가토의 ouabain 유발 부정맥에 미치는 acebuolol 및 carbamazepine의 영향 (The Effects of Acebutolol and Carbamazepine on the Ouabain-Induced Arrhythmias in Rabbits)

  • Adrenergic beta 1 수용체 봉쇄 약물인 acebutolol과 항 경련제로 사용되고 있는 carbamazepine은 실험적으로 ouabain유발 부정맥을 정상 심박동으로 환원시키는데 유효하다고 보고되었으나 그 상호 작용에 대해서는 밝혀진 바가 없다. 이에 본 실험에서는 가토에 ouabain 투여로 부정맥을 유발시킨 후 acebutolol과 carbamazepine을 단독 혹은 병용 투여하여 이 두 약물이 ouabain 유발 부정맥에 미치는 영향과 그 상호 작용을 규명하고자 하였다. 실험 결과 ouabain 유발 부정맥은 acebutolol 혹은 carbamazepine 단독 투여로 정상 심박동으로 환원되었으며 용량이 감소함에 따라 정상 심박동으로 회복하는데 요하는 시간이 연장되었다. 또 단독 투여시 항 부정맥 효과를 볼 수 없었던 용량을 병용 투여하였을 때 ouabain 유발 부정맥은 즉시 정상 심박동으로 환원되었으며 상기 용량의 두 약물을 병용하여 전처치함으로써 ouabain의 부정맥 유발 용량이 의의있게 증가되었다(P<0.01). 이상의 결과로 acebutolol과 carbamazepine은 ouabain 유발 부정맥을 용량 의존적으로 억제시키며 상협적인 상호작용(synergistic interaction)을 나타낸다고 사료된다.

환경재해 방지를 위한 생물정보로서의 사람 림프구 DNA 손상에 대한 방사선과 살충제의 상승작용 (Synergistic Interaction of Radiation with Pesticide on DNA Damage in Human Lymphocytes as Biological Information for Prevention of Environmental Disaster)

  • 병해충을 막기 위해 농업용 살충제가 광범위하게 사용되고 있다. 농약사용에 따른 생물학적 위해가 우려되며 농약이 또 다른 환경유해요인과 인체에 상승적으로 작용할 경우 농업재해로 이어질 가능성이 있다. 다양한 인자에 의한 DNA손상을 감지하는데 유용한 단세포 겔 전기영동법을 이용하여 살충제와 방사선에 의한 사람 림프구 DNA손상을 평가하였다. 각기 다른 농도로 살충제를 10분간 전처리한 림프구와 정상 림프구에 0-2.0 Gy의 방사선으로 조사한 다음 DNA 손상도를 평가하였다. DNA가닥 절단에 대한 표식인 tail moment의 증가는 감마선에 대해서 뚜렷한 선량-반응 관계를 나타내었다. 단세포 겔 전기영동법을 통한 평가결과 권장 사용농도 이상의 살충제는 림프구에 대한 유전독성을 나타내었을 뿐 아니라 방사선과 함께 상승작용을 일으켜 림프구 DNA손상을 더욱 증가시키는 것이 확인되었다. 이와 같은 결과는 환경재해의 방재 및 예방 조치에 필요한 생물정보를 사전에 제공하는 의미를 가진다.

Exploring Synergistic Effect in Metro Station Areas: A Case Study of Shanghai, China

  • In the process of exploring sustainable development, major cities in China are expanding metro systems as a strategy to reduce the negative environmental and social consequences of fast-paced motorization. A metro station is not only a transportation node, but also a place where diverse activities can be performed. Therefore, the realization of the spatial potential for human interaction is the essence of the strategy for integrated development in metro station areas. For this paper, 10 well-developed metro stations in Central Shanghai were selected to investigate the correlation between accessibility and spatial performance in station areas. The spatial performance in station areas is significantly affected by metro configuration. However, both vehicle and pedestrian accessibility show weak influence on spatial performance. A synergistic model was then developed to provide quantitative support for transit-oriented development in metro station areas.

Effect of the Combination of CI-988 and Morphine on Neuropathic Pain after Spinal Cord Injury in Rats

  • Cholecystokinin is known to be involved in the modulation of nociception and to reduce the efficacy of morphine analgesia. This study investigated the effects of intrathecal administration of morphine and the cholecystokinin type B antagonist CI-988 on below-level neuropathic pain after spinal cord injury in rats. We also examined the interaction of morphine and CI-988 in the antinociceptive effect. Both morphine and CI-988 given individually increased the paw withdrawal threshold to mechanical stimulation in a dose-dependent manner. The combination of ineffective doses of intrathecally administered CI-988 and morphine produced significant analgesic effects and the combination of effective doses resulted in analgesic effects that were greater than the sum of the individual effects of each drug. Thus, morphine showed a synergistic interaction with CI-988 for analgesia of central neuropathic pain.

