Identification and Characterization of an Oil-degrading Yeast, Yarrowia lipolytica 180

  • Among oil-degrading microorganisms isolated from oil-polluted industrial areas, one yeast strain showed high degradation activity of aliphatic hydrocarbons. From the analyses of 18S rRNA sequences, fatty acid, coenzyme Q system, G+C content of DNA, and biochemical characteristics, the strain was identified as Yarrowia lipolytica 180. Y. lipolytica 180 degraded 94% of aliphatic hydrocarbons in minimal salts medium containing 0.2% (v/v) of Arabian light crude oil within 3 days at 25$^{\circ}C$. Optimal growth conditions for temperature, pH, NaCl concentration, and crude oil concentration were 30$^{\circ}C$, pH 5-7, 1%, and 2% (v/v), respectively. Y. lipolytica 180 reduced surface tension when cultured on hydrocarbon substrates (1%, v/v), and the measured values of the surface tension were in the range of 51 to 57 dynes/cm. Both the cell free culture broth and cell debris of Y. lipolytica 180 were capable of emulsifying 2% (v/v) crude oil by itself. They were also capable of degrading crude oil (2%). The strain showed a cell surface hydrophobicity higher than 90%, which did not require hydrocarbon substrates for its induction. These results suggest that Y. lipolytica has high oil-degrading activity through its high emulsifying activity and cell hydrophobicity, and further indicate that the cell surface is responsible for the metabolism of aliphatic hydrocarbons.

일부 정신지체 장애인의 구강건강에 관한 연구 (A study on the oral health of some disabled people with mental retardation)

  • Purpose : The purpose of this study was to examine the oral health state of disabled people with mental retardation in an attempt to pave the way for oral health care planning geared toward the disabled. Subjects and Method : The subjects in this study were 46 mentally retarded people who attended rehabilitation centers for the disabled in the city of Wonju, Gangwon Province. After a survey was conducted, the collected data were analyzed with SPSS 13.0 program, and frequency analysis, percentage, ANOVA and t-test were utilized. Results : The findings of the study were as follows: 1. The simplified oral hygiene index of the mentally retarded people was 1.32, which was on the average. 2. The decayed teeth index and decayed teeth rate of the mentally retarded people were respectively 13.48 and 48.13 percent, which were above the average. 3. The decayed surface index and decayed surface rate of the mentally retarded people were respectively 27.17 and 17.39 percent. 4. The simplified debris index of the mentally retarded people significantly varied with gender(p<.01), and missing teeth index(p<.05) and missing surface index differed significantly with age. Filled surface index was significantly different according to the region. Conclusion : The findings of the study illustrated that their indexes related dental caries experience were high. In order to promote the oral health of the disabled with mental retardation, prolonged research should be implemented, and a dental checkup should be carried out on a regular basis by specialists. Preventive care and early treatment should be provided, and the development of customized oral health education programs tailored to different sorts of disabilities and oral health control methods is required.

핵연료 봉의 마찰변태구조 관찰과 프레팅 마멸 특성 (Observation of Tribologically Transformed Structures and fretting Wear Characteristics of Nuclear Fuel Cladding)

  • In this research, fretting tests were conducted in air to investigate the wear characteristics of fuel cladding materials with the fretting parameters such as normal load, slip amplitude, frequency and the number of cycles. A high frequency fretting wear tester was designed for this experiment by KAERI. After the experiments, the wear volume and the shape of wear contour were measured by the surface roughness tester. Tribologically transformed structures(TTS) were analysed by means of optical and scanning electron microscopes to identify the main wear mechanisms. The results of this study showed that the wear volume were increased with increasing slip amplitude, and the shape of wear contour was transformed V-type to W-type. Also, it was found that the critical slip amplitude was 168${\mu}{\textrm}{m}$. These phenomena mean that wear mechanism transformed partial slip to gross slip to accelerate wear volume. The wear depth increased with an increase of friction coefficient due to increase of normal load and frequency. The fretting wear mechanisms were believed that, after adhesion and surface plastic deformation occurred by relative sliding motion on the contact between two specimens, TTS creation was induced by surface strain hardening and wear debris were detached from the contact surface which were produced by the micro crack propagation and creation.

Optimal strategy for low surface brightness imaging with KMTNet

  • Most galaxies are believed to evolve through mergers and accretions. In particular, minor mergers and gas accretion appear to play an important role in galaxy evolution in the present-day Universe. Tidally-disrupted debris from such processes remain as diffuse, low-surface brightness structures because the dynamical timescale in the outskirts is significantly longer than that in the central regions. Although these structures will give us useful insight into the mass assembly history of galaxies, it is difficult to detect them due to their faint surface brightness. In order to investigate the structural properties of outskirts in nearby galaxies, we conduct deep and wide-field imaging survey with KMTNet. We present our observing strategy and an optimal data reduction process to recover faint extended features in the images of KMTNet. Using the imaging data of NGC 1291 obtained from KMTNet, we find that a peak-to-peak sky gradient can be reduced less than 0.4-0.6% of the original sky level in the entire image. We also find that we can reach the surface brightness of ${\mu}_{(B,1{\sigma})}$ ~ 29.5, ${\mu}_{(R,1{\sigma})}$ ~ 28.5 mag $arcsec^{-2}$ in one-dimensional profile, that is mainly limited by the uncertainty in the sky determination. It indicates that deep imaging data of KMTNet is suitable to study the extended faint features of nearby galaxies, such as stellar halos, outer disks, and dwarf companions.

