Symplocarpus koreanus (Araceae; Orontioideae), a new species based on morphological and molecular data

  • Symplocarpus koreanus (Araceae; Orontioideae) from Korea is described as new. Symplocarpus koreanus has long been considered to be conspecific with S. renifolius in Japan, but phylogenetic, morphological, and cytological data indicate its taxonomic distinction. Compared to S. renifolius, S. koreanus has a much smaller spathe and more spherical spadix with fewer, smaller flowers. Previous phylogenetic studies also suggested that S. koreanus is more closely related to nonthermogenic S. nipponicus than to S. renifolius in Japan. Like its nonthermogenic sister species, S. nipponicus, in Korea and Japan, S. koreanus is diploid (2n = 2x = 30), while S. renifolius in Japan is tetraploid (2n = 4x = 60). A detailed species description, geographical distribution, major morphological differences between the species and a dichotomous key to the species in eastern Asia are provided.

Comparison of Bacterial Community of Healthy and Erwinia amylovora Infected Apples

  • Fire blight disease, caused by Erwinia amylovora, could damage rosaceous plants such as apples, pears, and raspberries. In this study, we designed to understand how E. amylovora affected other bacterial communities on apple rhizosphere; twig and fruit endosphere; and leaf, and fruit episphere. Limited studies on the understanding of the microbial community of apples and changes the community structure by occurrence of the fire blight disease were conducted. As result of these experiments, the infected trees had low species richness and operational taxonomic unit diversity when compared to healthy trees. Rhizospheric bacterial communities were stable regardless of infection. But the communities in endosphere and episphere were significanlty affected by E. amylovora infection. We also found that several metabolic pathways differ significantly between infected and healthy trees. In particular, we observed differences in sugar metabolites. The finding provides that sucrose metabolites are important for colonization of E. amylovora in host tissue. Our results provide fundamental information on the microbial community structures between E. amylovora infected and uninfected trees, which will contribute to developing novel control strategies for the fire blight disease.

A report of 21 unrecorded bacterial species of Korea belonging to the phylum Bacteroidota isolated in 2021

  • During screening for indigenous prokaryotic species in Republic of Korea in 2021, a total of 21 bacterial strains assigned to the phylum Bacteroidota were isolated from a variety of environmental habitats including pine cone, seaweed, soil, sea sediment, brackish water and moss. Based on the 16S rRNA gene sequence similarity value of more than 98.7% and formation of a robust phylogenetic clade with the type strain of the closest bacterial species, it was found that the 21 strains belong to independent and recognized bacterial species. There has been no official report that the identified 21 species have been isolated in Republic of Korea up to date. Therefore, 16 species in six genera of two families in the order Flavobacteriales, two species in two genera of two families in the order Cytophagales, one species in one genus of one family in the order Chitinophagales and two species in one genus of one family in the order Sphingobacteriales are proposed as unrecorded species of the phylum Bacteroidota isolated in Republic of Korea. Their Gram reaction, colony and cell morphology, basic phenotypic characteristics, isolation source, taxonomic status, strain ID and other information are described in the species descriptions.

Oomycete pathogens, red algal defense mechanisms and control measures

  • Oomycete pathogens are one of the most serious threats to the rapidly growing global algae aquaculture industry but research into how they spread and how algae respond to infection is unresolved, let alone a proper classification of the pathogens. Even the taxonomy of the genera Pythium and Olpidiopsis, which contain the most economically damaging pathogens in red algal aquaculture, and are among the best studied, needs urgent clarification, as existing morphological classifications and molecular evidence are often inconsistent. Recent studies have reported a number of genes involved in defense responses against oomycete pathogens in red algae, including pattern-triggered immunity and effector-triggered immunity. Accumulating evidence also suggests that calcium-mediated reactive oxygen species signaling plays an important role in the response of red algae to oomycete pathogens. Current management strategies to control oomycete pathogens in aquaculture are based on the high resistance of red algae to abiotic stress, these have environmental consequences and are not fully effective. Here, we compile a revised list of oomycete pathogens known to infect marine red algae and outline the current taxonomic situation. We also review recent research on the molecular and cellular responses of red algae to oomycete infection that has only recently begun, and outline the methods currently used to control disease in the field.

