Inhibition of $\alpha$-Glucosidase Activity by Quercetin (Quercetin에 의한 $\alpha$-glucosidase 활성 저해)

  • Quercetin is a flavonoid molecule that is known to tie in various sources of natural products such as vegetables and fruits. It has been proven that quercetin plays a crucial role in the prevention of colon cancer as well as homeostasis as radical scavenger in human body. It is also well-known that glycosidases, including $\alpha$-glucosidase, are involved in a variety of degenerative metabolic disorders. In the course of screening useful $\alpha$-glucosidase inhibitors, we screened out quercetin as a $\alpha$-glucosidase inhibitor from chemical libraries. Quercetin was shown to be a reversible, slow-binding, and noncompetitive inhibitor of yeast a-glucosidase with a K$_i$ value of $6.3\times10^{-8}$ M when it was included with an enzyme mixture. Together, these results show that quercetin has potential in treating disorders including diabetes, although the further mechanistic study is needed.

Quality Characteristics of Strawberry Cultivated with Natural Organic Manure (천연 유기질 비료로 재배한 딸기의 품질 특성)

  • The use of artificially synthesized inorganic manure for the production of fruits and vegetables gives rise to soil contamination as well as noxious effects against human. Changes in quality of strawberry (SN) cultivated with a natural organic manure developed to solve such problems were studied during storage at 4$^{\circ}$C for 2 weeks. SN showed higher weight/volume ratio and lower weight loss during storage than strawberry (control) cultivated with commercial synthetic mature. Sweetness was higher in SN (9.2 Brix) than in control (8.2 Brix), and decrease in sweetness during storage was smaller in SN than in control. Redness (b) of strawberry was 32.1 in SN and 30.2 in control, and the redness decreased during storage showing 25.2 and 23.0, respectively, after 2 weeks. Hardness was higher in SN than in control, and decrease in hardness during storage was smaller in SN than in control. Vitamin C content was slightly higher in SN than in control, and mineral contents were generally higher in SN than in control.

Incidence Rates of Postharvest Fruit Rots and Detection Rates of Their Pathogens on New Kiwifruit Cultivars Bred in Korea (국내에서 개발된 참다래 신품종의 과실무름병 발생률과 병원균 검출 빈도)

  • The incidence rates of postharvest fruit rots of four kiwifruit cultivars which were cultivated under rain-proof tunnel house at a same orchard were examined. Among them, 'Halla-Gold', 'Jecy-Gold' and 'Jecy-Sweet' were new cultivars bred in Korea. The disease incidence was varied with cultivars; 74.8%, 65.3%, 57.1% and 16.2% for 'Hayward', 'Halla-Gold', 'Jecy-Sweet' and 'Jecy-Gold' cultivars, respectively. Two hundred and eighteen isolates were obtained from diseased fruits and identified by mycological and molecular biological methods. Three fungi, Botryspheria dothidea, Diaphorthe actinidiae and Botrytis cinerea, were identified as pathogens of the postharvest fruit rots with detection rates of 95.4%, 4.6% and 2.3%, respectively.

  • Rickets is a nutritional disorder which is caused either by deficiency of vitamin D or by a defective activation of vitamin D. In these days, even though the incidence of rickets has decreased through adequate nutritional support, we sometimes experience rickets in babies receiving a prolonged special diet as therapy for chronic diarrhea, or those subject to a in receiving the prolonged elimination of milk because of allergy. But there are no reports about rickets caused by absolute elimination of milk because of allergies in Korea. We report here a case of rickets developed after feeding on Sunsik( a mixture of several grain and fruits powder) during a seven months period in an 8-month-old male patient. This male infant manifested vomiting, poor feeding, decreased serum calcium and 25-hydroxycholecalciferol levels, and markedly increased serum alkaline phosphatase and parathyroid hormone levels. Skeletal X-rays showed cupping and fraying in distal metaphyses of radius and ulna, and generalized osteopenia. The patient improved with vitamin D and calcium therapy.

  • Management of obesity needs good rapport among pediatrician, parents and children. Through motivational interview, pediatrician should explore practical ways to modify the eating behaviors conducive to obesity. Imbalance between energy intake and consumption contributes to weight gain. Therefore decrease of sedentary behavior (screen time <2 hrs) and increase of physical activity (>60 minutes) is also important in management of obesity. The goal of management is the long-term healthy life though healthy behaviors. Creating a home environment that supports healthy habits and behaviors is an important key to modifying lifestyle behaviors. For example, increasing intake of vegetables and fruits, restriction of sweetened beverage, education of healthy selection of foods, and frequent family dinner may be advisable. Family members should change their own behaviors in order to help their children. Parents should be positive role model and monitor their children authoritatively but not restrictively.

