타액 면역글로부린과 치아우식의 상관관계에 관한연구 (A STUDY OF SALIVARY IMMUNOGLOBULIN AND DENTAL CARIES)

  • This study was undertaken to observe the relationship between salivary IgA, IgG, agglutinin titer & dental caries. The subjects divided into two groups, an active caries group (AC group) and caries-free and treated caries group (CFTC group). The AC group consisted of 36 subjects who had one or more carious lesions and the CFTC group of 12 subjects who had no evidence of caries or had filled teeth without present carious teeth for the last six months. The IgA, IgG and IgM levels in their saliva were measured by single radial immunodiffusion method using a disposable low-level immunodiffusion plate. The salivary agglutinin titers to Streptococcus mutans were measured by microtitration system. The results were as follow; 1. The mean value of IgA concentration in saliva of AC group was slightly higher than that of CFTC group, but its difference was slight. 2. The mean value of IgG concentration in saliva of AC group was slightly lower than that of CFTC group. The IgM concentration in saliva of both groups was neither below 1.1 mg/dl nor detected on LC partigen immunodiffusion plate. 3. There was no difference in the agglutinin titer to S. mutans antigen by serotypes, but low level agglutinin to type d was measured in both groups. 4. AC group showed low correlation between IgA, IgG & DMFT, but CFTC group revealed negative correlation. 5. The relationship between salivary IgA & agglutinin titers to S. mutans was low correlation in AC group, but CFTC group showed significant positive correlation. (P<0.05) 6. There were no specific correlations among the concentrations of salivary immunoglobulins, agglutinin titers to S. mutans, and the DMF teeth. They had no close concern to induce the dental caries.

태독이 장내 세균총에 미치는 영향을 규명하기 위한 임신쥐의 스트레스 및 식이에 따른 신생쥐의 장내 세균총 및 IgA 농도 분석 시험 (Study on Intestinal Flora and IgA Concentration Analysis in Newborn Mice by Stress and Diet in Pregnant Mice to Investigate the Effect of Taedok on the Intestinal Flora)

  • Objectives The purpose of this study is to look for pathological mechanism of disease development caused by Taedok, by studying whether stress and diet in pregnant ICR mice affect the intestinal flora and IgA (Immunoglobulin A) concentration. Methods The mice were divided into 4 groups (n=5 per group) based on the concept of Taedok: the control group (G1), stress group (G2), capsaicin diet group (G3), high fat diet group (G4). We collected and analyzed intestinal flora from maternal feces and cecal flora from neonatal mice by group. Then, IgA concentration in the maternal feces and sIgA (secretory Immunoglobulin A) concentration in the cecal contents of newborn mice were analyzed. In addition, serum corticosterone was analyzed before and after stress application. Results Changes in maternal intestinal flora and neonatal mice cecal flora by stress and diet were observed. There were no significant changes in the IgA concentration in maternal feces and the sIgA concentration in the cecal contents of neonatal mice. No significant changes compared to the control group were observed between groups before and after applying stress. However, when comparing within one subject, a significant increase was confirmed after stress application in the stress group (G2). Conclusions Based on the results, we observed stress and diet in pregnant mice affect the intestinal flora of maternal and neonatal. We were able to interpret the pathological mechanism of Taedok based on the principle of interaction between mother and newborn intestinal flora.

흰쥐에서 식이 단백질 수준이 수유를 통한 수동면역에 미치는 영향 (Effect of Dietary Protein Level on Immune Substances in Milk and its Transfer to Pups in Rats)

