The Formulation and Bioavailability of Oral Sustained Release Sulindac Delivery System (설린닥의 경구용 지속성 제제설계 및 생체이용율)

  • In order to design a 24 hr sustained release preparation of sulindac for oral administration, fast release pellet (FR), slow release pellet (SR) and two combined formulation (1 : 1 and 1 : 2) were prepared. The pharmacokinetic effect of such preparations has been evaluated using rabbits as a suitable in vivo model, and tested in man. Dose determination was carried out using curve fitting according to RSTPJP II program. In bioavailability test using rabbit, AUCs of sulindac in a few designed formulations were similar to each other. $C_{max}$- of RF and SR were 1.8 times and 1.2 times higher, respectively, compared to that of combined formulation (FR:SR=1:1). While plasma concentration of FR and SR decreased rapidly, that of combined formulation (FR:SR 1:1) lasted at the level close to $C_{max}$ for 24 hrs. Plasma concentration of sulfide form from the combined pellet(FR:SR=1:1) lasted for 24 hrs, and its AUC value was 1.4-fold, 2.7-fold. and 1.2-fold greater than FR pellet, SR pellet and combined pellet (FR:SR 1 : 2). Thus, the combined pellet of 1:1 ratio was found to be the most effective for oral sustained release formulation. Bioavailability test in human showed that AUC of sulfide from TSRP (1 : 1) was approximately 1.5 times greater than total AUC of Immbaron$^{\circledR}$ administered twice in a day. While $T_{max}$ of sulfide from lmmbaron$^{\circledR}$ was 4.33 +/- 1.37 hr (lst administration) and 3.33 ${pm}$ 0.82 hr (2nd administration), respectively, that of sulfide from TSRP increased to 7.17 ${pm}$ 2.86 hr. Plasma concentration of sulfide from TSRP was sustained at more, than 1.0 ${\mu}g{\cdot}$hr/ml until 24 hrs after one dose administration. In addition, TSRP may decrease local adverse reaction in the stomach, since plasma concentration of sulfide from the combined pellet was low within 2hrs in the stomach. In conclusion, it is suggested that TSRP formulation may be effective for oral 24 hr sustained release formulation of sulindac dosing 300 ~ 350mg once a day.

Prediction of cyanobacteria harmful algal blooms in reservoir using machine learning and deep learning (머신러닝과 딥러닝을 이용한 저수지 유해 남조류 발생 예측)

  • In relation to the algae bloom, four types of blue-green algae that emit toxic substances are designated and managed as harmful Cyanobacteria, and prediction information using a physical model is being also published. However, as algae are living organisms, it is difficult to predict according to physical dynamics, and not easy to consider the effects of numerous factors such as weather, hydraulic, hydrology, and water quality. Therefore, a lot of researches on algal bloom prediction using machine learning have been recently conducted. In this study, the characteristic importance of water quality factors affecting the occurrence of Cyanobacteria harmful algal blooms (CyanoHABs) were analyzed using the random forest (RF) model for Bohyeonsan Dam and Yeongcheon Dam located in Yeongcheon-si, Gyeongsangbuk-do and also predicted the occurrence of harmful blue-green algae using the machine learning and deep learning models and evaluated their accuracy. The water temperature and total nitrogen (T-N) were found to be high in common, and the occurrence prediction of CyanoHABs using artificial neural network (ANN) also predicted the actual values closely, confirming that it can be used for the reservoirs that require the prediction of harmful cyanobacteria for algal management in the future.

A Comparative Study on Injury Severity, Self esteem, Health Locus of control and Health Promotion Lifestyles between Helmeted and Nonhelmeted Motorcycle Accident Victims (오토바이 사고환자의 안전모 착용여부에 따른 뇌 손상비교와 자아존중감, 건강통제위 성격, 건강증진행위의 비교연구)