Sequence Divergence and Phylogenetic Investigation of the Nymphalidae (Lepidoptera: Papilionoidea) Occurring in South Korea

  • As a first step toward understanding the divergence and relationships of the Nymphalidae (Lepidoptera: Papilionoidea) occurring in South Korea, cytochrome oxidase subunit I (COI), 16S ribosomal RNA (16S rRNA), and elongation factor-$1{\alpha}$ (EF-$1{\alpha}$) that comprise 3,501-3,716 bp were either sequenced (55 species) or the sequences were obtained from GenBank (23 species). The concatenated sequence divergence of six nymphalid subfamilies ranked in the following order: Danainae (10.3%), Satyrinae (9.5%), Limenitidinae (8.0%), Apaturinae (7.0%), Nymphalinae (6.7%), and Heliconiinae (6.2%). As has been reported in previous large scale international studies, the subfamilial relationships of (((((Limenitidinae + Heliconiinae) + (Nymphalinae + Apaturinae)) + Satyrinae) + Libytheinae) + Danainae) were also confirmed, except for the switched positions between Danainae and Libytheinae, and supported all subfamilies and tribe monophylies. Unlikely consistent phylogenetic relationships among genera within the majority of tribes in Nymphalidae, a conflicting relationship within the subfamily Apaturinae was obvious, presenting Apatura as sister to either Mimathyma or (Mimathyma + (Sephisa + (Hestina + Sasakia))), and both of these relationships are unconventional. Within the subfamily Limenitidinae, the genus Neptis was consistently revealed as a paraphyletic with respect to the genus Aldania, requiring further taxonomic investigation of the genus. Although limited, current sequence information and phylogenetic relationships are expected to be helpful for further studies.

Morphological and Molecular Characterization of Thamnocalamus falconeri Hook f. ex. Munro

  • The economy of India and so also of many Asian countries depends on bamboos and their uses are not only in domestic items but also in rural housing and raw materials to several industries and germplasm characterization is an important link between the conservation and utilization of plant genetic resources. Classical taxonomic studies of the bamboos are based on floral morphology and growth habit, which can cause problems in identification due to erratic flowering coupled with different biotic agencies and environmental factors. Identification and genetic relationships among accessions of Thamnocalamus falconeri were investigated using morphology and random amplified polymorphic DNAs (RAPD) technique. Analysis started by using 51 vegetative characters and forty two 10-mer primers that allowed us to distinguish different genotypes hailing from different eco- zones of Garhwal Himalayas (India). The selected primers (12) were used for identification and for establishing a profiling system to estimate genetic diversity. A total of 79.33% polymorphism was estimated by using 12 selected primers. The genetic similar analysis was conducted based on binary digits i.e. presence (1) or absence (0) of bands, which revealed a wide range of variability among the species whereas genetic relatedness was quite high based on vegetative characters. Cluster analysis clearly showed two major clusters for both of the markers viz. morphology and RAPD belonging to 10 accessions of T. falconeri. Two major clusters were further divided into minor clusters. Cluster based on RAPD marker showed grouping of accessions of closed locality whereas analogy was reported for vegetative traits. The RAPD technique has the potential for use in species identification and genetic relationships studies of bamboo for breeding program.

  • 지금(只今)까지 한국(韓國)에는 담배, 오이, 참외, 인삼(人參) 등(等)에서 입고병(立枯病)을 일으키는 Pythium debaryanum을 보고(報告)한 것 뿐이였으나 1975년(年) 저자(著者)들은 3종(種)의 미기록(未記錄) Pythium 균(菌)을 동정(同定)할 수 있었다. P. spinosum Sawada는 양배추에서 P. myriotylum Drechl은 강낭콩에서 그리고 P. butleri Subr는 오이, 시금치, 팥, 무우등(等)의 기주(寄主)에서 분리(分離)하였다. P. myriotylum과 P. butleri는 병원성(病原性)이 인정(認定)되었으나 P. spinosum는 병원성(病原性)이 없는 것으로 나타났다. Cornmeal agar 배지(培地)에서는 대부분(大部分)의 분리균(分離菌)이 완전(完全)한 자실체형성(字實體形成)이 되지 않았으며 몇 개의 분리균(分離菌)은 같은 배지(培地)에서 계대배양(繼代培養)함에 따라 퇴화(退化)된 자실체(字實體)를 형성(形成)하였다. 그러나 해바라기 종자(種子)를 이용(利用)한 배지(培地)에서는 모든 분리균(分離菌)이 만족(滿足)할만한 자실체(字實體)를 형성(形成)하였다.

