This experimental Study was designed to investigate the effects of Acanthopanacis Cortex Extract(ACE) on the immunity, anti-cancer and obesity in mice. The results were as follows; ACE was significantly increased in the proliferation of thymocytes and splenocytes, and NO production from peritoneal macrophages in normal mice. ACE was significantly increased in the proliferation of thymocytes and splenocytes, and NO production from peritoneal macrophages in L1210 cells transplanted mice. ACE was significantly decreased in the proliferation of L1210 cells in L1210 cells transplanted mice. ACE was significantly inhibited body weight and tumor weight in S-180 cells transplanted mice. ACE was significantly increased in the mean survival days in S-180 cells transplanted mice. ACE was significantly decreased in the body weight in rats fed high fat diet. ACE was significantly decreased in the serum total cholesterol level, free fatty acid level, total lipid level, phospholipid level in rats fed high fat diet. According to above results, the authors suggest that ACE is able to be used for the herb of physiological-action.
Acanthopanax sessiliflorus SEEM extracts(AS) have been used to treat patient with diseases including cancer in Oriental countries. Recently, AS was known to have anti-cancer and immuno-stimulating activites. For these reasons, we investigated the effects of AS following gamma-ray irradiation on cytotoxicity for solid tumor cell line (S-180) and immune-potentiating ability such as proliferation of thymocytes and splenocytes. Finally we also investigated tumor weight and survival rate in tumor bearing mice. In our results, Treatment with AS suppressed proliferation of solid tumor cells (S-180) effectively. Treatment with AS accelerated thymocyte and splenocyte proliferation in tumor bearing mice. In addition, Treatment with AS reduced tumor weight and prolonged life of tumor bearing mice. In conclusion, we demonstrate that AS following gamma-ray irradiation is useful to treat patients with cancer, and also demonstrate that AS have both direct cytotoxic ability for cancer cells and indirect immune-stimulating action for thymocytes and splenocytes.
Objectives : This study was designed to estimate the antioxidative and neuroprotective effects of Torreyae Semen hot water extracts (TS). Methods : Torreyae Semen was extracted by hot water for 2 hours with a temperature of 105 degrees. Polyphenols and total flavonoid were measured and LC-MS/MS was used to certificate anticipated antioxidative compounds. The antioxidant activities of TS were measured as scavenging effects of 1,1-Diphenyl-2-picrylhydrazyl (DPPH) and Nitrite Oxides (NO). Cell viability and proliferation rate was measured MTT assay. The toxicities to thymocytes and splenocytes were evaluated by the proliferation rate of primary cultured cells of 7 weeks, male Balb/c mice. The antioxidant activities of TS on C6 mouse glioma cells were measured by the analysis of total glutathione contents variation. The neuroprotective effects against oxidative stresses were measured by MTT assay. Results : Polyphenols of TS was $92.00{\pm}1.24{\mu}g/mg$, and total flavonoids was $0.36{\pm}0.14{\mu}g/mg$. TS includes gallocatechin, epigallocatechin, gallocatechin gallate and epigallocatechin gallate. TS included gallocatechin, epigallocatechin, gallocatechin gallate, epigallocatechin gallate. TS showed DPPH and NO scavenging effects as dose-dependent manner at the concentrations of $0-10mg/m{\ell}$. In MTT assay, TS shows no significant toxicity to C6 cells, primary cultured thymocytes and splenocytes of Balb/c mice. TS increased the level of total glutathiones. TS increased cell viabilities of C6 cells against oxidative stresses such as $H_2O_2$, sodium nitroprusside (SNP), Rotenone at the concentrations of $0-0.063mg/m{\ell}$. Conclusions : TS shows the antioxidant and neuroprotecitive effects in these experiments.

Thymic atrophy is a complication that results from exposure to many environmental stressors, disease treatments, and microbial challenges. Such acute stress-associated thymic loss can have a dramatic impact on the host's ability to replenish the necessary naïve T cell output to reconstitute the peripheral T cell numbers and repertoire to respond to new antigenic challenges. We have previously reported that treatment with the orexigenic hormone ghrelin results in an increase in the number and proliferation of thymocytes after dexamethasone challenge, suggesting a role for ghrelin in restraint stress-induced thymic involution and cell apoptosis and its potential use as a thymostimulatory agent. In an effort to understand how ghrelin suppresses thymic T cell apoptosis, we have examined the various signaling pathways induced by receptor-specific ghrelin stimulation using a restraint stress mouse model. In this model, stress-induced apoptosis in thymocytes was effectively blocked by ghrelin. Western blot analysis demonstrated that ghrelin prevents the cleavage of pro-apoptotic proteins such as Bim, Caspase-3, and PARP. In addition, ghrelin stimulation activates the Akt and Mitogen-activated protein kinases (MAPK) signaling pathways in a time/dose-dependent manner. Moreover, we also revealed the involvement of the FoxO3a pathway in the phosphorylation of Akt and ERK1/2. Together, these findings suggest that ghrelin inhibits apoptosis by modulating the stress-induced apoptotic signal pathway in the restraint-induced thymic apoptosis.
