  • The measurement of strains in stamped sheet metal is essential to the design and manufacture of sound sheet metal products. The measured strains can also be used in verifying the reliability of the computer analysis such as finite element analysis. In most engineering applications, strains are measured from the deformed square grids or deformed circular grids in comparison with the initial undeformed grids. In such a case, however, strains are averaged in each grid and the localized strain in a region smaller than a grid size can not be measured. In the present study, the B-spline surface interpolation technique is introduced in order to measure the strains more exactly and effectively. The strains calculated by using the surface interpolation technique are compared with the strains calculated from the three-noded grids as well as with the finite element analysis.

Cell Surface Display of Four Types of Solanum nigrum Metallothionein on Saccharomyces cerevisiae for Biosorption of Cadmium

  • We displayed four types of Solanum nigrum metallothionein (SMT) for the first time on the surface of Saccharomyces cerevisiae using an α-agglutinin-based display system. The SMT genes were amplified by RT-PCR. The plasmid pYES2 was used to construct the expression vector. Transformed yeast strains were confirmed by PCR amplification and custom sequencing. Surface-expressed metallothioneins were indirectly indicated by the enhanced cadmium sorption capacity. Flame atomic absorption spectrophotometry was used to examine the concentration of Cd2+ in this study. The transformed yeast strains showed much higher resistance ability to Cd2+ compared with the control. Strikingly, their Cd2+ accumulation was almost twice as much as that of the wild-type yeast cells. Furthermore, surface-engineered yeast strains could effectively adsorb ultra-trace cadmium and accumulate Cd2+ under a wide range of pH levels, from 3 to 7, without disturbing the Cu2+ and Hg2+. Four types of surfaceengineered Saccharomyces cerevisiae strains were constructed and they could be used to purify Cd2+-contaminated water and adsorb ultra-trace cadmium effectively. The surface-engineered Saccharomyces cerevisiae strains would be useful tools for the bioremediation and biosorption of environmental cadmium contaminants.

기계 가공면의 소성스트레인에 관한 연구 (A Study on Plastic Strain in Machined Surface)

  • Typical plastic strains in the machined surface are very difficult to measure, since they are located within a very short distance from the surface and they change very rapidly. There is an alternative way to determine the residual strain in plastically deformed materials by measuring the grain size after a subsequent recrystallization precess. Although, this technique has been successfully applied by several researchers to find the plastic zone around notches and cracks in various materials and welding beads, few works have been reported using the recrystallization method to determine the residual strains in machined surface. Therefore, the purpose of this investigation is to explore the effectiveness of the recrystallization technique in machining applications and in particular, to find the effect of cutting parameters, i.e., depth of cut, rake angle, on the plastic strains. As the result, the recrystallization technique was succesfully applid to determine the plastic strain in machined surface.

  • It is well known that metal cutting leaves a plastically deformed layer in the machined surface. This residual phenomenon affects in various forms the physical properties of machined components such as the fatigue strength, the dimensional instability, microcracks, and the stress corrosion cracking. These physical properties, so called surface integrity, are very important for designing highly stressed and critically loaded components.   • Longitudinal strains({$varepsilon}_x$) of the core and skin layers in glass fiber reinforced plastic(GFRP) cross-ply composite laminates have been studied using the embedded optical fiber sensor of totally-reflected extrinsic Fabry-Perot interferometer(TR-EFPI). Foil-type strain gauges bonded on both the upper and lower surfaces were used for the measurement of the surface strains. Both TR-EFPI sensor and strain gauge bonded on the specimen surface showed excellent agreement within -0.0086 ~ +0.0302% strain. It was shown that values of {$varepsilon}_x$ in the interior of the surface layer and the core layer measured by embedded TR-EFPI sensor was significantly higher than that of the specimen surface measured by strain gauges. The experimental results were ascertained with finite element analysis. Embedded TR-EFPI optical fiber sensor could measure accurately the internal strains which were different from the surface.

확률론에 의한 Single Surface 구성모델의 변형률 예측능력 평가 (Probabilistic Evaluation on Prediction of the Strains by Single Surface Constitutive Model)

  • 본 문은 Lade의 Single surface 구성모델의 변형율 예측 능력을 평가하기 위해 백마강모래를 사용, 등방압축시험과 배수삼축시험을 반복 시행하여 모델에 필요한 각 토질매개변수값을 다수 구하여 통계처리 하였다. 그리고 1계근사법을 이용하여 이 구성모델의 변형율 예측능력을 확률론적으로 평가하였다. 그 결과 변동계수와 상관계수를 효과적으로 이용하여 토질매개변수의 수를 줄일 수 있을 것으로 기대되며 변동계수가 0.51 이하로서 이 구성모델의 변형율 예측 능력은 확률론적으로 매우 안정된 구성모델임을 알았다.

