Studies on the Development of the Yogurt Decreasing Blood Glucose (혈당 강하 요구르트 개발에 관한 연구)

  • This experiment was carried out to investigate the effect of hypoglycemic yogurt as a clinical test with person both who have diabetes and who don't have diabetes. Diabetes is a disease caused by unused and accumulated glucoses produced via the foods digestion due to the lack of insulin or lower tissue reaction to insulin. Patients with diabetes or complications of diabetes are on the rise annually. For the therapy and prevention of diabetes, a yogurt made from functional materials such as insulin-resistant FK-23 lactic acid bacteria, Pinitol, edible fiber which delays glucose-absorption, extract powder of glucose-absorption-inhibitory white bean and low-caloric Erythritol and banaba extract is effective for reduce or restrain the blood glucose levels. The blood glucose levels after a meal of non-diabetic persons were lower after drinking a hypoglycemic yogurt than they after drinking general yogurt. The blood glucose levels after a meal of 10 diabetic patients were checked after drinking the yoghurt. As a result, 19 mg/dL, 30 mg/dL, 31 mg/dL of blood glucose levels decreased in 30 min, 60 min and 120 min respectively after drinking the yogurt. Blood glucose inhibition rates of 80 % of the tested patients also decreased. The blood glucose level of the diabetic patients having drunk the yogurt for 30 days consistently decreased into 59 mg/dL in 120 min after a meal.

Physiological Characteristics and ACE Inhibitory Activity of Lactobacillus zeae RMK354 Isolated from Raw Milk (원유에서 분리한 Lactobacillus zeae RMK354의 생리적 특성 및 ACE 억제능)

  • In order to develop a new starter for fermented milk, 1037 bacterial strains were isolated from raw milk. The strain that showed excellent acid producing and angiotensin converting enzyme (ACE) inhibitory activity (88.6%) was selected and identified as a Lactobacillus zeae based on the result of API carbohydrate fermentation pattern and 16S rDNA sequence. Lactobacillus zeae RMK354 was investigated further to study its physiological characteristics. It showed strong ACE inhibitory activity compared with commercial LAB starters tested. The optimum growth temperature of L. zeae RMK354 was $40^{\circ}C$ and it took 10 hr to reach pH 4.3 under this condition. L. zeae RMK354 showed more sensitive to penicillin-G, bacitracin, novobiocin, in a comparison of 14 different antibiotics, and showed most resistance to polymyxin B and vancomycin. It showed higher esterase and leucine arylamidase activities compared with 16 other enzymes. It was comparatively tolerant to bile juice and able to survive at pH 2 for 3 hr. It showed inhibitory activity against Salmonella Typhimurium with the rate of 60%. Based on these and previous results, L. zeae RMK354 could be an excellent starter culture for fermented milk with high level of ACE inhibitory activity.

Effects of Dietary Fiber from Mozuku (Cladosiphon novae-caledoniae kylin) Residue on Antioxidant Activity and Anticancer in HT-29 Human Colon Cancer Cells according to Extraction Condition (추출법에 따른 큰실말(Cladosiphon novae-caledoniae kylin) 부산물 식이섬유의 항산화 효과 및 HT-29 대장암 세포에 대한 항암 작용)

  • In this study, the dietary fiber contents of Mozuku (Cladosiphon novae-caledoniae kylin) residue and the extraction condition (HCl, $H_2SO_4$, NaOH, $Na_2CO_3$, $Na_2EDTA$) of the dietary fiber was investigated. We examined that the contents of the total polyphenols and flavonoids in the dietary fiber from Mozuku residue, and the potent anti-cancer effect was also tested through the growth inhibition in human colon cancer cells (HT-29) in vitro. It was effective to extract soluble dietary fiber with 1.5% $Na_2EDTA$ and 0.05 N HCl in Mozuku residue. The extraction time and temperature affected the yields of soluble dietary fiber. The contents of the total polyphenols and flavonoids in the dietary fiber from Mozuku residue were the highest in 1% NaOH extract (Total polyphenols $34.4{\pm}0.055$ mg gallic acid/g dry basis, total flavonoids $34.7{\pm}0.023$ mg naringin/g extract dry basis). In DPPH radical scavenging activity, 1% NaOH extract showed the most potent antioxidant activity. In the result of viability in human colon cancer cells, growth inhibition was observed in D.W., 0.05 N HCl, and 0.5% $Na_2CO_3$ extracts in a dose-dependent manner. These results demonstrated that soluble dietary fiber from Mozuku residue significant antioxidant activity and anticancer in human colon cancer.

