인삼엽요병에서 효소활성도의 변화 (Study on the Enzyme Activity in Leaf-Burning Disease of Panax ginseng C.A. Meyer)

  • 인삼엽을 강광(100 KLuw) 및 고온($45^{\circ}C$, 암상태)에 처리하여 효소(glucose-6-phosphate dehydrogenase, acid phosphatase, catalase, peroxidase)의 활성도를 조사한 결과 두 처리구에서 공히 감소하는 경향이었으나, 특기 강광에서 활성도가 현저히 감소하였다. 이와 같은 활성도 감소는 효소의 thermal stabilities나 coagulation 등과 같은 광에 의한 2차적인 엽온상승 효과에 따른 inactivation이 아니며, proteolytic activity 증가로 인한 효소단백질의 함량감소로 확인되었다. 인삼엽에서 proteolytic activity가 강광에 의해 급속히 증가하는 것으로 보아 정상엽(normal leaf)에 inactive 상태로 내재(compartmentation)되어 있는 proteinase가 타 식물에 비해 많은 것으로 사료된다. 또한 chlorphyll bleaching과 효소의 inactivation을 유발시킬 수 있는 superoxide radical(${O_2}^{-}$)의 광화학적 생성율이 비교식물(Solanum nigrum)보다 높게 나타나고 crude saponin이 superoxide의 생성율을 촉진하는 것으로 보아 superoxide에 의한 pigment system의 광산화율이 타 식물에 비해 높을 것으로 사료된다.

  • An antioxidant was isolated from Korean mistletoe (Viscum album var. coloratum) by consecutive purification using silica gel, Sephadex LH-20, and RP-HPLC. The active principle was identified as homo-flavoyadorinin-B (3',7-dimethoxyluteolin-4'-O-[apiosyl $(1{\rightarrow}2)$ glucoside]) by spectral analyses. It inhibited 74.6% of hydroxyl radical and 30.6% of superoxide anion radical at 0.01 mM; however, th~compound did not show any scavenging activity against hydrogen peroxide radical. At 0.1 mM, above compound scavenged superoxide anion radical about twice as effective as positive controls, BHT and ${\alpha}-tocopherol$. Radical scavenging activities of homo-flavoyadorinin-B on DPPH, hydroxyl, and hydrogen peroxide radicals were almost same with those of positive controls.

간장조직의 활성산소 및 그 제거효소에 미치는 실크 피브로인의 영향 (Effect of Silk Fibroin on Oxygen Radicals and Their Scavenger Enzymes in Liver of SD Rats)

  • This study was designed to investigate the effect of silk fibroin (Mw 500)power (SFP) on oxygen radicals and their scavenger enzymes in liver membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (SFP-2.5 and SFP-5.0 groups) added 2.5 and 5.0 g/kg BW/day for 6 weeks. Hydroxyl radical (.OH) levels resulted in a considerable decreases (5.8% and 8.4%, 3.7% and 11.1%, respectively) in liver motochondria and micorsomes of SFP-2.5 and SFP-5.0 groups compared with control group, and $O_2$radical level was remarkably decreased about 15% and 20% in liver cytosol of SFP-2.5 and SFP-5.0 groups compared with control group. Lipid peroxide (LPO) levels were significantly decreased (8.3% and 18.0%, 13.4% and 18.4%, respectively) in liver mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group. Oxidized protein (OP) levels were remarkably decreased about 11.6% in liver mitochondria of SFP-5.0 group compared with control group. Mn-SOD activities were remarkably in creased (17.6% and 28.8%, respectively) in mitochondria of SFP-2.5 and SFP-5.0 groups, and Cu/Zn-SOD activities were also effectively in creased (about 14.4%) in liver cytosol of SFP-5.0 groups, but significant difference between GSHPx activity in liver cytosol of these two groups could be not obtained. These results suggest that anti-aging effect of silk fibroin may play an effective anti-aging role in a aattenuating a oxidative stress and increasing a scavenger enzyme activity in liver membranes.