Effects of essential oil (blended and single essential oils) on anti-biofilm formation of Salmonella and Escherichia coli

  • Background: Biofilms were the third-dimensional structure in the solid surface of bacteria. Bacterial biofilms were difficult to control by host defenses and antibiotic therapies. Escherichia coli (E. coli) and Salmonella were popular pathogenic bacteria that live in human and animal intestines. Essential oils are aromatic oily liquids from plant materials and well known for their antibacterial activities. Method: This study was conducted to determine effect of essential oil on anti-biological biofilm formation of E. coli and Salmonella strains in in vitro experiment. Two kinds of bacterial strains were separated from 0.2 g pig feces. Bacterial strains were distributed in 24 plates per treatment and each plates as a replication. The sample was coated with a Bacterial biofilm formation was. Result: Photographic result, Escherichia coli (E. coli) and Salmonella bacteria colony surface were thick smooth surface in control. However, colony surface in blended and single essential oil treatment has shown crack surface layer compared with colony surfaces in control. Conclusion: In conclusion, this study could confirm that essential oils have some interesting effect on anti-biofilm formation of E. coli and Salmonella strains from pig feces.

수종의 상악 총의치수지상 금속보강법에 관한 비교연구 (A COMPARATIVE STUDY ON THE SEVERAL METAL REINFORCEMENT METHODS OF MAXILLARY COMPLETE ACRYLIC RESIN DENTURE BASE)

  • A common site of fracture in maxillary complete denture is on the anteroposterior midline that coincides with the notch for relief of the labial frenum. Various approaches to reduce the incidence of this type of fracture have been suggested. The most widely used technique is the reinforcement of acrylic resin denture base with several solid metal forms. But few comparative studies on the efficacy of metal reinforcements have been reported. This study was conducted to compare reinforcing effects of commonly available metal reinforcements, which include wire, metal mesh embedded in the denture base and metal plate affixed to the impression surface of denture base by silicoating technique. This was load on the posterior. The strain gauges were oriented perpendicular to the anteroposterior midline of maxillary polished denture surface at one labial and the four palatal sites Non-renforced denture was used as control. The results were as follows : 1. In the non-reinforced denture group, only tensile strains on the palatal polished surface were observed. The tensile strains decreased in the order of incisive papilla, posterior denture border area, mid palatal area and rugae area. Compressive strain was observed on the labial polished surface. 2. As compared with the non-reinforced denture group, the metal plate or the metal mesh reinforced denture groups showed reduced palatal tensile strains,and the metal mesh reinforcement had a better reinforcing effect than the metal plate. But both reinforced denture groups showed no difference in the amount of compressive strain on the labial polished surface when compared to the non-reinforced denture group. 3. The metal wire positioned just above the labial notch decreased the compressive strain on the labial polished surface. But the presence of metal wires in the palatal polished surface caused increase in tensile strains in the area.

  • Xylose를 효소 유도물질로한 배지에서 glucose isomerase를 생산하는 균주를 분리하기 위하여 전국각지의 토양시료 150점에서 약 280주의 Streptomyces속 균주를 분리하여 그 중에서 효소 활성능이 높은 5주를 분리하였다. 이들 균주의 성질을 조사한 결과, 서로 다른 균종임을 확인하고 분류가 완성될 때까지 Streptomyces(st) spp. K-14, K-53, K-71, K-77 및 K-733 이라 부르기로 하였다. 특히 K-77과 K-733균주는 포자 표면이 침상형임으로 다른 균종과 완전히 구별되었다. 이들 균주 중에서 K-14균주는 효소 활성능이 가장 양호하였다.

Adhesion Properties of Indigenous Dadih Lactic Acid Bacteria on Human Intestinal Mucosal Surface

  • Dadih is Indonesian traditional fermented buffalo milk believed by the natives to have beneficial effects on human health. This may be due to the probiotic properties possessed by the lactic acid bacteria (LAB) involved in its fermentation process. It was discovered that ten strains of dadih lactic isolates possessed some probiotic properties in vitro. In this study, the adhesion properties of dadih LAB, in comparison with documented probiotic strains, were investigated in vitro by using mucin extracted from human faeces and Caco-2 cells as the models for human intestinal mucosal surface and intestinal cells respectively. The adhesion results showed the distinction of Lactobacillus reuteri IS-27560 in adhering to both mucus layer and Caco-2 cells. The competition assay for adhesion to the mucus layer between dadih LAB and selected pathogens indicated the competence of Lactococcus lactis IS-16183 and Lactobacillus rhamnosus IS-7257 in significantly inhibiting the adhesion of Escherichia coli O157:H7. Accordingly, these two strains may be potential candidates for use as probiotic strains. Overall, the adhesion properties of all dadih LAB strains were relatively comparable to that of Lactobacillus casei Shirota and Lactobacillus rhamnosus GG, the documented probiotic strains.
