우리나라 양돈장에서 Mycoplasma hyopneumoniae 유병율 조사 (Seroprevalence of Mycoplasma hyopneumoniae infection in Korean swine herds)

  • Serum samples of 1,175 pigs from 148 Korean swine farms not using Mycoplasma hyopneumoniae (M. hyo) vaccines were collected for seroepidemiological study of M. hyo infection by indirect ELISA method. Informations of each farm were provided about province where the farm was located and season when blood samples were collected. Then, the selected farms were divided into farm units which had 5 serum samples according to production stages : sow, suckling piglet (<30 days old), nursery pig (30-70 days old), and growing pig (>70 days old). Seroprevalence of M. hyo infection according to production stages, province, and season was investigated by using ELISA-positve rate of the selected samples for each study. This study showed that 85.34% (78.94-91.78%, 95% CI) of farms were positive to M. hyo infection and 34.81% (32.09-37.53%, 95% CI) among pigs were sero-positive to M. hyo infection in Korean swine farms. In the study of seroprevalence by production stage, most farms had sows and growing pigs which were sero-positive to M. hyo infection (sow: 83.05%, growing pigs: 87.72%) and most pigs seemed to be naturally infected by M. hyo at 8-10 weeks of age. Also, M. hyo infection showed seasonal pattern that most pigs were infected in late fall to early winter. However, in the study of seroprevalence by province, there was no significant correlation between province and M. hyo sero-positive rate.

축산물 잔류 sulfadimethoxine 검출용 ELISA kit 개발 (Development of an ELISA kit for the detection of residual sulfadimethoxine in edible animal products)

  • An enzyme linked immunosorbent assay (ELISA) was developed to screen residues of sulfadimethoxine (SDM) in edible animal products. An indirect competitive ELISA was allowed to compete with rabbit anti-SDM for binding to a limited amount of SDM-gelatin conjugate and SDM in serum samples. Sera was diluted 20 times with phosphate buffered saline (PBS) and boiled for 5 minutes to destruct immunoglobulins of serum. Detection limit of this competitive ELISA for SDM was 0.1 ppb or less. Among eight sulfonamide analogues tested for specifity, only sulfamonomethoxine showed significant cross-reaction in the assay. The EC-50 value for sulfamonomethoxine was 3.5 ppm. Recovery of SDM in spiked serum samples between 100 ppb and 500 ppb ranged from 110.7% to 128.9%.

Immortalization of Swine Umbilical Vein Endothelial Cells with Human Telomerase Reverse Transcriptase

  • Swine endothelial cells are commonly used as an in vitro model for studying features of the blood-brain barrier and some hemorrhagic diseases. However, primary cultures of swine cells have finite lifespans. To establish immortalized swine umbilical vein endothelial cells (SUVECs) using human telomerase reverse transcriptase (hTERT), the plasmid pCI-neo-hTERT was transfected into SUVECs by lipofection. Clones were selected for G418 resistance, and positive clones were amplified. One of the clones was cultured for up to 50 passages. Factor VIII-related antigen and CD34 were detected. The immortalized cells shared the properties of normal cells, such as contact inhibition, serum requirement and anchorage dependence. Karyotype analysis revealed that the immortalized cells were in the diploid range. In addition, both in vivo and in vitro assays of tumorigenicity showed no neoplastic transformation. Furthermore, NO, $PGI_2$, and ET-1 concentrations in the transfected cells were normal. These results suggest that the SUVECs immortalized by hTERT retain their original characteristics.

Predicting Methane Production Potential of Anaerobic Co-digestion of Swine Manure and Food Waste

  • Anaerobic co-digestion of swine manure and food waste for biogas production was performed in serum bottles at 2% volatile solids(VS) concentration and various mixing ratios of two substrates(swine manure: food waste = 100 : 0 $\sim$ 0 : 100). Through kinetic mode of surface methodology, the methane production was fitted to a Gompertz equation. The specific methane production potential of swine manure alone was lower than that of food waste. However, maximum methane production potential increased up to 1.09-1.22% as food waste composition increased up to the 80%. The maximum methane production value of food waste was 544.52 mL/g VS. It was observed that the maximum methane production potential of 601.86 mL/g VS was found at the mixing ratio of 40:60.

대체의학으로서 백화사설초 급여가 돼지 생산성에 미치는 효과 (Effects of Oldenlandidae Herba as an Alternative Medicine on the Swine Productivity)

  • As control of swine consumptive disease is the key of success in pig farming business, the effect of Oldenlandidae Herba on prevention of swine diseases as an alternative medicine was investigated in-vivo. As it contains substances such as ${\beta}$-sitisterol, ursolic acid and 3-O-${\beta}$-D-giucoside, it is effective on suppression of colonic tumor in rats and also, is widely used as an preventative medicine of cancer and inflammation in Chinese medicine. In this experiment, sows and piglets on two different farms were treated with Oldenlandidae Herba, and they recoded weaning rate greater than 80%, which were higher than national average 70% (PSY 68%). Meanwhile, neither organ accumulation nor toxicity has been detected on the basis of hematology and serum chemistry. In conclusion, the extracts of Oldenlandidae Herba is expected to be an good alternative medicines for swine productivity.

