Enhanced Production of Bacterial Cellulose in Komagataeibacter xylinus Via Tuning of Biosynthesis Genes with Synthetic RBS

  • Bacterial cellulose (BC) has outstanding physical and chemical properties, including high crystallinity, moisture retention, and tensile strength. Currently, the major producer of BC is Komagataeibacter xylinus. However, due to limited tools of expression, this host is difficult to engineer metabolically to improve BC productivity. In this study, a regulated expression system for K. xylinus with synthetic ribosome binding site (RBS) was developed and used to engineer a BC biosynthesis pathway. A synthetic RBS library was constructed using green fluorescent protein (GFP) as a reporter, and three synthetic RBSs (R4, R15, and R6) with different strengths were successfully isolated by fluorescence-activated cell sorting (FACS). Using synthetic RBS, we optimized the expression of three homologous genes responsible for BC production, pgm, galU, and ndp, and thereby greatly increased it under both static and shaking culture conditions. The final titer of BC under static and shaking conditions was 5.28 and 3.67 g/l, respectively. Our findings demonstrate that reinforced metabolic flux towards BC through quantitative gene expression represents a practical strategy for the improvement of BC productivity.

Fatigue Strength of In-plane Welded Attachments (면내 거셋 용접연결부의 피로강도)

  • This s paper presented the results of the three phases of fatigue tests to determine the fatigue strength of in-plane welded gusset joints, which are the most common details inevitably existing in the region of high stress range. A total of 57 fatigue specimens with varying thickness and strengths were made and tensile fatigue tests performed. One full-scale beam fatigue test was also performed. The validity of the fatigue strength of those details in the specification was confirmed, with the effects of thickness of flanges and welded attachments, length of attachments, and strength of applied steel examined. The characteristics of crack initiation and propagation were also observed. The test result was evaluated by comparing it with other test data and fatigue criteria of other countries. To determine the degree of stress concentration in the weld toe depending on geometric configuration such as attachment length and transition radius, analyses were performed. Compared to the present specification, analytical results indicate the need to revise and subdivide the detail categories.

  • In this study, we investigated the applicability of PCC(precipitated calcium carbonate) as a raw material for the manufacture of duplex-board. Papers were made with white-ledger stock collected from the actual duplex-board mill and PCC in a laboratory, and paper properties including bulk, ash content, tensile strength, burst strength, brightness and opacity were measured. The effect of PCC on the drying energy of duplex-board was also determined by measuring the moisture content of wet web and calculating drying energy reduction. PCC increased bulk and ISO brightness significantly, which means PCC can decrease the use of virgin pulp and recycled fibers. PCC decreased the moisture content of wet web, which means PCC can decrease drying energy consumption. However, paper strength decreased as addition levels of PCC addition increased. Therefore, the addition level of PCC must be determined considering the reduction of paper strengths.

Development of Microstructures and Mechanical Properties in Differential Speed Rolled Ni-30Cr Alloy (이주속압연된 Ni-30Cr 합금의 미세조직과 기계적 특성 발달)

  • We evaluated the developed microstructures and mechanical properties of a severely plastically deformed Ni-30Cr alloy. Normal rolling and differential speed rolling were used as deformation processes, and the thicknesses of the specimens were reduced to 68 % of the original thickness after holding at $700^{\circ}C$ for 10 min and annealing at $700^{\circ}C$ for 40 min to obtain a fully recrystallized microstructure. Electron backscattering diffraction was used to analyze the characteristic distribution of the grain boundaries on the deformed and annealed specimens. Differential speed rolling was more effective for refining grains in comparison with normal rolling. The grain size was refined from 33 mm in the initial material to 8.1 mm with normal rolling and 5.5 mm with differential speed rolling. The more refined grain in the differential-speed-rolled material directly resulted in increases in the yield and tensile strengths by 68 % and 9.0%, respectively, compared to normal rolling. We systematically explain the relationship between the grain refinement and mechanical properties through a plastically deformed Ni-30Cr alloy based on the development of a deformation texture. The results of our study show that the DSR process is very effective when used to enhance the mechanical properties of a material through grain refinement.

The Wetting and Interfacial Reaction of Vacuum Brazed Joint between Diamond Grit(graphite) and Cu-13Sn-12Ti Filler Alloy (다이아몬드 grit(흑연) / Cu-13Sn-12Ti 삽입금속 진공 브레이징 접합체의 젖음성 및 계면반응)

  • Various alloy system, such as Cu-Sn-Ti, Cu-Ag-Ti, and Ni-B-Cr-based alloy are used for the brazing of diamond grits. However, the problem of the adhesion strength between the diamond grits and the brazed alloy is presented. The adhesion strength between the diamond grits and the melting filler alloy is predicted by the contact angle, thereby, instead of diamond grit, the study on the wettability between the graphite and the brazing alloy has been indirectly executed. In this study, Cu-13Sn-12Ti filler alloy was manufactured, and the contact angles, the shear strengths and the interfacial area between the graphites (diamond grits) and braze matrix were investigated. The contact angle was decreased on increasing holding time and temperature. The results of shear strength of the graphite joints brazed filler alloys were observed that the joints applied Cu-13Sn-12Ti alloy at brazing temperature $940^{\circ}C$ was very sound condition indicating the shear tensile value of 23.8 MPa because of existing the widest carbide(TiC) reaction layers. The micrograph of wettability of the diamond grit brazed filler alloys were observed that the brazement applied Cu-13Sn-12Ti alloy at brazing temperature $990^{\circ}C$ was very sound condition because of existing a few TiC grains in the vicinity of the TiC layers.

