Bioconversion of onion extract to improve the bioavailability of quercetin glycoconjugate (쿼세틴 복합체의 생물학적 이용성 향상을 위한 양파 추출물의 유산균 발효)

  • Bioconversion and fermentation of onion extract by lactic acid bacteria were carried out to enhance the bioavailability of quercetin through the increase of quercetin recovery and aglycone formation. Lactobacillus casei, L. plantarum, and Kluyveromyces lactis were selected as the optimum strains for bioconversion. The environmental conditions to maximize the conversion ratio between glycoconjugate and quercetin aglycone have been evaluated. The concentrations of quercetin after fermentation of onion slurry by K. lactis and L. casei increased to 260% and 318%, respectively; however, the quercetin concentrations decreased after 48 hours of fermentation. Additionally, the quercetin hexose concentration increased to almost 141%. Controlling the initial pH of the onion juice increased the lactic acid production by L. casei and L. plantarum by more than two-fold. Meanwhile, the concentration of quercetin hexose decreased rapidly with the increased production of aglycones. The scale-up experiments showed the same fermentation efficiency; however, thermal sterilization reduced the quercetin glycone concentrations drastically.

Preparation of Porous Anti-Insect Repellent Powder Using Spray Drying of Medicinal Herbal Extracts Anti-Insect Repellent Silica Sol (분무건조법을 이용한 한약추출물 해충기피 실리카 졸의 다공성 방충입자의 제조)

  • Anti-insect repellent silica sol from mixture with silica and anti-insect repellent solution extracted from medicinal herbs was prepared. The micron size porous sphere powder with anti-insect repellent solution was prepared by the spray drying method. The characteristic of anti-insect repellent powder using spray drying method was analyzed by FE-SEM, PSA, TGA with the concentration of anti-insect repellent sol (anti-insect repellent solution and silica) and conditions of spray drier. The average particle size of 4, 7 wt% and 10 wt% of anti-insect repellent sol concentration were 8.3, $9.5{\mu}m$ and $11.7{\mu}m$, respectively. The particle size is increasing with high concentration of anti-insect repellent sol. Other hands, particle size as the temperature of inlet nozzle and velocity of sol injection were nearly same at high velocity of gas injection. Also, Anti-insect repellent impregnation in porous sphere powder were confirmed by TGA methode and its thermal property was stable up to $200^{\circ}C$. We expect that anti-insect repellent powder is applied for plastic compound and process of film manufacture.

Effect of Microwave Vacuum Heating on Inactivation of Enzymes (마이크로파 진공가열방법이 효소의 불활성화에 미치는 영향)

  • Microwave vacuum heating method (2450 MHz) was used for a low intensity of heat treatments. High vacuum under the microwave heating could bring low temperature condition. Inactivation of ${\alpha}-amylase,\;{\beta}-amylase$, glucoamylase and peroxidase by microwave vacuum heating were investigated at 60-$80^{\circ}C$. It was compared with conventional heating. The heating condition of microwave vacuum heating was confirmed by the destruction of ascorbic acid. When thermal inactivations of the enzymes by microwave vacuum heating were determined, it was less effective than that of conventional method at the initial stage of heating. It was due to a lag time of microwave heating. However, the heating time for complete inactivation of the enzymes by microwave vacuum heating could be reduced comparing with that of conventional heating. Optimum conditions for inactivation of the enzymes could be obtained by microwave vacuum heating.

Characteristics of Crude Protein-bound Polysaccharide from Agaricus blasei Murill by Extraction and Precipitation Conditions and Its Antitumor Effect (추출 및 침전조건에 따른 아가리쿠스버섯에서 분리한 조단백다당류의 특성 및 암세포 증식억제 효과)

  • Efficient extraction method of crude protein-bound polysaccharide (CPBP) from Agaricus blasei Murill was established. CPBP yields by ultrasonic and hot water extractions were 13.0 and 7.8%, respectively. Pressure extraction for 3 hr gave the highest ${\beta}-glucan$ content; no significant difference was observed between 2 and 3 hr extraction. Four volumes added ethanol gave the highest yields of CPBP and ${\beta}-glucan$ contents at 10.89 and 35.97%, respectively. Decomposition temperature of CPBP was $240-365^{\circ}C$, showing relatively good thermal stability. In SRB (sulforhodamine B) assay, CPBP treatment at $1,000\;{\mu}g/mL$ for 72 hr inhibited proliferations to A549, MCF-7, and AGS cancer cells by 43.9, 21.4, and 32.5%, respectively.

