Thermoresponsive Phase Transitions of PLA-block-PEO-block-PLA Triblock Stereo-Copolymers in Aqueous Solution

  • A series of PLA-PEO-PLA triblock stereo-copolymers with varying PLA/PEO and L-DL-LA ratios were synthesized via ring opening pelymerizations. Aqueous solutions of these copolymers undergo thermo-responsive phase transitions as the temperature monotonically increases. Further study shows that there is a critical gel concentration (CGC), and also lower and upper critical gel temperatures (CGTs), at which the thermo-responsive phase transition occurs. The CGC and CGTs are affected by various factors such as block length, as well as the compositions of the PLA blocks and of the additives. In particular, the changes in the phase diagram produced by varying the L-/DL-LA ratio in the PLA blocks were determined to be mainly due to consequent stereo-regularity changes in the PLA blocks.

  • Thermo-responsive polymers that exhibit phase transition in response to temperature change can be used as materials for smart windows because they can control solar light transmission depending on the outside temperature. The development of thermo-responsive polymers for smart windows that can be used over a wide temperature range is desirable. To obtain high performance smart windows materials, three-dimensional thermo-responsive poly(N-isopropylacrylamide) (PNIPAm) gels were prepared by free radical polymerization from monomer N-isopropylacrylamide, N, N'-methylenebis acrylamide (MBAm) as a crosslinking agent, ammonium persulfate (APS) as a strong oxidizing agent/tetramethylene diamine as a catalyst, and a mixture of two solvents (water/glycerol). This study examined the effect of glycerol content on the lower critical solution temperature (LCST), freezing temperature and the solar light transmittance of crosslinked PNIPAm gel films. The LCST and freezing temperature of PNIPAm gel films were found to be significantly decreased from 34.3 and $6.3^{\circ}C$ to 28.2 and $-6.5^{\circ}C$ with increasing glycerol content from 0 wt% to 10 wt%, respectively. It was found that the transparent PNIPAm gel films at $25^{\circ}C$ (temperature < LCST) were converted to translucent gels at higher temperature ($45^{\circ}C$) (temperature > LCST). These results suggested that the crosslinked PNIPAm gel materials prepared in this study could have high potential for application in smart glass materials.

Influences of ethanol and temperature on sucrose-evoked response of gustatory neurons in the hamster solitary nucleus

  • Taste-responsive neurons in the nucleus of the solitary tract (NST), the first gustatory nucleus, often respond to thermal or mechanical stimulation. Alcohol, not a typical taste modality, is a rewarding stimulus. In this study, we aimed to investigate the effects of ethanol (EtOH) and/or temperature as stimuli to the tongue on the activity of taste-responsive neurons in hamster NST. In the first set of experiments, we recorded the activity of 113 gustatory NST neurons in urethane-anesthetized hamsters and evaluated responses to four basic taste stimuli, 25% EtOH, and 40℃ and 4℃ distilled water (dH2O). Sixty cells responded to 25% EtOH, with most of them also being sucrose sensitive. The response to 25% EtOH was significantly correlated with the sucrose-evoked response. A significant correlation was also observed between sucrose- and 40℃ dH2O- and between 25% EtOH- and 40℃ dH2O-evoked firings. In a subset of the cells, we evaluated neuronal activities in response to a series of EtOH concentrations, alone and in combination with 32 mM sucrose (EtOH/Suc) at room temperature (RT, 22℃-23℃), 40℃, and 4℃. Neuronal responses to EtOH at RT and 40℃ increased as the concentrations increased. The firing rates to EtOH/Suc were greater than those to EtOH or sucrose alone. The responses were enhanced when solutions were applied at 40℃ but diminished at 4℃. In summary, EtOH activates most sucrose-responsive NST gustatory cells, and the concomitant presence of sucrose or warm temperatures enhance this response. Our findings may contribute to elucidate the neural mechanisms underlying appetitive alcohol consumption.

