISA result can indicate only that the tested horse was previously exposed to B. burgdorferi, with no certainty over the time of exposure. Since global warming is likely to increase the abundance of ticks in Korea, continuous monitoring of tick-borne diseases in Korean horses is needed.

Confirmed cases of severe fever with thrombocytopenia syndrome in companion cats with a history of tick exposure in the Republic of Korea

  • Severe fever with thrombocytopenia syndrome (SFTS) is a zoonotic disease, and its clinical information and prevalence are important. This study was conducted on 22 feline patients from the Republic of Korea (ROK), suspected to suffer from a tick-borne disease. Four cats were positive for SFTS, and genotypes B-1, B-3, D, and F were identified. Clinical symptoms, such as anorexia, jaundice, thrombocytopenia, leukopenia, and hyperbilirubinemia, were detected. This is the first report of SFTS virus genotypes B-1, D, and F from cats in the ROK. Moreover, our results suggest that jaundice may be an indicator of SFTS in cats.

Microbiome of Haemaphysalis longicornis Tick in Korea

  • Ticks can transmit pathogenic bacteria, protozoa, and viruses to humans and animals. In this study, we investigated the microbiomes of Haemaphysalis longicornis according to sex and life stages. The Shannon index was significantly higher for nymphs than adult ticks. Principal coordinates analysis showed that the microbiome composition of female adult and male adult ticks were different. Notably, Coxiella-like bacterium (AB001519), known as a tick symbiont, was found in all nymphs and female adult ticks, but only one out of 4 male adult ticks had Coxiella-like bacterium (AB001519). In addition, Rickettsia rickettsii, Coxiella burnetii, and Anaplasma bovis were detected in this study.

독일 셰퍼드 개에서 Ehrlichia와 Borrelia spp.의 감염 (Ehrlichia and Borrelia spp. Infection in German Shepherd Dogs in Korea)

  • 국내 독일 셰퍼드로부터 에를리히아증과 라임병에 대한 항체가 조사를 위하여 효소면역측정법을 시용하였으며, 중합효소연쇄반응을 이용하여 진드기매개성 병원체를 조사하였다. 2005년 10월부터 2006년 9월까지 국내 5개 지방으로부터 291마리 독일 셰퍼드를 선정하였다. 개 에를리히아 항체 조사를 위한 ELISA (Snap$^{(R)}$ 3Dx$^{(R)}$ test kit, IDEXX) 검사결과 전국적으로 7.56%(22마리)의 에를리히아 양성 항체 보균개와 1.72%(5마리)의 보렐리아 양성 항체 보균개로 조사되었다. 이 중 2마리에서는 두 가지 병원체 모두에 대한 양성 항체가 검출되었다. 에를리히아에 대한 양성항체를 지방별로 보면 경기도는 1.28%(78마리 중 1마리), 강원도는 12.64%(87마리 중 11마리), 충청도는 9.76%(41마리 중 4마리), 경상도는 8.93%(56마리 중 5마리), 전라도는 3.45%(56마리 중 5마리)로 조사되었다. Ehrlichia chaffeensis 유전자를 검출한 결과 전국 평균이 3.09%(291 마리 중 9마리), 강원도는 2.41%(291 마리 중 7마리) 그리고 충청도는 0.69%(291마리중 2마리)로 조사되었다. 중합효소연쇄반응으로 증폭된 유전자(SNU-EC3과 SNU-EC5)에 대한 유전자염기서열분석결과 한국(Haemaphysalis longicornis 진드기)과 중국 그리고 Arkansas 분리주인 E. chaffeensis와 매우 유사하였다. 이 연구의 결과 국내에서 사육되는 독일셰퍼드에서 에를리히아증의 병원체와 라임병에 대한 항체가 검출된 것으로 볼 때 임상 진단에 있어서 진드기매개전염병에 대한 중요성을 인식해야 할 것으로 판단된다. 따라서 혈소판감소증이나 백혈구감소증이 있는 경우 그리고 봄부터 가을까지 야외활동을 한 경력이 있는 환자에 대한 감별진단 항목에 반드시 포함시켜야 할 것을 제안한다.

