Prunus sargentii of Rosaceae familiy, has been reported to have radical scavenging activity and anti-inflammatory effect. On these facts, this research was conducted to evaluate pharmaceutical activities of the bark extracts P. sargentii. Free radical scavenging activities of fraction (Fr) -5${\sim}$10 isolated from P. sargentii was higher than 80% respectively at 10ppm. The superoxide dismutase (SOD)-like activity of Fr-5, 9 were about 97, 84%, respectively at 1,000 ppm. Xanthine oxidase inhibitory effect of Fr-9, 10 were about 75, 78%, respectively at 1,000 ppm. The tyrosinase inhibitory effect related to skin-whitening was 72, 68% in Fr-2, 9 isolated from Prunus sargentii R. at 1,000 ppm. Hyaluronidase inhibition activity related to the anti-inflammation effect was 98% for Fr-8 at 500 ppm. Isolation of the methanol soluble fraction from P. sargentii yielded two major phenol compounds, (-)-epicatechin and taxifolin. The structure of the compound was certainly determined by chemical analyses, as well as NMR and Mass spectroscopy. The present study was carried out in a search for new cosmetic material from the bark from P. sargentii. and, (-)-epicatechin and taxifolin were isolated as active principles. So P. sargentii R. methanolic extracts may be used for the cosmetic material.
The structures of six flavonoids isolated from heartwood of Prunus sargentii(Rosaceae) were analyzed by Mass and NMR spectrometry. These flavonoids were grouped into dihydroflavonol, flavanone, and flavanone glycoside, and identified as follows : 3,3',4',5,7-pentahydroxyflavanone(taxifolin) as a dihydroflavonol, 5-hydroxy-7-methoxyflavanone(pinostrobin), 4',5,7-trihydroxyflavanone(naringenin), 3',4',5,7-tetrahydroxyflavanone(eriodictyol), 5,7-dihydroxyflavanone(pinoccmbrin) as a flavanone and 7-hydroxyflavanone 5-O-${\beta}$-D-glucopyranoside(verecundin) as a flavanone glycoside.

Taxifolin-3-O-$\beta$-D-xylopyranoside and quercetin-3-O-$\alpha$-L-rhamnopyranoside were isolated from an EtOAc-soluble extract of the leaves of Chamaecyparis obtuse. Quercetin-3-O-$\alpha$-L-rhamnopyranoside was found to possess a potent inhibitory activity of human recombinant aldose reductase in vitro, its $IC_{50}$ value being $11.5\;{\mu}M$. Kinetic analysis showed that quercetin-3-O-$\alpha$-L-rhamnopyranoside exhibited uncompetitive inhibition against DL-glyceraldehyde. Also, quercetin-3-O-$\alpha$-L-rhamnopyranoside suppresses sorbitol accumulation in rat lens under high glucose conditions, demonstrating the potential to prevent sorbitol accumulation in vivo. These results suggest that this compound may be a promising agent in the prevention or treatment of diabetic complications.

In this paper, we have isolated six phenolic compounds, such as (+)-catechin (1), taxifolin (2), taxifolin-3-O-${\beta}$-D-(+)-xylose (3), quercetin (4), quercetin-3-O-${\alpha}$-L(+)-rhamnose (quercitrin) (5), apigenin-8-C-rhamnosyl-(1'''${\rightarrow}$2'')-glucoside (2''-O-rhamnosylvitexin) (6) from the EtOAc(Ethyl Acetate) and $H_2O$ soluble fractions of Japanese anise(Illicium anisatum L) leaves and twigs. Also, we have evaluated antioxidative and antiviral activity for each isolated compound. The antioxidative test was DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity. According to the experimental results, all of the isolated compounds indicated the increased radical scavenging activities as the concentration increases and most of the isolated compounds indicated generally good antioxidative values compare to the controls, ascorbic acid and ${\alpha}$-tocopherol. In the antiviral activities, all of the isolated compounds had no potentials in rhinovirus 1B (HRV 1B). But in enterovirus 71 (EV 71) and Influenza virus A/PR/8 (Influenza PR8), only quercetin (4) indicated the good antiviral activity compare to the control. Based on the above results, we found that the phenolic compounds of Japanese anise may be applied for one of the natural biomass sources that can be used as an antioxidant and an antiviral substance.

Hovenia dulcis is a herbal plant, which belongs to the Rhamnaceae family and is a native of Japan, China and Korea. Its fruit stalk is called 'Jiguja' in Korea. It has been traditionally used as a medicinal plant in East Asia. It was reported to have detoxification effects on alcohol poisoning, and antioxidant, antidiabetic etc. Sample of 5 g was extracted with 50 mL of 70% EtOH. The supernatant was filtered by 0.45 ㎛ membrane filter before analysis. The UPLC system was performed on Waters alliance UPLC HSS T3 column (2.1 × 100 mm, 1.7 ㎛) with a UV detector. The gradient system was a binary eluent of 0.1% formic acid in water(A) and 0.1% formic acid in acetonitrile(B) with gradient conditions as follows: Initial, 10% B; 1 min, 10% B; 4 min, 20% B; 10 min, 25% B; 12 min, 30% B; 14 min, 90% B; 17 min, 90% B; flow rate of 0.2 mL/min. The samples were injected by 2 µL and were detected at UV 355 nm. As a result of analysis, chromatographic patterns appeared in two cases: samples analyzed for ampelopsin and myricetin, and samples analyzed for taxifolin and quercetin. Among the four compounds, the largest regional difference was found to be taxifolin.
Comparative analysis of paeoniflorin, albiflorin and phenolic compound contents as bioactive components of peony was performed by Reverse Phase-High Performance Liquid Chromatography (RPHPLC) using the four- year-old peony which were different plant parts and pretreatment, such as removing or unremoving the cork layer of peony root before drying. The contents of paeoniflorin, albiflorin, (+)-taxifolin $3-O-{\beta}-D-glucopyranoside$, (+)-catechin and (-)-epicatechin were the highest in rhizome part, but those of gallic acid and benzoic acid in the leaves were higher than other parts. The contents of albiflorin, gallic acid, benzoic acid and (-)-epicatechin in the cork layer were higher than in those of the core, but the contents of paeoniflorin, (+)-taxifolin $3-O-{\beta}-D-glucopyranoside$ and (+)-catechin in the core were higher than those in the cork layer. In general, the rhizome part of peony root has been used only propagation purpose, but this part contained high contents of bioactive component. Therefore, it is needed that medicinal application of rhizome part in peony root was firmly investigated. Also, In the use of peony root for medicinal purpose, the use of peony root with cork layer can be efficient way on the practical use of useful components and the reduction of labor for removing the cork layer.

