Characterization of Cucumver mosaic virus Isolated from Hydrangea macrophylla for. otaksa (Sieb. et Zucc) Wils. (수국에서 분리한 Cucumber mosaic virus의 특성)

  • An isolate of Cucumber mosaic cucumovirus(CMV) was isolated from Hydrangea macrophylla for. otaksa(Sieb. et Zucc. ) Wils. showing mosaic symptoms, and designated as Hm-CMV. Hm-CMV was characterized by the tests of host range, physical properties, serological properties, RNA and coat protein compositions, and reverse transcription and polymerase chain reaction (RT-PCR) analysis. Twelve species in 4 families were used in the host range test of Hm-CMV and could be differentiated from Y-CMV used as a control CMV by the ringspot and line pattern on inoculated leaves of several tobacco plants. Thevirus produced local lesions on inoculated leaves of Chenopodium amarticolor, C. quinoa and Vigna unguiculata. The physical properties of the virus were as follows; thermal inactivation point(TIP) was 60$\^{C}$, dilution end point (DEP) was 10$\^$-3/, and longevity in vitro (LIP) was 3∼4 days. Hm-CMV was serologically identical to Y-CMV. SDS-polyaciylamide gel electrophoresis(SDS-PAGE) showed one major protein band of about 28 kDa. In RNA or dsRNA analysis, Hm-CMV consisted of four RNA or dsRNA species, but satellite RNA was not detected. In RT-PCR using CMV-common primer and CMV subgroup I-specific primer, bothe amplified expected size of about 490 bp and 200 bp DNA fragments from Hm-CMV, respectively. Restriction enzyme analysis of the 490 bp RT-PCR products using EcoR I and Msp I showed that Hm-CMV belonged to CMV subgroup I. However, Hm-CMV could be differentiated from other CMV subgroup I isolates by RNA fingerprinting by arbitrarily primed polymerase chain reaction (RAP-PCR).

  • Super-critical $CO_2$ ($S-CO_2$) Brayton cycle has been considered to replace the current steam Rankine cycle in Sodium-cooled Fast Reactor (SFR) in order to improve the inherent safety and thermal efficiency. Several austenitic alloys are considered as the structural materials for high temperature $S-CO_2$ environment.. Microstructural change after long-term exposure to high temperature $S-CO_2$ environment could affect to the mechanical properties. In this study, candidate materials (austenitic stainless steels and Alloy 800HT) were exposed to $S-CO_2$ to assess oxidation resistance and the change in tensile properties. Loss of ductility was observed for some austenitic stainless steels even after 250 h exposure. The contribution of $S-CO_2$ environment on such changes was analyzed based on the characterization of the surface oxide and carburization of the materials in which 316H and 800H showed different oxidation behaviors.

Characterization of Porcine Tissue Perforation Using High-Power Near-Infrared Laser at 808-nm Wavelength (808 nm 파장의 고출력 근적외선 레이저 조사 시 돼지 조직의 천공 특성 연구)

  • A fundamental study on laser-tissue interaction was conducted with the aim of developing a therapeutic medical device that can remove lesions on the intestinal wall by irradiating a high-power 808-nm infrared laser light incorporated in an endoscopic system. The perforation depth was linearly increased in the range of 1~4 mm in proportional to laser output (3~12 W) and irradiation time (5~20 s). We demonstrated that the perforation depth during laser irradiation was varied according to the tissue property of each extracted porcine organ. The measurement of the temperature distribution suggests that the energy is localized in the irradiation spot and transferred to deep tissue, which protects the surrounding tissue from thermal injury. These results can be used to set the driving parameters for a laser incision technique as an alternative to conventional surgical interventions.

  • PLA(polylactic acid), one of biodegradable polymers was blended with various amounts of wood pulp powder through solution blending technic to verify the effect of reinforcing pulp amount on the mechanical properties of blend films. Also these blend films were further modified with TDI(toluene diisocyanate) as crosslinking agent to introduce urethane functions by reaction of pulp hydroxyl groups and isocyanate. As a result, the tensile strength of blend film with 0.25 wt% pulp was increased from $565.25kg_f/cm^2$ for PLA film itself to $624.20kg_f/cm^2$. However, elongation of this film was decreased by 50% of that of PLA film itself. Only PLA/pulp blend film further modified with 500% of TDI/0.25 wt% pulp showed the slightly increased tensile strength but decreased elongation. Melting point and glass transition temperature of PLA/pulp blend films were confirmed by using Differential Scanning Calorimeter(DSC). Thermal stability of these blend films measured by TGA showed only a slight increase at temperature lower than $300^{\circ}C$.

