There has been growing concern about the toxicity of phthalate esters. Phthalate esters are being used widely for the production of perfume, nail varnish, hairsprays and other personal/cosmetic uses. Recently, exposure to phthalates has been assessed by analyzing urine for their metabolites. The parent phthalate is rapidly metabolized to its monoester (the active metabolite) and also glucuronidated, then excreted. The objective of this study is to evaluate the toxicity of phthalic acid (PA), which is the final common metabolic form of phthalic acid esters (PAEs). The individual PA isomers are extensively employed in the synthesis of synthetic agents, for example isophthalic acid (IPA), and terephthalic acid (TPA), which have very broad applications in the preparation of phthalate ester plasticizers and components of polyester fiber, film and fabricated items. There is a broad potential for exposure by industrial workers during the manufacturing process and by the general public (via vehicle exhausts, consumer products, etc). This review suggests that PA shows in vitro and in vivo toxicity (mutagenicity, developmental toxicity, reproductive toxicity, etc.). In addition, PA seems to be a useful biomarker for multiple exposure to PAEs in humans.
To obtain the standard compounds of metoprolol for a pharmacokinetic study, a convenient synthetic procedure to prepare enantiomers of metoprolol (3a) and its major metaboites, 2-4-(2-hydroxy-3-isopropylamino)propoxyphenylathanol (3b) and 4-(2-hydroxy-3- isopropylamino) pro-poxyphenylacetic acid (4), was developed from their respective starting materials, 4-(2-methoxyethyl)phenol (1a), 4-(2-hydroxyethyl)phenol (1b) and methyl 4-hydroxyphenylacetate (1c). These phenolic compounds (1a, b, c) were converted in situ to their corresponding phenoxides with sodium hydroxide treatment followed by (R)- or (S)-epichlorohydrin treatment. The resulting epoxides 2 were transformed to 3 through reaction with isopropylamine. Ester 3c was hydrolyzed to the metabolite 4. Measured using the HPLC method on chiral column without any derivatization, the optical purity of enantiomers of metoprolol and o-demethylated metabolite 3b ranged between 96-99 % ee and that of enantiomers of carboxylic acid metabolite 4 ranged 91% ee.
Mesenchymal stem cells (MSCs) have been recognized as a therapeutic tool for various diseases due to its unique ability for tissue regeneration and immune regulation. However, poor survival during in vitro expansion and after being administrated in vivo limits its clinical uses. Accordingly, protocols for enhancing cell survivability is critical for establishing an efficient cell therapy is needed. CDDO-Me is a synthetic C-28 methyl ester of 2-cyano-3,12-dioxoolean-1,9-dien-28-oic acid, which is known to stimulate nuclear factor erythroid 2-related factor 2 (Nrf2)-antioxidant response element (ARE) pathway. Herein, report that CDDO-Me promoted the proliferation of MSCs and increased colony forming units (CFU) numbers. No alteration in differentiation into tri-lineage mesodermal cells was found after CDDO-Me treatment. We observed that CDDO-Me treatment reduced the cell death induced by oxidative stress, demonstrated by the augment in the expression of Nrf2-downstream genes. Lastly, CDDO-Me led to the nuclear translocation of NRF2. Our data indicate that CDDO-Me can enhance the functionality of MSCs by stimulating cell survival and increasing viability under oxidative stress.
최근 건축구조물에 있어서 초고층화, 대형화의 요구에 의해 고강도 및 고성능 콘크리트의 사용이 증가하고 있다. 하지만 고강도 및 고성능 콘크리트는 화재 시 발생하는 폭렬현상에 취약한 문제가 있다. 폭렬은 화재 시 콘크리트 피복의 손실을 초래하여 내부콘크리트와 철근의 열전달률을 높여 콘크리트와 철근의 온도를 상승시키는 작용을 한다. 이러한 고강도 콘크리트의 화재 시 폭렬을 방지하기 위하여 많은 연구들이 진행되고 있으며, 대표적으로 폴리프로필렌(polypropylene)섬유, 강섬유를 콘크리트에 혼입한 연구들에 의해 폭렬제어성능이 입증되었으나, 섬유 혼입에 따른 유동성 저하 및 시공성 불량 등의 문제점이 제기되고 있다. 따라서 본 연구에서는 이와 같은 문제를 해결하기 위하여 에스테르계 윤활제 및 비이온성 계면활성제를 포함한 코팅액으로 코팅된 Polyamide 섬유를 콘크리트에 혼입하여 내화성능 및 시공성을 확보하였으며, 강도영역별 적정 조건을 실험을 통하여 도출하였다. 그 결과, 13mm의 polyamide 섬유 적용시 강도영역별로 60MPa에서는 $0.8kg/m^3$, 80MPa에서는 $1.0kg/m^3$, 100MPa는 $1.5kg/m^3$이상 혼입시 폭렬 제어 및 시공성 확보가 가능할 것으로 사료된다.
