The present study was conducted to investigate the antibiotic resistance of 24 Salmonella gallinarum isolated from 48 chicken samples of diagnosed fowl typhoid cases during the period from November 1998 to November 1999. And the isolates of S gallinarum were also tested for their invasion abilities to experimental infection as one of virulence tests, and the presence of virulence-related plasmid in S gallinarum isolates. The results obtained through this study were summarized as follows; 1. All of isolates from 48 cases of 24 farms were identified S gallinarum by biochemical and serological tests.2. Antimicrobial drug resistance test against 24 isolates showed that the isolates were resistant to Colistin(95.8%), and Penicillin(79.2%), Polymyxin B(75.0%), Streptomycin (65.2%), Gentamycin(54.2%), and Tetracycline(33.3%). 3. Mortality in chicken following peroral inoculation of four isolates of S gallinarum during 14-days inoculation pecked at 5 days(40%) after inoculation and all of experimental chickens died within 13 days after inoculation.4. Based on the pattern and number of isolated plasmid from each isolate, plasmid profiles were divided into five groups; group I with 3 plasmids, group II to group IV with 4 plasmids and group V with 5 plasmids.
17S-ribosomal RNA gene from the hygromycin resistant protozoan Tetrahymena thermophila hmr 3 was cloned on E. coli vector pBR 322 as part of study to work the 17S-rRNA structure and the mechanism of hygromycin resistance. The 17S-rDNA was inserted into the Hind 111 site of pBR 322. The clones having recombinant plasmid were selected by the method of colony hybridization with a 17S-rDNA probe of wild type B1868. The orientation of 17S-rDNA insert was located near the tetracycline resistant gene of pBR 322 in a clone 5-19 with the recombinant plasmid.
The chromosomal DNA of Klebsiella penumoniae KNF1 was partially digested with HindIII. pBR322 was completely digested with same enzyme with additional alkaline phosphatase treatment. Both DNA samples were ligated and transformed into E. coli KO60. Single coloneis of the transformed cells were isolated on N0free agar media. These colonies were ampicillin-resistant and tetracycline-sensitive. When the plasmids in transformants were cured, nitrogen-fixing activities were lost. Therefore, these transformants harbored recombinant plasmid including nif genes of K. pneumoniae. Nitrogenase activity of transformant was higher than or the same as that of K. pneumoniae KNF1.

The strain capable of growing on minimal medium containing aniline as a sole source of carbon was isolated from activated sludges and identified as Pseudomonas putida biotype A. The characterizations of the strain were determined. The optimum concentration for growth of the strain was 1-20 mM of aniline. No changes of pH were detected during cultivation. Some metabolic products of biodegradation of aniline were detected after cultivation of the strain on 10 mM aniline for 48 hours. The strain showed to be resistant to streptomycin, tetracycline, trimethoprim, and sulfanilamide. The strain was also capable of utilizing other aromatic compounds related to aniline as a sole source of carbon. One plasmid carried by this strain was detected. The properties of some of the mutant strains treated with mitomycin C were also discussed. The results suggest that separate, regulatory enzyme systems capable of degrading aniline may exist in plasmid DNA.

Shigella strains isloated in the Teagu area during the period from 1973 to 1985 were studied for species distribution, drug resistance, and R plasmids. Approximately 1,200 strains were isolated during this period, and most of them were classified into Shigella flexneri, S. sonnei occupied less than 20%, and S. dysenteriae and S. boydii were very rarely isolated. More than 95% of them were resistant to one or more of these drugs; chloramphenicol (Cm), tetracycline (Tc), streptomycin (Sm), sulfisomidine (Su), ampicillin (Ap), and trimethoprim (Tp). Strains resistant to kanamycin, nalidixic acid (Na), and rifampin (Rf) were rare, and no strain was resistant to cephaloridine, gentamicin, and amikacin. Approximately half of the isolates were resistant to drugs in 1973, but the rate of resistant strains increased to more than 95% from 1977. Strains resistant to the four drugs (Cm, Tc, Sm, and Su) occupied the majority of resistant strains until 1977, but the most prevalent multiplicity of drug resistance increased to six drugs (Cm, Tc, Sm, Su, Ap, and Tp) from 1978 with the marked increase of Ap- and Tp-resistant strains. Approximately 75% of them transferred resistance to Escherichia coli by conjugation, and the resistance was considered to be mediated by R plasmids. Almost all of them transferred the complete patterns of resistance to drugs except Na and Rf. However, among some strains of recent isolates, small numbers of segregants of transferred resistance were observed. The R plasmids in Shigella were mostly classified into Inc FII, and only small numbers into Inc B. Segregants were in most cases unclassified.