  • Data on 63 patients who had had motorcycle accidents and who were admitted to four general hospitals in the Chung Chung Nam Do area from July / 1993 to August 1993 were analyzed. The tool used for this study was a structured questionnaire which consisted of ten items on self- esteem, 18 items on health locus of control and 37 items profiling health prometion lifestyle. Injury severity scores were calculated bated based on data from the patients’ medical records. The collected data were analyzed using the SPSS, yielding descriptive statistics, t-test, ANOVA, Pearson’s Product Moment Correlation. The findings of this study are as follows. 1) Of the 63 injured motorcyclists, 35(55.6%) were helmeted and 28(44.4%) were nonhelmeted, and the nonhelmeted motorcyclists were predominantly young and male. The demographic variables for the helmeted and nonhelmeted groups were heterogeneous for age and occupation. 2) The results of the comparison between the two groups showed a statistically significant difference in the injury severity score(t=-4.70, p=0.000). The helmeted group had lower scores on injury severity score (9.00±3.93) than the nonhelmeted group(14.32土5.05). More than 60% of the nonhelmeted motorcyclists had brain injuries compared to only a third of the helmeted cyclists. 3) There .was a statistically significant difference between the two groups on self esteem(t=4.5, 000). The helmeted group had a higher mean score (31.27±2.72) than the nonhelmeted group(27.46±3.80). 4) The means for Internal health locus of control (IHLC), Powerful others health locus of control (PHLC), and Chance health locus of control (CHLC) in the two groups were similar to instrument norms reported in other literature. The mean scores on the IHLC in the two groups were higher than scores on the PHLC or the CHLC. However, there was a significant difference between the mean scores for the two groups on the PHLC (t=2.85, P=0.006). 5) The mean score for the helmeted group on the health promotion lifestyle profile was higher than the mean score for the nonhelmeted group(107.30±11.10, 96.57土 15.54 respectively), and there was a significant difference between the mean scores (t=3.64, p=0.001) . The highest score for helmeted group on the health promotion lifestyle profile was in the health care domain. However, for the nonhelmeted group the highest score was in the exercise domain and the lowest score was in the health care domain. 6) With regard to the relationship between health promotion lifestyle, health locus of control and self esteem in the two groups, the correlation coefficient between health promotion lifestyle and internal health locus of control for the helmeted group was 50(p〈0.01). For the nonhelmeted group, there was no correlation between health promotion lifestyle and internal health locus of control. However, there were significant correlation between health pro-motion lifestyle and external locus of control(r=0. 46, p〈0.01), and self esteem(r=0.495, p〈0.01). 7) Among the demographic variables, age and education had an impact on individual’s self-esteem The modifying factors of age made a contribution to explaining health - promoting lifestyle. In the present study, more than 40% rf the motorcyclists were riding without a helmet. The incidence of brain injury for patients riding without a helmet was nearly twice as high in the nonhelmeted rider as compared to the helmeted rider. The nonhelmeted motorcyclists in this study had lower self-esteem, obtained a higher score on the IHLC, and were not strongly engaged in performing health promotion activities as compared to the helmeted riders. However, some of the nonhelmeted riders who had a strong belief in PHLC were positively associated with engaging in health promotion activities. Based on the results obtained from this study, strategies to promote helmet usage for motorcyclists have to be developed.

Variation of Vital Sign according to Time in Full Immersion of Hot and Cool Bath (온.냉욕 전신침수욕시 기간에 따른 vital sign의 변화)

