Isolation and Characterization of Pepsin-soluble Collagens from Bones, Skins, and Tendons in Duck Feet

  • The objectives of this study were conducted to characterize pepsin-soluble collagen (PSC) extracted from bones (PSC-B), skins (PSC-S), and tendons (PSC-T) of duck feet and to determine their thermal and structural properties, for better practical application of each part of duck feet as a novel source for collagen. PSC was extracted from each part of duck feet by using 0.5 M acetic acid containing 5% (w/w) pepsin. Electrophoretic patterns showed that the ratio between α1 and α2 chains, which are subunit polypeptides forming collagen triple helix, was approximately 1:1 in all PSCs of duck feet. PSC-B had slightly higher molecular weights for α1 and α2 chains than PSC-S and PSC-T. From the results of differential scanning calorimetry (DSC), higher onset (beginning point of melting) and peak temperatures (maximum point of curve) were found at PSC-B compared to PSC-S and PSC-T (p<0.05). Fourier transform infrared spectroscopy (FT-IR) presented that PSC-S and PSC-T had similar intermolecular structures and chemical bonds, whereas PSC-B exhibited slight difference in amide A region. Irregular dense sheet-like films linked by random-coiled filaments were observed similarly. Our findings indicate that PSCs of duck feet might be characterized similarly as a mixture of collagen type I and II and suggest that duck feet could be used for collagen extraction without deboning and/or separation processes.

Permeation Flux of Ester Compounds through Hydrophobic Membrane by Pervaporation (투과증발에 의한 Ester 성분의 소수성막의 투과플럭스)

  • The objective of this work was to investigate the performance of pervaporation process for recovery of ester compounds from model aqueous solutions and how the fluxes of esters and water were affected by changes in feed concentration and temperature. The flux of ethyl acetate (EA), propyl acetate (PA), ethyl propionate (EP), butyl acetate (BA), and ethyl butyrate (EB) increased with an increase in feed concentration from 0.15 wt% to 0.60 wt%, and increased with temperature change from $30^{\circ}C$ to $50^{\circ}C$. The flux of esters (EA, PA, EP, BA, and EB) was in order of (EA) < (PA, EP) < (BA, EB). This result meant that the flux strongly depended on affinity between esters and membrane surface; EA is the least hydrophobic because it has one hydrophobic function group ($-CH_2-$), (PA, EP) have two ($-CH_2-$), and (BA, EB) are the most hydrophobic because these have three ($-CH_2-$). As well as such an influence of hydrophobicity of ester molecules on ester flux, the influence of hydrophobicity of membrane surface on ester flux needs further investigation. With increase in temperature, water flux of aqueous EA, PA, EP, BA, and EB solution increased. However, water flux of aqueous ester solutions did not change appreciably with increase in concentration. This experimental results may be used as fundamental data for pervaporation (PV) to improve the aroma recovery process as an alternative to thermal evaporation and distillation processes.

Review of Site Characterization Methodology for Deep Geological Disposal of Radioactive Waste (방사성폐기물의 심층 처분을 위한 부지특성조사 방법론 해외 사례 연구)

  • In the process of site selection for a radioactive waste disposal, site characterization must be carried out to obtain input parameters to assess the safety and feasibility of deep geological repository. In this paper, methodologies of site characterization for radioactive waste disposal in Korea were suggested based on foreign cases of site characterization. The IAEA recommends that site characterization for radioactive waste disposal should be performed through stepwise processes, in which the site characterization period is divided into preliminary and detailed stages, in sequence. This methodology was followed by several foreign countries for their geological disposal programs. General properties related to geological environments were obtained at the preliminary site characterization stage; more detailed site characteristics were investigated during the detailed site characterization stage. The results of investigation of geology, hydro-geology, geochemistry, rock mechanics, solute transport and thermal properties at a site have to be combined and constructed in the form of a site descriptive model. Based on this site descriptive model, the site characteristics can be evaluated to assess suitability of site for radioactive waste disposal. According to foreign site characterization cases, 7 or 8 years are expected to be needed for site characterization; however, the time required may increase if the no proper national strategy is provided.