Study on Efficient Carbonizing Conditions When Carbonizing Fish Offal (어류폐기물의 탄화처리시 효율적 탄화조건에 관한 연구)

  • Experiments on carbonization were conducted using fish offal generated from fish market for the purpose of resource recycling. Elemental composition of fish offal and effect of carbonation temperature on the overall yield were investigated. Carbon and hydrogen contents of fish offal were 51.1% and 7.6%, respectively in view of elemental composition. Particularly, nitrogen and sulfur contents were as high as 9.8% and 1.0%, respectively. These values suggests that odor problem of fish offal can be serious. Comparing elemental composition of fish offal with other waste materials, it is thought that carbon and hydrogen contents are considerably high. These implies that thermal disposal will be the best option for final disposal method of fish offal. As a results of carbonization experiments on Mackerel, Hairtail, Croaker and mixed sample of Mackerel, Hairtail and Croaker, carbonization patterns were quite similar irrespective of fish species. Carbonization yield was varied significantly depending on carbonization temperature at the carbonization time of 5 minutes and 10 minutes. When the carbonization time was maintained longer than 30 minutes, yield variation depending on time variation at each temperature was insignificant. Thus, it can be concluded that effect of carbonization time on overall yield was minor when the carbonization time was maintained longer than 30 minutes. Primary vaporization in carbonization conducted at the temperature of $400^{\circ}C$ was minor. Thus, difference of yield between temperature of $500^{\circ}C$ and $400^{\circ}C$ was appeared greatly. It can be concluded that yield difference depending on carbonization temperature can be neglected if the carbonizing temperature exceed $600^{\circ}C$ and carbonizing time exceed 10 minutes at the same time.

Remote field Eddy Current Technique Development for Gap Measurement of Neighboring Tubes of Nuclear Fuel Channel in Pressurized Heavy Water Reactor (중수로 핵연료채널과 인접관의 간격측정을 위한 원거리장 와전류검사 기술개발)

  • Liquid Injection Nozzle(LIN) tube and Calandria tube(CT) in pressurized Heavy Water Reactor (PHWR) are .ross-aligned horizontally. These neighboring tubes can contact each other due to the sag of the calandria tube resulting from the irradiation creep and thermal creep, and fuel load, etc. In order to judge the contact which might be the safety concern, the remote field eddy current (RFEC) technology is applied for the gap measurement in this paper. LIN can be detected by inserting the RFEC probe into pressure tube (PT) at the crossing point directly. To obtain the optimal conditions of the RFEC inspection, the sensitivity, penetration and noise signals are considered simultaneously. The optimal frequency and coil spacing are 1kHz and 200mm respectively. Possible noises during LIN signal acquisition are caused by lift-off, PT thickness variation, and gap variation between PT and CT. The simulated noise signals were investigated by the Volume Integral Method(VIM). Signal analysis on the voltage plane describes the amplitude and shape of LIN and possible defects at several frequencies. All the RFEC measurements in the laboratory were done in variance with the CT/LIN gap and showed the relationship between the LIN gap and the signal parameters by analyzing the voltage plane signals.

Design and Control of Ultra-precision Dual Stage with Air bearings and Voice coil motor for nm scanning system (나노 정밀도 스캐닝 용 공기베어링과 보이스 코일 모터의 초정밀 이중 스테이지 설계 및 제어)

  • In this paper, a decoupled dual servo (DDS) stage for ultra-precision scanning system with large working range is introduced. In general, dual servo systems consist of a fine stage for short range and a coarse stage for long range. The proposed DDS also consists of a $XY\theta$ fine stage for handling and carrying workpieces and one axis coarse stage. Its coarse stage consists of air bearing guide system and a coreless linear motor with force ripple. The fine has four voice coil motors(VCM) as its actuator. According to a VCM's nature, there are no mechanical connections between coils and magnetic circuits. Moreover, VCM doesn't have force ripples due to imperfections of commutation components of linear motor systems - currents and flux densities. However, due to the VCM's mechanical constraints the working range of the fine is about $25mm^2$. To break that hurdle, the coarse stage with linear motors is used to move the fine about 500mm. Because of the above reasons, the proposed DDS can achieve higher precision scanning than other stages with only one servo. With MATLAB's Sequential Quadratic Programming (SQP), the VCMs are optimally designed for the highest force under conditions and constraints such as thermal dissipations due to its coil, its size, and so on. And for their movements without any frictions, guide systems of the DDS are composed of air bearings. To get precisely their positions, a linear scale with 5nm resolution are used for the coarse stage's motion and three plane mirror laser interferometers with 5nm for the fine's $XY\theta$ motions. With them, on scanning the two stages have same trajectories. The control algorithm is named Parallel method. The embodied ultra-precision scanning system has sub 100nm following error and in-positioning stability.

