  • Diospyros lotus has been cultivated for its edible fruits, which are considered to have medicinal importance. The aim of this study was to evaluate the antioxidant and antipruritic activities of water-soluble, methanol extract, and ethyl acetate (EA) fractions from D. lotus leaves. The EA fraction showed the lowest $IC_{50}$ vale (DPPH: $5.3{\mu}g/mL$, ABTS: $53.8{\mu}g/mL$). Therefore, we further investigated anti-inflammatory and antipruritic effects of the EA fraction. TNF-${\alpha}$ production increased by PMA plus A23187 treatment was significantly inhibited by the EA fraction in a dose-dependent manner. The EA fraction also inhibited histamine release from rat peritoneal mast cells stimulated by compound 48/80, which promotes histamine release. Furthermore, EA fraction had inhibitory effects on scratching behavior induced by compound 48/80 in Balb/c mice. These results suggest that the EA fraction from D. lotus leaves has potential as ameliorative agent against oxidative stress and pruritus-related disease.

  • Six commercial rootstock varieties were tested for resistance to bacterial wilt (Ralstonia solanacearum) by dipping the roots of seedlings in the bacterial suspension at seedling stage. 'Shincheonggang' and 'B-Blocking' ranked top and second in resistance to bacterial wilt, respectively. For disease index of the grafted plants, 'Dotaerang' grafted onto 'Shincheonggang' was the least, followed by 'Ultra', 'Fighting', 'B-Blocking', 'Special', and 'Connection' grafts. Thus, disease development on the grafted plants was correlated with resistance of rootstocks. The effect of rootstocks on horticultural characteristics including days to flower of scion was minimum or negligible. In correlation between horticultural characteristics of rootstocks and scions grafted thereon, statistically significant negative correlation was observed in days to flower and fruit height but no significant correlation was observed in yield per plant, fruit diameter and soluble solid content in the fruits. Thus, selection of resistant rootstock varieties that support the growth and yield of scion well is necessary for effective control of the disease and yield increase.

  • Mulberry leaves contain many natural components and are considerable resource for natural antiokidants. The antioxidant effect of 'Cheongilppong' collected from Chuncheon, Suwon and Wanju regions were investigated with minilum L-100 device and ARAW-KIT (anti-radical ability of water-soluble substance), in comparison to the ascorbic acid. The measurement parts were mulberry leaves collected from different areas and some kinds of mulberry branches. We obtained that even if samples were same variety, antioxidant capacity of those showed different results according to collected regions. Also, antioxidant effect of mulberry leaves were higher than that of branches. The order of antioxidant effect in Spring was Chuncheon > Suwon > Wanju. The antioxidant effect of mulberry leaves collected from Chuncheon, Suwon and Wanju regions of just before the coloration stage with anthocyanin in fruits were 3627.5 nmol, 2620.0 nmol 1360.7 nmol (ascorbic acid equivalents), respectively. In Autumn, the antioxidant effect and hardness quality of mulberry leaves were increased gradually until just before fallen leaves stage. Therefore, we concluded that being the most suitable period for collecting mulberry leaves as a antiokidant resource is from the latter part of May to the first ten days of June.

  • In worldwide, viral diseases of apple plants has caused the serious problems like reduced production and malformation of fruits. Also, the damages of apple plants by virus and/or viroid infection (Apple chlorotic leaf spot virus, Apple stem grooving virus, Apple mosaic virus, and Apple scar skin viroid) were reported in Korea. However there is few report about the protection approach against the infection by apple viruses. Therefore, this paper introduced the experimental protocol for the development of virus-free apple cultivars (Danhong, Hongan, Saenara, Summerdream). Apple plants were treated at $37^{\circ}C$ for 4 weeks and shoot tips were cultured in vitro. After heat treatment, the detection of apple viruses was performed by RT-PCR using virusspecific detection primers in new apple cultivars. With the heat treatments followed by in vitro shoot tip culture, the proportion of virus-free stocks of 'Danhong', 'Hongan', 'Saenara', and 'Summerdream' was 28%, 16%, 12%, and 12%, respectively. Taken together, this approach can be a good tool for production of virus-free apple stocks.

  • Quercetin mainly exists in the skin of colored fruits and vegetables as one of flavonoids. Recent studies show that quercetin, like other flavonoids, has diverse pharmacological actions. However, relatively little is known about quercetin effects in the regulations of ligand-gated ion channels. In the previous reports, we have shown that quercetin regulates subsets of homomeric ligand-gated ion channels such as glycine, 5-$HT_{3A}$ and ${\alpha}7$ nicotinic acetylcholine receptors. In the present study, we examined quercetin effects on heteromeric neuronal ${\alpha}3{\beta}4$ nicotinic acetylcholine receptor channel activity expressed in Xenopus oocytes after injection of cRNA encoding bovine neuronal ${\alpha}3$ and ${\beta}4$ subunits. Treatment with acetylcholine elicited an inward peak current ($I_{ACh}$) in oocytes expressing ${\alpha}3{\beta}4$ nicotinic acetylcholine receptor. Co-treatment with quercetin and acetylcholine inhibited $I_{ACh}$ in oocytes expressing ${\alpha}3{\beta}4$ nicotinic acetylcholine receptors. The inhibition of $I_{ACh}$ by quercetin was reversible and concentration-dependent. The half-inhibitory concentration ($IC_{50}$) of quercetin was $14.9{\pm}0.8\;{\mu}M$ in oocytes expressing ${\alpha}3{\beta}4$ nicotinic acetylcholine receptor. The inhibition of $I_{ACh}$ by quercetin was voltage-independent and non-competitive. These results indicate that quercetin might regulate ${\alpha}3{\beta}4$ nicotinic acetylcholine receptor and this regulation might be one of the pharmacological actions of quercetin in nervous systems.