Quercetin (QT) and taxifolin (TF) are structurally similar plant-derived flavonoids that have antioxidant properties and act as free radical scavengers. The objective of this study was to investigate effects of QT and TF on nuclear maturation of porcine oocytes. Effects of TF at 0, 1, 10, and $50{\mu}g/mL$ on oocyte nuclear maturation (polar body extrusion) were investigated. After incubation for 44 h, there were no significant differences between the treatment and control groups except in the $50{\mu}g/mL$ group which was significantly lower (59.2%, p<0.05) than the other groups (control: >80%). After parthenogenetic activation, further in vitro development of QT- or TF-treated vs control oocytes was investigated. A significantly higher proportion of QT-treated ($1{\mu}g/mL$) oocytes developed into blastocysts compared to controls (24.3% vs 16.8%, respectively); however, cleavage rate and blastocyst cell number were not affected. The TF-treated group was not significantly different from controls. Levels of reactive oxygen species (ROS) and intracellular glutathione (GSH) in oocytes and embryos in a culture medium supplemented with QT or TF were measured. Both treatment groups had significantly lower (p<0.05) levels of ROS than controls, however GSH levels were different only in QT-treated oocytes. We conclude that exogenous flavonoids such as QT and TF reduce ROS levels in oocytes. Although at high concentration ($50{\mu}g/mL$) both QT and TF appear to be toxic to oocytes.

Korean red pine (Pinus densiflora) is one of the major Pinus species in Korea. Red pine bark is removed prior to the chipping process in the wood industry and discarded as waste. However, red pine bark contains a considerable amount of naturally occurring phenolics, including flavonoids, and therefore may have a variety of biological effects. In this study, we investigated if Korean red pine bark extract (KRPBE) could protect neuronal PC-12 cells from oxidative stress and inhibit cholinesterase activity. Analysis of reversed-phase high-performance liquid chromatography results revealed four phenolics in KRPBE: vanillin, protocatechuic acid, catechin, and taxifolin. The total phenolic and flavonoid contents of KRPBE were 397.9 mg gallic acid equivalents/g dry weight (DW) and 248.7 mg catechin equivalents/g DW, respectively. The antioxidant capacities of KRPBE measured using ABTS, DPPH, and ORAC assays were 697.3, 521.8, and 2,627.7 mg vitamin C equivalents/g DW, respectively. KRPBE and its identified phenolics protected against $H_2O_2$-induced oxidative cell death in a dose-dependent manner. Acetylcholinesterase and butyrylcholinesterase, which degrade the neurotransmitter acetylcholine to terminate neurotransmission in synaptic clefts, were inhibited by treatment with KRPBE and its identified phenolics. Taken together, these results suggest that KRPBE and its constituent antioxidative phenolics are potent neuroprotective agents that can maintain cell viability under oxidative stress and inhibit cholinesterase activity.
Opuntia cactus grows in the southern part of Korea, Geoje Island, Jeju Island, and South Coast. The flowers bloom around April to May of each year, and purple berries ripen around November to December. It has been widely used in home medicine since ancient times and it has great effects on blood circulation, detoxification, analgesia and antioxidant activity. In this study, we tried to demonstrate antioxidant, antidiabetic and antimicrobial activities of Opuntia monacantha Haw., and also grasp the value as a functional raw material. As a result, a strong antioxidative effect was confirmed by extracting taxifolin which is an antioxidative substance in Opuntia monacantha Haw. In the antidiabetic function test, a high level of antidiabetic function was confirmed when compared to that of using a commercially available diabetic drug. In addition, as a result of the antimicrobial effect measurement, it showed a strong antimicrobial activity on E.coli and Salmonella, and proved the excellent value of Opuntia manacantha Haw. as a functional ingredient.
Black locust(Robinia pseudoacacia) and poplar(Populus alba ${\times}$ glandulosa) trees were collected, extracted with acetone-$H_2O$(7:3, v/v) after drying, fractionated with hexane, chloroform and ethylacetate, and freeze dried to get some brown powder. Each fraction of the powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluting solvents. The wood extractives of black locust contained (+)-leucorobinetinidin as flavan, robtin as flavanone and dihydrorobinetin as flavanonol, and robinetin as flavonol. The poplar bark extractives contained various kinds of phenolic compounds : (+)-catechin as flavan, naringeoin, eriodictyol, sakuranetin, aromadendrin and taxifolin as flavanonol, salireposide as salicin derivative, and minor compounds such as aesculin and p-coumaric acid. However, aesculin has not been reported as a constituent of the poplar bark in Korea yet. NMR and FAB-MS analyses were done to elucidate the structures of isolated phenolic constituents.