Antioxidant Activities of Red Algae from Jeju Island

  • The aim of the present study was to evaluate the antioxidant activity of red algae in Jeju Island. The algal extracts were obtained with MeOH and fresh water at 20 and 70°C, and screened for antioxidant activities using hydroxyl radical (HO·), superoxide anion (O2–), hydrogen peroxide (H2O2) and DPPH free radical scavenging assays. Among them, Gracilaria verrucosa methanolic extract at 20°C (20ME, 96.85%), G. textorii aqueous extract at 20°C (20AE, 88.01%), Grateloupia filicina 20AE (85.35%), and Polysiphonia japonica 20ME (94.92%) exhibited the highest scavenging activities against HO·, O2–, H2O2, and DPPH free radicals, respectively. Moreover, P. japonica (20ME and 70ME) is correlated between DPPH free radical scavenging activity and polyphenolic contents. These results indicate that some red algae in Jeju Island could be potential candidates for development of antioxidants.

Phloroglucinol Attenuates Free Radical-induced Oxidative Stress

  • The protective role of phloroglucinol against oxidative stress and stress-induced premature senescence (SIPS) was investigated in vitro and in cell culture. Phloroglucinol had strong and concentration-dependent radical scavenging effects against nitric oxide (NO), superoxide anions ($O_2{^-}$), and hydroxyl radicals. In this study, free radical generators were used to induce oxidative stress in LLC-PK1 renal epithelial cells. Treatment with phloroglucinol attenuated the oxidative stress induced by peroxyl radicals, NO, $O_2{^-}$, and peroxynitrite. Phloroglucinol also increased cell viability and decreased lipid peroxidation in a concentration-dependent manner. WI-38 human diploid fibroblast cells were used to investigate the protective effect of phloroglucinol against hydrogen peroxide ($H_2O_2$)-induced SIPS. Phloroglucinol treatment attenuated $H_2O_2$-induced SIPS by increasing cell viability and inhibited lipid peroxidation, suggesting that treatment with phloroglucinol should delay the aging process. The present study supports the promising role of phloroglucinol as an antioxidative agent against free radical-induced oxidative stress and SIPS.

큰도둑놈의갈고리 지상부의 DPPH Free Radical 소거효과 (DPPH Free Radical Scavenging Effect of the Aerial Parts of Desmodium oldhami)

  • The antioxidant activity of Desmodium oldhami (Leguminosae) was determined by measuring the radical scavenging effect on DPPH (1,1-diphenyl-2-picrylhydrazyl). The methanolic extract of D. oldhami showed the strong antioxidant activity. Five compounds, kaempferol-3-O-rhamnopyranoside (afzeline) (1), quercetin-3-O-[$\alpha$-L-rhamnopyranosyl($1{\rightarrow}6$)-$\beta$-Dglucopyranoside] (rutin) (2), kaempferol-3-O-glucopyranoside (astragalin) (3), genistein-7-O-$\beta$-D-glucopyranoside (genistin) (4), kaempferol-3-O-rutinoside (5) were isolated from the active ethyl acetate soluble fraction of D. oldhami through repeated silica gel and Sephadex LH-20 column chromatography. Among them, compound 2 showed the most significant antioxidative effect on DPPH free radical scavenging test. Compounds 1-5 are reported for the first time from this plant.

Biphenyldimethyl dicarboxylate(DDB)가 염화 제2수은-유발 간독성 흰쥐에서의 지질 과산화와 Oxygen Free Radical 제거효소 활성도에 미치는 영향 (Effects of Biphenyldimethyl Dicarboxylate(DDB) on the Lipid Peroxidation and Oxygen Free Radical Scavenging Enzyme Activities in Mercuric Chloride-induced Hepntotoxic Rats)