Effect of Fermented Garlic Extract on Adhesion of THP-1 Cells to Bovine Aortic Endothelial Cells (혈관내피세포에 부착하는 THP-1에 대한 발효마늘추출물의 효과)

  • Fermented garlic extract (FGE) is a well-known functional food containing cycloalliin. Here we tested if FGE has an effect on the vascular system. We found that FGE had an effect on monocytic adhesion to endothelial cells, which occurs at the initial step of processes of inflammation. From dose-dependent experiments, 0.1 to $1{\mu}g/ml$ of FGE appeared to inhibit lipopolysaccharide (LPS)-enhanced heterotypic cell adhesion between THP-1 and BAECs. This result indicates that FGE blocks vascular inflammation. Then we found that FGE activates eNOS and Akt in BAECs. The phosphorylation of eNOS was maximally elevated 10 min after FGE treatment. Parallely, the phosphorylation of Akt was also maximally increased 10 min after FGE treatment. Consistently, it was found that FGE enhanced the production of nitric oxide. We then examined whether NO mediates THP-1 cell adhesion to BAECs. Both Akt and eNOS inhibitors appeared to reverse an inhibitory effect of FGE. These findings indicate that FGE inhibits LPS-enhanced heterotypic cell adhesion via Akt and eNOS. In conclusion, FGE plays an important role in prevention of inflammatory diseases.

Seismic Performance and Flexural Over-strength of Hollow Circular RC Column with Longitudinal Steel Ratio 2.017% (축방향철근비 2.017%인 중공 원형 RC 기둥의 내진성능과 휨 초과강도)

  • Three small scale hollow circular reinforced concrete columns with aspect ratio 4.5 were tested under cyclic lateral load with constant axial load. Diameter of section is 400 mm, hollow diameter is 200 mm. The selected test variable is transverse steel ratio. Volumetric ratios of spirals of all the columns are 0.302~0.604% in the plastic hinge region. It corresponds to 45.9~91.8% of the minimum requirement of confining steel by Korean Bridge Design Specifications, which represent existing columns not designed by the current seismic design specifications or designed by seismic concept. The longitudinal steel ratio is 2.017%. The axial load ratio is 7%. This paper describes mainly crack behavior, load-displacement hysteresis loop, seismic performance such as equivalent damping ratio, residual displacement and effective stiffness and flexural over-strength of circular reinforced concrete bridge columns with respect to test variable. The regulation of flexural over-strength is adopted by Korea Bridge Design Specifications (Limited state design, 2012). The test results are compared with nominal strength, result of nonlinear moment-curvature analysis and the design specifications such as AASHTO LRFD and Korea Bridge Design Specifications(Limited state design).

  • The proposed algorithm in this paper is based on the wavelet decomposition and the energy computation of composed blocks so the amount of calculation and complexity is minimized by adaptively replacing the DWT coefficients and managing the resources effectively. We are now living in the world of a lot. of multimedia applications for many digital electric appliances and mobile devices. Among so many multimedia applications, the digital image compression is very important technology for digital cameras to store and transmit digital images to other sites and JPEG2000 is one of the cutting edge technology to compress still images efficiently. The digital cm technology is mainly using the digital image compression features so that those images could be efficiently saved locally and transferred to other sites without any losses. JPEG2000 standard is applicable for processing the digital images usefully to keep, send and receive through wired and/or wireless networks. The discrete wavelet transform (DWT) is one of the main differences to the previous digital image compression standard such as JPEG, performing the DWT to the entire image rather than splitting into many blocks. Several digital images m tested with this method and restored to compare to the results of conventional DWT which shows that the proposed algorithm get the better result without any significant degradation in terms of MSE & PSNR and the number of zero coefficients when the energy based adaptive DWT is applied.

  • Experiments using real guided weapons for the development of the LADAR(Laser radar) are not practical. Therefore, we need computing environment that can simulate the 3D detections by LADAR. Such simulations require dealing with large sized data representing buildings and terrain over large area. And they also need the information of 3D target objects, for example, material and echo rate of building walls. However, currently used 3D models are mostly focused on visualization maintained as file-based formats and do not contain such semantic information. In this study, as a solution to these problems, a method to use a spatial DBMS and a 3D model suitable for LADAR simulation is suggested. The 3D models found in previous studies are developed to serve different purposes, thus, it is not easy to choose one among them which is optimized for LADAR simulation. In this study, 4 representative 3D models are first defined, each of which are tested for different performance scenarios. As a result, one model, "Body-Face", is selected as being the most suitable model for the simulation. Using this model, a test simulation is carried out.

