김치에서 tetracycline 내성 유산균의 분리 (Isolation of Tetracycline-resistant Lactic Acid Bacteria from Kimchi)

  • 대구지역에서 수집한 50점의 김치 중에서 10점의 김치로부터 tetracycline내성 세균을 분리하였다. 이 균주들의 tetracycline에 대한 MIC는 25-100 mg/l 이상의 범위로 분포하였으며 다른 항생제에 대한 내성도 다양하였다. tet(M), tet(O)특이적 primer를 이용한 PCR에서 HJ9 한 균주에서만 tet(M)유전자가 검출되었으며, tet(M)은 플라스미드상에 존재하는 것으로 나타났다. HJ9 균주의 tet(M)부분 염기서열을 분석한 결과 기존에 보고된 Gram양성균의 tet(M)의 DNA 염기서열 및 아미노산 서열과 각각 90-99%, 94-100%의 높은 상동성을 보였다. 165 rRNA 염기서열을 분석한 결과 HJ9 균주는 Lactobacillus sakei로 동정되었다. 김치를 통하여서도 항생제 내성 유전자의 전파 확산이 가능할 것으로 생각된다.

Antimicrobial Resistance of Seventy Lactic Acid Bacteria Isolated from Commercial Probiotics in Korea

  • In this study, lactic acid bacteria were isolated from 21 top-selling probiotic products on Korean market and their antimicrobial resistance were analyzed. A total 152 strains were claimed to be contained in these products and 70 isolates belonging to three genera (Bifidobacterium, Lactobacillus, and Lactococcus) were obtained from these products. RAPD-PCR showed diversity among isolates of the same species except for two isolates of Lacticaibacillus rhamnosus from two different products. The agar dilution method and the broth dilution method produced different MICs for several antimicrobials. With the agar dilution method, five isolates (three isolates of Bifidobacterium animalis subsp. lactis, one isolate of B. breve, one isolate of B. longum) were susceptible to all nine antimicrobials and 15 isolates were multi-drug resistant. With the broth microdilution method, only two isolates (one isolate of B. breve and one isolate of B. longum) were susceptible while 16 isolates were multi-drug resistant. In this study, only two AMR genes were detected: 1) lnu(A) in one isolate of clindamycin-susceptible and lincomycin-resistant Limosilactobacillus reuteri; and 2) tet(W) in one tetracycline-susceptible isolate of B. longum B1-1 and two tetracycline-susceptible isolates and three tetracycline resistant isolates of B. animalis subsp. lactis. Transfer of these two genes via conjugation with a filter mating technique was not observed. These results suggest a need to monitor antimicrobial resistance in newly registered probiotics as well as probiotics with a long history of use.

전통발효식품에서 분리한 유산균의 항균활성 및 프로바이오틱스 기능성 연구 (Antibacterial Activity and Probiotic Properties of Lactic Acid Bacteria Isolated from Traditional Fermented Foods)

  • The aim of this study was to investigate probiotic characteristics and fermentation profile of selected lactic acid bacteria (LAB) isolated from traditional fermented foods. Antibacterial activity against various pathogens, acid and bile salt tolerance, cell hydrophobicity, and antibiotic resistance were examined. 16S rRNA sequencing was carried out to identify eight presumptive LAB isolates. In general, all identified LAB (Enterococcus faecium MG89-2, Lactobacillus plantarum MG207, L. paracasei MG310, L. casei MG311, Streptococcus thermophilus MG510, L. bulgaricus MG515, L. helveticus MG585, and L. fermentum MG590) showed strong antimicrobial activity. Also, the selected strains were resistant to bile acid up to 3% and their autoaggregation rates were as high as 60%. All selected strains tested were sensitive to chloramphenicol, tetracycline, and ampicillin, whereas resistant to nalidixic acid and kanamycin.

