Evaluation of Mesoporous Alumina Adsorbent for the Purification of Paclitaxel (Paclitaxel 정제를 위한 메조기공 알루미나 흡착제 평가)

  • Several types of mesoporous alumina adsorbents with different physical properties were prepared by spray pyrolysis and were used for the separation/purification of the anticancer agent paclitaxel. The pore diameter of the adsorbents had a greater effect than did the surface area and the pore volume on the removal of plant-derived impurities. An appropriate pore diameter (~10.8 nm) was required for effective impurity removal. At a constant pore diameter, the surface area of the adsorbent affected not only the purity but also the yield of paclitaxel. Also, increasing the surface area of the adsorbent resulted in an increase in the adsorption of paclitaxel and impurities (biomass-derived tar and wax components). Removal of these impurities was confirmed by HPLC analysis of the absorbent after the treatment and TGA of the organic substances that were bound to the adsorbent.

Comparative evaluation of the mutagenicity and genotoxicity of smoke condensate derived from Korean cigarettes

  • Objectives Cigarette smoking is associated with carcinogenesis owing to the mutagenic and genotoxic effects of cigarette smoke. The aim of this study was to evaluate the mutagenic and genotoxic effects of Korean cigarettes using in vitro assays. Methods We selected 2 types of cigarettes (TL and TW) as benchmark Korean cigarettes for this study, because they represent the greatest level of nicotine and tar contents among Korean cigarettes. Mutagenic potency was expressed as the number of revertants per ${\mu}g$ of cigarette smoke condensate (CSC) total particulate matter whereas genotoxic potency was expressed as a concentration-dependent induction factor. The CSC was prepared by the International Organization for Standardization 3308 smoking method. CHO-K1 cells were used in vitro micronucleus (MNvit) and comet assays. Two strains of Salmonella typhimurium (Salmonella enterica subsp. enterica ; TA98 and TA1537) were employed in Ames tests. Results All CSCs showed mutagenicity in the TA98 and TA1537 strains. In addition, DNA damage and micronuclei formation were observed in the comet and MNvit assays owing to CSC exposure. The CSC from the 3R4F Kentucky reference (3R4F) cigarette produced the most severe mutagenic and genotoxic potencies, followed by the CSC from the TL cigarette, whereas the CSC from the TW cigarette produced the least severe mutagenic and genotoxic potencies. Conclusions The results of this study suggest that the mutagenic and genotoxic potencies of the TL and TW cigarettes were weaker than those of the 3R4F cigarette. Further study on standardized concepts of toxic equivalents for cigarettes needs to be conducted for more extensive use of in vitro tests.

  • This study provide Car driver with car sensor information which is car trouble code in engine and many sensors. This provide car manager with many information of tar sensors when we go to vehicle maintenance. This system consist of embedded linux system which uses RS232 and EML327 for CAN communication of car system. This System can control digital appliances in home network using wireless and ZigBee. Finally this is a lot of application for embedded linux system which get sensor informations of car control sensor system.

  • Rice straw is one of the main renewable energy sources in Korea, and bio-oil is produced from rice straw with a lab. scale plant equipped mainly with a fluidized bed and a char removal system. We investigated how the reaction temperature affected the production of bio-oil and the efficiency of a char removal system. To elucidate how the temperature depended on the production of bio-oil, experiment were conducted between $450^{\circ}C\;and\;600^{\circ}C$ with a feed rate of about 300g/h. The mass balance was established in each experiment, and the produced gas and oil were analyzed with the aid of GCs and a GC-MS system. The char removal system is composed of a cyclone and a hot filter. In the experiments, we observed that the optimum reaction temperature range for the production of bio-oil is between $450^{\circ}C\;and\;500^{\circ}C$.

Physical and Chemical Properties of Charcoal Added Paper for Cigarette Filter (활성탄 첨가에 따른 담배용 필터지의 이화학적 특성)

  • This study was carried to investigate optimum conditions of charcoal added paper in making cigarette filter for the removal of smoke contents of cigarettes. We investigated the physical properties of charcoal added paper according to the ratio of pulps, the amounts of characoal , the weight of sheets and crimping conditions in dry method, and then analyzed the smoke contents of cigarettes. The results obtained were as follows. 1. Sw-BKP was higher than Hw-BKP in methylene blue absorption by UV test. 2. Charcoal had an effect on stiffness for decreasing factor greatly , while the addition amounts of Sw-BKP and high weigth of sheets increased it, and the interaction of weight of sheets and charcoal was higher than other factors. 3. Tear index decreased by charcoal addition greatly, on the other hand high weight of sheets and Sw-BKP/Hw-BKP ratio increased ter, and the interaction of pulps and chacoal acted on major factors. 4. We estimated crimp index with sensory evaluation , and charcoal hand an effect on decreasing factor in crimping greatly, while the interaction of weight of sheets and pulp ratio was higher than others. 5. In the case of charcoal added paper filter, tar removal efficiency of smoke contents was higher but about 6% than cigarette filter. Especially vapour phase(aldehyde) in smoke contents was removed greatly.

