The surface-enhanced Raman scattering(SERS) of methoxybenzonitrile in a silver sol was investigated. All of ortho-, meta-, and para-substituted benzonitriles were found to adsorb on the silver surface via the nitrogen lone pair electrons. The benzene ring of meta derivative seemed likely to stand perpendicular to the silver surface, while the benzene rings of both otho and para derivatives assumed tilted stances with respect to the surface. The SERS technique appeared to be a useful means to study the substituent electronic effects.
We demonstrate the possible application of the sandwich type surface-enhanced Raman scattering (SERS) immunoassay using antigen-antibody binding for detection of prostate-specific antigen (PSA) in cancer cells. In this sandwich type of SERS immunoassay, to capture antigens onto the immobilized layer of antibodies on the gold substrate we prepared the monolayer of gold nanoparticles on the APTMS-derivatized surface of a glass slide by using the SAM technique. This sandwich type of SERS immunoassay in which antigens on the substrate specifically capture antibodies on a Raman reporter (DSNB coated gold nanoparticles with R6G) could successfully detect PSA at low levels. A strong SERS spectrum of Raman reporter was observed only with a substrate in which PSA is present.
Surface-Enhanced Raman Scattering (SERS) spectroscopy is a multifaceted surface sensitive methodology which exploits spectroscopy-based analysis for various applications. This technique is based on the massive amplification of Raman signals which were feeble previously in order to use them for appropriate identification at qualitative and quantitative in chemical as well as biological systems. This novel powerful technique can be utilized to identify pathogens such as bacteria and viruses. As far as SERS is concerned, one of the most studied problems has been functionalization of SERS active substrate. Metal colloids and nanostructures or microstructures synthesized using noble metals such as Au, Ag and Cu are considered to be SERS active. Silver and gold are extensively used as SERS active substrates due to chemical inertness and stability in air compare to copper. However, use of Cu as a suitable alternative has been taken into account as it is cheap. Herein, we have synthesized air-stable copper microstructures/nanostructures by chemical, electrochemical and microwave-assisted methods. In this paper, we have also discussed the use of as synthesized copper micro/nanostructures as inexpensive yet effective SERS active substrates for the fast identification of micro-organisms like Staphylococcus aureus and Escherichia coli.
Surfaced-enhanced Raman scattering (SERS) of 4,4'-biphenyldithiol (BPDT) has been investigated at a silver island film. Ordinary Raman (OR) spectra of neat sample in solid state and in basic solution have also been taken for comparison. The spectral feature in the SERS spectrum was similar to that for the OR spectrum in basic solution, except for the broadening of ring stretching bands indicative of the presence of surface-phenyl ring $\pi$ interaction. In contrast, only absence of the C-H stretching band with very small Raman scattering cross-section seemed not pertinent in judging the definitive orientation of molecule. The observed vibrational bands in the SERS spectrum have been assigned by referring to the normal modes and wavenumbers from density functional theory (DFT) calculations of the simple model as 4,4'-biphenyldithiolates bound to two Ag atoms at the both ends. Excellent agreement between the experimental and the calculated results was achieved, which is remarkable considering the level of theory applied.

This study reports a fiber-optic sensor detecting biomolecule by simultaneously monitoring localized surface plasmon resonance (LSPR) from gold nanoparticles (Au NPs) of ca. $50{\pm}5$ nm attached on one end of optical fiber and surface enhanced Raman scattering (SERS) of the reporter molecules adsorbed on the gold surfaces as an additional sensing tool. The sensor was fabricated by immobilizing Au NPs on one end of an optical fiber by chemical reaction. LSPR and SERS signals of the sensor were measured using various refractive indices solutions. Finally, the sensor was applied to observe real-time LSPR sensor-gram and SERS spectra of the reporter molecule of 4-aminothiphenol during the antibody-antigen reaction of interferon-gamma (IFN-${\gamma}$) as a proof-concept experiment of biological applications.
In this study, we synthesize silica-core gold-satellite nanoparticles (SGNPs) for the surface-enhanced Raman scattering (SERS) based sensing applications. They consist of gold satellite nanoparticles (AuNPs) fixed on the silica core nanoparticles, which sizes of AuNPs can be tunned by varying the amount of reactants (growth solution and reducing agent). Their surface plasmon resonance (SPR) properties were characterized by using UV-vis spectroscopy, showing that the growth of AuNPs on silica cores leads to the light absorption in the longer wavelength region. Furthermore, the size increase of AuNPs exhibited the dramatic change in SERS activity due to the formation of hot spots. The optimized SGNPs showing enhancement factor ${\sim}3.8{\times}10^6$ exhibited a detection limit of rhodamine 6G (R6G) as low as $10^{-8}M$. These findings suggest the importance of size control of SGNPs and their SPR properties to develop highly efficient SERS sensors.