혈액 및 생체반응물질등에 함유된 미량의 glucose를 효소적으로 간편하게 측량하기 위하여 먼저 고가의 glucose oxidase를 고정화 시켰다. 전리 방사선조사에 의하여 이 효소를 쉽게 고정화 시킬수 있는 방법과 그 알맞는 조건 및 높은 잔여 활성에 관한 결과는 다음과 같다. 1) 여러가지 monomer 및 polymer의 combination 중에서 AA-Bis, NK ester 23G, 물 (1:1:2)에 가용성 효소 1ml를 고정화 시켰을 경우를 비롯하여 GOD의 잔여활성이 50%이상인 여러 monomer Combination 찾았다 2) Carrier의 방사선중합에 필요한 선량은 100 krad 이상 이였으나, 400~500krad가 적당하였고, solvent는 toluene, n-haxane, petoleum ether chloroform 등을 이용할 때 고정화된 GOD의 잔여 활성 및 이화학적 성장이 좋았다. 3) GOD 고정화에 이용될 수 있는 완충 용액은 tris-glycerol buffer (pH 7.0)가 phosphate (pH 7.0) 보다 높은 활성을 보여 주었다. 4) 고정화된 GOD의 최적 pH는 6.0~6.5 또 온도는 30~4$0^{\circ}C$로서 가용성 효소보다 작용범위가 넓었고, pH 및 온도의 변화도 완만하였다. 본 연구에서는 내분비계 교란 물질로 알려진 프탈레이트계 가소제를 대체하기 위하여 동·식물성 오일로부터 추출되는 지방산과 고리를 포함하는 알코올을 이용하여 가소제 후보물질을 합성하고, 가소화 효율을 포함한 가소제로서의 특성을 평가하였다. 지방산 3종(caprylic acid, capric acid, lauric acid)과 알코올 3종(solketal, benzyl alcohol, cyclohexanol)의 에스터화 반응으로 합성한 9종의 화합물의 구조는 1H-NMR을 이용하여 확인하였다. 합성한 9종의 가소제 후보물질은 PVC 수지에 첨가하여 가소 특성을 평가하였고, 상용 가소제인 di-2-ethylhexyl phthalate (DEHP), di(isononyl)cyclohexane-1,2-dicarboxylates (DINCH), epoxidized soybean oil (ESO)과 비교·분석하여 대체 가소제로서의 가능성을 평가하였다. 결과에 따르면 가소화 효율은 DEHP 대비 0.96~1.02 배, ESO 대비 0.94~0.98 배로 비슷한 결과를 얻었으며, DINCH의 경우보다는 1.05~1.10 배 우수한 결과를 보였다. 열적 안정성은 DEHP, DINCH보다 다소 떨어지나 ESO의 경우보다는 우수한 결과를 나타내었다. 용출성 시험 결과, 수성용매인 물과 유성용매인 노말 헥세인에서 DEHP, DINCH보다 다소 높은 결과를 보였으나 ESO 대비 유사하거나 우수한 특성을 나타내는 것을 확인하였다.