Identification of Fruit-specific cDNAs in a Ripened Inodorus Melon Using Differential Screening and the Characterization of on Abscisic Acid Responsive Gene Homologue

  • Eight cDNAs corresponding to fruit-specific genes were isolated from ripened melon through differential screening. Sequence comparison indicated that six of these cDNAs encoded proteins were previously characterized into aminocyclopropane-1-carboxylate (ACC) oxidase, abscisic acid, stress and ripening inducible (ASR) gene, RINC-H2 zinc finger protein, pyruvate decarboxylase, or polyubiquitin. RFS2 and RFS5 were the same clone encoding polyubiquitin. The other cDNAs showed no significant homology with known protein sequences. The ASR homologue (Asr1) gene was further characterized on the cDNA and genomic structure. The deduced amino acid sequence had similar characteristics to other plant ASR. The Asr1 genomic DNA consisted of 2 exons and 1 intron, which is similar to the structure of other plants ASR genes. The promoter region of the Asr1 gene contained several putative functional cis-elements such as an abscisic acid responsive element (ABRE), an ethylene responsive element (ERE), a C-box or DPBf-1 and 2, Myb binding sites, a low temperature responsive element (LTRE) and a metal responsive element (MRE). The findings imply that these elements may play important roles in the response to plant hormones and environmental stresses in the process of fruit development. The results of this study suggest that the expressions of fruit specific and ripening-related cDNAs are closely associated with the stress response.

Fabrication and Characterization of Thermo-responsive Nanofibrous Surfaces Using Electron Beam Irradiation (전자선 조사에 의한 온도응답성 나노섬유 표면의 제조 및 특성분석)

  • We have fabricated a novel thermo-responsive nanofibrous surfaces by grafting PIPAAm by electron beam irradiation onto poly(3-hydroxybutyrate-co-3-hydroxyvalerate)(PHBV) nanofibrous mats. The electrospun PHBV nanofiber structures revealed randomly aligned fibers with average diameter of 400 nm. Increased atomic percent of nitrogen was observed on the PIPAAm-grafted PHBV mats after electron beam irradiation determined by ESCA. The amounts of PIPAAm-grafted onto PHBV films were $6.49{\mu}g/cm^2$ determined by ATR-FTIR. The PIPAAm-grafted surfaces exhibited decreasing contact angles by lowering the temperature from 37 to $20^{\circ}C$, while ungrafted PHBV surfaces had negligible contact angle change. This result indicates that PIPAAm surfaces, which are hydrophobic at the higher temperature, became markedly more hydrophilic in response to a temperature reduction due to spontaneous hydration of the surface-grafted PIPAAm. Thermo-responsive nanofibers showed good tissue compatibility. Cultured cells were well detached and recovered from the surfaces by changing culture temperature from 37 to $20^{\circ}C$.

An EST survey of genes expressed in liver of rock bream(Oplegnathus fasciatus) with particular interests on the stress-responsive and immune-related genes

  • EST analysis was performed to identify stress-responsive and immune-related genes from rock bream (Oplegnathus fasciatus). cDNA libraries were constructed with liver and randomly chosen 624 clones were subjected to automated sequence analysis. Of 624 clones sequenced in total, approximately 15% of ESTs was novel sequences (no match to GenBank) or sequences with high homology to hypothetical/unknown genes. The bioinforamtic sequence analysis including functional clustering, homology grouping, contig assembly with electronic northern and organism matches were carried out. Several potential stress-responsive biomarker and/or immune-related genes were identified in all the tissues examined. It included lectins, ferritins, CP450, proteinase, proteinase inhibitors, anti-oxidant enzymes, various heat-shock proteins, warm temperature acclimation protein, complements, methyltransferase, zinc finger proteins, lysozymes, macrophage maturation associated protein, and others. This information will offer new possibilities as fundamental baseline data for understanding and addressing their molecular mechanism involved in host defense and immune systems of this species.

