Taxonomic Studies on Korean Oaks-Quercus- by Pollen Protein Analysis (한국산 참나무속 식물의 화분단백질분석에 의한 분류학적 연구)

  • This research was conducted to determine if the obtained serological data would reveal: (1) grouping among basic Quercus species, (2) the degree of correspondence within and among any detected grouping, and (3) the comparative value of new serological technique, "rocket immunoelectrophoresis". Overall quantitative and qualitative data indicate the genus Quercus is distantly separated from the Fagus. Within the Quercus, the subgenus Cyclobalanopsis is separable from the Lepidobalanus. But the elevation of the Cyclobalanopsis to the rank of a separate genus is not warranted. Antisera produced to the species belonging to Lepidobalanus gave strong reactions with other experimental species in the same subgenus. Particularly, Q. aliena, Q. donarium, and Q. serrata are tied up each other in terms of serological similarity. Rocket immunoelectrophoresis used in this systematic research proved to be a valuable new technique for systematic serology. To obtain the degree of protein similarity (serological correspondence) this technique was used in plant systematic research and determined by sum of total rocket heights. The rocket immunoelectrophoresis illustrated the measurement of individual immunoprecipitin systems identical or partly identical to those in the reference-reaction as quantitation of all available rocket heights.t heights.

  • From Salix maximowiczii of the Korean Salix, morphological and palynological characters were investigated by stereoscopic, light, and scanning electron microscopes. Also, this study was eonducted to investigate the effect of meteorological factors on the disjunct distribution in Salix maximowiczii forests grown within the restricted region. 1. Morphological characters of S. maximowiczii clarified with the descriptions and figures. 2. The palynological description was made to the S. maximowiczii and identified distinctly species. 3. S. maximowiczii is distributed only in Baekdam and Hangyeryong valley of Mt. Sorak in Kangwon-do, and valley of height region in Kyeongseong-gun (Hamgyongbuk-do), Sinhueng-gun, and Pungsan-gun (Hamgyongnam-do) in North Korea. 4. Water factors was better than temperature factors in the influence of meteorological factors for disjunct distribution. 5. These ecological and taxonomical informations could be used in practical application for silvicultural prescription, such as erosion control forest, timber production, and development of green area in river bank.

Report on 24 unrecorded bacterial species of Korea belonging to the phylum Firmicutes

  • As an outcome of the study on the bacterial species diversity in Korea, we report 24 unrecorded bacterial species of Korea belonging to the phylum Firmicutes. The unrecorded species excavated through this study were assigned to 12 different genera of 7 families, namely Bacillus, Halobacillus, Lysinibacillus and Thalassobacillus of Bacillaceae, Brevibacillus and Paenibacillus of Paenibacillaceae, Viridibacillus of Planococcaceae, Salinicoccus and Staphylococcus of Staphylococcaceae, Enterococcus of Enterococcaceae, Lactobacillus of Lactobacillaceae, and Lactococcus of Streptococcaceae, respectively. The bacterial isolates were obtained from various ecosystems in Korea. The isolates were identified based on 16S rRNA gene sequences, and those exhibiting at least 99% sequence similarity with known bacterial species but never reported in Korea were selected as unrecorded species. The selected isolates were subjected to further taxonomic characterization including the analysis of cell shape and fine structure using electron microscope, colony color and shapes, enzyme activities and carbon source utilization. The descriptive information on the 24 unrecorded species are provided.

A report of 28 unrecorded bacterial species, phylum Bacteroidetes, in Korea

  • In order to investigate indigenous prokaryotic species diversity in Korea, various environmental samples from diverse ecosystems were examined. Isolated bacterial strains were identified based on 16S rRNA gene sequences, and those exhibiting at least 98.7% sequence similarity with known bacterial species, but not reported in Korea, were selected as unrecorded species. 28 unrecorded bacterial species belonging to the phylum Bacteroidetes were discovered from various habitats including wastewater, freshwater, freshwater sediment, wet land, reclaimed land, plant root, bird feces, seawater, sea sand, tidal flat sediment, a scallop, marine algae, and seaweed. The unrecorded species were assigned to 18 different genera in five families: Flavobacterium, Epilithonimonas, Dokdonia, Gillisia, Flavicella, Chryseobacterium, Algibacter, Aquimarina, Lacinutrix, Gaetbulibacter, Cellulophaga, Tenacibaculum, and Maribacter of Flavobacteriaceae, Dyadobacter of Cytophagaceae, Draconibacterium of Draconibacterium_f, Sunxiuqinia of Prolixibacteraceae, and Fulvivirga of Fulvivirga_f. The selected isolates were subjected to further taxonomic characterization including analysis of Gram reaction, cellular and colonial morphology, biochemical activities, and phylogenetic trees. Descriptive information of the 28 unrecorded species is provided.

Study on Radulae of Seven Species of the Family Pleuroceridae in Korea (한국산 다슬기 과 7 종의 치설 연구)

  • These studies were conducted to compare the external radulae of seven species (Semisulcospira gottschei, S. coreana, S. forticosta, S. tegulata, S. ibertina, Koreanomelania nodifila and Koreoleptoxis globus ovalis) of freshwater snails in three genera of the family Pleuroceridae in Korea. Under a microscope of 100 magnifications, observation of seven species can be distinguished sharply between three genera of Semisuicospira, Koreanomelania and Koreoleptoxis, and the morphological characters of radulae can be used as taxonomic tool in genus level of Pleuroceridae. Besides SEM photograph of the radulae based on the morphological characters in seven species can be verified a new criteria for classification of genus' unit grade in accord with firsthand drawing sketch under a microscope of 100 magnifications. But it is difficult that all of the species can be detected sharply the difference of interspecies by comparisons of radulae in the family Pleuroceridae in Korea. Therefore, the morphological characters of radulae can not be set a criteria for classification of species level.