밤, 고구마, 우엉 등의 일부 식물체를 오랜 시간 보관하거나 가공하는 과정에서 표면에 녹변화합물이 형성된다. 본 연구에서는 이때 형성되는 녹변화합물이 유전독성 가능성을 내포한 novel trihydroxy acridine 유도체로서, 녹변화합물에 대한 복귀돌연변이시험을 수행하였다. 시험은 Salmonella typhimurium TA98, TA100 균주를 이용하였고 대사활성계 적용 유무에 따른 시험을 모두 수행하였다. 시험에 사용된 녹변화합물은 알라닌, 아르기닌, 아스파트산, 글라이신, 라이신, 페닐알라닌과 클로로겐산을 사용한 합성 및 고구마를 매트릭스로 하여 녹변화합물을 형성한 후 추출하는 두가지 방법을 통하여 준비하였다. 합성 녹변화합물과 추출 녹변화합물의 최대 처리 용량은 생육저해시험과 용해도, 흡광도를 고려 하여 각각 2000 ${\mu}g$/lplate, 50 ${\mu}g$plate로 결정 하였다. 녹변화합물 합성 과정 후 잔존하는 아미노산들과 클로로겐산의 영향을 배제하고자 이들을 또 다른 시험군으로 설정하고 각각 250 ${\mu}g$/plate, 1750 ${\mu}g$/plate의 농도로 처리하여 복귀돌연변이시험을 수행하였다. 시험결과 두 균주에서 농도에 의존적으로 콜로니 수가 증가하는 경향은 관찰되지 않았으며, 음성대조군 수치와 비교하여 두 배 이상의 콜로니 수도 나타나지 않았다. 이상의 결과를 종합할 때, 합성 녹변화합물과 추출 녹변화합물은 본 시험 조건에서 복귀돌연변이를 유발하지 않는 것으로 확인되었다.
본 연구에서는 헤마토코커스 플루비알리스(H. pluvialis)의 파쇄세포(cracked cell)로부터 추출된 아스타잔틴의 HPLC 분석방법을 확립하고, UV/visible 및 FT-IR 분광분석법을 이용하여 헤마토코커스 추출물과 합성 아스타잔틴의 구조적 특성을 조사하였다. 아스타잔틴을 메탄올 용매에 녹이고, 45분 정도 초음파로 처리할 경우 교반처리하는 경우 보다 1.5배 정도 용해율이 높아지는 것을 확인하였다. 에스터 형태로 존재하는 추출물 중의 아스타잔틴의 분석을 위해 cholesterol esterase를 이용해 헤마토코커스 추출물을 가수분해처리를 하여 유리형으로 전환시킨 후 HPLC 분석을 수행하였다. 아스타잔틴의 함량을 분석하기 위해 역상칼럼(C18)과 UV/visible 검출기를 사용하였고, 이동상은 메탄올과 물(95:5)의 혼합용매를 사용하여 분석하였다. 효소처리 후, 헤마토코커스 추출물의 흡수스펙트럼이 합성 아스타잔틴과 유사한 패턴으로 변화하는 것을 확인함으로써 헤마토코커스로부터의 아스타잔틴의 추출 및 분석조건을 확립하였다.
This paper describes the preparation and characterization of two kinds of fluorinated polybenzimidazole (PBI)s which can be potentially used for phosphoric acid-doped, high-temperature polymer electrolyte membrane fuel cells. Two kinds of perfluorocyclobutane (PFCB)-containing monomers were prepared via following synthetic steps; after fluoroalkylation of methyl 3-(hydroxy) benzoate and methyl 4-(hydroxy) benzoate with 1,2-dibromotetrafluoroethane and subsequent Zn-mediated dehalogenation, these compounds were cyclodimerized at $200^{\circ}C$ affording the ester-terminated monomers containing PFCB ether groups. The synthesized intermediates and monomers were characterized using FT-IR, $^1H-NMR,\;^{19}F-NMR$, and mass spectroscopy. The fluorinated PBIs were then successfully prepared through the solution polycondensation of the monomers and 3,3'-diaminobenzidine in polyphosphoric acid. Compared with traditional PBI, the glass transition temperatures of the fluorinated PBIs were obtained at $262^{\circ}C\;and\;269^{\circ}C$ which are lower than that of PBI and their initial degradation temperatures were still high over $400^{\circ}C$ under nitrogen. The fluorinated PBIs showed higher d-spacing values and improved solubility in several organic solvents as well as phosphoric acid, which confirmed they could be good candidates for the high temperature fuel cell membranes.