  • In an attempt to define the effects of biphenyldimethyl dicarboxylate (DDB) on the lipid peroxidation and oxygen free radical scavenging enzymes activities in mercuric chloride-induced hepatotoxic rats, we studied malondialdehyde (MDA) level and the activities of catalase and superoxide dismutase (SOD) in liver of the rats at 24 hr after the injection of mercuric chloride. Sprague-Dalwey albino rats were injected subcutaneously with mercuric chloride (5 mg/kg) only and mercuric chloride (5 mg/kg) plus. DDB (200 mg/kg/day, p.o) is administered for 4 days prior to 3 days from the injection of mercuric chloride. The group treated with mercuric chloride showed significantly higher MDA level and lower catalase and SOD activities as compared with that of control group. The group treated with mercuric chloride plus DDB showed significantly lower MDA level and catalase activity and higher SOD activity as compared with that of mercuric chloride-treated group. These results suggest that the excessive oxygen free radicals resulting from the depression of superoxide dismutase activity is an important determinant in the pathogenesis of mercuric chloride-induced hepatotoxicity and DDB has antioxidant effects.

시호사물탕이 t-BHP로 유도된 간세포의 산화적 손상 및 자유기에 의한 지질과산화 반응에 미치는 영향 (Preventive Effects of Sihosamultang Extract on Lipid Peroxidation by Free Radicals and Oxidative Damage of Hepatocytes by tert-Butyl Hydroperoxide)

  • Sihosamultang(SST) has been used for the treatment of puerperal fever, liver disease in traditional medicine. The present study was carried out to evaluate the antioxidant effects of SST extract in vitro. The inhibitory effect of SST extract on lipid peroxidant was examined in the linoleic acid autoxidation system. In this test, SST extract significantly inhibited the time course of the lipid peroxidation. And SST extract showed about 73% scavenging effect on DPPH radical. And this extract inhibited not only the lipid peroxide formation induced by hydroxyl radical derived from $ H_{2}O_{2}-Fe^{2+}$ in the rat liver homogenate, but also the superoxide generation from xanthine-xanthine oxidase system in a dose-dependent manner. In addition, SST extract protected the hepatic cell death induced by tert-butyl hydroperoxide. These data indicated that SST might play a protective role against oxidative injury by free radicals.

누에분말이 혈청중의 활성산소 및 제거효소에 미치는 영향 (Effects of Silkworm Powder on Oxyen Radicals and Their Scavenger Enzymes in Serum of Rats)

  • This study was designed to investigate the effects of silkworm powder (SWP) on oxygen radicals and their scavenger enzymes in serum of rats. Sprague Dawley (SD) male rats (160${\pm}$10g) were fed experimental diets (SWP-200and SWP-400 groups)added 200 and 400mg/kg BW/day for 6 weeks. Triglyceride (TG)levels were significantly inhibited (10% and 25%) in SWP-200 and SWP-400 groups, but there were no singificant differences in total, LDL-and HDL-cholesterol levels in both SWP-200 and SWP-400 groups. Hydroxyl radical ($.$OH) formations resulted in a marked decreases (about 20%) in both SWP-200 and SWP-400 groups compared with control group. Superoxide radical (O2.-)and hydrogen peroxide formations resulted in a significant decreases in SWP-400 group compared with control group. Lipid peroxide (LPO)and oxidized protein(>C=O group) productions resulted in a significant decreases (SOD) and catalase (CAT) activiteies were remarkably increased (10∼25% and 40∼50%)in SWP-200 and SWP-400groups. Glutathione peroxidase(GSHPX) activities were significantly increased (about 10%) in SWP-400 groups compared with control group. These results suggest that anti-aging effect of silkworm powder (SWP) may play a pivotal role in attenuation a various chronic degenerative diseases age-related changes.

Lipid peroxide(LPO) levels were significantly decreased (14.4% and 9.1%, respectively) in liver mitochondria and microsomes of SWP-400 group only compared with control group. Oxidized protein (OP) levels were remarkably decreased about 12.7% and 16.3% in liver microsomes only of SWP-200 and SWP-400 groups, but significant difference between liver mitochondria could not obtained. Mn-SOD activities were remarkably increased (15.8% and 25.2%, respectively) in mitochondria of SWP-200 and SWP-400 groups, but significant difference between Cu, Zn-SOD activities in these group could be not obtained. GSHPx activity was significantly increased in liver cytosol of SWP-400 group compared with control group. These results suggest that silkworm powder may play an effective role in a attenuating a oxidative stress and increasing a scavenger enzyme activity in liver membranes.
