Isolation of N2-fixing Microorganism from the Root of Non-leguminous Crops (수종(數種)의 비두과(非豆科) 식물(植物)로 부터의 질소고정균(窒素固定菌)의 분리(分離)에 관(關)한 연구(硏究))

  • A number of nitrogen-fixing microorganisms were isolated from roots of non-legumious crops and evaluated for their nitrogen-fixing ability on their host crops. The results are summarized as follows: 1. Among nitrogen-fixing isolates obtained from 9 different crops, several isolates showed relatively higher level of acetylene reduction activity on Spinach, Chinese cabbage, Cucumber, mustard and Egg plant. 2. When the isolates were inoculated into 4 host crops, a number of isolates were found to grow well on Spinach, Chinese cabbage and Cucumber, but particularly well on the former. 3. Results of this study suggest that nitrogen-fixing a symbiotic microorganisms are also present and able to grow well on roots of non-legumious crops, and stimulate plant growth by promoting growth and differentiation of roots.

Control Effects of Micromonospora sp. AW050027 by Media Optimization and Microbial Treatment Against Pine Wood Nematode (Micromonospora sp. AW050027 균주의 배지최적화 및 미생물제제 처리에 의한 소나무재선충 방제효과)

  • Nematicidal activity against pine wood nematode, Bursaphelenchus xylophilus, was tested in the pot and field by the treatment of microbial formulation after media optimization. The optimized media composition was glycerol 10 g, soybean meal 10 g, NaCl 1 g, $CaCO_3$ 2 g, $K_2HPO_4$ 0.125 g per liter and microbial complex formulation was made with liquid and powder type. Most effective antibiotics against symbiotic microorganism with nematode, kanamycin, was added to the formulation. The control effects against pine wood nematodes were checked by pot test and field test. In the result of treatment by trunk injection, five times treatment was more effective than one time and the treatment with the formulation of concentrated culture supernatant was the most effective in the nematicidal activity showing below 10% mortality in pine tree.

Screening of Bacteria Producing Lipase from Insect Gut: Isolation and Characterization of a Strain, Burkholderia sp. HY-10 Producing Lipase (곤충 장내미생물로부터 lipase 생산능력이 우수한 Burkholderia sp. HY-10 균주의 분리 및 특성)

  • From the course of screening of useful enzyme producing microorganism from insect guts, we isolated 9 lipase producing strains and their lipase producing activities were tested. 16S rDNA sequence analysis showed that they were Gram negative bacteria grouped on Serratia sp., Pseudomonas sp., and Burkholderia sp.. Among them, an excellent lipase producing strain, Burkholderia sp. HY-10 identified by 16S rDNA analysis and biochemical methods, was further studied its lipase producing characteristics. It was isolated from a longcorm beetle, Prionus insularis and showed cell density dependent lipase producing activity in the culture media that contained olive oil as a carbon source. Maximum lipase production was achieved in the M9 media containing 0.5% yeast extract and 0.5% olive oil when cultured at $30^{\circ}C$ for 36-42 hrs.

  • Probiotics are live organisms that when administered in adequate amounts confer a health benefit on hosts. The administration of direct-fed microbials (DFM) such as lactobacilli and bacillus, may be a more direct approach to beneficially alter gastrointestinal microflora than altering dietary ingredients or supplementing with growth-promoting levels of antibiotics. It is apparent that microbes have an important influence on immune development and resistance to infection; that microbes are not static colonizers of our bodies, but are dynamic, symbiotic coresidents. And it can improve the surrounding environments; decrease the malodor caused by degrade the excrement. Recently, new paradigm such as environment protection and safe food have been settled. In domestic farm house, there is a great demand for probiotics as a substitute of antibiotics for the improvement of environmental quality and the production of a competitive goods. Probiotics circulated in a country have three categories: an animal medicine permitted by national veterinary research quarantine service (NVRQS), a support feed registered in city or country house, and not-registered goods. However, lots of unqualified goods were produced and circulated. And thus, it is in urgent need of evaluating the present situation and effect of probiotics. This study was conducted to evaluate the system of a probiotics as a livestock-environment improving agents for the alternation of antibiotics and quality control of it.