Preparation and Characterization of Acrylic Bone Cement with Poly(methyl methacrylate)/Montmorillonte Nanocomposite Beads (폴리(메틸 메타크릴레이트)/몬모릴로나이트 나노복합체를 이용한 아크릴계 골시멘트의 제조 및 특성)

  • Poly(methyl methauylate)/montmorillonite nanocomposites were incorporated into acrylic bone cement in order to improve the mechanical properties and reduce the exotherm of acrylic bone cement. The nanocomposites were prepared using a suspension polymerization and characterized by scanning electron microscopy, X-ray diffraction, trans-mission electron microscopy, gel permeation chromatography, particle size analyzer and electron dispersive spectroscopy. The acrylic bone cements with poly (methyl methacrylate)/nanocomposite s were prepared and their thermal and mechanical properties were characterized. The prepared polymeric beads were composed of polymer-intercalated nanocomposites with partially exfoliated MMT layers, and the mean diameter of them was $50\~60$ fm with the spherical shape. The maximum setting temperature of the acrylic bone cements decreased from 98 to $81\~87^{circ}C$. The mechanical strengths and moduli of the acrylic bone cement with 0.1 $wt\%$ MMT were increased. compared to that without MMT. However, the mechanical properties were generally decreased with increasing incorporated MMT amounts. It is presumably due to the bubbles in nanocomposite beads generated during polymerization.

  • Poly(vinylidene fluoride) has received much attention in the last several years for the lithium secondary batteries. In this study, to enhance the porosity, PVdF was prepared by phase inversion method using as an additive, PEG (poly(ethylene glycol)), with N,N-dimethylformamid as a solvent. The pores are generated during the solvent and non-solvent exchange process in the coagulation bath filled with non-solvent (distilled water). The surface and cross-section of the membranes were observed with a scanning electron microscopy (SEM). The mechanical property of the membrane was determined by using an universal testing machine (UTM) and thermal property was verified by heat shrinkage. Uniformed sponge structure of PVdF-PEG membrane for the lithium secondary batteries was prepared with 10 wt% of PEG concentration in the PVdF-PEG solution. Porosity, elongation and tensile strengh of the membrane were 87%, 75.45%, and 275. 27 MPa respectively.

Physical Characterization of GaAs/$\textrm{Al}_{x}\textrm{Ga}_{1-x}\textrm{As}$/GaAs Heterostructures by Deep Level transient Spectroscopy (DLTS 방법에 의한 GaAs/$\textrm{Al}_{x}\textrm{Ga}_{1-x}\textrm{As}$/GaAs 이종구조의 물성분석에 관한 연구)

  • The deep level electron traps in AP-MOCVD GaAs/undoped Al\ulcornerGa\ulcornerAs/n-type GaAs heterostructures have been investigated by means of Deep Level Transient Spectroscopy DLTS). In terms of the experimental procedure, GaAs/undoped Al\ulcornerGa\ulcornerAs/n-type GaAs heterostructures were deposited on 2" undoped semi-insulating GaAs wafers by the AP-MOCVD method at $650^{\circ}C$ with TMGa, AsH3, TMAl, and SiH4 gases. The n-type GaAs conduction layers were doped with Si to the target concentration of about 2$\times$10\ulcornercm\ulcorner. The Al content was targeted to x=0.5 and the thicknesses of Al\ulcornerGa\ulcornerAs layers were targeted from 0 to 40 nm. In order to investigate the electrical characteristics, an array of Schottky diodes was built on the heterostructures by the lift-off process and Al thermal evaporation. Among the key results of this experiment, the deep level electron traps at 0.742~0.777 eV and 0.359~0.680 eV were observed in the heterostructures; however, only a 0.787 eV level was detected in n-type GaAs samples without the Al\ulcornerGa\ulcornerAs overlayer. It may be concluded that the 0.787 eV level is an EL2 level and that the 0.742~0.777 eV levels are related to EL2 and residual oxygen impurities which are usually found in MOCVD GaAs and Al\ulcornerGa\ulcornerAs materials grown at $630~660^{\circ}C$. The 0.359~0.680 eV levels may be due to the defects related with the al-O complex and residual Si impurities which are also usually known to exist in the MOCVD materials. Particularly, as the Si doping concentration in the n-type GaAs layer increased, the electron trap concentrations in the heterostructure materials and the magnitude of the C-V hysteresis in the Schottky diodes also increased, indicating that all are intimately related.ated.