  • ZnO nanowire is known as synthesizable and good mechanical properties. And, stimuli-responsive polymer is widely used in the application of tunable sensing device. So, we combined these characteristics to make precise tunable sensing devise. In this work, we investigate the dependence of ZnO nanowire alignment and morphology on si substrate using nanosphere template with various conditions via hydrothermal process. Also, pH-temperature dependant tuning ability of nanostructure was studied. The brief experimental scheme is as follow. First, Zno seed layer was coated on a si wafer ($20{\times}20mm$) by spin coater. And then $1.15{\mu}m$ sized close-packed PS nanospheres were formed on a cleaned si substrate by using gas-liquid-solid interfacial self-assembly method. After that, zinc oxide nanowires were synthesized using hydrothermal method. Before the wire growth, to specify the growth site, heat treatment was performed. Finally, NIPAM(N-Isopropylacrylamide) was coated onto as-fabricated nanostructure and irradiated by UV light to form the PNIPAM network. The morphology, structures and optical properties are investigated by FE-SEM(Field Emission Scanning electron Microscopy), XRD(X-ray diffraction), OM(Optical microscopy), and WCA(water contact angle).

Clinical Identification of the Vertebral Level at Which the Lumbar Sympathetic Ganglia Aggregate

  • Background: The location and the number of lumbar sympathetic ganglia (LSG) vary between individuals. The aim of this study was to determine the appropriate level for a lumbar sympathetic ganglion block (LSGB), corresponding to the level at which the LSG principally aggregate. Methods: Seventy-four consecutive subjects, including 31 women and 31 men, underwent LSGB either on the left (n = 31) or the right side (n = 43). The primary site of needle entry was randomly selected at the L3 or L4 vertebra. A total of less than 1 ml of radio opaque dye with 4% lidocaine was injected, taking caution not to traverse beyond the level of one vertebral body. The procedure was considered responsive when the skin temperature increased by more than $1^{\circ}C$ within 5 minutes. Results: The median responsive level was significantly different between the left (lower third of the L4 body) and right (lower margin of the L3 body) sides (P = 0.021). However, there was no significant difference in the values between men and women. The overall median responsive level was the upper third of the L4 body. The mean responsive level did not correlate with height or BMI. There were no complications on short-term follow-up. Conclusions: Selection of the primary target in the left lower third of the L4 vertebral body and the right lower margin of the L3 vertebral body may reduce the number of needle insertions and the volume of agents used in conventional or neurolytic LSGB and radiofrequency thermocoagulation.

Intelligent and Responsive Window Opening-Closing Operation Process for Carbon Dioxide(CO2) Management of Secondary School Classroom (중등학교 교실의 이산화탄소(CO2) 관리를 위한 지능형 창호개폐 작동 프로세스)

  • The school classroom is a common living place where students spend 7 to 14 hours a day to prepare for their careers. Therefore, if the ventilation of the classroom is not properly performed, it may lead to the deterioration of learning ability due to the unclear air. The concentration of carbon dioxide in the classroom is reported to be high, and the increase in carbon dioxide concentration has a negative effect on the learner's academic performance. In this context, the purpose of this study is to propose a methodology for intelligent and responsive window opening-closing operation process that can reduce the concentration of $CO_2$ in the classroom in order to build a support space that can create an effective teaching-learning environment for adolescents. The specific objectives are as follows. First of all, we define the concept of window opening-closing operation. Secondly, twe develop the operation process of window opening-closing. Thirdly, we develop an algorithm for real-time window opening and closing (process) (Window Opening-Closing Operation Process). Finally, we verify the intelligent responsive window opening-closing operation process through developing examples of window opening-closing operation process using the parametric design program. This study is a preliminary study to develop algorithms necessary for window opening-closing operation. Based on the first-order algorithm, We simulated window opening-closing operations according to a hypothetical scenario. As a result, This study can show that the window is open and close depending on the $CO_2$ concentration, but the $CO_2$ concentration in the room is higher than outdoors. Consequentially, we suggest that it is necessary to develop an algorithm to supplement these results because window is often not working when the temperature difference between indoor and outdoor in winter is large.