  • This study was conducted to see variation of vital sign of hot and cool bath according to time, a questionnair survey and measurement was carried out for 32 students(sophomore) of department of physical therapy Andong Junior College on the 27th of June, 1995. The result were as follows: The average systolic blood pressure(SBP) of stability for 32 college students who were measured was 105.3mmHg, the average diastolic blood pressure(DBP) was 67.3mmHg, the average pulse frequency(PF) was 70.7(frequency/min), the average respiratory frequency (RF) was 15.6 (frequency/min), and the body temperature(BT) was $36.6^{\circ}C$. As time went on, SBP for 32 students who were measured in hot bath according to stability, 3 min, 6 min, 9 min, and 12 min was decreased(105.15 mmHg, 104.69mmHg, 104.24 mmHg, 103.03 mmHg, and 96.69 mmHg)(P=0.3006). SBP was decreased in cool bath, too(105.15 mmHg, 103.33 mmHg, 103.33 mmHg, and 100.91 mmHg), but it at 12 min was a little higher(l03.09 mmHg)(P=0.7566). As time went on, DBP according to stability, 3 min, 6 min, 9 min, and 12 minutes was decreased in hot bath(66.82 mmHg, 65.45 mmHg, 64.54 mmHg, 63.03 mmHg, and 59.39 mmHg)(P=0.0906). It was similar in cool bath(66.82 mmHg, 67.87 mmHg, 68.48 mmHg, 67.87 mmHg, and 68.78)(P=0.9654). As time went on, PF was significantly increased in hot bath(70.42 times, 86.96 times, 93.57 times, 99.30 times, and 101.78 times)(P=0.0001). It was a little increased in cool bath, too (70.42 times, 70.85 times, 71.63 times, 71.06 times, and 71.45 times)(P=0.9803). As time went on, RF was significantly increased in hot bath(15.75 times, 19.09 times, 22.09 times, 24.94 times, and 26.48 times)(P=0.0001). I t in cool bath of stability, 3 min, and 6 min was a little increased(15.75 times, 19.30 times, 19.39 times), but it in 9 min(18.67 times), and 12 min(18.09 times) was a little decreased(P=0.0176). As time went on, BT was significantly increased in hot bath($36.63^{\circ}C,\;37.45^{\circ}C,\;37.81^{\circ}C,\;38.12^{\circ}C,\;38.33^{\circ}C$)(P=0.0001). It was a little increased in cool bath of stability and 3 min($36.63^{\circ}C,\;37.40^{\circ}C$), but others are similar($37.33^{\circ}C,\;37.37^{\circ}C$, and $37.36^{\circ}C$)(P=0.0001). It was revealed by this study, SBP and DBP according to time in hot and cool bath were decreased. PF, RF, and BT in hot bath were higher, RF and BT in cool bath were higher too. but PF was similar.

Electrophoretic Patterns of Proteins from Paragonimus westermani in Early DEvelopmental Stages (초기발육단계 폐흡충에서 추출한 단백질의 전기영동상)

  • In order to observe the protein composltlOns of soluble extracts of P. westermani, and their changes during early developmental stages, the crude saline extracts of 4, 6, 8, 10 and 12 week-old worms which were harvested from experimentally infected dogs were analysed by disc-PAGE. The results were as follows: 1. A total of 15 bands were identified from electrophoregrams of respective developmental stages. Of them, 5 bands were recognized throughout the developmental stages. 2. The number and protein amount of identified bands changed according to the worm development from 4 weeks to 12 weeks. However, tLe bar::ding patterns of 4 and 6 week-old worms and 8 and 10 week-old worms were similar each other. 3. Of 15 identified bands, Band 1 was recognized only in 12 week-old worms whereas Bands 3, 4, 8, 9, 10, 11 and 15 gradually lowered their amount according to dcvelor:ment to disappear in 12 week-old. In addition, Band 5 became a major band in 12 week-old while Band 6 turned to a minor band at the same age. The possible relations of changing patterns of protein bands with worm development were discussed.

Head Motion Detection and Alarm System during MRI scanning (MRI 영상획득 중의 피험자 움직임 감지 및 알림 시스템)

  • Purpose : During brain MRI scanning, subject's head motion can adversely affect MRI images. To minimize MR image distortion by head movement, we developed an optical tracking system to detect the 3-D movement of subjects. Materials and Methods: The system consisted of 2 CCD cameras, two infrared illuminators, reflective sphere-type markers, and frame grabber with desktop PC. Using calibration which is the procedure to calculate intrinsic/extrinsic parameters of each camera and triangulation, the system was desiged to detect 3-D coordinates of subject's head movement. We evaluated the accuracy of 3-D position of reflective markers on both test board and the real MRI scans. Results: The stereo system computed the 3-D position of markers accurately for the test board and for the subject with glasses with attached optical reflective marker, required to make regular head motion during MRI scanning. This head motion tracking didn't affect the resulting MR images even in the environment varying magnetic gradient and several RF pulses. Conclusion: This system has an advantage to detect subject's head motion in real-time. Using the developed system, MRI operator is able to determine whether he/she should stop or intervene in MRI acquisition to prevent more image distortions.