  • This investigation was attemped to know fruit quality of apple 'Fuji' grown at three different areas of Taegu. Fruits were gathered from hill side area : Kyungsangpookdo yongchongun chongtongmynn synwonri, flat area : Kyungsangpookdo yongchongun synyoungmyun youujeongri and river side area : Kyungsangpookdo yongchongun kewmhoyeub naenhchundong. Results were as follows: 1. In fruit sampling, we made an effort to select the same size of fruit from all areas, However, the size at river side area was about 40g lighter and minimum length was shorter than those of the other areas. On the other hand, maximum and minimum diameter of the fruit at flat and river side areas were longer than hill side area. Accordingly, shape index of the fruit in river side area was flatter than the other areas. This difference may be due to temperture, soil moisture content, etc. In squeeze rate of the fruit, no significant difference was recognized among areas. 2. Though, rate of fructose in total sugar was almost 50% in all areas, sucrose, fructose and sorbitol concentration of hill side area were higher than the other areas. however glucose was reverse amd brix was almost same as total sugar concentration. 3. Rate of malic acid in total acid was almost 98% and citric acid was only 2% in all areas. Malic acid concentration of hill side area was higher than the other areas and titratable acid concentration was almost same as total acid.

  • The purpose of this study is to review the taxonomical position of Phytolacca insularis Nakai based on morphological characters and ITS sequences. Phytolacca insularis was similar to P. acinosa in the stem shape, inflorescence, apocarpous, eight pistils, eight stamens, pink anther, and eight fruits. But the unique difference between P. insularis and P. acinosa was leaf size and pistil color. Phytolaccri insularis and P. acinosa have a similar sculpturing patterns as an foveolate and microscabrate in pollen, but they differed from P. americana in having a bigger foveolae size in sculpture pattern. Phytolacca insularis and P. acinosa were similar in the seed size but different from P. americana by having a small size of seed. The seed surface of P. insularis and P. acinosa was similar with a irregularly undulate shape, but its feature of P. americana differed in having the reticulate shape. Phytolacc a insularis had identical ITS sequences with P. acinosa. Phytolacca insularis and P. acinosa have a close relationship in the morphological characters and ITS sequences. The examinations of morphology and ITS sequences suggest that P. insularis be a form of P. acinosa although it has been formerly treated as a distinct species.

  • This study investigated the anti-inflammatory activities and cell viability of Amelanchier asiatica (A. asiatica) 70% ethanol extracts against RAW 264.7 cells induced by lipopolysaccharide (LPS). Cell toxicity test on macrophage cells (RAW 264.7) was performed by 3-[4,5-dimethyl-thiazol-2-yl]-2, 5-diphenyl-tetrazoliumbromide (MTT) assay and results showed 96% cell viability at $1,000{\mu}g/mL$ concentration. Anti-inflammatory activity was examined via the inhibitory tests on the production of LPS induced NO in RAW 264.7 cells by Griess assay. The result showed that the extract inhibited NO production in concentration dependent manner. The iNOS and COX-2 protein expression inhibitory effects were confirmed by western blot and by reverse transcription-polymerase chain reaction (RT-PCR). From the former they were decreased by 84.3%, 56.2% at $500{\mu}g/mL$ concentration, respectively, and from the latter decreased by 89.8%, 84.9% at $500{\mu}g/mL$, respectively. In conclusion, this study showed the anti-inflammatory effects of A. asiatica extracts. Thus, this could be applied to an anti-inflammatory agent.

  • We investigated the optimum concentration of a $NaClO_2$ solution and the amount of gaseous $Cl_2$ for production of high yield and purity of aqueous $ClO_2$ by use of a gaseous chlorine-chlorite $ClO_2$ generator. This system produced lower concentrations of chlorine dioxide and is applicable for direct-use in food processing as a cleaner and sanitizer. The concentration of $NaClO_2$ solution and the amount of gaseous $Cl_2$ was varied from 0.01-0.1% and 100-1,000 g/hr, respectively. The concentrations of chlorite, chlorate, FAC (free available chlorine), and chlorine dioxide that were produced increased with increasing concentration of $NaClO_2$ solution and with the amount of gaseous $Cl_2$. The optimum concentration of $NaClO_2$ solution and amount of gaseous $Cl_2$ were 0.1% and 900 g/hr respectively. $ClO_2$ and FAC produced at these concentrations were 882.0 ppm and 8.0 ppm, with no detection of chlorite and chlorate. The yield and purity of $ClO_2$ were 97.0% and 96.0% respectively. Immersion-cleaning experiments showed that this protocol decreased the level of CFU/g by $10^3$- to $10^4$-fold, with a similar effect on fruit.