사료첨가용 생균제 개발을 위한 마늘 내성 유산균의 배양 조건 (Culture Conditions of Garlic Resistant Lactic Acid Bacteria for Feed Additives)

  • 파김치에서 분리한 마늘 내성 유산균인 Lactobacillus plantarum TJ-LP-002 균주의 균체생육과 항균활성에 영향을 미치는 배양조건 및 배지조건을 조사하였다. 선정 유산균의 배양 상등액 내에는 acetic acid, citric acid, lactic acid, tartaric acid와 같은 유기산이 존재하였고, 배양 중에 lactic acid와 acetic acid의 생성이 크게 증가하는 것으로 확인되었다. 단백질분해효소를 비롯한 각종 효소 처리에 의해 항균활성이 소실되지 않아, 선정 유산균이 생산하는 항균활성은 단백질성 물질이 아닌 산 생성에 의한 작용일 것으로 판단되었다. 항생제와 생균제의 병용 가능성을 확인하기 위하여 선정 유산균의 항생제 감수성을 조사한 결과, neomycin sulfate, spectinomycin dihydrochloride, lincomycin hydrochloride에 내성을 나타내었고, streptomycin sulfate에는 감수성을 나타내었으며, ampicillin trihydrate, chloramphenicol, erythromycin, tetracycline hydrochloride, kanamycin sulfate에는 중간 내성을 나타내었다. L. plantarum TJ-LP-002는 배양온도 30${^{\circ}C}$, 초기 pH 7.0, 24시간의 배양조건에서 최적의 균체생육과 항균활성을 나타내었으며, 탄소원은 glucose 3%(w/v), 질소원은 yeast extract 3%(w/v) 첨가 시에 균체생육과 항균활성이 높게 나타났다. 무기염류는 manganese sulfate와 ammonium citrate가 항균활성에 많은 영향을 주는 것으로 나타났으며, 각 성분을 단독 첨가하는 것보다 혼합 첨가하는 것이 더 우수한 영향을 나타내는 것으로 확인되었다.

  • 생균제를 제조할 목적으로 유아와 동물의 분변으로부터 내산성, 내담즙성 및 항생물질 내성균주를 분리하였다. 분리균주 중 TK-19 균주는 산에 대하여 강한 내성을 나타내었고, 500$\mu g$/ml의 담즙에서도 생육이 가능하였으며 kanamycin(30 $\mu g$/ml), streptomycin(10 $\mu g$/ml), nicin(50 $\mu g$/ml) 및 tetracycline(10 Mug/ml)에 대하여 내성을 나타내었다. TK-19 균주를 동정한 결과 Streptococcus faecium으로 판단되어 Streptococcus faecium TK-19로 명명하였다.

Selection of indigenous starter culture for safety and its effect on reduction of biogenic amine content in Moo som

  • Objective: The aims of this study were to select one strain of Lactobacillus plantarum (L. plantarum) for a potential indigenous safe starter culture with low level antibiotic resistant and low biogenic amine production and evaluate its effect on biogenic amines reduction in Moo som. Methods: Three strains of indigenous L. plantarum starter culture (KL101, KL102, and KL103) were selected based on their safety including antibiotic resistance and decarboxylase activity, and fermentation property as compared with a commercial starter culture (L. plantarum TISIR543). Subsequently, the effect of the selected indigenous safe starter culture on biogenic amines formation during Moo som fermentation was studied. Results: KL102 and TISIR 543 were susceptible to penicillin G, tetracycline, chloramphenicol, erythromycin, gentamycin, streptomycin, vancomycin, ciprofloxacin and trimethoprim (MIC90 ranging from 0.25 to $4{\mu}g/mL$). All strains were negative amino acid-decarboxylase for lysis of biogenic amines in screening medium. For fermentation in Moo som broth, a relatively high maximum growth rate of KL102 and TISIR543 resulted in a generation time than in the other strains (p<0.05). These strain counts were constant during the end of fermentation. Similarly, KL102 or TISIR543 addition supported increases of lactic acid bacterial count and total acidity in Moo som fermentation. For biogenic amine reduction, tyramine, putrescine, histamine and spermine contents in Moo som decreased significantly by the addition KL102 during 1 d of fermentation (p<0.05). In final product, histamine, spermine and tryptamine contents in Moo som inoculated with KL102 were lower amount those with TISIR543 (p<0.05). Conclusion: KL102 was a suitable starter culture to reduce the biogenic amine formation in Moo som.