  • 2,4-D 제초제(除草劑)의 상호작용(相互作用)을 조사(調査)키 위하여 2,4-D와 제초제(除草劑의 혼합처리(混合處理)가 수도(水稻)의 초기생육(初期生育)에 미치는 영향(影響)을 조사(調査)한 결과(結果)는 다음과 같다. 2,4-D 처리(處理)에 의한 신초생육(新鞘生育)이 더욱 억제(抑制)된 반면에 thiobencarb, butachlor 및 glyphosate 처리(處理)에 의한 IR28의 신초생육(新鞘生育) 억제(抑制)는 Taipei 309 보다 더욱 크게 나타났다. 2,4-D와 thiobencarb 혼합처리시(混合處理時)에 상호작용(相互作用)은 thiobencarb 농(濃度)가 낮은 처리구(處理區)의 신초(新鞘)길이에 대하여 두 품종(品種) 공(共)히 길항작용(拮抗作用)을 나타내었고 thiobencarb 고농도(高濃度)에서는 상승작용(相乘作用)을 보였다. 신초((新鞘)의 생체중(生體重)에 대한 2,4-D와 thiobencarb의 상호작용(相互作用)은 IR28의 경우 모든 처리구(處理區)에서 상승작용(相乘作用)을 나타낸 반면에 Taipei 309의 경우에는 2,4-D의 농도(濃度)가 낮을 때만 상승작용(相乘作用)이 나타났다. 2,4-D와 butachlor 혼합처리(混合處理)에 의해서는 IR28의 신초길이와 생체중(生體重)이 Taiperi 309보다 더욱 높은 억제현상(抑制現象)을 보여 상승작용(相乘作用)을 나타내었다. 2,4-D와 glyphosate 혼합시(混合時)에는 신초(新鞘)의 길이에서는 두 품종(品種) 공히 길항작용(拮抗作用)을 보였으나, 2,4-D와 glyphosate의 최고농도(最高濃度) 혼합시(混合時)에는 신초(新鞘)의 생체중(生體重)에 대해서 IR28은 상승작용(相乘作用)을 보인 반면에 Taipei의 309는 길항작용(拮抗作用)을 나타내었다.

Novel Genome-Wide Interactions Mediated via BOLL and EDNRA Polymorphisms in Intracranial Aneurysm

  • Objective : The association between boule (BOLL) and endothelin receptor type A (EDNRA) loci and intracranial aneurysm (IA) formation has been reported via genome-wide association studies. We sought to identify genome-wide interactions involving BOLL and EDNRA loci for IA in a Korean adult cohort. Methods : Genome-wide pairwise interaction analyses of BOLL and EDNRA involving 250 patients with IA and 296 controls were performed using the additive effect model after adjusting for confounding factors. Results : Among 512575 single-nucleotide polymorphisms (SNPs), 23 and 11 common SNPs suggested a genome-wide interaction threshold (p<1.25×10-8) involving rs700651 (BOLL) and rs6841581 (EDNRA). Rather than singe SNP effect of BOLL or EDNRA on IA development, they showed a synergistic effect on IA formation via multifactorial pair-wise interactions. The rs1105980 of PTCH1 gene showed the most significant interaction with rs700651 (natural log-transformed odds ratio [lnOR], 1.53; p=6.41×10-11). The rs74585958 of RYK gene interacted strongly with rs6841581 (lnOR, -19.91; p=1.64×10-9). Although, there was no direct interaction between BOLL and EDNRA variants, two EDNRA-interacting gene variants of TNIK (rs11925024 and rs1231) and FTO (rs9302654), and one BOLL-interacting METTL4 gene variant (rs549315) exhibited marginal interaction with BOLL gene. Conclusion : BOLL or EDNRA may have a synergistic effect on IA formation via multifactorial pair-wise interactions.

Synergistic Effect of Interleukin-18 on the Expression of Lipopolysaccharide-Induced IP-10 (CXCL-10) mRNA in Mouse Peritoneal Macrophages

  • Interleukin (IL)-18, a member of the family of IL-l cytokine, is one of the principal inducers of $interferon-{\gamma}(IFN-{\gamma})$ in T lymphocytes and natural killer cells. The objective of the present study was to evaluate the effect of IL-18 on the expression of chemokine IP-10 (CXCL-10) mRNA in mouse peritoneal macrophages. IL-18 had very weak direct effect or synergistic effect with IL-12 on the expression of IP-10 mRNA in C57BL/6 mouse peritoneal macrophages. However, IL-18 pretreatment was found to playa cooperative role in the expression of lipopolysaccharide (LPS)-induced IP-10 mRNA. For the expression of LPS-induced IP-10 mRNA, the synergistic effect was detected after 16 h of IL-18 pretreatment prior to LPS stimulation. The expression level of CD14 in cells stimulated with LPS was not changed by IL-18 pretreatment, and the level of $IFN-{\gamma}$ production during IL-18 pretreatment plus LPS stimulation was barely discernible ($0.36{\pm}0.31pg/ml$). Namely, the synergistic effect of IL-18 pretreatment was not related to a change of LPS receptor, CD14 expression, and the production of $IFN-{\gamma}$ by the interaction between IL-18 and LPS. The synergistic effect of IL-18 pretreatment on the expression of LPS-induced IP-10 was related to not NF-kB but AP-1 activation, and associated with the extracellular signal-regulated kinase (ERK) pathway, one of the mitogen-activated protein kinase signaling pathways. These results provide useful information that may elucidate the mechanisms underlying the effect of IL-18 on the expression of IP-10 mRNA.