The purpose of this research was to investigate the effects of Bojunglkgi-tang water extract (BE) on the specific immune response in BALB/c mice. When BE (500mg/kg) was administerd p.o. once a day for 7 days to BALB/c mice, the cell viability of splenocytes was increased and DNA fragmentation of splenocytes was decreased. But, BE did not affect the cell viability and DNA fragmentation of thymocytes. Also, BE increased the population of Thy1/sup +/ cells and TH cells in splenocytes. In addition, BE increased the production of γ -interferon from splenocytes. These results suggest that BE enhances the specific immune response via activation of TH1 cells in splenocytes.
The purpose of this research was to investigate the effect of Yonggak-san (YGS) on the immune reaction and apoptosis of leukemia cells. Administration of YGS(500 mg/kg) enhanced proliferation of splenocytes, thymocytes and mesenteric lymph node cells, and also YGS accelerated subpopulation of splenic Band T, thymic T and mesenteric lymph node-T lymphocytes, especially significantly increased CD4+-TH cells in BALB/c mice. YGS accelerated phagocytic activity and production of nitric oxide in peritoneal macrophages. YGS induced apoptosis of transplanted-L1210 cells in vivo, increased apoptotic cell death of cultured-L1210 and/or Molt4 human leukemia cells, decreased of mitochondrial transmembrane potential of both cells in vitro. These results suggest that YGS have an immune-regulatory effect and anti-cancer property.
The purpose of this research was to investigate the effect of Eunkyo-San(EKS) on the immune system. Administration of EKS(500 mg/kg) enhanced viability of splenocytes and thymocytes in BALB/c mice, and also EKS increased of splenic B, T lymphocytes and thymic T lymphocytes, significantly increased CD4 positive TH cells. EKS markedly enhanced the production of -interferon in mice serum. EKS accelerated the production of nitric oxide in peritoneal macrophages. These results suggest that EKS have an immune-enhancing activity.
The purpose of this research was to investigate the immunoregulatory effect, apoptosis of L 1210 leukemia cells of Comus officinalis Sieb.et Zucc. The proliferation of cultured splenocytes and thymocytes were enhanced by the addition of SSY. Splenic, thymic and mesenteric lymph node-T lymphocytes, especially TH cells was significantly increased in SSY-administered (p.o. for 7 days) mice. SSY treatment induced the apoptosis of L1210 mouse leukemia cells. In addition, SSY accelerated the phagocytic activity and nitric oxide production in peritoneal macrophages. These results suggest that SSY have an immuno-regulatory property and anti-cancer effect.
The purpose of this research was to investigate the effects of Chungjokupye-tang(CJKPT) on the anti-carcinogenic action. The cell viability of mouse spienocytes and thymocytes were enhanced by the addition of CJKPT. CJKPT were increased of splenic and thymic T lymphocytes, such as T/sub H/ cells were markedly increased by the treatment of CJKPT in vivo. CJKPT treatment induced the apoptotic cell death of Jurkat and HL60 leukemia cells. CJKPT reduced mitochondrial transmembrane potential and increased the expression of ICE, c-myc and p53 gene in Molt-4cells dose dependant manner. These results suggest that CJKPT have an anti-carcinogenic action via immunoregulatory mechanism.
Effects of Panax ginseng were tested on morphine-and steroids-induced immunosuppression, focusing on mechanism and identification of active components. To investigate overall effects of morphine and ginseng total saponin (GTS) on immune system, body weight and lymphoid organ weight were measured. Morphine significantly reduced body weight, spleen/body weight, and thymus/body weight ratio. GTS, at 100mg/kg (oral), restored spleen/body weight ratio. Because morphine is known to increase corticosterone level, serum corticosterone level was measured by radioimmunoassay. Serum corticosterone was increased by morphine and it was restored to the control level by GTS 100mg/kg (oral). In vitro proliferation studies were also conducted to study the effects of ginseng on steroids-induced immunosuppression. While ginsenoside Rg$_1$ and ginseng alkaloids were effective on proliferation and dexamethasone-induced death of thymocytes, 50% ginseng ethanol extract and polysaccharides were effective on splenocytes. In vivo mprohine-induced apoptosis of thymus was partly protected by GTS.
