Antioxidative and Antiaging Effects of Dietary Yellow and Black Soybean in Rats (노란콩 및 검정콩의 섭취가 흰쥐의 항산화 및 항노화 시스템에 미치는 영향)

  • To investigate antioxidative and antiaging effects of yellow and black soybeans in vivo system, male SD rats (n=24) were fed the diets containing casein (C), yellow soybean (YS) or black soybean (BS) for 8 weeks. Experimental groups showed the preventive effect on lipid and protein oxidation, especially protein oxidation of plasma was significantly inhibited in BS group. SOD and catalase activities were not different among the groups, but hepatic glutathione peroxidase (GSH-px) activity was significantly lowered in YS and BS groups compared with control group. The contents of superoxide anion radical in cytosol were significantly lowered in experimental groups compared with control group. And hydroxyl radical was slightly lowered in soybean groups. Lipofuscin accumulation on the heart and eyes of rats was effectively inhibited by yellow soybean and black soybean, respectively. In conclusion, these results suggest that yellow soybean and black soybean have the antioxidative/antiaging effect in vivo system and they have similar activities despite different color of seed coat.

The Inhibition of UVA-induced Matrix Metalloproteinase-1 in Human Dermal Fibroblasts and the Improvement of Skin Elasticity by Cirsium setidens Extract (고려엉겅퀴 추출물의 사람 섬유아세포에 있어서 자외선으로 유도된 MMP-1발현 저해와 피부 탄력 개선 효과)

  • In this study, we measured the anti-oxidative activity of Cirsium setidens and investigated its effect on UVA-induced MMP-1 expression in human dermal fibroblats. And then we examined possible improvement in skin elasticity by topical treatment with fomular including Cirsium setidens extract. The ethanol extract of C. setidens showed free anion radical scavenging effect(87.47 % at 1 mg/mL) and superoxide anion radical scavenging effect(61.71 % at 1 mg/mL) in the xanthine/xanthine oxidase system, respectively. At the concentration of 100 ${\mu}g/mL$, C. setidens extract showed 95.54% inhibition on lipid peroxidation of linoleic acid. UVA-induced MMP-1 expression in human dermal fibroblasts was reduced to 54.69 % by treatment with 100 ${\mu}g/mL$ of C. setidens extract. A human clinical study, in which oil-in-water emulsion with C. setidens extract was topically applied, showed significant increase in skin elasticity. These results suggest that the C. setidens extract can be effective anti-aging ingredient for cosmetics applications.

  • Glinus oppositifolius and Trianthema decandra belonging to the Ficoidaceae family were commonly used by tribal peoples for the treatment of liver disorders and cancer. The preliminary phytochemical screening of those plants showed the presence of flavonoids, terpenoids, tannins and saponins. The aim of this study was to evaluate the in vitro antioxidant activities of the methanol extracts of Glinus oppositifolius (MEGO) and Trianthema decandra (METD). The antioxidative capacities of MEGO and METD were determined by the following four complementary assay; DPPH radical scavenging assay, superoxide anion generation by xanthine-xanthine Oxidase assay, hydroxyl radical scavenging assay and $Fe^{2+}$-ascorbate induced by lipid peroxidation assay. The $IC_{50}$ values of the both extracts were calculated from the inhibition curve. The $IC_{50}$ MEGO and METD in DPPH, superoxide anion, hydroxyl radical scavenging and lipid peroxidation assay are 1.85, 7.31, 13.95, 22.82 and 2.21, 9.78, 14.87, 19.76 ${\mu}g/ml$ respectively. Both the extracts exhibited a significant antioxidant effects.

  • Hydrogen-donating capacity, scavenging activity of reactive oxygen including superoxide anion radical and hydrogen peroxide, metal-chelating activity, and reducing power of garlic extracts were investigated. All tested garlic extracts exhibited in vitro antioxidant activities, with Uiseong extract showing highest hydrogen-donating and hydrogen peroxide-scavenging activities, and reducing power, followed by Seosan and Samchek extracts, in proportion to total thiosulfinate contents. Higher scavenging activity of superoxide anion radical was observed in Uiseong than Seosan and Samchek extracts. Metal-chelating activity increased in order of Uiseong < Seosan < Samchek, showing inverse relations to total thiosulfinate content. Garlic extracts of Uiseong and Seosan showed weak prooxidant activities and that of Samchek showed strong antioxidant activity against $Cu^{+2}$-induced human LDL oxidation. Protective effects on peroxyl and hydroxyl radical-induced DNA strand damages were observed in all tested garlic cloves. These results indicate growing conditions of garlic cloves affect total thiosulfinate content and antioxidant activities.

