Relationship between Preharvest Factors and the Incidence of Storage Disorders in 'Fuji' Apples during CA Storage

  • This study was carried out to evaluate the effect of pre-harvest factors on fruit quality and the incidence of storage disorders in 'Fuji' apple during CA storage. Incidence of storage disorders varied, depending on the growing regions, field conditions, and altitude of the orchards. Results indicated that fruit maturity may play a crucial role in the incidence and severity of flesh browning and watercore. The incidence of these storage disorders increased with fruit maturity until 180 days after full bloom (DAFB). In addition, occurrence of watercore was correlated with the incidence of flesh browning during CA storage. The incidence of flesh browning was positively correlated with the sum of the diurnal temperature range from September through October and amount of precipitation from August to October in 1996-1999 growing years. These results suggested that 'Fuji' apple should be harvested no later than 180 DAFB because the late harvested fruits were prone to development of watercore which was correlated with the incidence of flesh browing during CA storage in Korea.

Suppressive Effects of Hesperidin on Th2-associated Cytokines Expression in RBL-2H3 Cells (RBL-2H3 세포에서 Hesperidin의 Th2 사이토카인 발현 억제 효과)

  • Hesperidin (HES), a flavonone glycoside isolated from the citrus fruits such as lemons and oranges, has been reported to have many biological properties including antiinflammatory, antioxidant, and antiallergy activities. In this study, we focused on the action of HES modulating Th2-associated cytokines such as IL-4 and IL-13 expression in PMA/ionomycin (PI)-stimulated rat basophilic leukemia (RBL-2H3) cells. The production of IL-4 and IL-13 was quantified by ELISA and the mRNA expression was detected by using RT-PCR assay. In addition, western blot analysis was performed to determine the transcription factors involved in the cytokine expression. We found that HES significantly decreased PI-induced IL-4 and IL-13 productions and also decreased the level of mRNA in a dose-dependent manner. Furthermore, western blot analysis of the transcription factors implied that HES down-regulated the protein level of c-Jun and c-Fos, which are the activating protein 1 (AP-1) family and nuclear factor-kappaB (NF-${\kappa}B$) characterized as a transcription factors related to the Th2-associated cytokine expression. Taken together, our data showed that the action of HES responsible for antiallergy activities is based on suppression of Th2-associated cytokines through inhibition of AP-1 and NF-${\kappa}B$ transcription factors.

An Analysis of Food Purchase Accessibility and Availability for Rural Households : The Cases of Bibong-myeon and Maesong-myeon, Hwaseong-si (농촌지역 가구의 식료품 구매 접근성과 이용가능성 분석 - 화성시 비봉면과 매송면을 중심으로 -)

  • This study examines the relationship between food purchase accessibility for 86 homemakers and that for 42 grocery stores. The Study area included Bibong-myeon and Maesong-myeon, Hwaseong-si, Gyeonggi-do, to represent a rural area. The analysis classified three regions (residence, outside of residence, and other areas) and seven types of grocery stores (LDS, CS, NCS, CC, NCC, TM, and other). The expansion of the area range increased the availability of food items and the accessibility of large-scale grocery stores as well as those for food procurement. The respondents had difficulty preparing family meals and buying food items because of a lack of food item variety and the small scale of grocery stores. In particular, fresh meat, fruits, and fish were difficult to purchase, causing some nutritional imbalance. These results suggest a need to develop meal composition samples and programs that can actively encourage the intake of meat and fish products with a diet based in self-produced vegetables for better dietary behaviors.

Shelf-life comparison of two Korean golden kiwifruit (Actinidia chinensis) cultivars under low temperature storage (국내 육성 황육계 키위푸르트 품종간 저장 특성 비교)