A 30 GHz Band Low Noise for Satellite Communications Payload using MMIC Circuits (MMIC 회로를 이용한 위성중계기용 30GHz대 저잡음증폭기 모듈 개발)

  • A 30GHz band low noise amplifier module, which has linear gain of 30dB and noise figure of 2.6dB, for 30GHz satellite communication transponder was developed by use of MMIC and thin film MIC technologies. Two kinds of MMIC circuits were used for the low noise amplifier module, the first one is ultra low noise MMIC circuit and the other is wideband and high gain MMIC circuit. The pHEMT technology with 0.15$mu extrm{m}$ of gate length was applied for MMIC fabrication. Thin film microstrip lines on alumina substrate were used to interconnect two MMIC chips, and the thick film bias circuit board were developed to provide the stabilized DC bias. The input interface of the low noise amplifier module was designed with waveguide type to receive the signal from antenna directly, and the output port was adopted with K-type coaxial connector for interface with the frequency converter module behind the low noise amplifier module. Space qualified manufacturing processes were applied to manufacture and assemble the low noise amplifier module, and space qualification level of environment tests including thermal and vibration test were performed for it. The developed low noise amplifier was measured to show 30dB of minimum gain, $\pm$0.3dB of gain flatness, and 2.6dB of maximum noise figure over the desired operating frequency range from 30 to 31 GHz.

Properties of Extracts from Extruded Root and White Ginseng at Different Conditions (압출성형 공정변수에 따른 건조수삼과 백삼 압출성형물의 침출속도 및 침출물 특성)

  • The comparison in release rate constant and properties of extracts from extruded raw ginseng and extruded white ginseng was conducted to apply extrusion process for manufacturing of released ginseng tea bag. Dry raw ginseng and white ginseng powder were extruded at 20∼30% moisture content and 200∼300 rpm by using an experimental twin-screw extruder. Browness and redness (both indicated the releasing of saponin and ginsenosides) were increased with the increase in the screw speed and the decrease of moisture content. Crude saponin and water solubility index (WSI) of both ginseng also share the same behaviour against the level of screw speed and moisture content, as well as browness and redness. The particle size effects of extruded raw ginseng at 20% and 28% moisture content on absorbance of released extract at 260 up to 560 nm, WSI, and water absorption index were determined. While particle size decreased from 800∼1000 nm to 200∼500 nm, absorbance and WSI are decreased. Absorbance and WSI shown increasing level while moisture content was decreased. In conclusion, the formation of pores by expansion and disruption of cell wall in extrusion cooking were obviously responsible to increase the amount of released extract of extruded ginseng and its WSI as well. The extrusion process turns out be the efficient process for manufacturing of commercial ginseng tea product than those of other thermal processes.

Status of Soil Remediation and Technology Development in Korea (국내 오염토양 복원 현황과 기술 동향)

  • Soil contamination in Korea has been accelerated every year. Because of their persistence and cumulative tendency in the environment, soil contaminants have potential long-term environmental and health concerns and it is estimated to cost enormous expense for clean-up. Korea government has legislated the law on conservation of soil environment in mid 1990s, and managed and treated hazardous wastes in contaminated sites as a remediation policy since then. Soil remediation technologies are classified into in-situ/ex-situ or biological/physico-chemical/thermal processes according to applied places or treatment methods, respectively. In Korea, clean-up of polluted sites has been mostly carried out at military areas, railroad-related sites and small-scale oil spilt sites. For these cases, in-situ remediation technologies such as soil vapor extraction (SVE) and bioventing were mainly used. In recent days, an environmental-friendly soil remediation emerged as a new concept - for example, a new soil remediation process using nanotechnology or molecular biological study and an integrated process which can overcome the limitation of individual process. To have better applicability of remediation technologies, comprehensive understandings about the pollutants and soil characteristics and the suitable techniques are required to be investigated. Above all, development of environmental technologies based on the sustainability accompanied by public attention can improve soil environment in Korea.

An Analysis of Dynamic Characteristics of RDX Combustion Using Rigorous Modeling (상세 모델링을 통한 RDX 연소 동특성 분석)

  • In the treatment of spent high energetic materials, the issues such as environmental pollution, safety as well as working capacity should be carefully considered and well examined. In this regard, incineration has been recommended as one of the most promising processes for the disposal of such explosives. Due to the fact that high energetic materials encompass various types and their different characteristics, the technology development dealing with various materials is not an easy task. In this study, rigorous modeling and dynamic simulation was carried out to predict dynamic physico-chemical phenomena for research department explosive (RDX). Plug flow reactor was employed to describe the incinerator with 263 elementary reactions and 43 chemical species. Simulation results showed that safe operations can be achieved mainly by controlling the reactor temperature. At 1,200 K, only thermal decomposition (combustion) occurred, whereas increasing temperature to 1,300 K, caused the reaction rates to increase drastically, which led to ignition. The temperature further increased to 3,000 K which was the maximum temperature recorded for the entire process. Case studies for different operating temperatures were also executed and it was concluded that the modeling approach and simulation results will serve as a basis for the effective design and operation of RDX incinerator.