  • Torque teno virus (TTV), a species of Anellovirus, is a non-enveloped single stranded DNA virus with a wide range of animal hosts. The incidence of TTV is quite ubiquitous throughout the world. A total of 235 serum samples obtained from 137 pigs and 98 cattle at slaughterhouses in Korea during April 2005 to May 2005 were tested by TTV-specific PCR as to monitor prevalence of TTV among swine and cattle. As a result, the prevalent rates of TTVs in pigs and cattle were 43.1% and 4.1%, respectively. It seems that TTV infection is quite prevalent in swine population.

Improving behavior characteristics and stress indices of gestating sows housed with group housing facility

  • This study was conducted to investigate the effects of group-housing facility (GHF), compared to an individual confinement stall (CON), on the reproductive performance, behavior, and stress hormones of gestating sows. A total of 50 primiparous sows (Landrace × Yorkshire) were randomly allocated into either CON (n = 25) or GHF (n = 25) during the gestation period. One week before parturition, the sows were transferred into conventional farrowing crates, and cross-fostering was conducted within 1 d of delivery. Blood was collected for analyses of stress indices at 75 d of gestation and postpartum. Reproductive performance was estimated during the period of birth to weaning. Behavior patterns were identified at 90 d of gestation. Litter size was not different between the CON and GHF treatments. Weaning to estrus interval, however, tended to be lower in the GHF than in the CON (p < 0.1). Activity, treating, belly nosing, and exploring behaviors were observed only in the GHF group, whereas rubbing was shown only with the CON. Serum cortisol concentration was lower in the GHF than in the CON at 75 d of gestation (p < 0.05). Sows housed in the GHF showed lower epinephrine and norepinephrine concentrations than those housed in the CON at postpartum (p < 0.05). The GHF sows demonstrated more natural behavior characteristics associated with stress relief than the CON sows with no adverse effects on reproductive performance. Therefore, these results suggest that GHF could be applied as an alternative housing facility to improve animal welfare on swine farms.

Seroprevalence of swine influenza and porcine reproductive and respiratory syndrome in Korea

  • A total of 501 serum samples were selected from blood samples that were submitted to Department of Veterinary Pathology, Kangwon National University from all provinces in Korea from September 2001 to August 2002. Their sera were examined for antibodies to swine influenza virus subtype H1N1 (SlV H1N1) and porcine repro-ductive and respiratory syndrome virus (PRRSV) according to the age of pig, season, and herd size using enzyme-linked immunosorbent assay. The seroprevalence of SIV H1N1, PRRSV, and dual infection were 39.12%, 61.48%, and 25.95%, respectively. The seroprevalence of SIV H1N1 according to herd size was not significant differences (p>0.05). The results showed that the PRRSV infection spread widely in swine herds throughout the country.

Potential Methane Production on Anaerobic Co-digestion of Swine Manure and Food Waste

  • Anaerobic co-digestion of swine manure and food waste for biogas production was performed in serum bottles at various volatile solids(VS) contents and mixing ratios of two substrates(swine manure:food waste=$100:0{\sim}0:100$). Through kinetic mode of surface methodology, the methane production was fitted to a Gompertz equation. The ultimate methane production potential of swine manure alone was lower than that of food waste regardless of VS contents. However, it was appeared that maximum methane production potentials in 80 : 20 of the mixing rate at VS 3% was enhanced at 144.7%, compared to its only swine manure. The potential increased up to 815.71 ml/g VS fed as VS concentration and food composition increased up to 3.0% and 20%, respectively. The ultimate amount of methane produced had significantly a positive relationship with that of methane yield rate. Overall, it would be strongly recommended that feeding stocks use 20% of mixing ratio of food waste based on VS 3% contents when operating the anaerobic reactor on site at $35^{\circ}C$ if not have treatment of its anaerobic waste water.

돼지 지방세포 원형질막 단백질에 대한 다클론항체의 생산 및 이용에 관한 연구 (Studies on the Development and Utilization of Polyclonal Antibodies Against Swine Adipocyte Plasma Membrane Proteins)

  • The objectives of the current study were to develop polyclonal antibodies in sheep against adipocyte plasma membrane(APM) proteins isolated from swine, to investigate tissue specificity, and to determine cytotoxic effects of antiserum on swine adipocytes. Plasma membrane proteins from adipocyte, brain, heart, kidney, liver, and spleen were isolated using a 32% sucrose gradient. Adult male sheep was immunized three times at three week interval with the purified swine APM proteins. Antiserum was taken from immunized sheep at 10, 12, and 14 days after the third immunization. Antiserum expressed strong reactivity with APM proteins determined by enzyme-linked immunosorbent assay(ELISA), and the reactivity could be detected at dilutions in excess of 1 : 81,000. Antiserum showed very low binding affinity with proteins isolated from brain, heart, kidney, liver, or spleen. Tissue specificity of the antiserum was reconfirmed by Western immunoblotting using anti-sheep immunoglobulin G•alkalinephosphatase conjugate as a secondary antibody. The reactivity of antiserum to the external surface of fixed swine adipocytes was confmned by an immunohistochemical technique using anti-sheep immunoglobulin G-FITC. Confluent swine adipocytes in culture were lysed by antiserum treatment and cytosolie lactate dehydrogenase(LDH) was released as a dose-dependent patterns while adipocytes treated with normal sheep serum maintained their integrity and expressed low level of LDH. These results implicate that fat contents in the pigs can be reduced by immunological methods.