대전 주택가 산책로 진드기의 인수공통전염병 병원체 감염실태 조사 (Isolation and identification of tick-borne pathogens in hard ticks collected in Daejeon)

  • In this study, a total of 9,449 hard ticks were collected once a month from April to October 2020 from a neighborhood park in Daejeon by flagging & dragging method and CO2 manned trap method. The collected ticks were classified according to the Yamagutsi search table using a stereoscopic microscope and molecular biological analysis of four pathogens (SFTSV, Anaplasma spp., Ehrlichia spp., Borrellia spp.). As a result of the study, Haemaphysalis longicornis were collected the most in all areas of the five boroughs at a rate of 82 to 96 percent, while adults were collected the most in May to July, nymphs were collected the most in April to June, and larvae from August to October at a rate of 78 percent to 98 percent. In pathogens, three cases of SFTSV were detected, showing a minimum infection rate (MIR) of 0.46%, while Anaplasma spp. and Ehrlichia spp. were detected one each, with 0.15% and Borrelia spp. with a minimum infection rate of 0.46%. The detected SFTSV showed 99.9% homogeneity with the KF781490 detected in Cheongwon-gun, Chungbuk Province, Anaplasma spp. showed 99.0% homogeneity with JN990105 detected in China, and Erhlichia spp. showed 98.9% genetic similarity with U96436 separated from the U.S. In this study, the distribution status and pathogen infection rate of the hard ticks in the Daejeon area are analyzed and provided as basic data for the prevention of the hard tick-borne infectious disease.

전라북도 동부지역 사슴에서 진드기매개성 병원체에 대한 감염 실태 조사 (A survey for tick-borne disease agents from farm deer in the eastern area of Jeonbuk)

  • Ticks cause economic losses to the deer industry by decreasing the growth and production of the farmed animals. The mediation of ticks affects humans and animals by causing contagious disease both directly and indirectly. Blood from farmed deer from the areas near Jangsu branch was collected for screening of infectious protozoa and rickettsial disease. Seventy deer blood samples were collected from 30 different deer farms located in Jinan, Jangsu and Muju. This blood samples were used for blood slide smear examination and hematological analysis. DNA from these samples was extracted and was used for PCR analysis for detection of gene fragments of Theileria spp, Babesia spp, Anaplasma spp and Ehrlichia spp. In the blood slide smear examination and PCR analysis all samples did not show presence of protozoal and rickettsial diseases. Eight blood samples showed anemia, 1 sample showed iron deficiency and 7 samples showed regenerative anemia. Results for PCR analysis showed 2 samples were positive for T orientalis. All DNA samples were negative for Babesia spp, Anaplasma spp, and Ehrlichia spp.

방목중인 한우에서 발생한 급성 타일레리아증 치료 (Treatment of acute bovine theileriosis in grazing Korean native cattle)

  • Bovine theileriosis caused by Theileria sergenti is a tick-borne hematoprotozoan disease that is characterized by chronic anemia and fever in cattle. In this study, results of microscopic examination and PCR detection confirmed 17 Korean native cattle with emaciation and fever as acute bovine theileriosis caused by T. sergenti. Buparvaquone was injected as treatment, but was proved to be an inappropriate measure according to our study. After 6 months of injection, clinical signs and hematological values were recovered, but T. sergenti was still identified in blood sample as a result of microscopic exam and PCR. These results suggest that continuous management is necessary to control bovine theileriosis. Therefore, findings of this study may provide significant guideline on the control of bovine theileriosis.

Molecular and serological surveillance of equine piroplasmosis in the Republic of Korea between 2016 and 2017

  • Equine piroplasmosis (EP) is caused by Babesia caballi and Theileria equi infection. We investigated antigen and antibody of EP in horses in the Republic of Korea during 2016-2017. Antigen and antibody of T. equi was detected 0.06% (1/1,650). Phylogenetic analysis of 18S rRNA revealed that the T. equi was highly homologous with the strains from China, Mongolia, and Spain. Two Theileria spp. were also detected and highly homologous with T. buffeli, T. luwenshuni, and T. orientalis.

국내 서식 한우에서 큐열 항체 양성율 조사 (Seroprevalence of Q-fever in Korean native cattle)

  • Q-fever is a vector-borne (Coxiella [C.] burnetii) zoonotic disease that is an increasing public health concern. To date, some research about Q-fever prevalence in dairy herds and human patients has been reported in Korea, but information about Korean native cattle is scarce. To measure the prevalence rates of C. burnetii in Korean native cattle, a total of 1,095 bovine serum samples collected during 2010~2013 were analyzed with an enzyme-linked immunosorbent assay. Sixty-eight heads of cattle were diagnosed as positive and while 19 heads were suspected (positive rate = 6.2%). Interestingly, Jeju province had a seropositivity rate six times greater than that of other provinces (18.9% vs. 3.2%). High seroprevalence might be caused by wide distribution of ticks in Jeju province compared to other regions. Based on these data, extensive monitoring of C. burnetii infection in cattle, tick distribution, and climate changes is required.