Cloning and Characterization of a Novel Mannanase from Paenibacillus sp. BME-14

  • A mannanase gene (man26B) was obtained from a sea bacterium, Paenibacillus sp. BME-14, through the constructed genomic library and inverse PCR. The gene of man26B had an open reading frame of 1,428 bp that encoded a peptide of 475- amino acid residues with a calculated molecular mass of 53 kDa. Man26B possessed two domains, a carbohydrate binding module (CBM) belonging to family 6 and a family 26 catalytic domain (CD) of glycosyl hydrolases, which showed the highest homology to Cel44C of P. polymyxa (60% identity). The optimum pH and temperature for enzymatic activity of Man26B were 4.5 and $60^{\circ}C$, respectively. The activity of Man26B was not affected by $Mg^{2+}$ and $Co^{2+}$, but was inhibited by $Hg^{2+},\;Ca^{2+},\;Cu^{2+},\;Mn^{2+},\;K^+,\;Na^+$, and $\beta$-mercaptoethanol, and slightly enhanced by $Pb^{2+}$ and $Zn^{2+}$. EDTA did not affect the activity of Man26B, which indicates that it does not require divalent ions to function. Man26B showed a high specific activity for LBG and konjac glucomannan, with $K_m,\;V_{max}$, and $k_{cat}$ values of 3.80 mg/ml, 91.70 ${\mu}mol$/min/mg protein, and 77.08/s, respectively, being observed when LBG was the substrate. Furthermore, deletion of the CBM6 domain increased the enzyme stability while enabling it to retain 80% and 60% of its initial activity after treatment at $80^{\circ}C$ and $90^{\circ}C$ for 30 min, respectively. This finding will be useful in industrial applications of Man26B, because of the harsh circumstances associated with such processes.

Isolation and Characterization of Mannanase Producing Bacillus amyloliquefaciens CS47 from Horse Feces (말 분변으로부터 mannanase를 분비하는 Bacillus amyloliquefaciens CS47의 분리 및 특성)

  • The mannanase-producing bacteria, designated CS47, was isolated from the fresh feces of three horses (from a farm in Jinju National University). The isolate CS47 was facultatively anaerobic and grew at temperatures ranging from $20^{\circ}C$ to $50^{\circ}C$ with an optimal temperature of $38^{\circ}C$. The DNA G+C content of the isolate CS47 was 44 mlo%. The major fatty acids were anteiso-15:0 (39.6%), 17:0 (7.6%), and iso-15:0 (37.8%). The 16S rRNA gene sequence similarity between the isolate CS47 and other Bacillus strains varied from 93% to 98%. In the phylogenetic analysis based on these sequences, the isolate CS47 and Bacillus amyloliquefaciens clustered within a group and separated from other species of Bacillus. Based on the physiological and molecular properties, the isolate CS47 was classified within the genus Bacillus as Bacillus amyloliquefaciens CS47. The optimal pH and temperature for mannanase activity of B. amyloliquefaciens CS47 were pH 6.0 and $50^{\circ}C$, respectively. The thermal stability of mannanase from B. amyloliquefaciens CS47 is valuable when using this enzyme in industrial application.

Effect of Corona Discharge on the Changes in Quality and Pasteurization of Ginseng Powder (코로나방전 처리 인삼분말의 살균효과 및 품질변화)

  • As a newly emerging pasteurization method for improving the shelf-life of powdered ginseng, the corona discharge generated at high voltage conditions was examined for its effects on microbial pasteurization, physicochemical characterization, and sensory evaluation. The pasteurizing effects of corona discharge on total bacteria, yeast, molds, and coliforms increased with increasing voltage. However, corona discharge treatment of 5 kv/cm did not show significant effects of pasteurization on the yeast, molds, and coliforms. Treatment with 20 kv/cm reduced about 95% of the total bacteria $(6.14{\pm}0.04{\rightarrow}\;4.59{\pm}0.04$) and coliforms ($2.43{\pm}0.05{\rightarrow}\;1.00{\pm}0.05$) and 99% of the yeast and molds ($4.65{\pm}0.06{\rightarrow}\;2.73{\pm}0.06$). The proximate analysis, minerals, free sugars, crude saponin, and colors of the control ginseng and the ginseng treated with 20 kv/cm were not significantly different at p<0.05. Corona discharge of up to 20 kv/cm did not affect the proximate analysis, minerals, free sugars, crude saponin, and colors of the powdered ginseng. Sensory evaluation scores on color, flavor, taste and overall acceptability between the control ginseng and ginseng treated with 20 kv/cm corona discharge were not significantly different at p<0.05 level. Consequently, corona discharge treatment is thought to be a good alternative pasteurization method for improving the shelf-life of powdered ginseng due to its effective pasteurization, maintenance of nutrients and good sensory characteristics.