Roles of Kermanshahi Oil, Animal Fat, Dietary and Non-Dietary Vitamin D and other Nutrients in Increased Risk of Premenopausal Breast Cancer: A Case Control Study in Kermanshah, Iran

  • Background: Kermanshahi oil is one the most favorable oils in Iran especially in Kermanshah province. We aimed to evaluate the role of usual intake of Kermanshahi oil and other kinds of dietary fats as well as different meats, vegetables and fruits, carbohydrates, cereals, grains, sweets, candy and lifestyle habits in risk of breast cancer. Materials and Methods: A case-control study with 47 consecutive, newly diagnosed premenopausal breast-cancer patients and 105 age and socioeconomic matched healthy women was conducted from 2013-2014 in Imam Reza hospital of Kermanshah using a standardized, validated questionnaire assessing various anthropometric, socio-demographic, lifestyle and dietary characteristics. Results: Kermanshahi oil intake was associated with a 2.1-fold (OR=2.123, 95% CI 1.332-3.38) (p=0.002) higher likelihood of having breast cancer, while daily intake of other solid animal fats also increased the likelihood by 2.8-fold (OR = 2.754, 95% CI 1.43-5.273) (p < 0.001), after various adjustments made. Lack of fish oil, white meat, vegetables, soy products, nuts and dairy products (especially during adolescence) in daily regimens and lack of sun exposure were significantly associated with premenopausal breast cancer risk in this region. Conclusions: This study suggested that animal fat increases the risk of premenopausal breast cancer but many other dietary and non-dietary factors including calcium and vitamin D deficiency are consistently associated with increased odds of breast cancer in this region.

  • Background: Nasopharyngeal carcinoma (NPC) is endemic in Southern Chinese and Southeast Asian populations. Geographical and ethnic clustering of the cancer is due to genetic, environmental, and lifestyle risk factors. This case-control study aimed to identify or confirm both genetic and non-genetic risk factors for NPC in one of the endemic countries, Malaysia. Materials and Methods: A panel of 768 single-nucleotide polymorphisms (SNPs) previously associated with various cancers and known non-genetic risk factors for NPC were selected and analyzed for their associations with NPC in a case-control study. Results: Statistical analysis identified 40 SNPs associated with NPC risk in our population, including 5 documented previously by genome-wide association studies (GWAS) and other case-control studies; the associations of the remaining 35 SNPs with NPC were novel. In addition, consistent with previous studies, exposure to occupational hazards, overconsumption of salt-cured foods, red meat, as well as low intake of fruits and vegetables were also associated with NPC risk. Conclusions: In short, this study confirmed and/or identified genetic, environmental and dietary risk factors associated with NPC susceptibility in a Southeast Asian population.

  • Total 31 samples from 21 African plant species in 13 families were tested for their insecticidal and acaricidal activities against Nilaparvata lugens (Stal) and Tetranychus urticae (Koch) adults through topical application an leaf-dipping methods, respectively. The insecticidal and acaricidal activities were both plant parts and species dependent. The methanol extracts from whole plants of Casia occidental is and Cassia tora (Caesalpinaceae), an stem of Prosopis chinensis (Mimosaceae) revealed potent insecticidal activity against N. lugens. Potnet acaricidal activity against T. urticae was obtained from the methanol extracts from whole plants of Celosia trigyna (Amaranthaceae) and Combretum micronthum (Combretaceeae), leaves of Combretm glutinotum, and leaves and fruits of prosopis chinensis.

  • In plants, color is a powerful tool to attract insects and herbivores which act as pollinators and vehicles of seed dispersion. In particular, flower color has held key post for human with aesthetic value. Horticultural industry has developed methods to produce new and attractive color varieties in flowering plants. Carotenoids are one of the main pigments being responsible for red, orange, and yellow colors. Their biosynthetic pathway has been considered as a major target of metabolic engineering for color modification of flowers and fruits. Here, we review the diverse efforts to modify pigment phenotype by the control of carotenogenic gene expression and enzyme levels. Recent reports about regulating degradation and storage of carotenoids will be also summarized to help the creation of engineered flower with novel color phenotype which is not existed in nature.

  • Antioxidative activities of ethanol extracts of fruits from three cultivars of Prunus mune with different harvest times were determined. Extracts of P. mune cutivar Okyoung showed the strongest activity at 97.5%(compared to BHT) using DPPH or thiocyanate method, although no statistically significant differences were observed among the cultivars. Antioxidative activities of the extracts measured with glutathione-S-transferase method was over 70% of that of BHT. Survival rates of cells in mouse spinal cord culture challenged by superoxide anion were 89, 79, and 62% at 40 mg/ml of Okyoung, Namgo, and Kochunme extracts, respectively, whereas 90% at 6mM glutathion.