Controlling the shape of Ag nanoparticles (NPs) is very difficult. In the present work, urchin Ag NPs with different sizes and pod length control have been synthesized successfully in high yield by the concentration of a reducing agent. Unique Ag NPs were observed by TEM and SEM. These nanocrystals exhibit tunable surface plasmon resonance properties from the visible to near-infrared regions. They were applied to surface-enhanced Raman scattering (SERS) substrates using rhodamine 6G (R6G), benzenethiol (BT), and 4-amino benznethiol (4-ABT) molecules. The enhanced local field effect due to the sharp pod length, size, and surface plasmon of the urchin Ag NPs resulted in enhanced SERS properties and can serve as high-sensitivity substrates for SERS measurements.

It has been recently reported that the SERS (Surface Enhanced Raman Scattering) effect of the silver mirror substrate made by Tollen's method is much superior to that of other substrates. In this study, the experiments for comparison of surface enhancement of silver sol and silver mirror substrates were done, where we checked the characteristics of silver mirror substrate made by Tollen's method. The surface enhancement of fluorescein was analyzed by silver sol and silver mirror substrates. We observed the SERS spectra of fluorescein. The assignments of the vibrational bands shown in SERS spectra are given based on both literature and the semi-empirical calculations at the PM3 methods. The surface enhancement properties for fluorescein showed that the silver mirror was more superior to sliver sol substrates. Spectra of fluorescein revealed that fluorescein was adsorbed on silver surfaces by a common oxygen atom. According to the 'surface selection rule', the vibrations in the band intensities reflect the adsorption orientation of the molecule on to the surface of SERS substrates. Therefore, we deduced that the adsorption orientation of fluorescein was little tilted perpendicular to the silver surfaces by using of the surface selection rules.

Raman spectra of a single layer graphene sheet placed in different gold substrates were obtained and are discussed in the context of surface enhanced Raman scattering (SERS). The gold substrates were composed of a combination of a thermally deposited gold film and a close-packed gold nanosphere layer. The SERS effects were negligible when the excitation wavelength was 514 nm, while the Raman signals were enhanced 3-to 50-fold when the excitation wavelength was 633 nm. The large SERS enhancement accompanied a spectral distortion with appearance of several unidentifiable peaks, as well as enhancement of a broadened D peak. These phenomena are interpreted as the local field enhancement in the nanostructure of the gold substrates. The difference in the enhancement factors among the various gold substrates is explained with a model in which the spatial distribution and polarization of the local field and the orientation of the inserted graphene sheet are considered important.
Surface-enhanced Raman scattering(SERS) spectroscopy was employed to analyze the genotoxic nucleobase adducts of benzo[a]pyrene(BP) formed through one-electron oxidation pathway. SERS spectroscopy provided sufficient resolution to distinguish if BP intermediate was bound to different nucleobases(e. g. adenine or guanine). Furthermore, SERS specroscopy was also able to detect the difference in the binding position of the adduct to the various sites of the nucleobase. The linearity of the calibration curve for N7Ade-BP ranged from 20 picogram to 800 nanogram per microliter and the detection limit under the current conditions was determined 20 picogram per microliter in a solution volume of 20 microliter.