자동차용 마찰재에 함유된 세라믹분말의 함량에 따른 마찰특성 (Friction Characteristics of Automotive Friction Materials with Ceramic Powder Contents)

  • The friction characteristics of automotive brake friction materials that contained different ceramic content were investigated. Several kinds of raw materials, such as resin-based binder, reinforcing fiber, friction restraint, abrasive, and filling materials were mixed, pressed, and heated in order to make the brake friction materials. The contents of SiC and $BaSO_4$ changed from 5 vol% to 20 vol%, respectively. In addition to this, the content of $Al_2O_3$ adjusted from 1 vol% to 16 vol%. The surface morphology of the SiC containing sample appeared rough while more debris was observed when the contents of SiC increased. This implies that the SiC containing brake composite was not adequate for the automobile. However, the relatively smooth surface was observed in samples that contained the $Al_2O_3$. But the roughness was low with a content of 11 vol% $Al_2O_3$ compared to the other samples. This is consistent with the abrasive properties of the samples. In the case of $BaSO_4$ containing samples, the smoothes surface was observed in the contents of 15 vol% $BaSO_4$. Thus, it was concluded that the 11 vol% $Al_2O_3$ and 15 vol% $BaSO_4$ containing composite would be the optimum content for the brake composite. Similar to the results of the surface morphology, the abrasion resistance consistently decreased when the content of SiC increased. On the contrary, the sample that contained 11 vol% $Al_2O_3$ and 15 vol% $BaSO_4$ showed the highest abrasion resistance compared to the other samples.

  • 지난 ~30년 동안 가속기질량분석기와 비활성기체질량분석기 기술의 진보와 함께, 우주선유발 동위원소도 지표면의 연대측정 분야에 광범위하게 적용되어 왔다. 우주선유발 동위원소를 이용한 세부적인 연대측정법으로는 단순노출연대측정, 수직단면연대측정, 매몰연대측정 등이 있어, 다양한 노출(또는 퇴적) 환경에 따라, 다른 접근법이 요구된다. 국내에 적용 가능한 대상으로는, 하안단구, 해안단구, 선상지, 화산지형, 구조지형(단층애), 다양한 암설지형(테일러스, 암괴류 등), 호안 또는 해안 파식대 그리고, 제3기의 퇴적분지층과 고고학유물층에도 적용이 가능하다. 아울러, 기존의 석영이 풍부한 화강암과 변성암 중심에서, 최근에는 석회암과 화산암에도 적용가능하게 되었다.

초음파나노표면개질기술로 제작된 마이크로 그루브가 미끄럼 마찰 특성에 주는 영향 (Effect of Micro-grooves Manufactured via Ultrasonic Nanocrystalline Surface Modification on Sliding Friction)

  • The surface texture produced via surface texturing is an important approach for controlling the tribological behavior of friction behavior of mechanical devices. The purpose of this study is to investigate the effect of grooves generated via ultrasonic nanocrystal surface modification (UNSM) technology on the tribological performance of AISI 4150 steel against stainless steel 316L. In the study, tribological tests are performed under two different regimes, namely mixed and hydrodynamic lubrication, by varying the applied normal load and reciprocating speed during the tests. According to the test results, the friction coefficient decreases as static load (10 N, 30 N, and 50 N) of UNSM technology increases in the mixed lubrication regime. Conversely, the friction coefficient increases as the static load (10 N, 30 N, and 50 N) of UNSM technology increases in the hydrodynamic lubrication regime. Hence, the results indicate that micro-grooves generate hydrodynamic pressure in the outlet, which increases the oil film thickness between the two mating surfaces. This potentially leads to a reduction in friction in the mixed lubrication regime due to the prevention of contact of asperities and debris. However, the results indicate an adverse effect in the hydrodynamic lubrication regime. In this regard, additional experiments should be performed to investigate the effect of grooves generated by UNSM technology at varying conditions on the friction behavior of AISI 4150 steel, which in turn can be controlled by the generated pressure and oil film thickness at the contact interface.