Antioxidative Activity of Lichen Thamnolia vermicularis in vitro

  • This study was aimed at evaluating the antioxidant activities of methanol extract of Thamnolia vermicularis. The antioxidant activity, reducing power, superoxide anion radical scavenging and free radical scavenging activities were studied. The antioxidant activity of the extract correlated with its concentration ($0.2{\sim}2\;mg/ml$) in the reaction mixtures containing linoleic acid. Upto 67% of lipid peroxidation was inhibited by 2 mg/ml of the lichen extract. The extract showed strong free radical scavenging activity similar to that of BRA (positive control) in a manlier of concentration dependent. The lichen extract also showed moderate effects on superoxide anoin scavenging activity and reducing power, which was not so effective as that of Quercetin and BHA used as positive controls. This study suggests that T. vermicularis lichen can be used as a novel source of natural antioxidant.

Antioxidative Activities of New Citrus Hybrid 'Hamilgam' Peel Extracts (가공용 감귤 신품종 하밀감 과피 추출물의 항산화 활성)

  • The major objective of this study was to investigate the usability as cultivar for processing of new Citrus hybrid 'Hamilgam'. We investigated various antioxidant activities, such as DPPH radical, ABTS radical, Hydroxyl radical and Superoxide anion radical effect, along with total polyphenol content and total flavonoid content of Citrus hybrid 'Hamilgam', C. unshiu, C. natsudaidai. The total polyphenol content and total flavonoid content were the highest in the Hamilgam. The major flavonoids of Hamilgam and Natsudaidai were naringin and neohesperidin, and those of Unshu were narirutin and hesperidin, as determined by HPLC. Especially, the neohesperidin content of Hamilgam showed much higher (100 times over) than that of Unshu. However, the aglycone form was not found. DPPH and ABTS radical scavenging activities were the highest in Hamilgam peels. Hydroxyl radical scavenging activity was high in the order of Unshu, Hamilgam and Natsudaidai. Superoxide anion radical scavenging of 3 cultivars peels displayed low activities compared to DPPH and ABTS radical scavenging. Based on these results, Hamilgam peel extracts possess antioxidant activities and may thus serve as potential sources of functional food, cosmetic products, etc.

A Study of Effects of Suaeda glauca (Bunge) Bunge Extract and Its Fractions (나문재(Suaeda glauca (Bunge) Bunge) 추출물과 분획물의 효능에 관한 연구)

  • In this study, S. glauca (Bunge) Bunge extract was testified total polyphenol contents, electron donating ability on DPPH, and scavenging ability of superoxide anion radical and hydrogen peroxide. Total polyphenol contents of S. glauca (Bunge) Bunge extract were 7,053.30±915.93 ㎍ GAE/mL. In the fractions, fraction by ethanol was the highest content of 10,973.30±1,000.24 ㎍ GAE/mL. Fractions of ethyl acetate and water were 2,386.70±166.53 ㎍ GAE/mL, 2,413.30±41.63 ㎍ GAE/mL respectively. It was shown that total content of polyphenol according to solvent was significant relation at p<0.05. In the experiment of the electron donating ability, 70% ethanol extract and methylene chloride fraction were -246.15±24.17%, -254.01± 16.54% respectively. In case of ethyl acetate fraction, it was the highest electron donating ability to DPPH radical, 39.06±0.34%, and then water fraction of 36.71±5.55%, ethanol fraction of 29.77±2.57%, gradually. Electron donating abilities revealed significant difference (p<0.05) between the solvents. The superoxide radical scavenging ability of standard material was 0.029±0.0011; 70% ethanol extract of 0.022±0.00052, methylene chloride fraction of 0.027±0.00031, ethyl acetate fraction of 0.024±0.0011, ethanol fraction of 0.021±0.00024 and water fraction of 0.024±0.00019. Hydrogen peroxide scavenging ability of 70% ethanol extract was -0.0029±0.00040 and the others were as follow; methylene chloride fraction (-0.0042±0.00058), ethyl acetate fraction (-0.003± 0.0041), ethanol fraction (-0.0029±0.0015) and water fraction (-0.0028±0.00090).