Forty gentamicin-resistant isolates of Enterococcus faecalis were selected from various clinical materials, determined their antimicrobial susceptibility, and studied there R-plasmid characteristics and polypeptide patterns. All of the isolates were susceptible to vancomycin. The MICs($\mu$/ml) of antimicrobial agents to the isolates were as follows; the MIC of gentamicin was 128 and $\geq$2040, ampicillin 1 and 1, chlorarmphenicol 2 and 8, erythromycin 32 and 256, and vancomycin 1 and 2. E. faecalis HL-1 strain had 8 plasmid DNA elements, HL-2 and HL-3 strains had 6, HL-4 had 7, HL-5 had 4, and HL-6 had 5. The 51.7 Kb of gentamicin resistance plasmid DNA was conjugally transferred from two strains of E. faecalis HL-1 and HL-6 to S. aureus SK 982. The plasmid transfer frequency between S. aureus SK 982 and E. faecalis HL-1 or E. faecalis HL-6 was 6.3$\times10^{-4} and 3.7$\times10^{-5}$, respectively. Plasmid curing ratio after the treatment of ethidium bromide(10$\mu$/ml) to E. faecalis tarnsconjugants R-1 and R-6 were about 51% and 67%, respectively. The tetracycline gene was located in 2.15 Kb plasmid of E. faecalis HL-1, but it was not found in the E. faecalis HL-6 by Southern blot analyses. The antigenic components of E. faecalis HL-1, HL-6, R-1 and R-6 strains were analyzed by SDS-PAGE and immunoblotting. The E. faecalis strains had 7 to 16 polypeptide bands, however their major proteins were 97.8 and 26.8 Kd. At the Immunoblotting, 97.8, 95.8, 74.8, 63.5, 33.7 and 26.8 Kd polypeptides of the strains showed major antigenic activities with patient's sera infected intra-abdominally with an E. faecalis strain.
The incidence of verotoxin-producing Escherichia coli(VTEC) was surveyed in domestic foods including hamburger, raw meats and vegetables from 1997 to 1999. The molecular biological characteristics of the isolates were analyzed. Three VTEC strain were isolated from 1,700 samples. Serotypes of those isolates were 0157 : H7, 026 H4, and 056 : Hl2, respectively. Serotype O26 : H4 produced VT I and VT II, and 055 Hl2 isolate produced VT I, however the 60 MDa plasmid DNA and eae gene were not found from both strains. One 0157 : H7 isolate produced VT II and harbour 60 MDa plasmid DNA, however eae gene was not found in the strain. Although they produced VT, it seemed that the virulence of two strains were relatively weak because of the lack of the eae gene. In addition, the serotype O157 : H7 isolate resistant to ampicillin and streptomycin, while isolates of serotype O26 : H4 and O55 : Hl2 were multi-resistant to antibiotics including ampicillin, carbenicillin , cephalothin, trimethoprim/sulfamethoxazole and tetracycline. The antimicrobial susceptibility, genotypes of enterotoxins(LT, STa) and pili(K88, 987P), and plasmid profiles were investigated with 102 Escherichia coli isolated from piglets showing diarrhea in Korea. Almost of them were susceptible to ceftiofur(99%), cefquinone(97.1%). However they showed resistance to bacitracin(100%), streptomycin(98%), vancomycin(97%), trimethoprim/sulfamethoxazole(87.2%), tetracycline(84.3%) in antimicrobial susceptibility test. Moreover, all of the isolates demonstrated resistance to more than 2, and 78% of them were resistant to more than 8 of total 17 drugs. Multiplex PCRs for genotyping of enterotoxins(LT, STa) and pili(K88, 987P) were established with primers designed based on sequences from Genebank. Seventeen strains(16.6%) of the isolates had STa gene, 11 strains(10.8%) of them had both STa and LT genes, and 18 strains(17.8%) had K88 gene. But none of the isolates harbored a gene exclusively encoding LT. The gene encoding 987P pili was not found in all isolates. Fifty-four strains of 102 isolates(52.9%) had plasmid with various sizes ranging from 125kb to 1.1kb. Numbers of plasmid per isolates were also various, from 1 to 9. Distinctive relationship between plasmid profiles and genotypes of enterotoxins and pili in the isolates was not found. These results might provide the basic knowledge to establish strategies for the treatment and prevention of colibacillosis in piglets.