  • This study was performed to determine the effect of maternal protein intake on 1) the concentration of immune substances in milk 2) degree of passive immunity to pups via lactation, and 3) specific antibody production to a specific antigen, $\beta$-lactoglobulin(BLG). 4) the effect of passive immunity that pups received from mother during lactation on the production of antibodies when the pups were challenged to the same antigen. Part of the female rats were immunized with BLG before and during pregnancy. The pregnant rats were placed into either 25% or 10% isolated soy protein diet throughout gestation and lactation. After weaning, pups from each group continued to be fed the same diet. At 18 weeks of age, all the pups were challenged with BLG. Total IgA and IgG, lysozyme, BLG-specific IgA and IgG were measured in dam's serum, dam's milk, and pup's serum. Total IgG, and lysozyme in dam's serum and milk were higher in high protein group. Total IgA and IgG in pup's serum remained higher in high protein group from 5 to 18 weeks of age. BLG-specific antibodies were found in the milk and serum of immunized dams, and in serum of pups born to immunized dams but not in the non-immunized group. BLG-specific IgA and IgG were again higher in high protein group and declined with time. The concentration decreased faster in the low proetein group than in the high protein groups. After immunization the pups with LBG, serum BLG-specific antibodies were not differ between rats born to immunized dams and those born to non-immunized dams. Therefore passive immunity rats received via milk as a pup had no effect on the BLG-specific antibody production later in life. This study shows the importance of protein status of mother and strongly support to the endorsement of breast feeding.

  • The stabilizing effect of fructooligosanharide (FO) on hen's egg yolk immunoglobulin (yIgG) by heat and acid was investigated. The heat stability of yIgG at 70∼80$^{\circ}C$ was enhanced in a concentration-dependent manner by adding 0∼50% (w/v) FO to a yIgG solution. Acid-induced inactivation of yIgG was also suppressed in a concentration-dependent relationship by addition of FO. Addition of 50% FO almost completely stabilized yIgG at pH 3. The remarkable stablizing effect of FO on yIgG may enhance the use of yIgG as functional food ingredients.

Variations of Immunoglobulins in Colostrum and Immune Milk as Affected by Antigen Releasing Devices

  • This work was conducted to examine the variation of immunoglobulins (Igs) in serum, immune milk, normal milk and colostrum upon implantation of a new Antigen Releasing Device (ARD). The core of each ARD housed an immunostimulating complex (ISCOM) that was made of adjuvant Quil A and type XIII lipase from a Pseudomonas sp. Each ARD was coated with polylactic acid, known as polylactide, that controls antigen release. Twenty lactating Chinese Holstein cows were divided into 2 groups (n = 10): test group and control group. All cows in the test group were implanted with a single injection in the right iliac lymph node with 3 types of ARDs, which were designed to release the antigens at d 0, 14 and 28 post-implantation. Blood and milk samples were collected from both groups, and colostrum samples were also collected from other post-partum cows in the same farm. Concentrations of $IgG_1$, IgA and IgM in whey and serum were measured by sandwich ELISA. The results showed that the $IgG_1$, IgA and IgM concentrations in serum and whey from the test group were higher than from the control group. Among the three Igs measured, the $IgG_1$ concentration in serum was significantly higher at d 40 after ARD implantation, and the $IgG_1$ concentration in whey peaked at d 9, 17 and 30, which corresponded with release of the antigen. Based on Pearson's correlation between Ig concentration and production parameters, IgA concentration in normal milk was positively correlated with lactation period, which reflected IgA changes during the lactation period in immune milk. In colostrum, $IgG_1$, IgA and IgM decreased abruptly from d 0 to 3, and then decreased slightly. In conclusion, serum $IgG_1$ concentration can be affected by controlled release of the ARD, while whey IgA levels are primarily affected by lactation period. These results may be useful in future studies designed to regulate concentrations of Igs in immune milk.

특이혈청항체(特異血淸抗體) IgG분획(分劃)이 Streptococcus mutans의 평활면(平滑面) 부착(附着)에 미치는 영향(影響)에 관(關)한 연구(硏究) (Effect of specific serum IgG antibody against Streptococcus mutans on the adherence of S. mutans to smooth surface in vitro)