Study on the Characteristics of the Hybrid Parylene Thin Films (하이브리드 타입 패럴린의 박막 특성 연구)

  • The mechanical properties and surface characteristics of parylene thin film were improved using Xylydene-based dimers (DPX-C, DPX-D, and DPX-N). A single-parylene-C, D, N film and a hybrid chemical and physical parylene thin films in which two types are mixed were manufactured for each dimer by adjusting the deposition conditions and the thickness of the thin film by input. Parylene was deposited by chemical vapor deposition (CVD) and the thermal characteristics of the single thin film and the hybrid thin film were compared by thermal analysis. The mechanical properties of the thin films were characterized by tensile strength, elongation, and tear force tests, and the surface characteristics of the thin films were evaluated by contact angle and surface energy measurements. The hybrid chemical parylene thin film in which two types are mixed can complement the strengths and weaknesses of the different dimers, while the physical parylene thin film can freely adjust the thin film characteristics of the coated surface and the opposite surface.

Bond Strength of Band on Zirconia Crown with Compomer and Resin Cement (지르코니아 기성관에 컴포머와 레진 시멘트로 접착한 교정 밴드의 결합력)

  • The aim of this study was to evaluate the compomer cement and resin cement as an orthodontic band cement on zirconia crown. A total of 30 specimens were prepared. Preformed stainless steel crowns and zirconia crowns of upper right second primary molar were used. Orthodontic bands were cemented on stainless steel crowns (Group I, n = 10) and zirconia crowns (Group II, n = 10) with compomer cement. The other bands were cemented on zirconia crowns with resin cement (Group III, n = 10). The tensile loads were applied to band to measure the bond strength. The mean of bond strengths of group I, II and III were 0.79 MPa, 1.09 MPa and 1.56 MPa respectively. Bond strength of group II is significantly higher than group I. There was no significant difference between group II and III. Compomer cement and resin cement containing functional monomers showed favorable bond strength of orthodontic bands.

  • The precast concrete (PC) method has many advantages in fast construction, quality control, etc. In domestic construction market, however, its application has been quite limited because of the concerns about structural integrity and seismic performances due to the discrete connections between precast concrete members. By applying the post-tensioning method, the precast beam-column connection can be well tightened, allowing improved structural integrity, and proper seismic performances can be also achieved. In this study, reversed cyclic tests have been conducted on the beam-column connection specimens, where the test variables included the compressive strength of grouting mortar and the tensile strengths of prestressing strands, based on which their seismic performances have been examined in detail. The post-tensioned PC beam-column connections showed good seismic performances comparable to that of the monolithic reinforced concrete connection specimen. When 2400 MPa prestressing strands are applied to the beam-column connection, it is preferable to adjust the prestress level similar to that applied for the 1860 MPa prestressing strands to avoid premature local crushing failures at the beam-column connections.

A Study on the Mechanical Properties of Duplex Stainless Steel Weldment According to Mo Contents

  • This study investigated changes in phase fraction caused by the addition of Mo, as well as the subsequent behaviour of N and its effect on the mechanical properties of welded 24Cr-N duplex stainless steel weld metals. Filler metal was produced by fixing the contents of Cr, Ni, N, and Mn while adjusting the Mo content to 1.4, 2.5, 3.5 wt%. The delta ferrite fraction increased as the Mo content increased. In contrast, the ${\gamma}$ fraction decreased and changed from a round to an acicular shape. Secondary austenite (${\gamma}^{\prime}$) was observed in all specimens in a refined form, but it decreased as the Mo content increased to the extent that it was nearly impossible to find any secondary austenite at 3.5 wt% Mo. Both tensile and yield strengths increased with the addition of Mo. In contrast, the highest value of ductility was observed at 1.41 wt% Mo. At all temperatures, impact energy absorption showed the lowest value at 3.5 wt% Mo, at which the amount of ${\delta}$-ferrite was greatest. There was no significant temperature dependence of the impact energy absorption values for any of the specimens. As the fraction of ${\gamma}$ phase decreased, the amount of N stacked in the ${\gamma}$ phase increased. Consequently, the stacking fault energy decreased, while the hardness of ${\gamma}$ increased.

Inclusion and mechanical properties of ODS-RAFM steels with Y, Ti, and Zr fabricated by melting

  • Two groups of oxide dispersion-strengthened reduced-activation ferritic/martensitic steels (A and B) were prepared by adding Y, Ti, and Zr into steels through vacuum induction melting to investigate the inclusions, microstructures, mechanical properties of the alloys. Results showed that particles with Y, Ti, and Zr easily formed. Massive, Zr-rich inclusions were found in B steel. Density of micron inclusions in A steel was 1.42 × 1014 m-3, and density of nanoparticles was 3.61 × 1016 m-3. More and finer MX carbides were found in steel tempered at 650 ℃, and yield strengths (YS) of A and B steel were 714±2 and 664±3.5 MPa. Thermomechanical processing (TMP) retained many dislocations, which improved the mechanical properties. YSs of A and B treated by TMP were 725±3 and 683±4 MPa. The existence of massive Zr-rich inclusions in B steels interrupted the continuity of the matrix and produced microcracks (fracture), which caused a reduction in mechanical properties. The presence of fine prior austenite grain size and inclusions was attributed to the low DBTTs of the A steels; DBTTs of A650 and A700 alloy were -79 and -65 ℃. Tempering temperature reduction and TMP are simple, readily useable methods that can lead to a superior balance of strength and impact toughness in industry applications.