Forty-seven Salmonella Typhimurium (33 zoonotic, 14 clinical) strains were tested for antimicrobial resistance using the standard disk diffusion method. The presence of relevant resistance genes and class 1 integrons were investigated by using PCR. Pulsed-field gel electrophoresis (PFGE) and plasmid profiling were carried out to determine the genomic diversity of Salmonella Typhimurium. Approximately 57.4% of the S. Typhimurium strains were multidrug resistant (MDR) and showed high resistance rates to tetracycline (70.2%), sulfonamides (57.4%), streptomycin (53.1%), ampicillin (29.7%), nalidixic acid (27.6%), kanamycin (23.4%), chloramphenicol (21.2%), and trimethoprim (19.1%). Resistance towards cephalosporins was noted for cephalothin (27.6%), cephradine (21.2%), amoxicillin clavulanic acid (17.0%), and cephalexin (17.0%). Resistance genes, $bla_{TEM}$, strA, aadA, sul1, sul2, tetA, tetB, and tetC, were detected among the drug-resistant strains. Thirtythree strains (70.2%) carried class 1 integrons, which were grouped in 9 different profiles. DNA sequencing identified sat, aadA, pse-1, and dfrA genes in variable regions on class 1 integrons. Thirty-five strains (74.4%) were subtyped to 22 different plasmid profiles, each with 1-6 plasmids (2.0 to 95 kb). PFGE subtyped the 47 strains into 39 profiles. In conclusion, high rates of multidrug resistance were found among the Malaysian Salmonella Typhimurium strains. The emergence of multidrug-resistant Salmonella Typhimurium to cephalosporin antibiotics was also observed. The strains were very diverse and no persistent clone was observed. The emergence of MDR Salmonella Typhimurium is a worldwide problem, and this report provides information for the better understanding of the prevalence and epidemiology of MDR S. Typhimurium in Malaysia.

Multiply resistant Shigella strains isolated in Taegu area were subjected for the characterization of R plasmids. All strains isolated in 1984 and 1985 were susceptible to gentamicin, amikacin, and cephalothin, and most strains were susceptible to kanamycin (Km) and rifampin by agar dilution antimicrobial susceptibility test. The resistance frequency of S. flexneri against ampicillin (Ap) was higher than that of S. sonnei. The strains resistant to sulfisomidine (Su) and trimethoprim (Tp) were found at higher frequency in S. sonnei than in S. flexneri. The most prevalent resistance pattern of S. flexneri was chloramphenicol (Cm) tetracycline (Tc) streptomycin (Sm) Ap, followed by the pattern of CmTcSmSuApTp, CmTcSmSuApTp nalidixic acid, and CmTcSmSuAp in the decreasing order. The antibiogram of CmTcSmSuTp was found to be the most frequent pattern in S. sonnei. The ratio of conjugal transfer of S. flexneri was 47% and 75% of S. sonnei. The average number of plasmid harboring in Shigella was 4 and the size of plasmid ranged 1.3 to 134 megadalton (Mdal). Most S. flexneri carried plasmids of 2 to 3 Mdal and S. sonnei carried those of 3 to 4 Mdal size. The sizes of conjugative plasmids ranged 40-90 Mdal. The incompatibility group (Inc) F II plasmids (54-59 Mdal) were most frequent and rare Inc B plasmids (60 Mdal) of isolates in 1979 and 1980 and Inc FI (87 Mdal) of 1983 isolates were able to be classified by the colony test with standard reference plasmids. The R plasmids of known Inc group were tested for the restriction endonuclease analysis. The pattern of plasmids digested by EcoRl were apparently different by the Inc group but there was no significant difference between species or by the resistance patterns. Nonconjugative plasmids and their phenotypes were identified by transformation test. The transformants were resistant to less than two drugs. Colicin producing transformants carried the Col plasmid of 3.7 or 3.9 Mdal size. $Ap^r$ plasmids derived from S. sonnei were found to be mobilized by transfer factor RT641 to E. coli #CS100. $Ap^r$ plasm ids of same size shared by S. flexneri, S. sonnei, and E. coli were digested with Pstl. All of them showed two restriction fragments of 2.8 kilobase(kb) and 0.7kb. Other plasmids ($Sm^r\;Su^r$) derived from S. flexneri, S. boydii, and S. sonnei were digested with Pstl and they showed same restriction fragment patterns of 3.1kb and 2.9kb. The plasmid profiles of three strains of S. sonnei producing colicin and showing same resistance pattern of CmTcSmSuApTpKm appeared to be similar. Restriction patterns by EcoRl and the behavior of plasmids in conjugation or transformation process were also similar between those plasmids. The restriction patterns were significantly different between the plasmids of Inc FI group and those of unclassified Inc group.