Measurement of EMC/PCB Interfacial Adhesion Energy of Chip Package Considering Warpage (휨을 고려한 칩 패키지의 EMC/PCB 계면 접합 에너지 측정)

  • The adhesion reliability of the epoxy molding compound (EMC) and the printed circuit board (PCB) interface is critical to the quality and lifetime of the chip package since the EMC protects PCB from the external environment during the manufacturing, storage, and shipping processes. It is necessary to measure adhesion energy accurately to ensure product reliability by optimizing the manufacturing process during the development phase. This research deals with the measurement of EMC/PCB interfacial adhesion energy of chip package that has warpage induced by the coefficient of thermal expansion (CTE) mismatch. The double cantilever beam (DCB) test was conducted to measure adhesion energy, and the spring back force of specimens with warpage was compensated to calculate adhesion energy since the DCB test requires flat substrates. The result was verified by comparing the adhesion energy of flat chip packages come from the same manufacturing process.

Occupational Heat Stress Impacts on Health and Productivity in a Steel Industry in Southern India

  • Background: Workers laboring in steel industries in tropical settings with high ambient temperatures are subjected to thermally stressful environments that can create well-known risks of heat-related illnesses and limit workers' productivity. Methods: A cross-sectional study undertaken in a steel industry in a city nicknamed "Steel City" in Southern India assessed thermal stress by wet bulb globe temperature (WBGT) and level of dehydration from urine color and urine specific gravity. A structured questionnaire captured self-reported heat-related health symptoms of workers. Results: Some 90% WBGT measurements were higher than recommended threshold limit values ($27.2-41.7^{\circ}C$) for heavy and moderate workloads and radiational heat from processes were very high in blooming-mill/coke-oven ($67.6^{\circ}C$ globe temperature). Widespread heat-related health concerns were prevalent among workers, including excessive sweating, fatigue, and tiredness reported by 50% workers. Productivity loss was significantly reported high in workers with direct heat exposures compared to those with indirect heat exposures ($x^2=26.1258$, degrees of freedom = 1, p < 0.001). Change in urine color was 7.4 times higher among workers exposed to WBGTs above threshold limit values (TLVs). Conclusion: Preliminary evidence shows that high heat exposures and heavy workload adversely affect the workers' health and reduce their work capacities. Health and productivity risks in developing tropical country work settings can be further aggravated by the predicted temperature rise due to climate change, without appropriate interventions. Apart from industries enhancing welfare facilities and designing control interventions, further physiological studies with a seasonal approach and interventional studies are needed to strengthen evidence for developing comprehensive policies to protect workers employed in high heat industries.

A Study on Formation and Thermal Decomposition Kinetics of PU Elastomers by Dynamic DSC and TGA Analysis (Dynamic DSC 및 TGA 열분석을 이용한 PU Elastomer의 중합반응 및 열분해 반응 Kinetics에 관한 연구)

  • Reaction kinetics of polyurethane elastomers (PU) were studied using dynamic DSC and TGA for three PU samples of general purpose (Sample A), high temperature cross-likable CASE purpose with MOCA (Sample B), and RT cross-likable CASE purpose grade (Sample C). From DSC results, sample with MOCA(Sample B) showed lower shift of peak temperature, while showing broader thermograms than those of general purpose grade (Sample A). On the other hand, RT cross-linkable PU grade (Sample C) showed an interesting double mode reaction patterns, i.e., a lower temperature reaction at about $70\;^{\circ}C$, and a higher temperature reaction in the range of $140{\sim}170\;^{\circ}C$, indicating that it requires 2-step reaction process in order to complete the reaction. Once the cross-linking reaction completed, however, TGA results showed that all the samples would be considered to have similar chemical structures, showing similar decomposition processes. Sample C, especially, had showed decomposition properties of both Sample A and Sample B. Formation activation energies calculated from Kissinger method showed 10.39, 65.85, 36.52(Low $T_p$) and 18.21(High $T_p$) kcal/mol, while decomposition activation energies were 31.94, 30.84, 24.16 kcal/mol, respectively.