Effect of Aging Period on Chromatic, Chemical and Organoleptic Properties of Burley Leaf Tobacco (버어리종 잎담배의 숙성기간이 색상, 화학성분 및 끽미에 미치는 영향)

  • This study was carried out to examinate the effect of aging period on chromatic, chemical and organoleptic characteristics, and to evaluate of optimum aging period for each grade in burley leaf tobacco. The leaf tobaccos were produced in 2000, and threshed, redried and packed in carton box under the current methods. Four grades of processed leaf(A3T, B1T, C1W and D3W) were stored during 24 months (May 10, 2001 to April 31, 2003) in warehouse of Chungju Leaf Tobacco Processing Factory. The leaf tobaccos were sampled at three month intervals for analysis of chromatic, chemical and organoleptic properties. pH values of four grades, and Yellow(b) of A3T and BIT were also significantly decreased during the aging. Filling values, tar, nicotine and CO contents of tobacco smoke, and puff number of cigarettes were not significantly changed during the aging. Positive correlation coefficients were significantly observed between irritation of the calculated attributes from contents of volatile oil components in leaf tobacco and that of the panel sensory attributes. The ratio of maximum change in irritation attribute was larger than that in taste attribute during aging. The calculated irritations of 18~24 months aging for A3T and B1T were significantly lower than zero-aging, while those of C1W and D3W were not significant among aging periods. The panel irritations of 12~15 months aging for four grades were low tendency. The results suggest that decrease of aging period for thin leaf from 21~24 to 12~15 months may be beneficial to save storage cost in burley tobacco.

Filling values, tar, nicotine and CO contents of tobacco smoke, and puff number of cigarettes were not significantly changed during the aging. Positive correlation coefficients were significantly observed between taste and irritation of the calculated attributes from contents of volatile oil components in leaf tobacco and those of the panel sensory attributes. The ratio of maximum change in taste attribute was larger than that in irritation attribute during aging. The optimum aging periods estimated by taste for A3O, B1O, C1L and D3L were 17.8, 14.9, 10.8, and 9.8 months, respectively. The thin leaf(Primings and Cutters) undergo satisfactory aging earlier as compared to bodied leaf(Leaf and Tips). The results suggest that decrease of aging period for thin leaf from 18~21 to 9~12 months may be beneficial to save storage cost in flue-cured tobacco.

Changes in Unprotonated Nicotine Concentration in Cigarette Mainstream Smoke with Three Machine-Smoking Conditions

  • This study was conducted to determine the amount of unprotonated nicotine in cigarette mainstream smoke and to investigate its relationship to level of filter ventilation and machine smoking conditions. Unprotonated nicotine from TPM trapped on a Cambridge filter pad(CFP) was adsorbed by Carboxen/PDMS SPME fiber, thermally desorbed and determined by GC/MS. 2R4F reference cigarette, twelve commercial brands from the Korean market and five test cigarette samples, which had the same tobacco blend with different levels of filter ventilation, were analyzed for unprotonated nicotine. In commercial brands, the amount of unprotonated nicotine changed slightly depending on the pH values of smoke, and decreased as the tar level increased. filter ventilation in these commercial cigarettes was $28{\sim}80%$ and the higher filter ventilation increased relative unprotonated nicotine levels, but not significantly. However, in five test cigarettes with different filter ventilation$(0{\sim}70%)$, unprotonated nicotine levels increased almost linearly with the level of filter ventilation. Concentrations of unprotonated nicotine in mainstream smoke generally increased in order $HC\;<\;ISO\;{\leq}\;MDPH$ machine smoking conditions. The ratio of unprotonated nicotine to total nicotine among $cigarettes({\alpha}_{fb})$ increased in order RC < MDPB < ISO conditions. Concentrations of unprotonated nicotine varied with three machine smoking conditions.

Differential expression of microRNAs in the saliva of patients with aggressive periodontitis: a pilot study of potential biomarkers for aggressive periodontitis

  • Purpose: The aim of this study was to compare microRNA (miRNA) gene expression in saliva using miRNA polymerase chain reaction (PCR) arrays in healthy and aggressive periodontitis (AP) patients. Methods: PCR arrays of 84 miRNAs related to the human inflammatory response and autoimmunity from the saliva samples of 4 patients with AP and 4 healthy controls were performed. The functions and diseases related to the miRNAs were obtained using TAM 2.0. Experimentally validated targets of differentially expressed miRNAs were obtained from mirTarBase. Gene ontology terms and pathways were analyzed using ConsensusPathDB. Results: Four downregulated miRNAs (hsa-let-7a-5p, hsa-let-7f-5p, hsa-miR-181b-5p, and hsa-miR-23b-3p) were identified in patients with AP. These miRNAs are associated with cell death and innate immunity, and they target genes associated with osteoclast development and function. Conclusions: This study is the first analysis of miRNAs in the saliva of patients with AP. Identifying discriminatory human salivary miRNA biomarkers reflective of periodontal disease in a non-invasive screening assay is crucial for the development of salivary diagnostics. These data provide a first step towards the discovery of key salivary miRNA biomarkers for AP.

Development of an E. coli Expression Cassette for the Efficient Production of a Harmful Protein

  • In order to produce a harmful protein more efficiently, this expression cassette, dubbed pCol-MICT, is directed by the colicin promoter, and was constructed by the insertion of a $rrnBT_1T_2$ fragment of pEXP7, and a MxelnteinCBD fragment of pTXB3, into pSH375. To test whether harmful proteins, including proteolytic enzymes, could be effectively produced by this cassette, the carboxypeptidase (CPase) Taq gene was inserted into the pCol-MICT cassette to yield pCol-CPase Taq-MICT. E coli W3l 10 tells harboring pCol-CPase Taq-MICT produced a large quantity of this enzyme, as much as 47.2 mg of purified from per liter of culture, when cultured in the presence of mitomycin C ($0.4{\mu}g/mL$). This indicates that the colicin promoter-controlled E, coli expression cassette was able to produce almost 8 times of protein than the conventional tar promoter-based system, and that this cassette may be useful in the Synthesis of other harmful proteins.