김치유래 젖산균의 균체지방산 분석을 이용한 분류학적 연구

  • Two hundreds and thirty lactic acid bacteria, mostly isolated from Kimchi, including type strains were used for analysis of cellular fatty acids. The 230 test strains were recoverd in 7 major and 1 single clusters defined at Euclidian distance of 17.5. These aggregate taxa were equivalent to the genus Leuconostoc (aggregate group A, B, C and D), the genus Lactobacillus (aggregate group F), the genera Lactobacillus and Pediococcus (aggregate group E) and the genera Leuconostoc and Lactobacillus (aggregate group G). It is concluded as evident that FAMEs (Fatty Acid Methyl Esters) profile of cell can be used as a criterion in classification of lactic acid bacteria from kimchi. Additional comparative taxonomic studies need to be carried out on well chosen representative strains to determine the most appropriate methods of value.

AB3217-A and B, herbicidal compounds related to anisomycin from Streptomyces sp. ME-13 (Streptomyces sp. ME-13 균주가 생산하는 anisomycin계 AB3217 화합물의 제초활성)

  • During the screening of herbicidal substances from microbial secondary metabolites using photoautotrophic cells, a strain of ME-13 with strong herbicidal activity was isolated from soil. Based on the taxonomic studies, the strain was identified as Streptomyces. Two active compounds were purified from the culture broth through the column chromatographies using active charcoal, silica gel, MCI gel, and ODS HPLC. The compounds were identified as AB3217-A and B, respectively, related to anisomycin by spectroscopic methods. AB3217-A and B completely suppressed the germination of radish and barnyard grass at 25 ppm. In comparison to anisomycin, they showed the 6 times higher inhibitory activities against the growth of shoot and root of radish and barnyard grass with EC5O of around 6 ppm.

Isolation, Production, and Characterization of Protease from Bacillus subtilis IB No. 11

  • A potent protein degrading bacterium was isolated from soil samples of different environments. Polyphasic taxonomic studies and phylogenetic 16S rRNA sequence analyses led to identify the isolate IB No. 11 as a strain of Bacillus subtilis. The isolated strain was recognized to produce protease constitutively, and the maximum production (1.64 units/ml) was attained in a shake flask culture when the isolate was grown at $40^{\circ}C$, for 32 h in basal medium supplemented with starch (0.25%) and gelatin (1.25%) as sole carbon and nitrogen source, respectively. The optimum pH and temperature for the protease activity were determined to be pH 7.0 and $50^{\circ}C$, respectively. $Ca^{2+}$ and $Mn^{2+}$ enhanced remarkably the protease activity but neither showed positive effect on the protease's thermal stability. In addition, it was observed that the protease was fairly stable in the pH range of 6.5-8.0 and at temperatures below $50^{\circ}C$, and it could be a good candidate for an animal feed additive. The inhibition profile of the protease by various inhibitors indicated that the enzyme is a member of serine-proteases. A combination of UV irradiation and NTG mutagenesis allowed to develop a protease hyper-producing mutant strain coded as IB No. 11-4. This mutant strain produced approximately 3.23-fold higher protease activity (6.74 units/mg) than the parent strain IB No. 11 when grown at $40^{\circ}C$ for 32h in the production medium. The protease production profile of the selected mutants was also confirmed by the zymography analysis.

Pollen Morphology of Euphorbia Subsection Meleuphorbia (대극속 Meleuphorbia아절의 화분형태학적 연구)

  • Pollen morphological studies using light and scanning electron microscopy were conducted to determined the sub-sectional boundary and species relationship of 12 species of subsect. Meleuphorbia and related taxa. Based on the sculpturing patterns of pollen grains and UPGMA tree from numerical analyses using 8 quantitative pollen characters 12 species were divided into two types: type I, having microperforate pollen grains in E. epicyparissias, E. inermis, E. pillansii, E. obesa, E. bubalina, E. meloformis, E. nesemanii, and E. jansenvillensis, and type II, having fossulate sculpturing pattern in E. longituberculosa, E. monteiri, E. caput-medusae and E. grantii. The resulting pollen data suggest that the subsect. Meleuphorbia should not be a natural group, but closely related to the species from subsect. Anthacantha and sect. Treisia and sect. Medusea. Thus, the pollen morphological data recommended to combine above taxa as a single taxonomic group.

Analysis of Summative Evaluation Objectives in Middle School Biology based on Bloom's Revised Taxonomy of Educational Objectives (Bloom의 신 교육목표분류에 기초한 중학교 생물 영역 총괄 평가 문항의 목표 분석)

  • The purpose of this study was to find out taxonomic characteristics of objectives infered from questions of summative evaluation by analyzing those objectives with Bloom's revised taxonomy of educational objectives. 1,711 questions of midterm and final examinations collected from 25 middle schools were analyzed to classify objective of each question. The major findings of the study were as follows: first, from the analysis of objectives in the knowledge dimension, the assessment of factual knowledge was most prevailing(67.6%) in the biology summative evaluation. In the cognitive process dimension, memory assessment was most dominant(76.1%). Thus, the main objectives of evaluation leaned toward particular classes in the Bloom's revised taxonomy, which was different from the findings of earlier studies on the weights of evaluation areas. Second, the level of the objectives should be determined in consideration of the grade of middle schoolers, as their cognitive level improves with grade. In fact, however, there was no difference among three grades in characteristics of objective taxonomy.