A Study on the Anti-Oxidation Effects of Ulmi Cortex and Hemipteleae Cortex (유피(楡皮)와 자유피(刺楡皮)의 항산화(抗酸化) 효능에 대한 연구)

  • Objectives : This studies were carried out to evaluate the effects on anti-oxidation activities in different parts as trunk bark and root bark of Ulmus devidiana Planchon. var. japonica Nakai and Hemipteleae davidii (Hance) Planchon. Methods : For evaluation of antioxidative effects, scavenging activity on superoxide anion radical and DPPH radical were measured. Also, inhibitory activity on LDL oxidation and linoleic acid peroxidation were measured in each samples. In the in vivo test, inhibitory activity on TBARS production, GSH contents in rat liver were measured. SOD, Catalase, GSH-px and ALDH activity were analysed in ethanol extracts of Ulmi Radicis Cortex. Result : 1. Scavenging activity on superoxide anion radical was higher in water extract than in ethanol extract even in low concentration of 50ppm as over 90%. 2. There was no difference between water extract and ethanol extract in the scavenging activity on DPPH radical but, Ulmi Raclicis Cortex and Ulmi Trunci Cortex showed high effect even in low concentration of 10ppm. 3. GSH reduction was prevented and antioxidative activity such as Mn-SOD, GSH-px in the rat liver recovered in the treatment of ethanol extracts of Ulmi Radicis Cortex. Conclusion : Ulmi Radicis Cortex recorded as Ulmi Trunci Cortex in official regulation book. But, it was known that Ulmi Radicis Cortex was more effective than Ulmi Trunci Cortex in most physical activities.

Purification, Chemical Composition, and in vitro Antioxidant Activity of Two Protein-bound Polysaccharides from Rapeseed Meal

  • Crude polysaccharides from rapeseed meal (PRM) were extracted with 0.3% NaOH aqueous solution, followed by further purifications and 2 fractions, namely PRM1 and PRM2, were separated with a DEAE-cellulose DE-52 column. Their primary compositions were analysed and antioxidant activity was determined, including scavenging activity toward superoxide anion radicals, hydroxyl radicals, and nitric oxide radicals, reducing power, and inhibitory effects against the microsomal lipid peroxidation, compared to that of L-ascorbic acid. The results indicated that PRM1 and PRM2 exhibited not only good reducing power and inhibitory effects on the microsomal lipid peroxidation, but also strong scavenging activity toward superoxide anion radicals, nitric oxide radicals, and hydroxyl radicals. In addition, positive correlations were also observed between the superoxide anion radical scavenging activity and the protein contents of the polysaccharides, and the reducing power and the sulfate contents. These findings thus clearly suggest the polysaccharides possess direct and potent antioxidant activity.

Experimental Evidence of the Mobility of Hydroperoxyl/Superoxide Anion Radicals from the Illuminated TiO2 Interface into the Aqueous Phase

  • The understanding of behaviors of hydroperoxyl/superoxide anion radicals (${H_2O_2}^./{O_2}^{-.}$) generated from a photoirradiated $TiO_2$ surface is essential to improve the efficiency of $TiO_2$ photocatalytic reactions by decreasing the recombination of photoinduced electron-hole ($e^--h^+$) pairs. In contrast with previous studies, we found that ${H_2O_2}^./{O_2}^{-.}$ generated on the surface of illuminated $TiO_2$ particles are mobile. ${H_2O_2}^./{O_2}^{-.}$ formed by the photocatalysis of $TiO_2$ particles immobilized onto the inner surface of a coil-quartz tube were forced under a continuous flow through a knotted tubing reactor (KTR) and into the aqueous phase completely separated from the $TiO_2$ particles, and were measured by a chemiluminescence (CL) technique using 2-methyl-6-(p-methoxyphenyl)-3,7-dihydroimidazo[ 1,2-a]pyrazin-3-one (MCLA) as the reagent. The initial concentration of the ${H_2O_2}^./{O_2}^{-.}$ stream entering the KTR was determined by its half-life (98 s) at pH 5.8. We suggests that the efficiency of $TiO_2$ photocatalytic reactions may be further improved by utilizing the mobility of ${H_2O_2}^./{O_2}^{-.}$.