  • The copolymerization of ethylene (E) and norbornene (N) was examined by using various metallocene catalysts and modified-MAO(MMAO) cocatalyst. For $C_2$-symmetry catalysts such as rac-Et(Ind)$_2$ZrC $l_2$, M $e_2$Si(Ind)$_2$ZrC $l_2$, M $e_2$Si(Cp)$_2$ZrC $l_2$ and Cs-symmetrical iPr(FluCp)ZrC $l_2$ as well as CGC and di-bridged zirconocene, the effects of catalyst structure and [N]/[E] feed ratio on catalyst activity, thermal property and [N] content of copolymer (COC) was investigated. For rac-Et(Ind)$_2$ZrC $l_2$ catalyst of a constant [N]/[E] feed ratio, the appropriate conditions of [Al]/[Zr] mole ratio, polymerization temperature and cocatalyst structure were found to be 3000, 4$0^{\circ}C$, MMAO cocatalyst, respectively. As [N]/[E] feed ratio increased, the incorporation of norbornene to copolymer increased while, the activity of catalyst decreased except for iPr(FluCp)ZrC $l_2$ With consideration of catalyst activity as well as N content, it was found that rac-Et(Ind)$_2$ZrC $l_2$/MMAO system exhibited relatively high activity and controllable $T_{g}$.   • In this thesis, both soap and enzymatic degumming method were adopted and the optimum degumming conditions were obtained. Difference between the two degumming methods in silk fiber state was investigated and analyzed on the basis of the results of physical testings, polarizing microscopy, scanning electron microscopy, viscosity measurement, (${\alpha}$$\varepsilon$) amino group contents measurement, birefringence measurement, amino acid analysis, thermal analysis, infrared spectroscopy and x-ray diffraction analysis. The results obtained were summarized as follows; Physical test results of the degummed silk fiber showed that the tenacity and the elongation of enzymatic degummed silk fiber were lower than those of soap degummed fiber. But SEM observation and amino acid analysis showed almost the same tendency in the two degumming methods. The viscosity of enzymatic degummed silk fiber was lower than that of soap degummed fiber, but (${\alpha}$$\varepsilon$) amino group contents was higher in the enzymatic degummed fiber. It can be suggested that the enzymatic degummed silk fibroin was more degraded than the soap degummed fibroin. The birefringence, endothermic temperature of DSC spectrum, IR crystallinity and X-ray lateral order factor of enzymatic degummed silk fiber were higher than those of soap degummed fiber. It seems that the enzymatic degummed silk fiber has the higher crystallinity than that of soap degummed one according to the above results. However, it can be inferred that these differences between soap and enzymatic degummed fiber would be lessened if pretreatment and aftertreatment were included in the enzymatic degumming process.

Studies on the cellulase properties of Aspergillus clavatus from the Cellulose-Cultural Properties (섬유질 문화재로부터 분리된 Aspergillus clavatus의 섬유소분해호소에 관한 연구)

  • Cellulolytic mocrooganisms were isolated from the cellulose-cultural properties. Among them, Aspergillus clavatus with the highest cellulase activity was identified by the morphological characteristics and it's enzyme activities were compared on the various cultural conditions. It was found that the induction of carboxymethylcellulase(CMCase), avicelase and salicinase in CMC liquid media showed the highest enzyme acitivity on five day's cultivation at $30^{\circ}C$ and thereafter their activities were gradually decreased with time. After crude extracellular enzymes precipitated with the 70% saturated ammonium sulfate solution were dialyzed with 20 mM acetate buffer pH 6.0, each crude enzyme was examined. The optimal activities of CMCase and avicelase were both found to be at $50^{\circ}C$ and pH 6.0. Their thermal stability was appeared at the $50^{\circ}C$. CMCase and avicelase had the maxima activities with 1.5% and 2.2% substrate concentration, respectively. The concentration of 5 mM $Mg^{++}$ or $Ca^{++}$ was found to have a maximum cellulase activity and its activity was inhibited with more than 5 mM $Cu^{++}$ and $Zn^{++}$ concentration. Cellulase activity was also inhibited sensitively by the inhibitors such as 2-mercaptoethanol, iodine and sodium azide.