Fatty acid analysis as a tool to infer the diet in Illinois river otters (Lontra canadensis)

  • Fatty acids (FA) have recently been used in several studies to infer the diet in a number of species. While these studies have been largely successful, most have dealt with predators that have a fairly specialized diet. In this paper, we used FA analysis as a tool to infer the diet of the nearctic river otter (Lontra canadensis). The river otter is an opportunistic predator known to subsist on a wide variety of prey including, fishes, crayfish, molluscs, reptiles and amphibians, among others. We analyzed the principle components of 60 FA from otters and 25 potential prey species in Illinois, USA. Prey species came from 4 major taxonomic divisions: fishes, crayfish, molluscs and amphibians. Within each division, most, but not all, species had significantly different profiles. Using quantitative FA signature analysis, our results suggest that, by mass, fish species are the most significant component of Illinois River otters' diet ($37.7{\pm}1.0%$). Molluscs ranked second ($32.0{\pm}0.8%$), followed by amphibians ($27.3{\pm}4.3%$), and finally, crayfish ($3.0{\pm}0.6%$). Our analysis indicates that molluscs make up a larger portion of the otter diet than previously reported. Throughout much of the Midwest there have been numerous otter reintroduction efforts, many of which appear to be successful. In regions where mollusc species are endangered, these data are essential for management agencies to better understand the potential impact of otters on these species. Our analysis further suggests that quantitative FA signature analysis can be used to infer diet even when prey species are diverse, to the extent that their FA profiles differ. Better understanding of the otter's metabolism of FA would improve inferences of diet from FA analysis.

Difference of gut microbiota composition based on the body condition scores in dogs

  • Microorganism residing in the gut has been known to have important roles in the animal body. Microbes and host microenvironment are highly related with host's health including energy metabolism and immune system. Moreover, it reported that gut microbiome is correlated with diseases like obesity in human and dogs. There have been many studies to identify and characterize microbes and their genes in human body. However, there was little information of microbiome in companion animals. Here, we investigated microbiota communities in feaces from twenty - four Beagles (aged 2 years old) and analyzed the taxonomy profile using metagenomics to study the difference among gut microbiome based on body condition score (BCS). gDNA was isolated from feaces, sequenced and clustered. Taxonomy profiling was performed based on the NCBI database. BCS was evaluated once a week according to the description provided by World Small Animal Veterinary Association. Firmicutes phylum was the most abundant followed by Bacteroidetes, Fusobacteria, Proteobacteria and Actinobacteria. That main microbiota in gut were differently distributed based on the BCS. Fusobacteria has been known to be associated with colon cancer in human. Interestingly, Fusobacteria was in the third level from the top in healthy dog's gut microbiome. In addition, Fusobacteria was especially higher in overweight dogs which had 6 scales of BCS. Species Fusobacterium perfoetens was also more abundant when dogs were in BCS 6. It implied that F. perfoetens would be positively related with overweight in dogs. These finding would contribute to further studies of gut microbiome and their functions to improve dog's diets and health condition.

  • 분류학에서 이종으로 취급되고 있는 Forsythia속의 종간교잡을 실시하여 화주장의 분리비, 양화주간의 수정양식, $F_1$의 임성, ♀♂성 기타 분류학상의 제문제제를 구명한바 그 요지는 다음과 같다. (1) 화주장은 L:S=1:1로 분리한다. (2) 이종간의 적, 부적법수분인데도 양친 L, S의 수정양식은 일 일반형예식물 동일종내의 2형화와 전혀 동일하다. (3) $F_1$의 L, S간의 수정양식도 일반형예식물과 동일하다. (4) 종간교잡은 잘되고 $F_1$의 임성도 이상없다. (5) 교잡제일식의 형태, 생리적변이는 심하다. (6) S주도 적법수분에서는 임성이 양호하고 Forsythia가 ♀, ♂ 이주라는 것은 잘못이다. (7) $F_1$에는 영양생장 및 생식기관이 빈약한 왜형이 나타난다. (8) 본실험에 사용한 두종은 이종이라기보다 동일종내의 이화형개체라고 하는 것이 타당하다.