  • Physiological characteristics of two Korean golden kiwifruit cultivars, 'Halla Gold' and 'Haehyang', were compared to determine the storage potential of fruit. The soluble solid levels of the fruit were 8.9 and 6.9 oBrix in 'Halla Gold' and 'Haehyang' at harvest, respectively but increased up to 15.4 in 'Halla Gold' and 17.5 oBrix in 'Haehyang' after 2 months of storage. Major sugars were fructose and glucose, and sucrose content was relatively low regardless of cultivar. The edible quality of 'Haehyang' was better than 'Halla Gold' because of higher amount of sugars. Firmness of the fruits gradually decreased as the increase of storage period in 'Halla Gold' in both flesh and core tissue. Th firmness loss of 'Haehyang' fruit was faster in the first 2 months and then became slow. After 75 days of storage, the firmness of 'Haehyang' fruit was only 5.2% at harvest. Core tissue was soften enough to eat at ripe stage. Wall modifying enzyme activities including xylanase, ${\alpha}$-L-arabinofuranosi-dase and ${\beta}$-galactosidase were consistently higher in 'Haehyang' and the activity of pectate lyase was more increased than 'Halla Gold' after 2 months of storage. Respiration rate of 'Haehyang' was higher than 'Halla Gold' and further increased after 2 months of storage. Weight loss was much higher in 'Haehyang' which showed higher rate of the firmness loss. The storage potential of golden kiwifruit was estimated to be about 2 months for 'Haehyang' and 3 months for 'Halla Gold' when determined on the basis of the fruit firmness.

  • Tomato spotted wilt virus (TSWV) was occurred on the three vegetables of egg plant (Solanum melongena), whole radish (Raphanus acanthiformis) and sugar loaf (Cichorium intybus) at Anyang area infested with TSWV. Whole radish was produced the symptoms of necrotic spots on the leaves, and necrosis and malformation on the roots by TSWV. Egg plant was induced the symptoms of typical multiple ring spots on the leaves and necrotic rings on the fruits. Sugar loaf was infected severely with the typical symptoms of ring spots on the leaves and stunt. The three isolates of TSWV could infect locally on the indicator plants of Chenopodium amaranticolor, C. quinoa and Nicotiana debney, and systemically on N. glutinosa, N. benthamiana and Datura stramonium. Two TSWV isolates from egg plant and sugar loaf were very similar in virulence. However, the virulence of TSWV from whole radish was very different as local infection on 5 Nicotiana species including N. tabacum 'Xanthi NC'.

Identification of Verticillium tenerum Isolated from Potato and Pumpkin in Korea (감자와 단호박에서 분리한 Verticillium tenerum의 동정)

  • Verticillium tenerum was detected from the samples of potato (Solanum tuberosum L.) and pumpkin (Cucurbita maxima Duch.) in Korea. The samples on which this species was detected were potato tubers collected from Pyungchang and Gimjae, and fruits and seeds of pumpkin from Yeonchon in 2003. The conidiophores of the fungus produced mainly three to five phialides in verticil. The phialides were narrow, flask-shaped, only very slightly swollen at the base, and taper subulately to a narrow neck. They were hyaline, $10{\sim}26{\times}2{\sim}5\;{\mu}m$. The conidia were formed in slimy heads, and were oval to ellipsoidal, pale reddish-brown, $2.5{\sim}7.5{\times}2{\sim}4\;{\mu}m$. The colony on the substrata was unique, showing brick-red color. This is the first report on the distribution of Verticillium tenerum in Korea.

CALIBRATION TRANSFER FROM REFLECTANCE TO INTERACTANCE-REFLECTANCE WITHOUT STANDARDS: USE OF MATHEMATICAL PRETREATMENTS

  • The use of fibre optic probes for NIR quality control in the industry is becoming very important, as it provides a powerful tool to reduce sample analysis time and it facilitates the implementation of on-line analyses. However, most of the applications of fibre optics and probes have been done on suspensions, clear liquids and films, chemical and pharmaceutical products and also on fruits and animal products. Traditional applications of near infrared spectroscopy in agriculture have been developed in reflectance mode and calibration transfer could be an interesting way to reduce efforts. Classical methods for calibration transfer between different instruments involve the use of sealed reference cups, but, as fibre optic analysis does not use cups, it is necessary to develop new methods for calibration transfer without standards (Blank et al., 1996). In this paper, we have studied how the most used mathematical pretreatments (three methods of Multiplicative Scatter Correction, Standard Normal Variate, Detrending and derivatives) and their combinations applied to calibration development can contribute to reduce spectral differences between instruments. Calibration equations were obtained for three sets of cereals (barley, wheat and maize) scanned in reflectance mode and then they were validated with samples analysed in reflectance and interactance-reflectance mode (fibre optic). Preliminary results show how some combination of pretreatments reduce the differences in the predicted values, measured as standard error of differences, facilitating the use of calibrations obtained in reflectance for samples analysed by interactance-reflectance. However, the application of pretreatments is not enough to satisfy the control limits for calibration transfer suggested by Shenk et al. (1992), and it should be necessary to combine them with a specific algorithm for instruments standardization.