  • In order to demonstrate the effect of specific serum IgG antibody on the adherence of Streptococcus mutans to smooth surface and the mechanism of effective adherence inhibition by IgG antibody, in the present study authors obtained purified IgG from different immunogen preparations of S. mutans NCTC 10449(serotype c) and observed the effect of each IgG preparation on the adherence of each S. mutans strain cultured in different conditions. In addition, the present study was undertaken to observe the cross-reactivity of IgG and the effect of sucrose concentration on the adherence of S. mutans in vitro non-growth condition. The adherence of S. mutans to glass surface was effectively inhibited by serum IgG antibody. At the same IgG concentrations, anti-2% fructose grown/1N NaCl washed S. mutans NCTC 10449 cell showed greater adherence inhibitory effect to S. mutans strains than anti-2% sucrose grown and anti-S. mutans NCTC 10449 cell wall, and the greater inhibitory effects of IgG preparations were observed in assay using 2% fructose grown S. mutans cell preparations than using 0.1% sucrose grown cell preparations. These results suggest that the more effective adherence inhibition by serum IgG antibody is due to the reaction with S. mutans cell surface antigens rather than glucan and cell-associated glucosyltransferase. The greatest adherence inhibitory effect of IgG to S. mutans strains was observed on homologous NCTC 10449 strain and the inhibition cross-reactivities were observed between serotype c, e, and f strains. More pronounced cross-reactivity of adherence inhibition of IgG to S. mutans was observed in assay using anti-2% fructose grown/1N NaCl washed cell than using other IgG preparations, and observed in assay using 2% fructose grown S. mutans cell preparations than 0.1% sucrose grown cell preparations. It was interested that low, but adequate concentration of reactive IgG antibody significantly increased the adherence ability of S. mutans. This result may be due to the formation of small cell aggregates resulted in a increase in the numbers of organisms which adhered to glass surface. The adherence of S. mutans to glass surface was possible in the absence of glucan-synthetic activity. Low level of sucrose significantly increased the adherence ability of S. mutans to glass surface, but excessive amount of sucrose induced large cell aggregates resulted in a decrease in the numbers of organism which adhered.

DSS로 유도된 염증성 장 질환 마우스 동물모델에서 생식이 장관 임파조직내 면역조절 기능에 미치는 영향 (Immunoregulatory Effects of Saengshik on DSS-Induced Inflammatory Bowel Disease in Mouse Model System)

  • 본 연구의 목적은 DSS로 유발시킨 대장염 동물모델에서 생식섭취가 장관 내 면역조절에 미치는 영향에 대하여 검토해보고자 했다. 그 결과, 생식섭취가 DSS로 유발시킨 대장염에서 normal군보다 IFN-$\gamma$의 함량을 증가시키고, IL-4와 IL-10의 함량은 감소시킴이 관찰되었다. 또한, 증가된 Th1 세포의 cytokine과 감소된 Th2 세포의 cytokine은 염증이 유발된 후, 생식을 통한 치료로써 변환시킬 수 있음을 증명하였다. 이러한 연구 결과를 볼 때 DSS로 유발시킨 대장염에서 생식 섭취를 통한 치료는 IL-4와 IL-10과 같은 염증성 cytokine의 억제기능을 통하여 면역시스템을 강화시키고, 손상된 염증을 완화시켜 줄 것으로 판단된다. 생식의 면역조절에 관한 자세한 기전은 아직 밝혀져 있지 않지만, 염증성 장질환에서 생식섭취가 면역학적 역할에 대한 새로운 양상들에 대한 연구가 더욱더 필요할 것으로 사료된다.

Streptococcus mutans 균의 면역과 사료첨가제가 충치 예방용 계란의 생산에 미치는 영향 (Effect of Diet Supplements on the Production of Anti-Dental Caries Hen's Eggs by Immunization of Streptococcus mutans)