Analysis on the Assist Characteristics for the Knee Extension Motion of Lower Limb Orthosis Using Muscular Stiffness Force Feedback (근육 강성도 힘 피드백을 이용한 하지 보조기의 무릎 신전 운동 보조 특성 분석)

  • The lower limb orthosis with a pneumatic rubber actuator, which is intended for the assistance and the enhancement of muscular activities of lower limbs was developed in this study. Compared to other knee extension assistive devices being developed by other researchers, our device is designed especially for the elderly people and intended only for slight assistance so that the subjects can keep their muscular strength. For the effectiveness of system, muscular activities of major muscles in lower limbs during sit-to-stand (STS) and squat motion were measured and analyzed. Subjects were performed the STS and squat motion with and without lower limb orthosis. We made comparison muscular activities between with and without lower limb orthosis. Lower limb orthosis was controlled using muscular stiffness force feedback that is controlled by muscular activities of the measured muscle from force sensor. For analysis of muscular activities, electromyography of the subjects was measured during STS and squat motion, and these were measured using MP 150(BIOPAC Systems, Inc.). Muscles of interest were rectus femoris(RF), vastus lateralis(VL), vastus medialis(VM) and vastus intermedius(VI) muscles in lower limbs of the right side. A biodex dynamometer was used to measure the maximal concentric isokinetic strength of the knee extensors of wearing and not wearing orthosis on right side. The test were performed using the concentric isokinetic mode of test with the velocity set at 60°/s for muscles around the knee joints. The experimental result showed that muscular activities in lower limbs wearing orthosis using muscular stiffness force of a vastus medialis muscle was reduced and knee extension torque of an knee joint wearing lower limb orthosis was increased. With this, we confirmed the effectiveness of the developed lower limb orthosis.

The Effects of Wearing Roller Shoes on Muscle Activity in The Lower Extremity During Walking (롤러신발과 일반신발의 착용 후 보행 시 하지근의 근전도 비교)

  • The purpose of this study was to compare muscle activity in the lower extremity during walking wearing jogging and roller shoes. Twelve male middle school students (age: 15.0 yrs, height 173.7 cm, weight 587.7 N) who have no known musculoskeletal disorders were recruited as the subjects. Seven pairs of surface electrodes (QEMG8, Laxtha Korea, gain = 1,000, input impedance >$1012{\Omega}$, CMMR >100 dB) were attached to the right-hand side of the body to monitor the rectus femoris (RF), vastus medialis (VM), vastus lateralis (VL), biceps femoris (BF), tibialis anterior (TA), and medial (GM) and lateral gastrocnemius (GL) while subjects walked wearing roller and jogging shoes in random order at a speed of 1.1 m/s. An event sync unit with a bright LED light was used to synchronize the video and EMG recordings. EMG data were filtered using a 10 Hz to 350 Hz Butterworth band-passdigital filter and further normalized to the respective maximum voluntary isometric contraction EMG levels. For each trial being analyzed, five critical instants and four phases were identified from the recording. Averaged IEMG and peak IEMG were determined for each trial. For each dependent variable, paired t-test was performed to test if significant difference existed between shoe conditions (p<.05). The VM, TA, BF, and GM activities during the initial double limb stance and the initial single limb stance reduced significantly when going from jogging shoe to roller shoe condition. The decrease in EMG levels in those muscles indicated that the subjects locked the ankle and knee joints in an awkward fashion to compensate for the imbalance. Muscle activity in the GM for the roller shoe condition was significantly greater than the corresponding value for the jogging shoe condition during the terminal double limb stance and the terminal single limb stance. Because the subjects tried to keep their upper body weight in front of the hip to prevent falling backward, the GM activity for the roller shoe condition increased. It seems that there are differences in muscle activity between roller shoe and jogging shoe conditions. The differences in EMG pattern may be caused primarily by the altered position of ankle, knee, and center of mass throughout the walking cycle. Future studies should examine joint kinematics during walking with roller shoes.

The Effects of Tasks Setting for Mathematical Modelling in the Complex Real Situation (실세계 상황에서 수학적 모델링 과제설정 효과)

  • The purpose of this study was to examine the effects of tasks setting for mathematical modelling in the complex real situations. The tasks setting(MMa, MeA) in mathematical modelling was so important that we can't ignore its effects to develop meaning and integrate mathematical ideas. The experimental setting were two groups ($N_1=103$, $N_2=103$) at public high school and non-experimental setting was one group($N_3=103$). In mathematical achievement, we found meaningful improvement for MeA group on modelling tasks, but no meaningful effect on information processing tasks. The statistical method used was ACONOVA analysis. Beside their achievement, we were much concerned about their modelling approach that TSG21 had suggested in Category "Educational & cognitive Midelling". Subjects who involved in experimental works showed very interesting approach as Exploration, analysis in some situation ${\Rightarrow}$ Math. questions ${\Rightarrow}$ Setting models ${\Rightarrow}$ Problem solution ${\Rightarrow}$ Extension, generalization, but MeA group spent a lot of time on step: Exploration, analysis and MMa group on step, Setting models. Both groups integrated actively many heuristics that schoenfeld defined. Specially, Drawing and Modified Simple Strategy were the most powerful on approach step 1,2,3. It was very encouraging that those experimental setting was improved positively more than the non-experimental setting on mathematical belief and interest. In our school system, teaching math. modelling could be a answer about what kind of educational action or environment we should provide for them. That is, mathematical learning.