인공강우 시 나지교란사면 토사유출에 미치는 다짐처리의 영향 (Effects of Surface Compaction Treatment on Soil Loss from Disturbed Bare Slopes under Simulated Rainfalls)

  • 토양의 다짐은 강우 시 지표유출 및 토사유출에 큰 영향을 끼친다. 다짐은 체적밀도 증가, 전단강도 증가, 공극률 변형, 투수계수 등과 같은 토양특성 변화를 야기하기 때문이다. 본 연구는 인공강우 실험을 활용하여 개발지의 사면조건과 유사한 나지교란사면에서 표면의 다짐처리가 지표유출 및 토사유출에 미치는 영향을 파악하였다. 표면처리(다짐, 비다짐), 강우강도(68.5mm/hr, 95.6mm/hr), 사면경사($5^{\circ}$, $12.5^{\circ}$, $20^{\circ}$)의 각 조건별 3회 반복하여 총 36회의 강우모의에 따른 지표유출 및 토사유출을 측정하였다. 연구결과, 다짐처리 후 토양의 체적밀도 및 전단강도는 유의적으로 증가하였다. 그러나 이러한 물리적 특성의 변화가 지표유출에 미치는 영향은 강우강도와 사면경사에 따라 다르게 반응하였다. 평균 토사유출량은 강우강도와 사면경사가 증가함에 따라 유의적으로 증가하였다. 또한, 토사유출량은 강우강도와 사면경사 별 다짐처리 유 무에 따라 다른 반응을 보였다. 완경사($5^{\circ}$)에서는 다짐처리에서 더 많은 토사가 유출되었으나, 급경사($20^{\circ}$)에서의 다짐처리는 토사유출을 감소시키는 역할을 한 것으로 나타났다. 나지교란사면에서는 토사유출에 대한 다짐효과의 천이구간이 존재하는 것으로 파악되며, 본 연구의토양조건 및 강우조건에서 천이구간은 완경사와 급경사 사이로서 사면경사 $10{\sim}15^{\circ}$ 범위에 존재하는 것으로 판단된다.

의성 빙계계곡 일대의 지형적 특성 -테일러스를 중심으로- (Geomorphic Features of Bing-gye Valley Area(Kyongbuk Province, South Korea) -Mainly about Talus-)

  • 연구지역에서 발달하는 테일러스들은 테일러스의 일반적인 특성을 잘 반영하고 있다. 우리나라에 분포하는 테일러스가 보통 rock fall talus이듯이 본 연구지역의 테일러스도 동일한 유형으로 판단된다. 또 테일러스의 사면형태, 형성시기 발달과정, 미지형적 특성 등은 지금까지 발표된 여러 연구결과들과 거의 일치하고 있다. 그러나 인접한 테일러스간의 경계 부분에서는 암설입경의 상대적 크기에 따라 약간의 지형적 차이점을 인식할 수 있었다. 특히 본 연구에서는 테일러스 암설 최적층의 내부구조를 국내 처음으로 확인하여 3가지의 뚜렷한 층위를 구분할 수 있었다. 즉, 암설로만 구성된 표층과 암실과 세립물질(실트질 모래)이 혼재된 중간층 그리고 과거의 토양층인 기저층 등으로 구분된다. 또 암설 퇴적층에 대한 내부구조 인식은 테일러스 형성 당시로 부터 현재까지의 기후환경과 암설의 퇴적양상을 밝히는데 매우 중요하며 앞으로 이 분야의 연구에 큰 도움을 줄 것으로 생각된다. 한편 국지적으로 특수기상이 나타나는 빙혈이 있는 빙계계곡의 경우 그것의 형성에는 단층작용의 역할이 중요한 것으로 판단된다. 그러나 이 곳을 흐르는 쌍계천이 유독 빙계계곡에서만 협곡(빙계계곡의 폭은 주변 곡쪽의 $1/8{\sim}1/10$정도임)을 형성하는 이유에 대해서 정곡의 가능성을 제시했지만 입증할 만한 자료가 없는 것이 유감이다.

나노기공구조를 가진 알루미나필름의 트라이볼로지 특성 (Tribological Properties of Nanoporous Structured Alumina Film)

  • Tribological properties of nanoporous structured alumina film was investigated. Alumina film (AAO: anodic aluminum oxide) of $60{\mu}m$ thickness having nanopores of 45 nm diameter with 105 nm interpore-diatance was fabricated by mild anodization process. Reciprocating ball-on-flat sliding friction tests using 1 mm diameter steel ball as a counterpart were carried out with wide range of normal load from 1 mN to 1 N in an ambient environment. The morphology of worn surfaces were analyzed using scanning electron microscopy. The friction coefficient was strongly influenced by the applied normal load. Smooth layer patches were formed on the worn surface of both AAO and steel ball at relatively high load (100 mN and 1 N) due to tribochemical reaction and compaction of wear debris. These tribolayers contributed to the lower friction at high loads. Extremely thin layer patches, due to mild plastic deformation of surface layer, were sparsely distributed on the worn surface of AAO at low loads (1 mN and 10 mN) without the evidence of tribochemical reaction. Delaminated wear particles were generated at high loads by fatigue due to repeated loading and sliding.