Shear-Rate Dependent Ring-Shear Characteristics of the Waste Materials of the Imgi Mine in Busan (부산 임기광산 광미의 전단속도에 따른 링 전단특성 연구)

  • Abandoned mine deposits are exposed to various physico-chemical geo-environmental hazards and disasters, such as acid mine drainage, water contamination, erosion, and landslides. This paper presents the ring shear characteristics of waste materials. The ring shear box with a rotatable O-ring was used in this study. Three tests were performed: (i) Shear stress-time relationship for given normal stress and shear speed, (ii) shear stress as a function of shear speed, and (iii) shear stress as a function of normal stress. For a given normal stress (50 kPa) and speed (0.1 mm/sec), the materials tested exhibit a strain softening behavior, regardless of drainage condition. The peak and residual shear stresses were determined for each normal stress and shear speed. The shear stress was measured when shear speed is equal to 0.01, 0.1, 1, 10, 50, 100 mm/sec or when normal stress is equal to 20, 40, 60, 80, 100, 150 kPa. From the test results, we found that the shear stress increases with increasing shear speed. The shear stress also increases with increasing normal stress. However, different types of shearing mode were observed in drained and undrained conditions. Under drained condition, particle crushing was observed from the shearing zone to the bottom of lower ring. Under undrained condition, particle crushing was observed only at the shearing zone, which has approximately 1 cm thick. It means that a significant high shear speed under undrained condition can result in increased landslide hazard.

An Empirical Study on Service Quality and Patient Satisfaction in Specialty and General Hospitals (전문병원과 일반병원의 서비스의 질과 환자만족도에 관한 실증적 분석)

  • The objective of this study is to examine the effectiveness of the strategy of hospital specialization by analyzing the differences in expected and perceived services, perceived service quality, satisfaction, and intentions to revisit and recommend the hospital to others between general and specialty hospitals. Data were collected using self-administered questionnaire from patients admitted to four study hospitals: two speciality and two general hospitals. The questionnaire was developed based on SERVQUAL to measure five dimensions of service quality. Four hundreds questionnaires were distributed to inpatients or their guardians and 282 returned questionnaires were used in the analyses. The significance of the differences in study variables between specialty and general hospitals were tested by t-test and $x^2$-test. The factor analysis result confirmed the construct validity of 28 questions asked to measure service quality and resulted in four dimensions of service quality: reliability, assurance, tangible and empathy/responsiveness. Cronbach's Alpha ranged from .9013 to .9358, that confirmed the internal consistency of answers. The study results indicated that patients who used specialty hospitals had higher levels of expected and perceived service, a higher level of perceived service quality, and higher levels of service satisfaction than patients who used general hospitals. Percents of patients who had the intention to revisit the hospital and to recommend the hospital to others were higher among patients in specialty hospitals. The most frequent reason to choose the hospital was the excellence of doctors in both general(29.9%) and specialty(43.8%) hospitals, that was followed by convenient transportation(15.3%) and someone know works at the hospital(15.3%) in general hospitals and other's recommendation(14.6%), and nice amenities(13.1%) in specialty hospitals. Although there were no significant differences in clinical department, age, and sex of patients between general and specialty hospitals, patients who visited speciality hospitals had higher levels of education and income than their counter part in general hospitals. These results suggested that specialty hospitals performed better than general hospitals. Specialization could be a viable strategy to tide over recent financial difficulties experienced by hospitals, particularly small- and medium-sized hospitals.

Optimal Culture Conditions and XAD Resin on Tropane Alkaloid production in Scopolia parviflora Hairy Root Cultures (미치광이풀 모상근의 배양조건 구명 및 XAD Resin 처리에 의한 Tropane Alkaloid 생산)

  • The optimum culture conditions for tropane alkaloid production in hairy root cultures of Korea native Scopolia paviflora Nak. were investigated. Hairy root was induced from the rhizome of the mother plant on B5 medium containing 1.0 mg/L IBA. Among the culture media examined, 1/2 B5 medium was the best for tropane alkaloid production, whereas the growth of hairy root increased in SH medium. The best result on the growth of hairy root was obtained in 1.0 mg/L NAA, and tropane alkaloid production was obtained in plant growth regulator-free medium. Of the carbone sources tested, 3% sucrose promoted the growth of hairy root, whereas 5% sucrose increased tropane alkaloid production. Optimum inoculum densities for root growth and tropane alkaloid production were 0.5 g and 1 g, respectively. The addition of XAD resins (1 % w/v) to hairy root cultures led to increases in tropans alkaloid production, and the release of alkaloid into the medium and its adsorption by the resin accounted for about 50 to 80% of total production. It is concluded that optimized culture conditions and the addition of XAD resins could be used in the development of a bioprocess for tropane alkaloid production in hairy root cultures of S. paviflora Nak.