Side-Wall 공정을 이용한 WNx Self-Align Gate MESFET의 제작 및 특성

  • 초고주파 집적회로의 핵심소자로 각광을 받고 있는 GaAs MESFET(MEtal-emiconductor)은 게이트 형성 공정이 가장 중요하며, WNx 내화금속을 이용한 planar 게이트 구조의 경우 임계전압(Vth:threshold voltage)의 균일도가 우수할 뿐만 아니라 특히 Side-wall을 이용한 self-align 게이트는 소오스 저항을 줄일 수 있어 고성능의 소자 제작을 가능하게 한다.(1) 본 연구의 핵심이 되는 Side-wall을 형성하기 위하여 PECVD법에 의한 SiOx 박막을 증착하고, 건식식각법을 이용하여 SiOx side-wall을 형성하였다. 이 공정을 이용하여 소오스 저항이 낮고 임계전압의 균일도가 우수한 고성능의 self-aligned gate MESFET을 제작하였다. 3inch GaAs 기판상에 이온주입법에 의한 채널 형성, d.c. 스퍼터링법에 의한 WNx 증착, PECVD법에 의한 SiOx 증착, MERIE(Magnetic Enhanced Reactive Ion Etcing)에 의한 Side-wall 형성, LDD(Lightly Doped Drain)와 N+ 이온주입, 그리고 RTA(Rapid Thermal Annealing)를 사용하여 활성화 공정을 수행하였다. 채널은 40keV, 4312/cm2로, LDD는 50keV, 8e12/cm2로 이온주입하였고, 4000A의 SiOx를 증착한 후 2500A의 Side-wall을 형성하였다. 옴익 접촉은 AuGe/Ni/Au 합금을 이용하였고, 소자의 최종 Passivation은 SiNx 박막을 이용하였다. 제작된 소자의 전기적 특성은 hp4145B parameter analyzer를 이용한 전압-전류 측정을 통하여 평가하였다. Side-wall 형성은 0.3$\mu\textrm{m}$ 이상의 패턴크기에서 수직으로 잘 형성되었고, 본 연궁에서는 게이트 길이가 0.5$\mu\textrm{m}$인 MESFET을 제작하였다. d.c. 특성 측정 결과 Vds=2.0V에서 임계전압은 -0.78V, 트랜스컨덕턴스는 354mS/mm, 그리고 포화전류는 171mA/mm로 평가되었다. 특히 본 연구에서 개발된 트랜지스터의 게이트 전압 변화에 따른 균일한 트랜스 컨덕턴스의 특성은 RF 소자로 사용할 때 마이크로 웨이브의 왜곡특성을 없애주기 때문에 균일한 신호의 전달을 가능하게 한다. 0.5$\mu\textrm{m}$$\times$100$\mu\textrm{m}$ 게이트 MESFET을 이용한 S-parameter 측정과 Curve fitting 으로부터 차단주파수 fT는 40GHz 이상으로 평가되었고, 특히 균일한 트랜스컨덕턴스의 경향과 함께 차단주파수 역시 게이트 바이어스, 즉 소오스-드레스인 전류의 변화에 따라 균일한 값을 보였다. 본 연구에서 개발된 Side-wall 공정은 게이트 길이가 0.3$\mu\textrm{m}$까지 작은 경우에도 사용가능하며, WNx self-align gate MEESFET은 낮은 소오스저항, 균일한 임계전압 특성, 그리고 높고 균일한 트랜스 컨덕턴스 특성으로 HHP(Hend-Held Phone) 및 PCS(Personal communication System)와 같은 이동 통신용 단말기의 MMICs(Monolithic Microwave Integrates Circuits)의 제작에 활용될 것으로 기대된다.