Endothelium Independent Effect of Pelargonidin on Vasoconstriction in Rat Aorta

  • In this study, we investigated the effects of pelargonidin, an anthocyanidin found in many fruits and vegetables, on endothelium-independent vascular contractility to determine the underlying mechanism of relaxation. Isometric contractions of denuded aortic muscles from male rats were recorded, and the data were combined with those obtained in western blot analysis. Pelargonidin significantly inhibited fluoride-, thromboxane A2-, and phorbol ester-induced vascular contractions, regardless of the presence or absence of endothelium, suggesting a direct effect of the compound on vascular smooth muscles via a different pathway. Pelargonidin significantly inhibited the fluoride-dependent increase in the level of myosin phosphatase target subunit 1 (MYPT1) phosphorylation at Thr-855 and the phorbol 12,13-dibutyrate-dependent increase in the level of extracellular signal-regulated kinase (ERK) 1/2 phosphorylation at Thr202/Tyr204, suggesting the inhibition of Rho-kinase and mitogen-activated protein kinase kinase (MEK) activities and subsequent phosphorylation of MYPT1 and ERK1/2. These results suggest that the relaxation effect of pelargonidin on agonist-dependent vascular contractions includes inhibition of Rho-kinase and MEK activities, independent of the endothelial function.

A Study on Selection and Measures of the Apprehensive Areas of Soil Loss in the So-ok Stream Watershed (소옥천유역 토양유실 우심지역 선정 및 대책에 관한 연구)

  • This study aims to find out critical areas of a soil loss and propose feasible measures to reduce the water quality deterioration by a soil lose. As a study area, the So-ok stream catchment locating at the upper area of Daecheong Reservoir is selected and intensive field observation was carried out. RUSLE model is applied to assess the impact of the pollution migration by a soil ross from the critical areas during storms on the water quality of Chusori embayment. As results, total amount of the soil ross assessed against to the critical area on which major type of land use is a orchard for fruits is 54.3 ton/ha/yr and that of an abandoned mine site is about 200 ton/ha/year. In particular as effective measures, a plantation of an appropriate species of fruit trees and an application of ecologic restoration schemes are proposed against to the orchard and the abandoned mine site, respectively.

A study on Living Culture of Korea through accounting records written by Song, Whasun at Hongcheon-Up in early 20th century (홍천읍 송화선(宋化善) 장기(掌記)를 통해 본 20세기 초 한국의 생활 문화 연구)

  • An assortment of daily supplies have been documented in and accounting book that Hwa-sun Song, a wholesale dealer in Hongcheon, Gangwon-do, sent to Young-hui Sin, a customer. This study analyzed a total of 163 documentations in the accounting book between 1910 to 1916, which includes types of daly supplies, trading volume, and prices, maintained accounting between. Consequently, we are able to indentify companies that produced the applicable goods, names of products, units by which goods were counted, and the lowest and highest prices prevailing, along with kinds of goods patronized in everyday life in Hongcheon in the early 20th century. Paper had the maximum trading volume. The second, most traded were cigarettes, a symbol of the new culture. These were traded under various brand names, such as Kkotpyo, Guksyu, Sanhopyo, Syonghak, and Joil. Foodstuffs, were the third most traded items, including fish, fruits, sugar, Waeddeok, Chilwaeddeok, Color candies and Okchyun candies. Our results indicate that the snack food business had developed since the 19th century. Lighting equipment, oil, candles, matches as well as traditional oil lamps and flints cornered the fourth largest stock being traded. Medications were fifth, with prescriptions written for Insohwan, Hoechyungsan and Siungo, including quinine, a medicine for malaria. Other trades included kitchen appliances such as soup bowls, porcelain bowls, kettles, and drinking cups, and a variety of daily supplies such as mirrors, mats, umbrellas, Geumjiwaemil, hair oil imported from Japan, and soap.