  • 난황 항체 강화를 위하여 켈프밀 $4\%$ 처리구와 계피 $0.3\%$와 박하 $2\%$가 첨가된 특수사료를 급여하고 Streptococcus mutans를 2주 간격의 5회 면역처리를 복합적으로 실시하면서 40주령 산란계를 사양하였다. 면역처리를 하지 않은 처리군에서는 박하 첨가군의 산란율이 높았다. 면역한 후에 발생되는 산란율 저하는 대조구와 계피 첨가군에서 적게 나타났다. 계란의 무게는 면역처리에 따른 효과가 나타나지 않았으나 사료에 따라 차이가 나타났다. 면역처리를 하지 않은 처리구에서 계피, 박하, 켈프밀 모두 총 IgY 함량이 대조구보다 높게 나왔으며, 특히 켈프밀의 경우 대조구에 비해 $5.4\%$ 가량 상승하였다. 면역 한 처리구끼리 각각 비교하여 보면 대조구에 비해서 다른 처리구에서 모두 높았으며, 특히 켈프밀 처리구가 월등히 높아 대조구에 비해 $7.9\%$ 정도의 상승이 있었다. S. mutans로 면역처리된 닭으로부터 얻은 계란에서의 충치균에 대한 특이(specific) 총 IgY 함량을 분석한 결과, 항충치균 IgY는 1차 면역하고 4주정도 후에 나타나기 시작하였다. 마지막 immunization후 5주 이상 경과하더라도 specific IgY의 양이 상당히 높은 수준을 유지하는 것을 볼 수 있었다. 총 실험기간 동안의 specific IgY 함량의 평균을 보면 켈프밀 처리구는 다른 식이에서 보다 훨씬 높은 specific IgY 역가 보여주었고 대조구에 비해 특이 항체가 약 $8.5\%$ 가량 증가되어 켈프밀의 첨가는 면역처리를 통한 특수 IgY 생산에 효과적임을 나타내었다

  • Mice were fed semi-purified diets containing 0, 2, 10 and 25 ppm(mg/kg) deoxynivalenol over 8 weeks and were assessed for effects on bodyweight gain, serum immunolglobulin levels and surface immunoglobulin bearing lymphocyte ratio. 1. The rate of body-weight gain was significantly reduced (p<0.05) at the 10 and 25 ppm of DON, whereas the mice ingesting the diet containing 2 ppm DON was not. 2. IgA in serum immunolglobulin was significantly increased (P<0.05) at the 10 and 25 ppm of DON, but IgG, IgM were decreased, whereas exposure to 2 ppm DON was not change. 3. Concentration of IgA from Peyer's patch of mice fed DON exhibited increased at 10, 25 ppm. 4. Lymphocytes surface marker studies revealed that IgA, IgG and IgM were 2.2%, 0.4% and 1.5% respectively. These results suggest that dietary exposure to DON alters regulation of IgA production

다운증후군 환자의 치아우식증과 치아우식 원인균 및 타액내 면역항체의 연관성 (THE DENTAL CARIES, CARIOGENIC MICROORGANISMS AND LEVELS OF SALIVARY IMMUNOGLOBULIN IN SUBJECTS WITH DOWN'S SYNDROME)

  • 다운증후군 환자의 치아우식증과 구강내 여러 요인들과의 연관성을 평가하고자 9-18세의 다운증후군 환자 25명과 정상인 63명을 대상으로 치아우식경험지수, 치태지수를 조사하고 효소면역분석법을 이용하여 타액의 면역항체 IgA와 뮤탄스 연쇄상구균-특이 IgA농도를 측정하고, Dentocult 키트를 이용하여 뮤탄스 연쇄상구균과 유산간균의 수, 타액의 완충능을 측정하여 두 군의 각 값들을 독립 t-test와 카이제곱 검정을 시행하였다. 다운증후군 환자에서 유치우식경험은 정상인에 비해 유의하게 낮았으나(p<0.001), 영구치우식경험은 유의한 차이를 보이지 않았다. 치태지수와 타액의 IgA의 농도는 두 군간에 차이를 보이지 않았으며, 뮤탄스 연쇄상구균 특이-IgA 농도와 타액 완충능은 다운증후군환자에서 낮았다(p<0.05). 뮤탄스 연쇄상구균 수와 유산간균 수는 두 군간에 유의한 차이는 없었다. 9-11세 군에서 뮤탄스 연쇄상구균의 수는 다운증후군 환자에서 정상인보다 낮았으며(p<0.001), 뮤탄스 연쇄상구균 특이-IgA는 다운증후군 환자에서 유의하게 낮았다(p<0.05). 유치의 치아우식경험치면지수는 완충능, 뮤탄스 연쇄상구균의 수와 유의한 상관관계를 보였다.
